Isolation and Biocontrol Potential of Bacillus amyloliquefaciens Y1 against Fungal Plant Pathogens

  • This study was performed to investigate thermophilic bacteria from soil having broad antifungal spectrum against Rhizoctonia solani, Colletotrichum gloeosporioides, Phytophthora capsici, Fusarium oxysporum f.sp. lycopersici, and Botrytis cinerea. One isolate selected could resist heat shock of $60^{\circ}C$ for one hour, and had broad antifungal activity in dual culture assay against all tested fungal pathogens and was identified as Bacillus amyloliquefaciens Y1 using 16S rRNA gene sequence. Further investigation for antifungal activity of bacterial culture filtrate (BCF) and butanol crude extract (BCE) of various concentrations showed broad spectrum antifungal activity and fungal growth inhibition significantly increased with increasing concentration with highest growth inhibition of 100% against R. solani with 50% BCF and 11 mm of zone of inhibition against R. solani with 4 mg BCE concentration. Treatment of butanol crude extract resulted in deformation, lysis or degradation of C. gloeosporioides and P. capsici hyphae. Furthermore, B. amyloliquefaciens Y1 produced volatile compounds inhibiting growth of R. solani (70%), C. gloeosporioides (65%) and P. capsici (65-70%) when tested in volatile assay. The results from the study suggest that B. amyloliquefaciens Y1 could be a biocontrol candidate to control fungal diseases in crops.

  • This study was conducted to evaluate the characteristics of installation dimensions of speed control humps based on the theoretical dynamic analyses. The field surveys of speed control humps were performed first to compare their sizes with those suggested in the installation standard. Then, the displacement response spectra under impact loads were obtained using the single degree of freedom models where the vehicle and human were assumed to combine completely. The analysis results showed that the human perception became larger as the width and height of the humps increased, but the extremely higher uncomfortableness should be avoided. In addition, the ratio between the width and height should be considered when the humps are designed and installed because the ratio governs the vertical acceleration magnitude.

  • Sonoluminescence (SL) characteristics such as pulse shape, radiance and spectrum radiance from submicron bubbles were investigated. In this study, a set of analytical solutions of the Navier-Stokes equations for the gas inside bubble and equations obtained from mass, momentum and energy equations for the liquid layer adjacent the bubble wall were used to estimate the gas temperature and pressure at the collapse point, which are crucial parameters to determine the SL characteristics. Heat transfer inside the gas bubble as well as at the liquid boundary layer, which was not considered in the most of previous studies on the sonoluminescence was taken it into account in the calculation of the temperature distribution inside the bubble. It was found that bremsstrahlung is a very possible mechanism of the light emission from either micron or submicron bubbles. It was also found that the peak temperature exceeding $10^{6}$ K in the submicron bubble driven at 1 MHz and 4 atm may be due to the rapid change of the bubble wall acceleration near the collapse point rather than shock formation.

  • Purpose: Satisfying the environmental regulations, the automobile manufacturer should install urea tank, which is a key component of the urea system. However, due to the limitations of existing layouts, it may be mounted which is disadvantageous to vibration and shock resulting in durability robust. analyze the factors affecting the durability life of urea tank and the vibration characteristics through RLDA. In this study, clarify the limit of the current practice test method of urea tank and analyze the possibility of the new vibration test method in the system unit reflecting the characteristics of actual use condition. Methods: Analyzing the factors affecting the durability life of urea tank and the vibration characteristics through PSD & FDS of RLDA that actual vehicle driving data on durability test road. Results: The limit of the uniform width/single frequency test method of urea tank is clarifed and the positive prospects of the new test method are discovered. Conclusion: The vibration durability test with PSD method in system unit effectively reflects the magnitude and frequency characteristics of field vibration.

  • Using the T3 free-piston shock tunnel in ANU, the cavity frequency and flow characteristics of no mass-injection, inclined mass-injection before the cavity, parallel or reverse mass-injection in the cavity are investigated in the case of Mach 3.7 inflow condition. No mass-injection doesn't have the harmonic frequencies but has high amplitude of pressure spectrum at 10 kHz. Inclined mass-injection attenuates the cavity flow fluctuation as disturbing the shear layer reflection at the trailing edge. Parallel mass-injection flow reflects at the trailing edge of the cavity directly hence, increases the cavity flow fluctuation at high injection pressure.

  • The objective of this study is to investigate tank container to be used as fuel tank for LNG fueled ship. Feasibility of tank container to the fuel tank of LNG fueled ship is addressed and the advantage of tank container as fuel tank of ship is investigated. Conceptual configuration of the tank container is designed as well as structural analyses based on finite element method are carried out to meet the design regulation suggested by shipping register. Static loading is considered by structural analysis and impact test is performed. It is necessary to require SRS(shock response spectrum) in order to investigate structural safety which can meet.

