Evaluation of serum immunoglobulin G4 concentrations in canine pancreatitis

  • The goal of this study was to measure immunoglobulin G4 (IgG4) concentrations and to evaluate the significance of these values in the management of canine pancreatitis. The medical records of 24 dogs that visited the Veterinary Medical Teaching Hospital between December 2016 and June 2018 were retrospectively reviewed to identify dogs that had been diagnosed with pancreatitis. The serum C-reactive protein and serum IgG4 concentration in the affected dogs were highly increased compared to the healthy group. Particularly, serum IgG4 measured significantly higher in dogs with pancreatitis and concurrent immune-mediated disease (p < 0.05). In conclusion, increased serum IgG4 concentrations are a characteristic finding in dogs with pancreatitis. The results of this research indicate that an elevation in IgG4 has the potential of being used as a tool for the diagnosis of pancreatitis and concurrent immune-mediated disease.

The Effects of the Low Power Wavelengths of Helium-Neon Infra- Red Laser on the Changes of the Serum Immunoglobulin Concentration in Burn Rats (저 에너지 레이저 파장이 화상쥐 혈청면역물질 농도 변화에 미치는 영향)

  • This studies were to investigate the effects of low power wavelengths Helium-Neon Infra-Red(He-Ne 1R) laser on the changes of the serum immunoblobulin(Ig) concentrations in bum rats, The thirty Spraque-Dawley adult male rats were assigned to the 5 groups: the experimental groups(3). the bum control group(1) and the control group(1). There was made three degree burn by the 290 mW IR on the back oreach rats, from 3 days after being burned. the experimental laser groups were irradiated low power wavelengths(292Hz- 1168Hz, 4672Hz) He-Ne IR laser far 5 minutes every day during the 7days. The results were as follows; 1. The concentrations of immunoglobulin G(Ig G) in serum of all experimental groups on the treated with the low power wavelengths of the He-Ne IR laser during the 5 minutes far 7 days were significantly lower than those of bum groups, but those 1168 Hz group were significantly higher than those of the 292 Hz and 4672 Hz groups. 2. There were significantly decreased the change of the level of immunoglobulin M(Ig M) in serum of all experimental groups on the treated with 5 minutes laser fer 7 days to the burn control group. and were significantly increased on treated with more higher wavelengths groups to those of lower groups. 3. The concentrations immunoglobulin E(Ig E) in serum of the 292Hz wavelengths groups were significantly lower than those in bum control group. As above results. the changes of the immunoglobulin in serum levels on the healing process have meaningful effected with the low power wavelengths role on the treated with He-Ne IR laser.

  • In fish vaccinology, the secondary antibodies against fish immunoglobulins (Igs) are necessary to measure specific humoral immune responses in immunized fish. In the present study, polyclonal antiserum against olive flounder (Paralichthys olivaceus) IgM heavy chain was generated by intramuscular immunization of rabbit with Escherichia coli/eukaryotic shuttle vector containing open reading frame (ORF) of olive flounder IgM heavy chain. Western blot analysis demonstrated the specific activity of the rabbit antiserum with reduced olive flounder serum H chain at dilutions up to 1:1000. Titer of immunized rabbit serum against olive flounder serum was significantly higher than that of pre-immunized rabbit serum when determined by ELISA.

  • lgM-like immunoglobulin was purified from the immune serum of M albus which immunized with bovine Serum albumin(BSA) as an antigen(Ag) and characterized. The Ag-specific antibody activity of the immune serum was increased after the immunization. The purified lgM-like immunoglobulin had a tetrameric structure which had a molecular weight of 800 kD and the monomer of IgM-like Ig had a mass of 199 kD which was composed of two heavy chains (Mol. wt. 70 kD) and iwo light chains (Mol. wt. 29.5 kD). The IgM-like Ig showed hemaggluti nating activity to mammalian RBC slightly.

  • Dietary conjugated linoleic acid(CLA) has been shown to affect immune function. Thus, the objective of this study was to investigate the effects of CLA on the mice that treated prednisone. Mice were randomized into 6 groups and fed diet containing either 0(control, P), 0.5%(CLA1, CP1) or 1.5%(CLA2, CP2) CLA for Sweets. Before 1 week of finishing diet supplement CP1, CP2, and P group treated the prednisone by subcutaneous injection. The levels of serum immunoglobulin A, G, E, gut lumen s-IgA, MLN immunoglobulin A, body weight, mucosal protein was compared. The level of serum IgA in CLA1, CLA2, CP1, and CP2 group increased, while which of P group was decreased. The level of serum IgG in CLA 1 group increased, while which of the other group no differences. Serum IgE level showed no difference and the immunoglobulin production in MLN lymphocyte in CLA 1 group increased. The level of gut lumen s-IgA in P group showed decreased, while which of the other group showed no differences. These results support the view that CLA supplement partially enhance the cell-mediated immunity and overcome the immunosuppressive effect of prednisone.

