Anatomical Characteristics of Hyperhydric Shoots Occuring in In Vitro Culture of Peace Poplar (Peace포플러의 기내 배양시 발생하는 과수화 식물체의 조직적 특성)

  • We investigated the anatomical aspects of vitrification in peace poplar. Comparisons were made with regard to characteristics occurring between hyperhydric and normal shoots in shoot proliferation cultures on MS medium containing 0.2 mg/L BA. Compared with normal plants, hyperhydric plants had thick, curled, and dark green leaves. Hyperhydric stems were thicker and shorter than those of normal stems. When examined under the microscopes, the mesophyll palisade cells of hyperhydric leaves were vacuolated, whereas those of normal leaves contained normal and enriched vacuole with cytoplasm. Generally, the hyperhydric leaves showed poorly developed palisade parenchyma, and revealed irregular and bigger sized intercellular structures in both palisade and spongy parenchyma as well as epidermis cells compare to those of normal leaves. In addition, the hyperhydric leaves had lower stomatal density and bigger sized cell. Vascular tissues of hyperhydic stems were less differentiated because of poorly lignified xylem tissue. The greatly expanded cortical cells and pith appeared to be the main cause of thick stems as compared with normal stems.

Plant Regeneration from Explant Types and Cultivars of Boxthorn (Lycium chinense Mill.) (구기자나무의 절편체 부위와 품종 간 재분화 특성)

  • Callus and shoot formation of leaf explants from in vitro propagated shoots and field grown plants depending on the position of leaf, and four boxthron cultivars were investigated. Callus formation of explants from both in vitro shoot and field grown plants as easily achieved at the cut surfaces of explants but the callus formed from leaf of in vitro shoots was hardened as the duration of culture was proceed. Calli were effectively induced from leaves detached from the middle position of both in vitro and in vitro plants on MS medium containing 0.5 mg/L NAA with 0.2 mg/L BA, and the growth of calli were better in field grown leaves than in vitro grown leaves. Shoot formation were effectively induced from leaves detached from the upper position in vitro plants, and the middle parts of in vitro plants on MS medium containing 0.01 mg/L (NAA with 0.2 mg/ BA. There was difference on the frequency of shoot formation among four different cultivars; 'Jindojaerae' was the best for shoot formation followed by 'Cheonyang', 'Younghagukija' and 'Cheongyangjaerae'.

Production and Rooting of Shoots in Bioreactor Culture of Scrophularia buergeriana Miquel (생물반응기에서 현삼의 신초 형성과 발근)

  • This study was carried out to know the factors affecting on shoot formation and rooting for stable and routine production of plantlets in bioreactor culture of Scrophularia buergeriana. Multiple shoots were formed effectively when explants were transplanted on the MS media with decreased concentration of $NH_4NO_3$ as 413mg/ l . Three hundred stem explants (0.8-1.0cm) was appeared as proper inoculation size in bioreactor culture. IBA (0.05mg/L) was more effective for rooting of the shoots in liquid as well as solid media. Six weeks long culture of explants in bioreactor gave better shoot shape for rooting on solid half-strength MS media.

Varietal Difference in Plant Regeneration from Cotyledon Culture of Capsicum annuum L. (고추 자엽에서 식물체 재분화의 품종간 차이)

  • Effects of genotype and culture medium on plant regeneration from cotyledon segments of red pepper(Capsicum annuum L.) was investigated. Among combinations of IAA(0.25 and 0.50 mg/L) and zeatin(2.0 and 4.0 mg/L) added to MS medium, combination of 2.0 mg/L zeatin and 0.25 mg/L IAA was shown to be the best for shoot differentiation from cotyledon segments. Shoot regeneration from cotyledon explants took 9 to 25days, depending on genotypes and culture media. Early shooting was observed in Yeongyangjaelae, Putgochw, Karkovskij-A-35, Gris I-A-1 on MS medium containing 2.0 mg/L zeatin and 0.25 IAA mg/L. Percent of explants producing shoots, as also influenced by genotypes and culture media, were over 90% for 621, Yeongyangjaelae, Putgochw, Nikko jacksacgmulgochw, Ch-6-Num-216, and Kajenskij-A-35 when cultured on MS medum supplemented with 2.0 mg/L zeatin and 0.25 mg/L IAA and for Fresno chile, PI 169126, Kajenskij-A-35, jacksacgmulgochw, and PI 297438 on MS medium including 2.0 mg/L BA and 1.0 mg/L IAA.

