Nitric oxide (NO) produced from endothelial cells plays a critical role in vascular physiology. The regulation of endothelial NO synthase (eNOS) involves various mechanisms including multiple Ser/Thr phosphorylations. Recently, eNOS-Ser617 was newly recognized to be phosphorylated in response to humoral factors including vascular endothelial growth factor. However, it remains unknown whether and how eNOS-Ser617 phosphorylation is stimulated by shear stress, the primary stimulus of endothelial NO production. This issue was explored in the present study using cultured bovine aortic endothelial cells (BAECs). Over-expression of a constitutively active protein kinase B(Akt) mutant in BAECs increased Ser617 phosphorylation while constitutively active protein kinase A mutant had no effect. When BAECs were subjected to an arterial level of laminar shear stress, eNOS-Ser617 phosphorylation was clearly increased in a time-dependent manner. Shear stress also stimulated Akt phosphorylation at Thr308, one of the key regulatory sites. The time courses of eNOS-Ser617 and Akt-Thr308 phosphorylations appeared to be very similar. These results suggested that eNOS-Ser617 phosphorylation, mediated by Akt, is a physiological response to the mechanical shear stress, involved in the regulation of NO production in endothelial cells.
전난류에서 파와 해류가 합성하였을 때 발생하는 해저면 마찰력을 계산하는 방법을 고찰하였다. 전난류에서 일방향 흐름에 의한 마찰력의 산정방법으로 절점조정법을 제시하였으며, Bijker의 관측자료와 비교하여 절점조정치를 산정하였다. 파와 해류의 합성류에 의한 마찰력 계산방법으로 수정된 Bkjker 모형(BYO Model)과 수정된 Fredsoe 모형(FY Model)을 Bijker의 관측자료에 적용하였으며, BYO 모형에서 최대마찰력을 산정하는데 있어 새로운 개선책을 제시하였다.
An optimization method of patch shape was developed in this study, in order to improve repair of cracked plates. It aimed to minimize three objectives: stress intensity factor, patch volume and shear stresses in the adhesive film. The choice of these objectives ensures improving crack repair, gaining mass and enhancing the adhesion durability between the fractured plate and the composite patch. This was a multi-objective optimization combined with Finite elements calculations to find out the best distribution of patch height with respect to its width. The implementation of the method identified families of optimal shapes with specific geometric features around the crack tip and at the horizontal end of the patch. Considerable mass gain was achieved while improving the repair efficiency and keeping the adhesive shear stress at low levels.
전단응력에 의한 전단강도 및 모드 II 파괴인성은 이산화탄소 지중저장에서의 덮개암 및 주입층의 안정성 평가에 활용되는 중요한 인자들이다. 본 연구에서는 짧은 보 압축시험을 이용하여 코코니노 사암의 전단강도 및 모드 II 파괴인성을 측정하였다. 측정된 평균 전단강도는 23.53 MPa이며, 모드 II 파괴인성은 1.58 MPa${\surd}$m이다. 응력확대계수(stress intensity factor)는 변위외삽법(displacement extrapolation method)을 이용한 유한요소법으로 결정하였다. 또한 이축응력(biaxial stress)과 수분포화(water saturation)가 모드 II 파괴인성에 미치는 영향을 분석하였다. 그 결과 이축응력이 증가할수록 파괴인성도 증가하였고, 완전포화된 시험편의 파괴인성은 건조상태의 파괴인성보다 대략 11.4% 감소하였다.

Background In addition to vascular endothelial cells, vascular smooth muscle cells (VSMCs) are subject to continuous shear stress because of blood circulation. The angiogenic properties of VSMCs in extracranial arteriovenous malformations (AVMs) may exceed those of normal blood vessels if the body responds more sensitively to mechanical stimuli. This study was performed to investigate the hypothesis that rapid angiogenesis may be achieved by mechanical shear stress. Methods VSMCs were obtained from six patients who had AVMs and six normal controls. The target genes were set to angiopoietin-2 (AGP2), aquaporin-1 (AQP1), and transforming growth factor-beta receptor 1 (TGFBR1). Reverse-transcriptase polymerase chain reaction (RT-PCR) and real-time PCR were implemented to identify the expression levels for target genes. Immunofluorescence was also conducted. Results Under the shear stress condition, mean relative quantity values of AGP2, AQP1, and TGFBR1 in AVM tissues were 1.927±0.528, 1.291±0.031, and 2.284±1.461 when compared with neutral conditions. The expression levels of all three genes in AVMs were higher than those in normal tissue except for AQP1 under shear stress conditions. Immunofluorescence also revealed increased staining of shear stress-induced genes in the normal tissue and in AVM tissue. Conclusions Shear stress made the VSMCs of AVMs more sensitive. Although the pathogenesis of AVMs remains unclear, our study showed that biomechanical stimulation imposed by shear stress may aggravate angiogenesis in AVMs.
