Effect of Cyclohexanone Treatment on the Activities of Oxygen Free Radical Metabolizing Enzyme in the Liver Damaged Rats (급성 간손상 실험동물에 Cyclohexanone투여가 Oxygen Free Radical 대사효소 활성에 미치는 영향)

  • Effect of cyclohexanone treatment on the activities oxygen free radical and cyclohexanone metabolizing enzyme in acute liver damaged rats, was investigated. Acute liver damage was induced in rats with pretreatment of 50% $CCl_4$ in olive oil(0.1ml/100g body wt) intraperitoneally 3 times every other day. Cyclohexanone(1.56g/kg body wt, i.p.) was administered to the animals 24 hours after the last Pretreatment of CC1$_4$. Rats were sacrificed at 4 hours after injection of cyclohexanone. On the basis of liver weight/body weight(%), serum levels alanine aminotransferase activity and hepatic protein content, cyclohexanone treatment to acute liver damaged animals led to the more enhanced liver damage. On the other hand, injection of cyclohexanone to the rats led to the increased activities of hepatic cytochrome P-450 dependent aniline hydroxylase and xanthine oxidase. Furthermore, by treatment of cyclohexanone to the acute liver damaged rats hepatic xanthine oxidase activity was more increased than the $CCl_4$ treated rats. In case of oxygen free radical scavenging system, the hepatic glutathione content and the activities of hepatic glutathione S-transferase, catalase, superoxide dismutase were generally increased by injection of cyclohexanone to rats, and the hepatic glutathione content, catalase and alcohol dehydrogenase activities were more decreased in liver damaged rats by the treatment of cyclohexanone. In conclusion, the cyclohexanone treatment to acute liver damaged rats led to enhancement of liver damage that may be due to oxygen free radical together with cyclohexanone.

  • To evaluate the effects of ingestion of alcoholic drinks on the toxicities of industrial compounds, cyclohexane (CH) was intraperitoneally administrated to rats (1.56g/kg body weight), which had been ingested 15% ethanol for up to 6 weeks,4 times by once a day and every other day. Following the last treatment of ethanol or CH, blood and liver tissues were collected after 4 hours prior to sacrifice of animals. By the injection of CH, liver weight (% of body weight) and xanthine oxidase activity in serum were increased, and glucose-6-phasphatase (G6P) activity in liver was decreased compared to them of control group. The activities of CH metabolizing enzymes, such as cytochrome P450 dependent aniline hydroxylase (CYPdAH) and alcohol dehydrogenase (ADH), were significantly increased by injection of CH, and those activities were the highest in CH-injected group after pretreated with alcohol. Ultrastructurally. both of alcohol treatment and CH injection induced transforming into the smooth-endoplasmic reticulum from rough-endoplasmic reticulum, the those rate was the highest in case of CH-injection after pretreated with alcohol. From these results, it is suggested that alcohol intake on a level without alcoholic degeneration of hepatocytes could enhance the CH metabolism of liver.

Effect of Circadian Rhythms on the Xylene Metabolizing Enzyme Activities in Rats (Xylene 대사 효소 활성에 미치는 주.야 시차의 영향)

  • To evaluate an effect of circadian variation on the xylene metabolizing enzyme activities, 50% m-xylene in olive oil(0.25 $m\ell$/100g body weight) was intraperitoneally administered to the rats every other day for 6 days both in the night; 24:00 and the day; 12:00. Then animals were sacrigiced at 8hr after last injection of m-xylene. Hepatic microsomal cytochrome p450 contents were more increased both in control and xylene treated rats of night phase than those of day phase. But the activity of hepatic alcohol dehydrogenase(ADH) in control of night phase showed the similar value with that in those of day phase and xylene treated rats of day phase showed an increasing tendency of hepatic ADH activity as those of night phase showing similar activity. Furthermore, control rats of night phase than those of day phase. And by xylene treatment, enzyme activities of rats of day phase were higher tendency in rats of control but those of night phase were somewhat inhibited. Besides, xylene-treated animals of night phase showed increasing tendency of urinary methylhippuric acid concentration compared with those of day phase. On the other hand, liver weight per body weight(%), hepatic lipid peroxide content and serum xanthine oxidase activity were higher in night phase. And the activities of hepatic oxygen free radical metabolizing enzymes such as xanthine oxidase, gluthathione S-transferase, and xylene-treated rats of night phase than those of day phase. In conclusion, it can be hypothesized on the basis of the results that the accumulation rate of m-xylene intermediate metabolite, i.e. m-methylbenzaldehyde in liver tissus may be higher in night phase than in day phase and it may be responsible for higher liver toxicity in bight phase than in day phase.

