Hymenoxys brachyactis의 화학성분에 관한 연구(II) (The study of chemical substances in Hymenoxys brachyactis(II))

  • Hymenoxys brachyactis지상부의 디클로로메탄 추출액으로부터 3개의 sesquiterpene lactone들과 새로운 lactone화합물인 isohymenograndin을 확인하였다. 그리고 2개의 sesquiterpene lactone축합물인 biennine C와 독성이 있는 것으로 알려진 flavone화합물인 hispidulin을 분리하여 각각을 고자장 NMR, Mass등의 기기를 이용하여 구조결정 하였다. Biennine C는 gas chromatography와 Mass분석을 통하여 독성화합물 hymenoxon과 화합물 8과의 Diels-Alder축합물임을 확인하였다. biennine C를 포함한 이상의 lactone화합물들은 ${\alpha},{\beta}$-unsaturated lactone으로서 독성화합물군으로서 알려진 hymenovin과 공통적인 구조를 갖고 있어서 생체독성을 나타낼 가능성이 높은 화합물들로 사료된다.

  • The chromatographic separation of a methylene chloride extract of Artemisia rubripes led to the isolation of a new sesquiterpene lactone (3), together with four known compounds, a coumarin (2) and three terpenes (1, 4, and 5). Their structures were characterized to be $1{\beta},6{\alpha}$- dihydroxy-4(15)-eudesmene (1), scopoletin (2), $1{\alpha},4{\beta}-dihydroxy-8{\alpha}$-acetoxy-guaia-2,10(14), 11(13)-triene-6,12-olide (3), $1{\alpha},4{\beta}$ -dihydroxy-8${\alpha}$-acetoxy-guaia-2,9,11(13)-triene-6,12-olide (4), and $\beta$ -sitosterol-3-O-${\beta}$-D-glycoside (5) by spectroscopic means.

  • To elucidate the molecular mechanisms for the suppression of LPS-induced nitric oxide (NO) production by a dehydrocostus lactone (DL) from Saussurea lappa, we examined the preventive effect of this compound on $NF-{\kappa}B$ activation in LPS-treated RAW 264.7 macrophages and U937 human monocytic cells. The results suggest that the suppression of NO production is mediated by the inhibitory action on the i-NOS gene expression through the inactivation of $NF-{\kappa}B$ and this sesquiterpene lactone can act as a pharmacological inhibitor of the $NF-{\kappa}B$ activation.

  • Nine sesquiterpene lactones, which were isolated from Hemisteptia lyrata Bunge. Chrysanthemum zawadskii Herbich var. latilobum Kitamura and Chrysanthemum boreale Makino were evaluated for the farnesyl-protein transferase (FPTase) inhibitor, conducted by the scintillation proximity assay (SPA). The angeloylcumambrin B and tigloylcumambrin B inhibited a recombinant rat FPTase with $IC_{50}$ value of $78\;{\mu}g/ml$ $(225\;{\mu}M)$ and $90\;{\mu}g/ml$ $(260 \;{\mu}M)$, respectively.

  • Three known sesquiterpene lactones were isolated from the n-BuOH traction of the roots of Ixeris sonchifolia. Their structures were identified as macrocliniside A (1), glucozaluzanin C (2), and ixerin H (3), by spectral analyses. Compounds 1 and 2 are guaiane-type, while compound 3 is a germacrane-type sesquiterpene lactone. Compounds 1-3 are first isolated from I. sonchifolia.

  • The phytochemical study of the aerial parts of Artemisia sylvatica (Asteraceae) led to the isolation of nine sesquiterpene lactones, l$\alpha$-hydroperoxy-rupicolin A acetate (1), rupicolin B acetate (2), rupicolin A acetate (3), l$\alpha$-hydroxy-4$\alpha$-hydroperoxy-bishopsolicepolide (4), 1$\alpha$-hydroperoxy-4$\beta$-hydroxy-8$\alpha$-acetoxy-guaia-2,9,11 (13)-triene-6$\alpha$,12-oxide (5), 1$\alpha$-hydroperoxy-4$\alpha$-hydroxybishopsolicepolide (6), l$\alpha$,4$\beta$-dihydroxy-8$\alpha$-acetoxy-guaia-2,9,11(13)-triene-6$\alpha$,12-olive (7), rupicolin A (8) and l$\alpha$,4$\alpha$-dihydroxy-bishopsolicepolide (9). Their structures were established by chemical and spectroscopic methods.

Cytotoxic Effects on Human Cancer Cells and Apoptosis of a Sesquiterpene Lactone from Saussure lappa

  • In order to study the cytotoxic properties of sesquitepenes, dehydrocostus lactone (DL) and costunolide from Saussurea lappa, cytotoxicity was measured by SRB method using various human cancer cell lines. Dehydrocostus lactone(DL) and costunolide exhibited significant cytotoxicity against A-549, SK-OV-3, SK-MEL-2, XF-498 and HCT 15 cells. The U937 human leukemia cells treated with DL showed several apoptotic evidences like chromosome condensation and formation of apoptotic bodies. From the results of FACS analysis, early apoptosis was observed by phosphatidylserine externalization detected by annexin V-FITC. Furethermore, these studies determined hypodiploid contents and effects on the cell phase distribution of DL-treated U937 cells. After exposure of U937 cells to $30\mu\textrm{M}$ DL effectively led to G2/M modified cell cycle distribution within 24hr. These observations suggest that DL can be used efficiently for the cancer treatment.

  • The young leaf of Perilla species was cultured by two stage culture system using the medium containing mevalonic acid lactone. The growth rate and productivity of essential oil of callus were increased. The essential oil from intact plant and callus was also analysed. Sesquiterpene hydrocarbons and one sesquiterpene alcohol were identified in essential oils of callus.