Decreased Serum Immunoglobulin in Recurrent Otitis Media with Effusion (재발성 삼출성 중이염과 혈청 면역글로불린 농도의 저하)

  • Background: Defective or immature antibody responses to pathogens in children may explain the increased susceptibility to acute otitis media. However, there is no study in Korea patients whether a correlation exists between otitis media with effusion and the levels of serum immunoglobulins, IgG subclasses, IgA, IgM and IgE. Methods: 45 children with otitis media with effusion more than 4 episodes in 12 months or 3 episodes in 6 months, 62 children with otitis media with effusion less than 3 episodes in 12 months and 102 children for control group took part in the study at the Department of Otorhinolaryngology of the KyungHee University from May 2004 to Feburary 2007. Serum immunoglobulin levels were determined by nephelometry. And then the relationship between otitis media with effusion and serum immunoglobulin level was evaluated. Results: In otitis media prone group, serum IgG1, IgG2, IgG4, and IgA level was lower than those level of control group, it was significantly decreased (p<0.05). In otitis media group, serum IgA, IgE, and IgG4 level was lower than those level of control. But it was not statistically significant (p>0.05). Conclusion: Lower immunoglobulins in children with otitis media with effusion suggest a generalized decreased antibody responses. Lower levels of serum IgG1, IgG2, IgG4, and IgA may be related with chronicity or intractability of otitis media with effusion.

  • This studies were to investigate the effects of low power intensity Helium-Neon Infra-Red(He-Ne IR) laser on the changes of the serum immunoglobulin(1g) components in burn rats, The fourty eight Spraque-Dawley adult male rats were assigned to the 8 groups: the experimental groups(4), the burn control groups(2) and the control groups(2) There was made three degree burn by the 250mW IR on the back of each rats, from 3 days after being burned, the experimental laser groups were irradiated low power He-Ne IR laser for 3 minutes and 4.5 minutes every day during the 7days and 21 days. The results were as follows: The concentrations of immunoglobulin G(Ig G) in serum of burn rats on the treated with during the 3minutes laser for 7 days were higher than those of burn and 4.5 minutes laser groups. There were significantly decreased the change of the level of immunoglobulin M(Ig M) in serum of burn rats on the treated with 3 minutes laser for 7 days to the control group, but were significantly increased on the 3minutes and 4.5 minutes laser groups for 21 days to the those of burn and control groups, and the levels of the Ig M on the 3minutes laser group for 21 days were higher than those of 7 days. The concentrations of immunoglobulin E(Ig E) in serun of the burn rats on the experimental and burn groups were significantly lower than those in control group. As above results, The changes of the level of immonoglobulin components in serum have meaningful role in the healing process on the treated with the low pewer intensity of HE-Ne IR laser.

Avidity of serum immunoglobulin G antibodies to Porphyromonas gingivalis in the elderly with chronic periodontitis

  • The present study aimed at evaluating serum immunoglobulin G (IgG) avidity to Porphyromonas gingivalis in elderly patients with mild and severe chronic periodontitis. The avidity of antibodies against P. gingivalis present in the sera of 18 patients with mild chronic periodontitis and 18 patients with severe chronic periodontitis was evaluated using an ammonium thiocyanate-dissociated enzyme-linked immunosorbent assay (ELISA). The results showed that the mean absorbance value in serum IgG antibody titers was significantly higher in the severe chronic periodontitis group than in the mild chronic periodontitis group ($198{\pm}35ELISA$ unit [EU] vs. $142{\pm}32EU$, p < 0.01). However, there was no significant difference between the two groups in antibody avidity ($65{\pm}57EU$ vs. $54{\pm}27EU$). These findings suggest that humoral immune responses to P. gingivalis between mild and severe chronic periodontitis in elderly patients are characterized by the differences in the quantity rather than the quality of the antibodies.

A Biotin-avidin Labeled Enzyme Immunoassay for the Quantitation of Serum TSH Using Protein-layered Solid Phase

  • A sensitive enzyme immunoassay for serum TSH has been developed utilizing the tight binding between biotin and avidin, and three layered protein polystyrene beads as solid phase. To increase binding capacity of TSH and sensitivity of the assay, the polystyrene beads were coated sequentially with mouse immunoglobulin as first layer, rabbit antimouse immunoglobulin as second layer and monoclonal anti-TSH as third layer. A serum sample was incubated simultaneously with a monoclonal anti-TSH immobilized polystyrene beads and a second monoclonal anti-TSH covalently attached to biotin. After washing, the antibody bound serum TSH-anti-TSH-biotin complex is reacted with horseradish peroxidase (HRP)-labeled avidin. Following second wash, the bound HRP activity was measured calorimetrically. Reproducible results were obtained within 4 hours for serum TSH in the range between $0{\mu}\textrm{IU}$ml and ${50}{\mu}\textrm{IU}$ml with detection limit of $0.1{\mu}\textrm{IU}$ per test.

Effects of Aerobic Exercise and a Protein Diet on Serum Lipid Profiles, Liver Function, and Immunoglobulin in Rats (유산소 운동과 단백식이가 흰쥐의 혈중지질, 간기능 및 면역글로불린에 미치는 영향)

  • The purpose of this study was to analyze the effects of aerobic exercise and a protein diet on serum lipid profiles, liver function, and immunoglobulin levels in rats. The subjects, 24 male Sprague-Dawley rats, were grouped into an exercise with protein diet group (A, n=6), a protein diet group (B, n=6), an exercise group (C, n=6), and a control group (D, n=6). The exercise with protein group trained for 4 weeks (20-30 min/day, speed at 14~17 m/min) and was given a 40% protein diet for 4 weeks. The variables of serum lipid profiles, liver function, and immunoglobulin levels were measured in all of the subjects at the end of the 4 weeks of treadmill exercise. Serum TC and LDL-C levels were significantly decreased in the exercise with protein diet group compared to the other groups and serum HDL-C, GOP, and IgG were significantly higher than in the other groups. However, no significant differences were found for the IgA and IgM levels. In conclusion, aerobic exercise with a protein diet was effective in improving serum lipid profiles, liver function, and immunoglobulin.
