OBSERVATIONS OF STAR FORMATION INDUCED BY GALAXY-GALAXY AND GALAXY-INTERGALACTIC MEDIUM INTERACTIONS WITH AKARI

  • Nearby spiral galaxies M101 and M81 are considered to have undergone a galaxy-galaxy interaction. M101 has experienced HI gas infall due to the interaction. With AKARI far-infrared (IR) photometric observations, we found regions with enhanced star forming activity, which are spatially close to regions affected by the interaction. In addition, the relation between the star formation rate (SFR) and the gas content for such regions shows a significant difference from typical spiral arm regions. We discuss possible explanations for star formation processes on a kiloparsec scale and the association with interaction-triggered star formation. We also observed the compact group of galaxies Stephan's Quintet (SQ) with the AKARI Far-infrared Surveyor (FIS). The SQ shows diffuse intergalactic medium (IGM) due to multiple collisions between the member galaxies and the IGM. The intruder galaxy NGC 7318b is currently colliding with the IGM and causes a large-scale shock. The 160 micron image clearly shows the structure along the shock ridge as seen in warm molecular hydrogen line emission and X-ray emission. The far-IR emission from the shocked region comes from the luminous [CII]$158{\mu}m$ line and cold dust (~ 20 K) that coexist with molecular hydrogen gas. Survival of dust grains is indispensable to form molecular hydrogen gas within the collision age (~ 5 Myr). At the stage of the dusty IGM environment, [CII] and $H_2$ lines rather than X-ray emission are powerful cooling channels to release the collision energy.

Purification and Structural Characterization of Cold Shock Protein from Listeria monocytogenes

  • Cold shock proteins (CSPs) are a family of proteins induced at low temperatures. CSPs bind to single-stranded nucleic acids through the ribonucleoprotein 1 and 2 (RNP 1 and 2) binding motifs. CSPs play an essential role in cold adaptation by regulating transcription and translation via molecular chaperones. The solution nuclear magnetic resonance (NMR) or X-ray crystal structures of several CSPs from various microorganisms have been determined, but structural characteristics of psychrophilic CSPs have not been studied. Therefore, we optimized the purification process to obtain highly pure Lm-Csp and determined the three-dimensional structure model of Lm-Csp by comparative homology modeling using MODELLER on the basis of the solution NMR structure of Bs-CspB. Lm-Csp consists of a ${\beta}$-barrel structure, which includes antiparallel ${\beta}$ strands (G4-N10, F15-I18, V26-H29, A46-D50, and P58-Q64). The template protein, Bs-CspB, shares a similar ${\beta}$ sheet structure and an identical chain fold to Lm-Csp. However, the sheets in Lm-Csp were much shorter than those of Bs-CspB. The Lm-Csp side chains, E2 and R20 form a salt bridge, thus, stabilizing the Lm-Csp structure. To evaluate the contribution of this ionic interaction as well as that of the hydrophobic patch on protein stability, we investigated the secondary structures of wild type and mutant protein (W8, F15, and R20) of Lm-Csp using circular dichroism (CD) spectroscopy. The results showed that solvent-exposed aromatic side chains as well as residues participating in ionic interactions are very important for structural stability. Further studies on the three-dimensional structure and dynamics of Lm-Csp using NMR spectroscopy are required.

가연성 기체 혼합물에서 복잡한 구조에 따른 화염 가속 모델링 (Modeling of Flame Acceleration Considering Complex Confinement Effects in Combustible Gas Mixture)

  • 본 연구는 가연성 기체로서 에틸렌-공기 혼합물로 채워져 있는 관에서 장애물과 굽은 관에 의한 지형적 효과에 따라 변화하는 충격파와 화염의 상호 작용, 화염 가속, 연소폭발천이 현상을 수치적으로 살펴보았다. 여기서 사용되는 모델은 지배방정식으로 Navier-Stokes 방정식과 경계조건 처리 방법으로 ghost fluid 기법을 사용하였으며 지형적 영향을 달리한 여러 모델링을 통하여 화염과 강한 충격파의 충돌에 의한 열점 생성과 화염 전파의 지연 혹은 가속 현상을 확인하였다. 추가적으로 평균 화학적 에너지 발생률이 대략 20 MJ/($g{\bullet}s$)에서 폭굉으로 천이한다는 사실을 확인하였다. 그리고 동일 위치 열점 생성에도 불구하고 폭굉의 발생 시기가 반응물의 부재와 화염면 전방의 온도와 압력 차에 의해 지연될 수 있음을 확인하였다.

