발효홍차 음료가 흰쥐가 혈청과 간의 지질대사에 미치는 영향 (Effect of Tea Fungus/Kombucha Beverage on Serum and Liver Lipids Metabolism in Rats)

  • We investigated the effect of tea fungus/kombucha(TF) beverage on serum and liver lipid metabolism in growing male rats. The rats(Sprague-Dawley) were divided into five groups; control(TF free water), 20 or 40% TF groups(20 or 40% TF in water) and 20 or 40% TFS groups(20 or 40% TF autoclaved in water) according to the level of TF supplementation. The rats were fed ad libitum each of the experimental beverage for 7 weeks. The body weight gain of all the TF and TFS groups were similar to that of the control group. The total lipid and triglyceride concentrations of serum in the 40% TF group were increased more than those in the control group, but those in the 20% TF and 20% TFS groups were similar to that of the control group. The total cholesterol, HDL-cholesterol concentrations, HDL-cholesterol / total cholesterol ratio and atherogenic index of serum in all the TF and TFS groups were similar to those of the control group. The LDL-cholesterol concentration of serum was significantly lower in the 20% TF group than that in the control group. The LDL-cholesterol concentration of serum was significantly lower in the 20% TF group than that in the control group, but those of the 40% TF, 20% TFS and 40% TFS groups were similar to that of the control group. The liver weights of all the TF and TFS groups were heavier than that of the control group. The concentrations of hepatic triglyceride in the 20% TF and 20% TFS groups were significantly decreased, compared with that in the control group. The concentrations of hepatic total cholesterol in all the TF and TFS groups were significantly more decreased than that in the control group. These results indicated that the liver weights of tea fungus/kombucha beverage feeding rats were increased, also the serum triglyceride and total cholesterol concentrations of 40% TF feeding rats were increased. but the serum LDL-cholesterol, hepatic triglyceride and cholesterol levels of 20% TF feeding rats were decreased.

The Effects of a High-fat or High-sucrose Diet on Serum Lipid Profiles, Hepatic Acyl-CoA Synthetase, Carnitine Palmitoyltransferase-I, and the Acetyl-CoA Carboxylase mRNA Levels in Rats

  • The purpose of this study was to investigate the effects of altering relative intakes of fat and carbohydrates on serum lipid profiles, hepatic acyl-CoA synthetase (ACS), carnitine palmitoyltransferase-I (CPT-I), and the acetyl-CoA carboxlyase (ACC) mRNA level in Sprague-Dawley rats. For four weeks the rats were fed either an AIN-76 diet or one of its modified diets that were supplemented with 20% beef tallow (high-fat diet, HF) and 66.3% sucrose (highsucrose diet, HS). The HS group had significantly higher serum triglyceride and total cholesterol concentrations when compared with the other groups. Serum LDL-cholesterol concentrations in the HS and HF groups were significantly higher when compared to the normal diet (ND) group. Serum HDL-cholesterol levels of the ND and HS groups were significantly higher than those of the HF group. The hepatic total lipid level of the HF group was significantly higher than those of other groups; triglyceride levels of the HS and HF groups were significantly higher than those of the ND group. Hepatic ACS mRNA levels of the HF group were significantly higher than those of the ND group. Hepatic CPT-I mRNA levels were higher in the HF group than other groups. Also, ACC mRNA levels in the liver increased in the HF group. In conclusion, changes in the composition of dietary fat and carbohydrates could affect the hepatic ACS, CPT-I, and ACC mRNA levels. These results facilitate our understanding of the coordinated regulation of the ACS, CPT-I, and ACC mRNA levels and will serve to enhance our understanding of the molecular mechanisms that underlie the regulation of fatty acid metabolism.

Hypolipidemic and Hepatoprotecitive Effects of Picrorrhiza Rhizoma in High Fat Diet Supplied Mice. A Pevention Sudy.

