Ceftizoxime sodium is a new synthetic ${\beta}$-lactam antibiotic combining potent antibacterial activity with high stability to a wide range of bacterial ${\beta}$-lactamase. This experiment was achieved to evaluate the antibacterial activities of ceftizoxime sodium againist Gram negative enteric bacteria isolated from in outpatient visiting Yeungnam university hospital and to study the emergence of drug induced bacterial varients which resist to ceftizoxime in vitro. The antibacterial activity of the ceftizoxime was compared with that of antibiotics and its effect on population of normal intestinal flora in mice was observed. The results are summarized as follows : 1. Highly effective antibacterial activity of ceftizoxime against Gram negative enteric bacilli was demonstrated and this antibacterial activity was superior to that of ampicillin. 2. Several test strains shows multiple antibiotic resistence. Among 15 strains of Escherichia coli, 1 strain was resistent to ampicillin, cefadroxyl, gentamicin, tetracycline, and 2 strains were resistent to ampicillin, cefadroxyl, tetracycline, five strains of Escherichia coli and Enterobacter cloacae was resistent to amplicillin, tetracycline and Shigella dysenteria was resistent to ampicillin, gentamicin, tetracycline. 3. The frequency of in vitro emergence of resistent varients among ceftizoxime sensitive bacteria in the presence of increasing concentrations of the compound was found to be low. 4. Plasmid was isolated in 6 of 9 strains (6 strains of Escherichia coli, Shigella dysenteriae, Enterobacter cloaceae and Salmonella typhi) That showed different antibiotic resistance. They were 5 strains of Escherichia coli and 1 strain of Shigella dysenteriae. However, plasmid could not be considered as a hallmark for antibiotic resistance by this. Further studies with curing experiment are to be accomplished for this purpose. 5. Changes in the bacterial count of normal intestinal flora following 25mg/kg/day administration of ceftizoxime over S consecutive days were not significant. In conclusion, ceftizoxime appeared to be a drug of choice in the treatment of Gram negative enteric bacilli infection.

This study was performed to investigate the antimicrobial effect of the Hedyotis diffusa extracts against food-borne pathogens. First, the Hedyotis diffusa was extracted with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of the Hedyotis diffusa extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Hedyotis diffusa showed the highest antimicrobial activity against Staphylococcus aureus and Shigella flexneri. Synergistic effect in inhibition was observed when Hedyotis diffusa extract was mixed Sophora subprostrata extract as compared to each extracts alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of Hedyotis diffusa against Staphylococcus aureus and Salmonella Typhimurium. The methanol extract of Hedyotis diffusa had strong antimicrobial activity against Staphylococcus aureus at the concentration of 4,000 ppm. At this concentration, the growth of Staphylococcus aureus was retarded more than 36 hours and up to 24 hours for Salmonella Typhimurium. In conclusion, the methanol extracts of Hedyotis diffusa inhibit efficiently Staphylococcus aureus and Salmonella Typhimurium.
The viability of Shigella sonnei, a significant cause of gastroenteritis in Korea, on TSA plates was determined after sublethal heating treatments and NaCl treatments. In addition, recovery levels of sublethally injured cells on TSA plates containing different concentrations of NaCl (TSAS) were investigated. The viability decreased significantly with increasing degree of sublethal heating treatments, but increases in NaCI treatment concentration from 0 to 6% had little effect on the viability. After being sublethally treated at $55^{\circ}C$ for 30 min, bacterial populations were reduced by 7.58, 7.83 and 7.93 log CFU/mL on 2, 4, and 6% TSAS, respectively. After being sublethally treated at $60^{\circ}C$ for 30 min, bacterial populations were reduced by 6.71, 6.73, and 6.73 log CFU/mL on 2, 4 and 6% TSAS, respectively. Decimal reduction times (D-values) decreased with increasing NaCl treatment concentrations after sublethal heating at 55 or $60^{\circ}C$. These data imply that the S. sonnei cells sublethally injured by insufficient heating processes had a lower recovery rate with increasing NaCl concentrations in the recovery media.
Diarrheal diseases constitute one of the major causes of morbidity and mortality in infants and young children globally. One of the main microorganisms causing diarrheal diseases is Campylobacter jejuni. For treatment of these diseases, Portulaca oleracea has been widely used as a folk remedy for a long time. This study was performed to investigate the antimicrobial activity of P. oleracea against gastroenteritis pathogens including C. jejuni. P. oleracea was extracted with petroleum ether, chloroform, ethylacetate, methanol, and hot water. The antimicrobial activity of the P. oleracea extracts was determined using the paper disc method, minimum inhibitory concentration, and the liquid culture method. The 10 $mg/m{\ell}$ ethylacetate extract showed the strongest antimicrobial activity against Salmonella typhimurium, Salmonella enteriditis, and Shigella spp.. The hot water extract from P. oleracea showed the highest anti-microbial activity against C. jejuni at 10~20 $mg/m{\ell}$. The hot water extract of P. oleracea retarded the growth of C. jejuni for 36 hr at $42^{\circ}C$.