The use of fracture mechanics has traditionally concentrated on crack growth under an opening mechanism. However, many service failures occur from cracks subjected to mixed-mode loading. Hence, it is necessary to evaluate the fatigue behavior under mixed-mode loading. Under mixed-mode loading, not only the fatigue crack propagation rate is of importance, but also the crack propagation direction. In modified range 0.3$\leq$a/W$\leq$0.5, the stress intensity factors (SIFs) of mode I and mode II for the compact tension shear (CTS) specimen were calculated by using elastic finite element analysis. The propagation behavior of the fatigue cracks of cold rolled stainless steels (STS304) under mixed-mode conditions was evaluated by using K$\_$I/ and $_{4}$ (SIFs of mode I and mode II). The maximum tangential stress (MTS) criterion and stress intensity factor were applied to predict the crack propagation direction and the propagation behavior of fatigue cracks.
An assessment of liquefaction potential is made in principle by comparing the shear stress induced by earthquake to the liquefaction strength of the soil. In this study, a modified method based on Seed and Idriss theory is developed for evaluating liquefaction potential. The shear stress in the ground can be evaluated with seismic response analysis and the liquefaction strength of the soil can be investigated by using cyclic triaxial tests. The cyclic triaxial tests are conducted in two different conditions in order to investigate the factors affecting liquefaction strength such as cyclic shear stress amplitude and relative density. And performance of the modified method in practical examples is demonstrated by applying it to liquefaction analysis of artificial zones with dimensions and material properties similar to those in a typical field. From the result, the modified method for assessing liquefaction potential can successfully evaluate the safety factor under moderate magnitude(M=6.5) of earthquake.
The general solution of the anti-plane shear problem for the curved interfacial crack between viscoelastic foam and composites was investigated with the complex variable displacement function and Kelvin-Maxwell model. The Laplace transform was applied to treat the viscoelastic characteristics of foam in the analysis. The stress intensity factor near the interfacial crack tip was predicted by considering both anisotropic and viscoelastic properties of two different materials. The results showed that the stress intensity factor increased with increasing the curvature of the curved interfacial crack and it also increased and eventually converged to a specific value with increasing time.
Exact solutions for stresses, strains, displacements, and the stress concentration factors of a rectangular plate perforated by an arbitrarily located circular hole subjected to in-plane pure shear loading are investigated by two-dimensional theory of elasticity using the Airy stress function. The hoop stresses, strains, and displacements occurring at the edge of the circular hole are computed and plotted. Comparisons are made for the hoop stresses and the stress concentration factors from the present study and those from a rectangular plate with a circular hole under uni-axial and bi-axial uniform tensions and in-plane pure bending moments on two opposite edges.
본 연구는 식생호안의 안정성 평가에 중요한 인자인 식생 근모량에 따른 식생호안의 전단강도와 침식특성을 분석하였다. 식생호안의 평균근모량은 자체 제작한 채취기를 이용하여 측정하였다. 본 연구에서 사용된 식생은 식생호안에서 다소 우점종인 달뿌리풀이다. 흐름 특성별 실험결과 근모량이 증가함에 따라 토양의 전단강도가 증가함을 확인하였고, 아울러 근모량의 증가에 따라 침식율은 지수함수적으로 감소함을 확인하였다. 식생토양의 전단강도가 증가함에 따라 침식율이 지수함수적으로 감소되어 근모량에 기인한 전단강도의 증가는 침식 저항능력의 증가를 가져왔다. 아울러 근모량에 따른 전단강도와 침식율의 상관분을 실시하여 높은 상관계수를 가진 식을 제시하였다. 식생을 가진 호안의 수리적 안정성에는 근모량의 증가에 의한 전단강도와 유수의 특성인 Froude수에 지배적임을 확인하였다.
