Herpes simplex virus type 2에 의한 신생아 수막뇌염 (Neonatal Meningoencephalitis caused by Herpes Simplex Virus Type 2)

  • 신생아 HSV 감염은 주로 HSV type 2에 의해 발생하는 것으로 알려져 있으나, 국내에서는 신생아 HSV 감염의 역학 자료뿐만 아니라, HSV type 2가 증명된 신생아 수막뇌염 증례 보고조차 없었다. 저자들은 열과 경련을 주소로 내원한 생후 16일 신생아에서 PCR 검사를 통해 HSV type 2에 의한 수막뇌염을 진단하고 acyclovir 및 항경련제 치료를 시행하였다. 환자는 뇌 MRI 검사에서 뇌연화증이 있어 성장과 발달에 대해 추적 관찰 중이다. 우리나라 여성에서 30대에 HSV type 2 초감염이 가장 빈번히 발생하고 최근 평균 초산 연령이 증가하여 30대로 진입함에 따라 향후 HSV type 2에 의한 신생아 감염이 증가될 것으로 예상되어, HSV type2 감염은 열이 나는 신생아에서 반드시 고려해야 할 것이다.

  • 저자들은 건강했던 12세 남아가 발열, 연하통, 삼킴 곤란 증세를 호소하여 시행한 내시경 상 식도의 궤양과 수포성 병변이 보이고 헤르페스 I형 항체 양성반응으로 진단된 헤르페스 식도염을 항바이러스제와 proton pump inhibitor 등 보존적 치료로 치유시킨 1예를 경험하였기에 문헌고찰과 함께 보고하는 바이다.

Inhibition of Herpes Simplex Viruses, Types 1 and 2, by Ginsenoside 20(S)-Rg3

  • Infections by herpes simplex viruses have an immense impact on humans, ranging from self-limiting, benign illness to serious, life-threatening diseases. While nucleoside analog drugs are available, resistance has been increasing and currently no vaccine exists. Ginsenosides derived from Panax ginseng have been documented to inhibit several viruses and bolster immune defenses. This study evaluated 12 of the most relevant ginsenosides from P. ginseng for toxicities and inhibition of herpes simplex viruses types 1 and 2 in Vero cells. The effects of test compounds and virus infection were determined using a PrestoBlue cell viability assay. Time course studies were also conducted to better understand at what points the virus life cycle was affected. Non-toxic concentrations of the ginsenosides were determined and ranged from 12.5 μM to greater than 100 μM. Ginsenoside 20(S)-Rg3 demonstrated the greatest inhibitory effect and was active against both HSV-1 and HSV-2 with an IC50 of approximately 35 μM. The most dramatic inhibition-over 100% compared to controls-occurred when the virus was exposed to 20(S)-Rg3 for 4 h prior to being added to cells. 20(S)-Rg3 holds promise as a potential chemotherapeutic agent against herpes simplex viruses and, when used together with valacyclovir, may prevent increased resistance to drugs.

사면안정해석에 대한 Constrained Simplex Method 적용 (A Constrained Simplex Method for Slope Stability Analysis)

  • 본(本) 연구(硏究)에서는 기존(旣存)의 절편법의 단점(短點)을 보완(補完)하기 위하여 Gussmann의 일반절편법을 안전율(安全率)로 정의하고, 또한 예상파괴면(豫想破壞面)을 구하기 위하여 Box가 제안(提案)한 Constrained simplex method를 최적화기법(最適化技法)으로 이용함으로써 안전율(安全率)과 한계파괴면(限界破壞面)을 동시에 구할 수 있는 사면안정해석(斜面安定解析) 프로그램 CSLOP을 개발(開發)하였다. 무한사면(無限斜面)과 일반사면(一般斜面)을 대상(對象)으로 사면해석을 한 결과 CSLOP에서 구한 안전율(安全率)은 Bishop의 간편해(簡便解)와 별 차이가 없었으며, 한계 파괴면을 찾는데 있어서 좋은 수렴(收斂)결과를 보였다.