  • The preventive hypolipidemic effect of the aqueous extracts of Picrorrhiza Rhizoma (PR) was observed in a high fat diet (HFD) feeding hyperlipidemic mouse with their hepatoprotective effects. PR extracts (50, 100 and 200mg/kg) were orally dosed once a day for 12 weeks initiated with HFD supply, and changes on body weight and gains, liver weight, serum aspartate transferase (AST) and alanine transferase (ALT) levels were monitored with serum low density lipoprotein (LDL), high density lipoprotein (HDL), triglyceride and total cholesterol levels. The efficacy of test articles was compared to that of 10mg/kg of simvastatin (SIMVA). Dramatic decrease of both absolute and relative liver weight was dose-dependently observed in all PR extract dosing groups as compared with HFD control group. The serum AST and ALT levels were dose-dependently decreased in PR extract dosing groups. The serum LDL, triglyceride and total cholesterol levels were dose-dependently decreased in PR extract dosing groups compared to that of HFD control group. The serum HDL levels were slightly but dose-dependently increased in PR extract dosing groups as compared with control group. The efficacy on the serum lipid levels of PR extracts was slighter than that of SIMVA. Based on these results, it is concluded that water extract of PR has a relatively good favorable preventive effects on the HFD inducing hyperlipidemia and hepatopathy.

  • This study was undertaken to investigate effects of alcohol and dietary protein levels on serum lipids and enzyme activities in 15 week-old male rats given a normal diet. Rats were divided into 8 groups : control group (16% protein 16PC) and 8%(8PE) 16%(16PE) and 24% protein groups(24PE) to which was given 5% ethanol mixed into their drinking water after 4 weeks and 10 weeks. Body weight organ weight and various blood components were determined at 4 and 10 weeks. Body weight gain organ weight hemoglobin concentration and hematocrit value were not influenced by ethanol and dietary protein levels. The levels of total cholesterol HDL-cholesterol and phospholipid in serum were not affected by ethanol consumption. Serum triglyceride concen-trations after 10 weeks were significnatly increased ethanol-treated group compared with that of control group and the effect was greater in low protein group than control group. Serum ALP activity was significantly higher in 8PE group than other group but there was no influence by ethanol consumption.

어유의 Eicosapentaenoic Acid의 식이섭취가 성숙쥐의 혈청 및 간의 지질조성에 미치는 영향 (Effect of Dietary Eicosapentaenoic Acid on Serum and Liver Lipids Patterns of Male Rat)

  • The dietry effects of marine n-3, plant n-3 and plant n-6 fatty acid on serum lipids levels, liver phospholipid fatty acid composition in rat were investigated. Four groups of male Sprague-Dawley rats, 30 weeks old, were fed on one of 4 different experimental diets for 4 weeks. The diets were composed of 15% fat(w/w) of either concentrated EPA oil(20:5, n-3 : 65%), fish oil(20:5, n-3 : 19%, 22:6, n-3 : 18%), perilla oil(18:3, n-3 : 60%) or corn oil(18:2, n-6 : 49%). Blood was initially taken before experimental feeding and also taken after 2 weeks and 4 weeks feeding the diet respectively and then examined for the levels of serum lipids. Rats were sacrificed at 4 weeks after the diet for the analysis of liver phospholipid fatty acid. EPA feeding remarkably decreased the serum levels of triglyceride, total cholesterol, HDL-cholesterol and total phospholipid than any other oil feeding. Fish oil feeding decreased serum HDL-cholesterol level comparable to the effect of EPA feeding and decreased total cholesterol and phospholipid less than but close to the effect of EPA feeding. Perilla oil feeding did not change serum levels of triglyceride and phospholipid, but it decreased serum total cholesterol a lot and HDL-cholesterol a little. Corn oil feeding did not affect triglyceride and total cholesterol while it increased serum level of HDL-cholesterol and total phospholipid. Serum HDL-cholesterol level was increased only in corn oil group. But contrary to the result of serum total phospholipid, liver phospholipid level found to be higher in fish oil and EPA groups than in perilla oil and corn groups. The fatty acid composition of liver phospholipid, phosphatidylcholine(PC) and phosphatidyl ethanolamine(PE) turned out to be affected by dietary fatty acid. 18:2 of liver PC was the lowest in FO group following CO group. The ratio of 20:4/18:2 was lower in PO group than in EPA group in consequence of higher 18:2 and lower 20:4 in PO group and vise versa in EPA group. In the liver PC and PE, similar trends in the ratios of n-6/n-3 and 20:4/18 were found showing higher ratios with CO and EPA group over FO and PO group. EPA group showed the lowest level of 20:5 and lower level of 20:6 than group. Fish oil was more efficient than EPA oil and PO in lowering the ratio of n-6/n-3 in consequence of the highest 22:6, and the lowest 18:2 in liver phospholipid. But PO lowers the ratio or 20:4/18 more than FO. In conclusion, EPA oil was more effective in lowering serum lipids than FO and PO. Reviewing the dietary effect of fatty acid on eicosanoids composition in rats, it is considered that more possibility was with FO than PO in the effectiveness of atherosclerosis prevention and more with PO than with EPA oil. It was also found that FO showed more effective than EPA oil for atherosclerosis prevention. It was hardly found that CO had any effect on lowering serum lipids and on eicosanoids composition in liver phospholipid for the prevention of atherosclerosis.