The antimicrobial substance produced by Pseudomonas aeruginosa KLP-2 strain was purified and identified. The substance was identified as a pyocyanine by the fast atom bombardment mass(FAB-MS). In physic-chemical properties, the pyocyanine was dark blue needles, and was soluble in various organic solvents such as chlorogorm, methanol, ethanol and ethyl acetae. The pyocyanine possessed a ultraviolet absorbance spectrum in methanol, 0.1 M HCl, and chlorogorm. The maximum absorption peak of the pyocyanine showed at 318 mm in methanol. The molecular formula of the pyocyanine was determined to the $C_{13}$ H$_{10}$ N$_{2}$O and protonate molecular ion species (M+H)$^{+}$ was observed at m/z 211 by FAB-MS. The pyocyanine showed antimicrobial against Bacillus cereus, Bacillus subtilis, Micrococcus luteus, Rodococcus equi, Staphylococcus aureus, Streptococcus faecalis, E. col, Legionella pneumophila, Shigella flexneri Shigella boydii, shgella sonnei, NAG Vibrio cholerae, Vibrio parahaemolyticus, Vibro vulnificus, Yersinia enterocolitica, and Saccharomyces cerevisiae. However, Salmonella spp. Shigela dysenteriae, 3 strains of Pseudomonas aeruginosa, Klebsiela pneumoniae, and Aspergillus niger were resistant to the pyocyanine. The pyocyanine showed the highest antimicrobial activity aganist Legionella pneumophila based on the size of inhibition zone by the disk contained 0.5 $\mu\textrm{g}$ of the pyocyanine.e.
Widespread distributions of repetitive DNA elements in bacteria genomes are useful for analysis of genomes and should be exploited to differentiate food-borne pathogenic bacteria among and within species. Enterobacterial repetitive intergenic consensus (ERIC) sequence has been used for ERIC-PCR genomic fingerprinting to identify and differentiate bacterial strains from various environmental sources. ERIC-PCH genomic fingerprinting was applied to detect and differentiate four major Gram-negative food-borne bacterial pathogens, Esherichia coli, Salmonella, Shigella, and Vibrio. Target DNA fragments of pathogens were amplified by ERIC-PCR reactions. Dendrograms of subsequent PCR fingerprinting patterns for each strain were constructed, through which relative similarity coefficients or genetic distances between different strains were obtained numerically. Numerical comparisons revealed ERIC-PCR genotyping is effective for differentiation of strains among and within species of food-borne bacterial pathogens, showing ERIC-PCR fingerprinting methods can be utilized to differentiate isolates from outbreak and to determine their clonal relationships among outbreaks.

The development of polymerase chain reaction (PCR) technology has the potential to solve for isolating pathogenic microorganisms from environmental samples than traditional plate counting methods. We have been detected pathogenic bacteria from raw water and water treatment process in Busan metropolitan city by PCR method. According to the result of survey from July 2004 to October 2005, 80 out of 92(87.0%) were positive for bacterial pathogens in raw water samples and positive rate of Shigella spp., Yersinia spp., Salmonella spp. and Legionella spp. were 46.2%, 40.7%, 17.6% and 9.9%, respectively. Pathogenic bacteria in raw water was mainly distributed through the lately Autumn to the winter and more highly detected Maeri than Mulgum region. During the period of survey in water treatment process, Shigella spp. was highly detected but all of bacterial pathogens were entirely removed after in post-ozonation step. These suggest that waters supplied in Busan metropolitan city may be safe against the pathogenic bacteria.
Antibacterial activities in each part of watercress(Oenanthe javanica D.C.) grown under different culture conditions were measured to determine the possibility to use watercress as a resource to develop the antibacterial substance. The leaves of watercress were extracted with methanol and the methanol extract was further fractionated with various organic solvents. Antibacterial activities against Shigella dysenteriae ATCC 9361 in all fractions were determined according to the agar diffusion method using paper disc. Methanol extract of watercress leaves was more effectively inhibited the growth of the tested bacteria than the extracts of roots or stems at the concentration of 0.5 g eq./disc, and the extract of watercress from Hwasoon was the most effective one as compared to others. Phenolic and neutral fractions fractionated from methanol extract of watercress had a considerable inhibiting activity on the growth of the bacteria, but acidic and basic fractions did not show any inhibitory effect. Minimum inhibitory concentrations of phenolic and neutral fractions against Shigella dysenteriae ATCC 9361 were $400\;{\mu}g/disc$ and $550\;{\mu}g/disc$, respectively.
1. At selected time intervals, viable cell plate counts were run. After the 12 hours incubution the number of viable cells was maximum. 2. The morphology of L. acidophilus in tomato juice was thicker rods than those of in milk media. 3. In the assay of antimicrobial activity; (1) The antimicrobial activity of acidophilus tomato juice showed bigger inhibitory zone than acidophilus milk in the well-cup assay technique. (2) The inhibitory zone against Shigella dysenteriae was the biggest among the other pathogenic organisms in the disc assay. 4. The antimicrobial activity of sephadex G-50 gel filtration chomatography showed in the 5th fraction. 5. The spectra of ultraviolet absorption exhibited around maximum peak at 270nm.