열충격 및 ${\alpha}$-amanitin과 cycloheximide의 처리를 통한 초기 계배의 체절 형성 기작에 대한 발생학적 연구 (Embryological Studies on Somitogenesis of Early Chick Embryos by heat shock and treatments of ${\alpha}$-amanitin and cycloheximide)

  • In order to investigate the factors of the control mechanism of somitogenesis, early chick embryos (H-H stage $8{\sim}13$) were treated with heat shock, ${\alpha}$-amanitin and cycloheximide and morphological changes of somite were examined by light microscopy, transmission and scanning electron microscopy. In normal chick embryo, somites were formed from the somitomere which preexisted in segmental plate. Somites were wrapped with extracellular collagen fibrils and connected with neural tube, notochord and ectoderm. And then, somites were differentiated to sclerotome, dermatome and myotome by the interaction of nervous tissue. Abnormal somites were observed after formation of six or seven so mites in heat shock treated group. Amounts of collagen fibrils were obviously decreased in this group. In cycloheximide treated group, most so mites were smaller and neural tube formation was incomplete. Chromatins were condenced and formed several heterochromatins in the nucleus of somite cells. Lipid like cytoplasmic dense mass and lipid droplets were also observed. Segmentation of somites seemed to be normal progress in ${\alpha}$-amanitin treated group. Center of somite, however, hollowed in longitudinal sectioned samples. These results suggested that so mites were already existed in the segmental plate as the form of somitomere. Segmented somites were contacted with neural tube or notochord and the somites were tightly connected with each other by the extracellular collagen fibrils which were secreted from neuroepithelium and somite cells. Somites are thought to differentiate into sclerotome, dermatome and myotome by these interactions.

  • 에틸렌 연료의 이중모드 스크램제트 연소기에서 연소와 충격파 열 발생의 과도 과정을 고해상도 기법을 이용하여 수치적으로 연구하였다. 연료 분사 이후 질량 공급에 의한 아음속 유동 감속을 위하여 연소기 확장부에 조절용 공기를 공급한다. 공기와 연료가 충분히 혼합된 수 ms 이후 점화가 이루어지며, 압력 상승은 격리부에 흡입구 노즐까지 전진하는 충격파 열을 형성한다. 이후 후방 공기공급을 중단하면 배출 과정이 진행되면서 후방 공기 공급 이전 상태로 서서히 복원된다. 본 연구의 결과는 이중모드 스크램제트 연소기에서 작동 영역과 특징의 이해를 돕는 상세 과정을 보여주었다.

Equilibrium Binding of Wild-type and Mutant Drosophila Heat Shock Factor DNA Binding Domain with HSE DNA Studied by Analytical Ultracentrifugation

  • We have investigated binding between wild-type and mutant Heat Shock Factor (HSF) DNA binding domains (DBDs) with 17-bp HSE containing a central 5'-NGAAN-3' element by equilibrium analytical ultracentrifugation using multi-wavelength technique. Our results indicate that R102 plays critical role in HSE recognition and the interactions are characterized by substantial negative changes of enthalpy (${\Delta}H^0_{\theta}=-9.90{\pm}1.13kcal\;mol^{-1}$) and entropy (${\Delta}S^0_{\theta}=-12.46{\pm}3.77cal\;mol^{-1}K^{-1}$) with free energy change, ${\Delta}G^0_{\theta}$ of $-6.15{\pm}0.03kcal\;mol^{-1}$. N105 plays minor role in the HSE interactions with ${\Delta}H^0_{\theta}$ of $-2.54{\pm}1.65kcal\;mol^{-1}$, ${\Delta}S^0_{\theta}$ of $19.28{\pm}5.50cal\;mol^{-1}K^{-1}$ and ${\Delta}G^0_{\theta}$ of $-8.35{\pm}0.05kcal\;mol^{-1}$, which are similar to those observed for wild-type DBD:HSE interactions (${\Delta}H^0_{\theta}=-3.31{\pm}1.86kcal\;mol^{-1}$, ${\Delta}S^0_{\theta}=17.38{\pm}6.20cal\;mol^{-1}K^{-1}$ and ${\Delta}G^0_{\theta}=-8.55{\pm}0.06kcal\;mol^{-1}$) indicating higher entropy contribution for both wild-type and N105A DBD bindings to the HSE.