  • This study was conducted to examine the effects of mulberry fruit tea (MFT) on the levels of serum lipid profiles and serum cardiovascular disease markers in sixty middle-aged people(30 males and 30 females) who consumed MFT for 4 weeks. Anthropometric measurements, serum lipid profile levels, and serum cardiovascular disease markers were analyzed before and after consumption of MFT. After consumption of MFT, no significant differences were observed in anthropometric measurements, levels of serum aspartate transaminase (AST) and alanine transaminase (ALT) in males and females. Among serum lipid profiles, triglyceride and LDL-cholesterol were decreased significantly, whereas HDL-cholesterol was significantly increased.

흰쥐의 담즙 상실이 혈청 지질과 Gastrin 대사에 미치는 영향 (Effect of Bile Deprivation on Serum Lipids and Gastrin in the Rat)

  • The effect of bile deprivation on serum lipid and gastrin contents was investigat\ulcornered after choledocho-urinary cystostomy in Sprague-Dawley rats. Bile deprivated rats were compared with sham operated control group. Gastrin levels in serum and antral tissue were measured and serum lipid concentrations were also measured. Gastrin levels of serum and tissue after bile deprivation were increased significantly compared with those of the controL At the end of 1st and 2nd week after bile deprivation, serum cholesterol and triglyceride contents were significantly lower han those of the control. By 4th week, there was no significant difference between two groups. Increases in serum and antral gastrin levels temporarily coincided well with decreases in serum lipid contents after bile deprivation. These results suggest that there is increase in biosynthesis and release of gastrin and decrease in fat absorption at early stage of bile deprivation.

식이지방이 흰쥐의 혈청 지질상태 및 조직 지방산분포에 미치는 영향 (Effect of Dietary Fat and Oils on Serum Lipid Status and Fatty Acid Composition in Tissues of Rat)