SNR 0104-72.3: A remnant of Type Ia Supernova in a Star-forming region?

  • We report our 110 ks Chandra observations of the supernova remnant (SNR) 0104-72.3 in the Small Magellanic Cloud (SMC). The X-ray morphology shows two prominent lobes along the northwest-southeast direction and a soft faint arc in the east. Previous low resolution X-ray images attributed the unresolved emission from the southeastern lobe to a Be/X-ray star. Our high resolution Chandra data clearly shows that this emission is diffuse, shock-heated plasma, with negligible X-ray emission from the Be star. The eastern arc is positionally coincident with a filament seen in optical and infrared observations. Its X-ray spectrum is well fit by plasma of normal SMC abundances, suggesting that it is from shocked ambient gas. The X-ray spectra of the lobes show overabundant Fe, which is interpreted as emission from the reverse-shocked Fe-rich ejecta. The overall spectral characteristics of the lobes and the arc are similar to those of Type Ia SNRs, and we propose that SNR 0104-72.3 is the first case for a robust candidate Type Ia SNR in the SMC. On the other hand, the remnant appears to be interacting with dense clouds toward the east and to be associated with a nearby star-forming region. These features are unusual for a standard Type Ia SNR. Our results suggest an intriguing possibility that the progenitor of SNR 0104-72.3 might have been a white dwarf of a relatively young population.

The Hearing Ability of Coralfish Chromis notatus to Low Frequency Sound 2. The Auditory Critical Ratio and Hearing Index (저주파음에 의한 자리돔의 청각 능력 2. 청각 임계비 및 청각능력지수)

  • In order to obtain the fundamental data on the auditory thresholds of fishes for catching method using low frequency sound, the auditory thresholds of coralfish Chromis notatus were measured in the presence of masking noise in the spectrum level range of 73~83dB re l$\mu$Pa/√Hz by heartbeat conditioning technique using pure tones coupled with a delayed electric shock. Critical ratios were about 23~41dB at measurement frequency, The critical ratio increased almost linearly with increasing frequency from 500Hz. The noise spectrum level at the start of masking was about 60~65dB. This suggests that hearing of coralfish is masked in the natural environment with the noise spectrum level above 60dB. The sound pressure level of which the signal sound of 300Hz is recognized by coralfish under the ambient noise is above 88dB and the critical ratio of them is above 23dB. The hearing index of coralfish with ambient noise was 81.

  • Nocardia farcinia, an aerobic, gram-positive bacilli actinomycetes of the genus Nocardia, is an uncommon pathogen found in humans. The most common Nocardia infection sites are the lung, central nervous system, and skin. Even though hematogenous dissemination can occur, isolation of the organism from blood cultures is very rare. We report a case of Nocardia infection that was isolated on blood cultures. A 59-year-old male with a medical history that includes a liver transplantation 6-years prior due to hepatocellular carcinoma secondary to chronic hepatitis B, developed pneumonia and was transferred to Severance Hospital. At the time of admission, the patient's initial exam showed hypothermia, tachypnea, and hypotension. His chest radiograph showed severe pneumonia and a large abscess on left upper lobe. Under the presumptive diagnosis of bacterial pneumonia or other opportunistic infection, we started broad spectrum antibiotics. However, he developed Nocardia sepsis, rapidly deteriorated, and subsequently died.

  • The only law related to airblast and ground vibration control in Korea is the Noise and Vibration Control Act enforced by the Ministry of Environment. But this law mainly deals with the annoyance aspects of noises and vibrations in ordinary human life. Hence, the law defines the safety criteria of ground vibration as the vibration level (VL) of dB(V) unit. The ground vibrations produced from blasting, however, have the unique characteristics that can be shown in shock vibrations, and the duration is also very short compared to the vibrations from machinery, tools or facilities. Hence, vibration regulations for blasting operations usually define the safety criterion as the peak particle velocity (PPV) considering the effect of ground vibrations to structural damage. Notwithstanding, there are several attempts that predict VL from PPV or estimate VL based on the scaled distances (SD; in unit of $m/kg^{1/2}$ or $m/kg^{1/3}$) without considering their frequency spectra. It appears that these attempts are conducted mainly for the purpose of satisfying the law in blasting contracts. But, in principle there could no correlation between peaks of velocity and acceleration over entire frequency spectrum. Therefore, such correlations or estimations should be conducted only for the waves with the same or very similar frequency spectra.