수종 Flavonoid의 항허피스바이러스효과 (Antiviral Activity of Some Flavonoids on Herpes Simplex Viruses)

  • To search for less toxic antiherpetic agents, the inhibitory effects of twelve kinds of flavonoids including chrysin, quercetin, quercitrin, rutin, fisetin, gossypin, kaempferol, morin, naringenin, naringin, hesperetin and hesperidin on the plaque formation of herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2) in Vero cells were examined by plaque reduction assay in vitro. Some flavonoids tested in this study showed potent antiherpetic activity, reducing intracellular replication of herpes simplex viruses when Vero cell monolayers were infected and subsequently cultured in medium containing flavonoids. Naringenin showed the most potent antiviral activity against HSV-1 with selectivity index (SI) of 19.1 and hesperetin showed the most potent antiviral activity against HSV-2 with SI of 9.8. These results suggest that some flavonoids may be a potential therapeutic agent for the treatment and prevention of herpes simplex virus infections.

시험관내에서 이버멕틴, 도라멕틴, 에타놀에 대한 아나사키스 유충의 운동성 억제효과 (Larval migration inhibition activity of ivermectin, doramectin and ethanol against Anisakis simplex in vitro)

  • This experiment has been investigated in order to examine larval migration inhibition activity of ivermectin, doramectin and ethanol against Anisakis simplex (A. simplex) in vitro. A. simplex larvae were obtained from the mackerel acquired from the fish market of Cheongju. They were divided into many groups and placed in culture dishes (40 larvae each) containing RPMI-1640, in the absence or presence of different concentrations of ivermectin, doramectin and ethanol. Ivermectin had a complete inhibition of larval migration at 72 h in all groups ($10-300{\mu}g/ml$). Ethanol reduced the migration of the larvae, its maximum activity being an high doses (7.5%, 10% ethanol) when it was 100% efficacy at 4 h. Doramectin had no efficacy in vitro. Being needed that further studies with ivermectin and doramectin, it is recommended that in vivo test with laboratory animals should be carried.

  • Herpes simplex virus(HSV) infections of the CNS are associated with significant morbidity and mortality even when appropriate antiviral therapy is administered. HSV infections of the brain can be subdivided into two categories : neonatal HSV infections, which usually are caused by HSV type 2, and herpes simplex encephalitis(HSE), which occur in patients over 3 months old and is nearly uniformly caused by HSV type 1. The clinical presentation of HSE is one of the focal encephalopathic process associated with altered levels of consciousness, fever, focal seizures and hemiparesis. But because of the lack of pathognomic clinical presentation and diagnostic procedure, the efforts to develop alternative diagnostic procedure have led to the use of new diagnostic technique such as polymerase chain reaction(PCR). We report a case of HSV type 1 encephalitis in 13 month old male infant who presented with altered level of consciousness, fever and focal seizures. With the use of the PCR, HSV-1 DNA was detected in cerebrospinal fluid from the patient. The symptoms and signs of encephalitis subsided by treatment with acyclovir in 14 days.

Comparison of Automatic Calibration for a Tank Model with Optimization Methods and Objective Functions

  • Two global optimization methods, the SCE-UA method and the Annealing-simplex (A-S) method for calibrating a daily rainfall-runoff model, a Tank model, was compared with that of the Downhill Simplex method. The performance of the four objective functions, DRMS (daily root mean square), HMLE (heteroscedastic maximum likelihood estimator), ABSERR (mean absolute error), and NS (Nash-Sutcliffe measure), was tested and synthetic data and historical data were used. In synthetic data study. 100% success rates for all objective functions were obtained from the A-S method, and the SCE-UA method was also consistently able to obtain good estimates. The downhill simplex method was unable to escape from local optimum, the worst among the methods, and converged to the true values only when the initial guess was close to the true values. In the historical data study, the A-S method and the SCE-UA method showed consistently good results regardless of objective function. An objective function was developed with combination of DRMS and NS, which putted more weight on the low flows.

  • 단순포진 바이러스 감염을 유발하는Herpes simplex2형 바이러스의 감염에 관여하는 항원들과 중화항체 생산을 유발하는 주요 항원들의 위치를 확인하였다. Vero cell에 감염하였을 때 48시간 동안 31, 43, 59, 69 kDa 바이러스 항원들이 지속적으로 발현되었으며, 감염된 쥐에서 생산한 항체와의 반응에서는 51 kDa 항원이 가장 강한 반응을 보였다. 면역전자현미경으로 위치를 확인한 결과 colloidal gold가 바이러스 표면에 발견되는 것으로 보아 이 항원이 바이러스 표면에 존재하고 있는 것으로 확인되었다. 형광현미경 분석은 이 항원들이 감염된 세포 내에서 전반적으로 발견되었고 특히 세포 표면에서 많이 발현되고 있었다.