  • In order to establish tissue lipid status in animal on feeding of various dietary fat and oils, each group of rats was fed a semisynthetic diet containing 10%(w/w) mackerel oil (MO), eel oil (EO), soybean oil (SO), rapeseed oil (RO) or beef tallow (BT) for 1, 2 and 4 weeks, After each feeding period, levels of cholesterol, triglyceride and phospholipid were measured in serum. Fatty acid ${\leftarrow}$ composition was also investigated in serum and tissue lipids. Levels of total serum cholesterol were lower but HDL-cholesterol were higher in fish oil groups, which resulted in significantly higher ratio of HDL to total cholesterol in the fish oil groups. Fish oil groups, in general, also had lower levels of serum triglyceride and phospholipid than other groups, but S0 group maintained as low phospholipid levrl as fish oil groups. Fatty acid composition of dietary fat was reflected in all the tissues investigated but with varying degrees. Very long chain fatty acids, specific components exclusively found in fish oils were most well reflected in liver and relatively well in serum, whereas linoleic acid and erucic acid of SO and RO in the diet were better shown up in heart and adipose tissue. It attracted a particular attention that major proportions of long chain monoenoic acids $(C_{22:1})$ occurring both in MO and RO were detected separately in liver and heart plus adipose tissue, the result of which strongly indicates that there is a significant difference in metabolism between isomers $(C_{22:1}\;w\;11\;and\;C_{22:1}\;w\;9,\;respectively)$. It is suggested from this study that differences in lipid status as well as in the levels of serum lipids result from uniqueness in metabolism of each different fatty acid and give rise to distinguishable change in serum lipoprotein pattern, followed by diet with different fat sources.

서목태(Rhynchosia nulubilis) 추출물이 고지방 및 고콜레스테롤 식이 급여 흰쥐의 혈청 지질 농도에 미치는 영향 (Effects of Methanol Extracts from Rhynchosia nulubilis on serum Lipid Concentrations in Rats fed High Fat and High Cholesterol Diet)

  • The effects of the Rhynchosia nulubilis extracts on serum lipid concentrations were evaluated in rats. Forty-two male Sprague-Dawley rats weighing $100{\pm}10g$ were divided into six groups and fed high fat and high cholesterol diets for four weeks. Experimental groups were administered with following diets; basal fat diet(control) and basal and high fat diets with 1% cholesterol and Rhynchosia nulubilis extract(40mg/kg, 400mg/kg and 4000mg/kg). The concentrations of serum triglyceride in rats fed the Rhynchosis mulubilis extract, high fat, 1% cholesterol, and RN-HFC(-40, -400, -4000mg/kg) groups were lower than those in other groups. The concentrations of total cholesterol in the RN-HFC (-40, -400, -4000mg/kg) groups were lower than those in fat diet groups. The concentrations of HDL-cholesterol in serum of the RN-HFC(-40, -400, 4000mg/kg) groups were significantly higer than those of other groups. The levels of LDL-cholesterol in serum of the RN-HFC(-40, -400, -4000mg/kg) groups were tended to be lower than those of other groups. These results suggest that Rhynchosia nulubilis extract may reduce elevated levels of serum lipid concentrations in rats fed high fat diets.

스테비아(Stevia. Rebaudian Bertoni)추출물 보강이 만성 알코올 섭취 흰쥐의 지질대사 및 간기능에 미치는 영향 (The Effect of Stevia (Stevia. Rebaudian Bertoni) Extract Supplementation on Lipid Metabolism and Liver Function of Rats Administered with Ethanol)

  • To investigate the effects of the supplementation of Stevia. Rebaudian Bertoni on serum and hepatic lipid levels and enzyme activities in rat administered with ethanol chronically. Sprague-Dawley male rats were orally treated with either an AIN-76 diet(C, control), a control diet plus ethanol(E, 4g/kg bw), E plus stevia extract-1(ES1, 1ml/kg bw), or E plus stevia extract-2(ES2, 2ml/kg bw) for 7 weeks. Serum triglyceride levels were increased in the E group and were decreased in the ES 1 group. Liver triglyceride levels were decreased significantly in the ES2 groups and Total-cholesterol were decreased in the ES1, ES2 groups compared with the E group. Liver $\gamma-GTP$ levels were decreased significantly in the ES1, ES2 groups compared with the E group. In addition, we have evaluated the serum or liver carnitine levels in those groups. Liver TCNE levels were increased significantly in the ES1, ES2 compared with the E group. These results may suggest that supplementation of Stevia. Rebaudian Bertoni has effects on the recovery of chronic ethanol-related diseases.