Effects of Corni Fructus Extract on the Progression of Diabetes and Renal Function in STZ-induced Diabetic Rats

  • This study investigated the effect of Corni Fructus(Cornus officinalis Sieb. et Zucc.) extract on hyperglycemia and renal function in streptozotocin-induced diabetic rats. Male Sprague-Dawley rats were divided into three groups including normal control(NC), diabetic control(DC), and diabetic treatment with Corni Fructus(DCF). Over a 4-week experimental period, Corni Fructus aqueous extract was administered orally at 500 mg/kg BW/day. The final fasting serum glucose, serum urea nitrogen, triglyceride, urinary total protein level, and relative weight of the left kidney in the DCF group were significantly lower than the DC group. Serum insulin level in the DCF group was higher than the DC group by 23%. The renal xanthine oxidase and superoxide dismutase activities in the DCF group were significantly lower than the DC group. The renal catalase activity in the DCF group was significantly higher than the DC group. In conclusion, these results indicated that Corni Fructus can reduce glucose level and prevent or retard the development of diabetic complication via its antioxidative effect and protecting against diabetic renal damage in streptozotocin-induced diabetic rats.

Effect of Tectorigenin Obtained from Pueraria thunbergiana Flowers on Phase I and -II Enzyme Activities in the Streptozotocin-induced Diabetic Rat

  • Tectorigenin has an apoptosis-inducing ability and immunosuppressive activity. We investigated the effect of tectorigenin on Phase I and II enzyme activities to elucidate the pharmacological action of the immunosuppressive tectorigenin in the diabetic rat. This compound was obtained from the hydrolysis of tetoridin isolated from the flower of Pueraria thumbergiana (Leguminosae). This crude drug (Puerariae Flos) has been used as a therapeutic for diabetes mellitus in traditional Korean medicine. Tecotrigenin inhibited the formation of malondialdehyde (MDA) and hydroxy radicals in serum and liver but promoted superoxide dismutase (SOD) activity. Low MDA contents and low xanthine oxidase and aldehyde oxidase activities were observed in the tectorigenin-treated rats, suggesting that such Phase I enzyme activities are the major source of lipid peroxidation. However, tectorigenin increased Phase II enzyme activities such as SOD, glutathione peroxidase and catalase, suggesting the activation of free radical-scavenging enzymes. The activities of tectorigenin were comparable to those of glibenclamide, which was employed as a positive control. These results suggest that tectorigenin may share some biological properties with glibenclamide in insulin-dependent-diabetes mellitus (IDDM).

Quality Characteristics of Chitosan-ascorbate Treated Kwamaegi Prepared by Vacuum Drying, and Lowering Effect of Serum Lipids in Rats Fed High Fat Diets (Chitosan-ascorbate 처리 감압건조 과메기의 품질특성과 고지방식이 흰쥐의 혈청지질에 미치는 영향)

  • Quality characteristics of Kwamaegi (semi-dried saury) prepared by treatment of chitosan-ascorbate (CA) and vacuum drying at $40{\sim}60^{\circ}C$(VDK), and the effect of the Kwamaegi on serum lipid profiles and anti-oxidation-related enzyme activity in rats fed high fat diets were investigated. The preparation periods were $4.5{\sim}8.3$ hr in VDK, while naturally dried Kwamaegi (NDK) took 360480 hr. Total microbe contents of VDK and NDK were $0.2{\sim}0.5$ and 8.2 log CFU/g, respectively. There was no significant difference in amino-nitrogen content. Compared with NDK, the acid and peroxide value, and fishy flavor of VDK40 (dried at $40^{\circ}C$) were significantly lower, and the texture, color and overall acceptability were higher. In animal experiments, weight gain, content of LDL-cholesterol and lipid peroxide, activities of total (T) and O type (O) xanthine oxidase, and the O/T ratio (%) were significantly lower in the VDK40 diet group than in the NDK diet group. The content of HDL-cholesterol in the VDK40 diet group was higher than in the NDK diet group. These results suggest that preparing CA-treated Kwamaegi with vacuum-drying at $40^{\circ}C$ can be applied throughout the year, and may shorten preparation time and improve its microbiological safety and nutritional values.