Effects of Explant Parts and Plant Growth Regulators on the in vitro Propagation of Lycoris squamigera (상사화의 기내증식에 미치는 배양부위와 생장조절물질의 영향)

  • This study was carried out to investigate the influence of medium composition for in vitro mass propagation of Lycoris squamigera Max. After the disks of short stems, segments of leaf within bulb and scale were cultured on MS basal medium supplemented with various plant growth regulators, they were examined for the extent of callus formation, shoot and root regeneration. In the culture of stem disks, adventitious shoots were regenerated from the basal tissue of bulb scales, and combined medium of 1.0 mg/L 2,4-D or NAA+2.0 mg/L BA or kinetin showed the the best response and 4∼6 shoots per explant formed. In the culture of leaf segments within bulbs, both MS medium supplemented with 1.0 mg/L NAA+2.0 mg/L TDZ and with 1.0 mg/L 2,4-D+1.0∼2.0 mg/L BA were produced callus profusely on the base of leaf tissue and 3∼6 shoots were regenerated per explant. In the scale segments culture, calli were produced on the basal tissue on medium with 1.0 mg/L 2,4-D+1.0∼2.0 mg/L BA. The best result were shown on MS medium with 1.0 mg/L NAA+2.0 mg/L TDZ, and 1.0 mg/L 2,4-D+1.0∼2.0 mg/L BA. Maximum number of regenerated shoots was up to 10∼12. Adventitious root formation from explants were formed profusely on MS medium with 1.0 mg/L NAA+2.0 mg/L kinetin. The most desirable method for mass propagation of plantlets was the shoot regeneration from scale segments then subsequently subcultured on medium for rooting.

Position Effect of Axillary Buds on Shoot Multiplication and Rooting in Bud Culture of Quercus acutissima (상수리나무 기내(器內) Axillary Bud의 치상부위(置床部位)에 따른 다경(多莖) 및 발근유도(發根誘道) 효과(効果))

  • This study was conducted to examine the position effect of axillary buds on shoot multiplication and rooting with 1-year-old seedlings of Quercus acutissima. Shoot multiplication was greatly affected by axillary bud position: Productivity of multiple shoots were decreased in the order of top, mid and basal explants respectively. The best shoot multiplication (mean 6.1 shoots per explant) was obtained on WPM medium containing $1.0mg/{\ell}$ BAP and $0.1mg/{\ell}$ NAA using basal explant after 4-week-culture. Rooting was also greatly influenced by position. Its percentage was increased in the order of top, mid and basal explant respectively. Root initiation was better and more rapid on 1/2MS medium than GD medium. High rooting percentage (100%) was obtained on 1/2MS medium containing $0.2mg/{\ell}$ IBA after 15 days culture. Sucrose concentrations did not effect on rooting. However root development and shoot growth were greatly affected by them. Root was developed shortly on 1-2% levels and shoot growth was getting retarded, whereas both of them did not show significant difference at 3-6% levels. Rotting was decreased on 7-8% levels gradually, but shoot and leaf condition was better than any other concentrations. Survival rate of rooted explants in pot was varied according to the position of explants. Seedlings of top part were survived up to about 50% but most of mid and basal part seedlings did not survive over 4 weeks even in high humidity condition. Seedlings in pots showed normal growth over 10 months but most of them showed the condition of premature leaf shedding.

Development of Herbicide(BIALAPHOS) Tolerant Tobacco through Tissue Culture (제초제(除草劑) BIALAPHOS에 대(對)한 연초(煙草)의 내성(耐性) 증대(增大))

  • This study was conducted to level up the tolerance of tobacco plant against bialaphos herbicide through tissue culture. The relatively good shoot regeneration from the subcultured calli treated with bialaphos at 0.5 ppm was observed in old the tobacco varieties tested such as NC 82, BY 4 and KA 101. However, at the treatment of bialaphos 1.0 ppm, shoot regeneration was only made in KA 101 variety, showing better regeneration than that of untreated one, When these shoots were transfered to the medium containing of bialaphos 10.0 ppm, the percentage of living shoots (i.e. tolerant plant) was very low, showing 2.43% in NC 82, 2.76% in KA 101 and 0.78% BY 4. Calli were induced and multiplied from leaf petiole of the above tolerant plants even under 2.5ppm of bialaphos, showing an average of 9% in NC 82 and 16% in KA 101 as compared with the untreated control. No calli were induced from tolerant plants as bialaphos concentration increased up to 5.0 ppm. Direct shooting from leaves of the above tolerant plants, that is selected at 10.0ppm of bialaphos treatment, was observed even under 10.0ppm of bialaphos treatment both in NC 82 and in KA 101 varieties, indicating that tolerance of tobacco plants against bialaphos can be greatly increased.

