Studies on Peroxidase Isozymes of Tobacco and Korean Radish: implication of Specific Isoperoxidase in Shoot Formation (담배와 무 Peroxidase Isozyme들의 세포 생화학적 연구: 조직 특이성 발현과의 연관성)

  • In an attempt to clarify the physiological functions of individual isoperoxidases, we have studied enzymatic and immunological properties as well as cellular distribution of isoperoxidases from tobacco callus and Korean radish. The gene expression patterns of isoperoxidases in shoot and non-shoot-forming tobbaco callus were also examined by rabbit reticulocyte lysatein vitro translation system. These results indicate that fraction of translatable poly(A)-isoperoxidase mRNA was increased considerably in shoots. At the present time, at least 6-7 isoperoxidases could be detected from the translation mixture of total cellular RNA, among which only one cell wall localized anodic isoperoxidase (named A3) mRNA was bimorphic mRNA. These data suggest the possible regulation of peroxidase activity during shoot formation by altering the polyadenylation state of mRNA. In case of Korean radish seedlings, poly(A)- peroxidase mRNA were also increased depending upon aging.

Establishment of Efficient Regeneration System Through In Vitro Culture of Lettuce (Lactuca sativa)

  • An efficient regeneration system was established by using in vitro plantlets of germinated seedlings from different cultivars of lettuce (Lactuca sativa cv. Chongchima, Chongchuckmyun, Jeokchima, Jeokchuckmyun). Shoot formation were observed from all cultivars on MS medium supplemented with 0.1 mg/L NAA and 0.5 mg/L BA. In all cultivars, when cotyledon was cultured, the number of shoot per explant was more greater than that hypocotyl and leaf disc were cultured. Shoot formation rate (91.7%) was high in a cotyledon culture of cultivar, Chongchukmyun. The growth of multiple shoots derived from the cultivar, Chongchukmyun, was most effective on medium containing 0.5 mg/L BA and 1.0 mg/L GA$_3$. When shoots were transferred on MS medium without plant growth regulators, roots were effectively differentiated. Rooted plantlets were acclimated on pots for further propagation.

Effect of Explant Source and Plant Regulator on Callus Formation and Shoot Regeneration in vitro Culture of Brassica napus L. (식물부위(植物部位)와 생장조절제(生長調節濟)가 유채(油菜)(Brassica napus)의 기관분화(器官分化)에 미치는 영향(影響))

  • Culture condition for callus formation and plant regeneration were optimized by the selection of explants and the manipulation of hormonal combination in the culture medium. The calli induced from seed, cotyledon, hypocotyl and mesophyll segments were more vigorously proliferated under dark condition than those under continuous light condition. Hypocotyl-and cotyledon-derived calli were more regenerative as compared with those of seed and mesophyll. Callus formation from hypocotyl and cotyledonary explants was enhanced on MS medium with 1.0 mg/${\ell}$ 2, 4-D and 0.1 to 0.5 mg/${\ell}$ kinetin or BAP. The combination of 0.1 mg/${\ell}$ NAA and 2.0 to 4.0 mg/${\ell}$ kinetin was the most effective for shoot regeneration from the callus. The maximum frequency (24.0%) of shoot regeneration was obtained from the hypocotyl-derived callus transferred to MS medium supplemented with 0.1mg/${\ell}$ NAA and 4.0 mg/${\ell}$ kinetin. The capacities for callus, root and shoot formation from cotyledon and hypocotyl explants were remarkably different among cultivars of B. napus tested. The calli induced from hypocotyl produced more shoots than those from cotyledon.

Plant regeneration via direct and indirect adventitious shoot formation and chromosome-doubled somaclonal variation in Titanotrichum oldhamii (Hemsl.) Solereder

  • The gesneriaceous perennial plant Titanotrichum oldhamii has beautiful foliage and attractive bright yellow flowers. However, breeding of T. oldhamii by conventional sexual hybridization may be difficult because sexual reproduction of this species is very rare. In the present study, plant regeneration systems via both direct and indirect formation of adventitious shoots from leaf explants were established as the first step toward breeding T. oldhamii by using biotechnological techniques. Adventitious shoots were formed efficiently on medium containing $0.1mg\;l^{-1}$ benzyladenine. Histological observation showed that shoot formation on this medium occurred directly from leaf epidermal cells without callus formation. On the other hand, leaf explants formed calluses on medium containing $0.1mg\;l^{-1}$ 2,4-dichlorophenoxyacetic acid. The calluses could be maintained by monthly subculturing to fresh medium of the same composition. When the calluses were transferred to plant growth regulator-free medium, they formed adventitious shoots. Directly and indirectly formed shoots rooted well on medium containing $0.1mg\;l^{-1}$ indole-3-butyric acid. Plantlets thus obtained were successfully acclimatized and grew vigorously in the greenhouse. Flow cytometry analysis indicated that no variation in the ploidy level was observed in plants regenerated via direct shoot formation, whereas chromosome doubling occurred in several plants regenerated via indirect shoot formation. Regenerated plants with the same ploidy level as the mother plants showed almost the same phenotype as the mother plants, whereas chromosome-doubled plants showed apparent morphological alterations: they had small and crispate flowers, and round and deep green leaves.