  • 추진기관은 노즐을 통해 추력을 발생하며 축소-확대 형상의 초음속 노즐에서는 노즐의 설계 팽창비가 내부 유동의 전압력과 배압의 압력비보다 매우 클 때 충격파의 발생과 함께 경계층 박리를 유발한다. 노즐 내부에서의 충격파 발생과 유동의 박리는 주어진 유동의 압력비에 가장 적합한 노즐형상을 구현하는 것으로 실제의 구조적 노즐의 형상보다 짧은 노즐에서 나타나는 유동과 같은 현상을 보인다. 수치 해석적 방법으로 고정된 형상의 2차원 노즐 내부의 충격파와 경계층 박리 현상에 관한 연구를 수행하였고 Hunter가 행한 실험적 연구와 비교하였다. 수치해석은 TVD 기법을 이용한 압축성 유체 해석 코드와 SST 2방정식 난류 모델을 이용하여 수행되었다. 낮은 압력비에서의 충격파와 경계층과의 상호작용에 의한 $\lambda$형태의 충격파 시스템을 잘 보여주고 있고 추력 값의 비교를 통해 고정된 형상의 노즐을 이용하여 필요한 운용범위를 충족할 수 있음을 알 수 있었다.

  • 본 연구는 생존성 측면에서 잠수함의 내충격 설계시 고려해야 하는 수중폭발 충격응답해석 기법에 관하여 고찰하였다. 생존성을 고려하는 것은 현대의 모든 함정설계에 필수요건이며, 특히 잠수함의 수중폭발에 대한 내충격 설계는 반드시 고려되어야 할 사항이다. 기존의 수중함에 대한 내충격 설계는 가정된 이론에 의한 충격가속도를 정적해석의 결과를 통하여 단순하게 적용하였으나 본 연구에서는 직접해석법에 수중폭발 충격응답해석을 수행함으로써 기존의 방법보다 매우 합리적이고 신뢰성 높은 결과를 제시하였다. 특히 본 연구에서는 직접해석법 중에서 기존 수상함에서 널리 사용하고 있는 LS-DYNA/USA code일 적용보다 LS-DYNA code만으로 사용자가 보다 손쉽게 접근할 수 있는 ALE기법을 이용하여 잠수함 액화산소탱크를 대상으로 수중폭발해석 방식을 제안하고자 하였다.

Effects of Extracorporeal Shock Wave Therapy on Ankle Function, Range of Motion, and Dynamic Balance in Patients with Chronic Ankle Instability

  • Purpose: This study investigated the short-term effectiveness of extracorporeal shock wave therapy (ESWT) on pain, the ankle instability, the ankle function, dorsiflexion range of motion (ROM), and dynamic balance in patients with chronic ankle instability (CAI). Methods: Eighteen participants were divided into an experimental (n=9) and control group (n=9). The ESWT in the experimental group was applied to the lateral collateral ligament in combination with the tibialis anterior whereas the ESWT was applied to the lateral collateral ligament of the ankle alone in the control group. Pain, the ankle instability, the ankle function, dorsiflexion ROM, and dynamic balance were measured using the Visual analog scale, Cumberland ankle instability tool, American Orthopedic Foot and Ankle Society ankle-hindfoot score, weight-bearing lunge, and Y-balance test, before and after ESWT intervention. Results: Significant interactions (group × time) and time effects were observed in the dorsiflexion ROM and dynamic balance. Bonferroni's post-hoc analysis showed that the experimental group revealed a more significant change in dorsiflexion ROM and dynamic balance than the control group. There was a significant time effect in the pain, the ankle instability, and the ankle function, but no significant interaction (group × time) was observed. Conclusion: The ESWT could improve the pain, ankle instability, ankle function, dorsiflexion ROM, and dynamic balance in patients with CAI. Furthermore, the ESWT combined with lateral ankle ligaments and tibialis anterior more improves the dorsiflexion ROM and dynamic balance.

Interaction of a 22 kDa Peptidyl Prolyl cis/trans Isomerase with the Heat Shock Protein DnaK in Vibrio anguillarum

  • Peptidyl prolyl cis/trans isomerases (PPIases) catalyze the cis/trans isomerization of peptidyl-prolyl peptide bonds preceding prolines. We investigated the protein-protein interaction between a 22 kDa PPIase (VaFKBP22, an FK506-binding protein) and the molecular chaperone DnaK derived from Vibrio anguillarum O1 (VaDnaK) using GST pull-down assays and a bacterial two-hybrid system for in vivo and in vitro studies, respectively. Furthermore, we analyzed the three-dimensional structure of the protein-protein interaction. Based on our results, VaFKBP22 appears to act as a cochaperone of VaDnaK, and contributes to protein folding and stabilization via its peptidyl-prolyl cis/trans isomerization activity.