Effect of Teminalia chebula Extract on Liver in Rat (가자(Terminalia chebula) 추출물이 흰쥐의 간장 활성에 미치는 영향)

  • In this study, we investigated the effect of Teminalia Chebula (TC) water extract on liver in Rat. Treatment of TC water extract was orally administered 200, 300 mg/kg daily for one week and two weeks. The clinical parameters of serum, values of AST, ALT showed significantly higher than in normal group. Xanthine oxidase and aldehyde oxidase activities were significantly increased comparison with normal group. Microsomal enzymes, aminopyrine N-demethylase and aniline hydroxylase were not affected. Water extract of TC also increased hepatic rnalondialdehyde formation and reduced glutathione content. We also found that water extract of TC decreased activities of glutathione S-transferase and glutathione reductase but was not affected activities of $\gamma$-glutamylcysteine synthetase Thus, it seems that the water extract of TC induced decrease of oxygen free radical scavenger, glutathione content by inhibition of glutathione reductase which may reform oxidized glutathione to reduced glutathione.

  • It was attempted to observe the effect of garlic on the hepatic purine metabolizing enzymes in this study. The activities of adenosine deaminase, guanase, and uricase in rats were not changed significantly following the feeding of 5% garlic juice. Whereas, garlic juice inhibited significantly the hepatic xanthine oxidase activity compared to control group with the lapse of treated-period. The orate level of serum in rats was significantly decreased by the treatment of garlic juice. The above inhibitory effect of garlic was greater in boiled garlic juice than fresh garlic juice-treated group. These results indicated that, according to the chemical properties of allicin which is unstable in heat, other components than allicin in garlic may regulate the hepatic purine metabolizing enzymes.

Effect of Cyclohexane and Xylene Mixture Treatment on the Liver Damage in Rats

  • To investigate the cyclohexane and xylene mixture treatment on the liver damage, the rats were treated by the mixture of cyclohexane and xylene (CH+X) and then, liver damage was demonstrated by liver function findings based on liver weight/body weight, serum level of alanine aminotransferase (ALT), xanthine oxidase (XO) and then compared with cyclohexane treated group (CH group) and xylene-treated group (X). The CH+X group showed merely severer liver damge than CH or X group. On the other hand, CH+X group showed lower activity of hepatic cytochrome P-450 dependent aniline hydroxylase (CYPdAH) than CH or X group, but no statical differences were demonstrated among three experimental groups. Especially the hepatic GSH content was merely declined than CH or X group and the activity of hepatic GST was higher in CH+X group than CH or X group. In conclusion, cyclohexane and xylene mixture treated animals showed merely severer liver damage than cyclohexane or xylene treated group and such a fact may be caused by inhibition of cyclohexane or xylene metabolism and oxygen free radical.

Effect of Cyclohexane Treatment on Serum Level of Glutathione S-Transferase Activity in Liver Damaged Rats ($CCl_4$ 에 의한 간손상 모델 실험동물에 있어서 cyclohexane 투여가 혈청 glutathione S-transferase 활성에 미치는 영향)

  • To evaluate the effect of cyclohexane(CH) treatment on the serum levels of glutathion S-transferase(GST) activity in liver damaged animals, damaged liver was induced with pretreatment of 50% $CCl_4$ dissolved in olive oil (0.1 m1/100g body weight) intraperitoneally 17 times every other day. To $CCl_4$-treated rats, CH (1.56 g/kg body weight, i.p) was injected once and then the animals were sacrificed at 4 hours after injection of CH. The $CCl_4$-treated animals were identified as severe liver damage on the basis of liver functional findings, 1,e, increased serum levels of alanine aminotransferase(ALT), alkaline phosphate(ALP) and xanthine oxidase(XO) activities. On the other hand, $CCl_4$-treated animals injected with CH once($CCl_4$-pretreated animals) showed more decreased serum levels of ALT and XO, and more increased those of ALP rather than $CCl_4$-treated animals. In case of comparing the GST with ALT activity in liver, both $CCl_4$-treated and pretreated animals showed similar changing pattern of enzyme actvity. Especially $CCl_4$-pretreated animals showed significantly increased serum level of GST actvity compared with the $CCl_4$-treated those, whereas those of ALT showed reversed tendency. In aspects of GST enzyme kinetics, $CCl_4$-pretreated animals showed higher Vmax of liver GST enzyme than $CCl_4$-treated animals. In conclusion, injection of CH to the liver damaged rats led to enhanced liver damage and more increased activity of serum GST which may be chiefly caused by the enzyme induction.