Efficient Procedures for Direct Shoots Regeneration from Leaf Explants of Rehmania glutinosa Lib. (지황 잎조직 절편으로부터 신초 형성)

  • Adventitious shoots were directly induced from leaf explants of R. glutinosa, an important medicinal plant. Proliferating shoot cultures were obtained by culturing leaf discs on Murashige and Skoog(MS) medium alone or combination with auxins and cytokinins. MS medium supplemented with 1 $mg/{\ell}\;BA\;and\;2\;mg/{\ell}$ IAA was the most effective, providing high shoot bud formation frequency without formation of intervening callus. The effect of leaf age on adventitious shoot formation was also investigated. The maximum shoot bud production (93.4%) was achived using 3rd leaf from apex of 6 weeks old plantlets after seed germination. Plantlet were rooted on an half-strength MS (1/2MS) medium containing 0.1 $mg/{\ell}\;IBA$. This prtocol is useful for clonal propagation and Agrobacterium-mediated transforamtion in R. glutinosa.

Effect of TDZ (Thidiazuron) on Shoot Proliferation of Peace Poplar (Peace 포플러(Populus koreana X P. trichocarpa)의 줄기형성에 미치는 Thidiazuron 효과)

  • Shoot formation was investigated from in vitro cultivation of exotic hybrid poplar (Populus koreana ${\times}$ P. trichocarpa) with a specific stomatal character occurring both upper and lower surface of leaves. Two different explants (stem and leaf segment) of Peace poplar were cultured on half strength Murashige and Skoog (MS) basal medium supplemented with the various concentrations of thidiazuron as a plant growth regulator. Most adventitious shoots were produced from excised ends of stem or mid-veins of leaf segments. The highest average numbers of shoots were 7.1 and 5.3 with the treatments of 0.02mg/L TDZ in both explants of stem and leaf segment. The highest shooting rates were achieved to 83.3% and 47.6% with the concentrations of 0.01mg/L and 0.02mg/L TDZ by axillary bud and leaf cultures, respectively.

In vitro Growth of Shoot Derived from Shoot Tip in Asparagus cochinchinensis (Lour.) Merr. (천문동 경정 유래 신초의 기내생장)

  • This study was carried out to investigate the optimal conditions for efficient in vitro growth of Asparagus cochinchinensis (Lour.) Merr. shoot derived from shoot tip. Shoots were successfully cultured in MS medium. It was found that $23^{\circ}C{\sim}25^{\circ}C$ were suitable for shoot growth. The growth of shoot was greatly influenced by cytokinins. The shoots derived from shoot tip were well elongated on MS medium supplemented with BA and zeatin. In vitro shoots were very poor or no growing. Especially, 3.0 mg/l BA on MS medium was very effective in elongation of shoot. Root formation from in vitro growth of plant let was achieved on MS medium supplemented with 0.5 mg/l IBA. The results suggest that selection of plant growth regulator could be important factor to achieve an efficient in vitro growth.

In Vitro Propagation of Lilium Asiatic Hybrid 'Hae Hwa' via the Formation of Shoot Clusters (신초 Cluster 형성에 의한 Lilium Asiatic Hybrid 'Hae Hwa'의 기내번식)

  • This experiment was conducted to micropropagate bulblets via shoot cluster formation and massproduce normal bulblets from the sections of proliferated shoot clusters in Lilium asiatic hybrid 'Hae Hwa'. The induction of shoot clusters from the culture of bulblet sections was more effective than that of bulb scales on MS medium with 1.0 mg/L BA and 0.5 mg/L IAA. Proliferation of shoot clusters from the formed shoot cluster sections was the most favorable on medium containing 5.0 mg/L BA and 0.5 mg/L IAA. The formation and the growth of bulblets from shoot cluster sections were achieved effectively on medium with 60∼90 g/L sucrose. The leaves derived from shoot clusters grew vigorously but the bulblets from shoot clusters grew very poor in 5L air-lift bioreactor culture. By the addition of 30 mL fresh liquid medium containing doulble strength MS salts, 250 g/L sucrose and 5 g/L activated charcoal after 8 weeks in the shoot cluster culture on MS medium with 5.0 mg/L BA and 0.5 mg/L IAA, the number of bulblets was increased in light condition, but the growth of bulblets was not affected by light. Bulblet production was possible with the bulblet product at 53 to 68 mg in fresh weight by liquid medium addition after the proliferation of shoot cluster.

Systematic Propagation of High Quality Garlic (Allium sativum L.) Through Shoot Apical Meristem Culture II. Effects of Sucrose Concentration and Nitrogen Source on In Vitro formation of Bulblets (생장점배양에 의한 우량마늘 체계적 증식 II 기내 인경 비대에 미치는 질소 및 Sucrose의 영향)

  • The effects of sucrose concentration and nitrogen source on shoot growth and in vitro formation of garlic (Allium sativum L. cv Seosan) bulblet were investigated in order to systematize propagation of high quality garlic through a shoot apical meristem culture. Shoot differentiation was not affected by sucrose concentration and nitrogen source, but plantlets which contain medium of NH$_4$- N or NH$_4$ + NO$_3$ were vigorous and healthy in .appearance. Shoot growth was vigorous in changeing of nitrogen source. The best quality of in vitro bulblets was obtained in culture on the medium containing 8% sucrose and NH$_4$ - N, and the formation of bulblet was more effective when plantlets were subjected to cold treatment before use. NH$_4$-N was a major factor for shoot growth and bulblet development, but NO$_3$-N was not and suppressed $K^{+}$absorption. The level of ethylene production was not affected by different nitrogen sources, however this production was enhanced in medium containing a higher concentration of sucrose.e.