Plant Regeneration via Multiple Shoots Formation from Sucker Explants of Rubus fruticosus L. (블랙베리(Rubus fruticosus L.)의 맹아절편체로부터 다경유도를 통한 식물체 재분화)

  • This study was carried out to induce plant regeneration via shoot formation from sucker explants of Rubus fruticosus L. To induce adventitious shoots, sucker explants were sterilized in $1.2\%$ NaOCl solution, and cultured on the MS solid medium supplemented with kinetin (0.5, 1.0, 3.0 mg/L) and BA (0.5, 1.0, 3.0 mg/L), respectively. As above, to induce adventitious shoots, sucker explants were cultured on the MS solid medium supplemented with IBA (0, 0.1, 1.0 mg/L) and BA (0, 0.1, 1.0, 2.0 mg/L). After 4 weeks of culture, the highest frquency $(100\%)$ of shoot formation from sucker explants was obtained from the medium with 1.0 mg/L BA. The highest shoot number per explant from in vitro shoot explants was 5.3. After 10 weeks of culture, the number of shoot per explant was increased. The highest frequency $(85\%)$ of root formation was obtained at 0.5 mg/L glycine medium, when the explant with shoot were cultured on the MS medium containing glycine at various concentrations from 0 to 2.0 mg/L. The survival rate of the plantlets after transfer to plastic pots containing sand, soil, and vermiculite (1:1:1, vol.) was $95\%$. The results indicate that micropropagation procedure can be applied for an efficient mass propagation of Rubus fruticosus.

In vitro shoot propagation of Ranunculus kazusensis Makino, an endangered aquatic plant (멸종위기 수생식물인 매화마름(Ranunculus kazusensis Makino)의 기내 신초 증식)

  • A micropropagation method via callus for Ranunculus kazusensis Makino, an endangered species, was established. When stem segments were cultured on MS media supplemented with 1.0 mg/L IAA, NAA, IBA and 2,4-D, the highest frequency of callus induction was achieved on MS medium supplemented with 1.0 mg/L NAA. Multiple shoot per explant was obtained, the MS medium containing 1.0 mg/L BA and 0.5 mg/L NAA. Additionally, effect of activated charcoal (AC) and sucrose on shoot growth in in vitro culture were examined. The most suitable conditions for shoot growth after 4 weeks of culture were the MS medium with AC and sucrose. This in vitro propagation protocol will be valuable for conservation and mass propagation of this endangered plant.

Influence of Nitrogen, Phosphorous, Potassium and Sucrose on the Shoot and Bulb Growth of Allium victorialis var. platyphyllum (산마늘의 신초와 구근생장에 미치는 질소, 인, 칼륨 및 당의 영향)

  • Effects of nitrogen (N), phosphorous (P) and potassium (K) on the shoot and bulb growth of wild garlic ($Allium$ $victorialis$ var. $platyphyllum$) were studied by adopting $in$ $vitro$ culture. These macronutrients influenced the growth of both the shoot and bulb of garlic depending upon their application doses. A minimum of 3% potassium nitrate ($KNO_3$) as a source of nitrogen was found to be critical for shoot elongation while higher concentrations were inhibitory. Garlic bulb growth was profuse on the usual $KNO_3$ strength and sucrose (7%), followed by $KNO_3$ (9.4 mM) supplement. On providing 41.22 mM ammonium nitrate ($NH_4NO_3$) as nitrogen source highest shoot growth was observed while 82.45 mM $NH_4NO_3$ as a source of nitrogen supported high bulb growth. With regard to potassium a good shoot growth was observed in medium that contained 0.31 mM $KH_2PO_4$ and 3% sucrose, while bulb growth was high on 2.5 mM $KH_2PO_4$ and 7% sucrose. These experiments may thus direct the development of excellent growth conditions for the commercial production of edible wild garlic.
