Isolation and sequence analysis of a small cryptic plasmid from Lactobacillus farciminis KCTC3681 (Lactobacillus farciminis로부터 미지의 작은 플라스미드의 분리와 염기서열 분석)

  • From the extensive screening for small cryptic plasmid among about 23 lactic acid bacteria (LAB), 2.4 kb of cryptic plasmid was isolated from Lactobacillus farciminis strain KCTC 3681 and named as pLF24. The plasmid pLF24 was a circular molecule of 2,396 base-pairs in length with a G+C content of 38%. Two protein-coding sequences could be predicted. ORF1 and ORF2 showed homologies to plasmids of gram-positive bacteria. The replication protein coded by ORF2 and the plus origin, were similar to replication regions of other gram-positive bacteria as shown in plasmids such as pLH2, pLS141-1 and pLC2. The nucleotide sequence of pLF24 was deposited into Genbank data base with an accession number of EU429343. The newly isolated plasmid can be used for construction of shuttle vector in Lactobacillus bacteria.

  • This paper analyzes matched-filter acquistion performance of a direct-sequence spread-specturm multiple-access system in a Rayleigh fading channel. For an application of multiple access system, multiple access interference is considered in the performance analysis. A signal flow graph technique is used to derive mean acquisition time in terms of detection and false alarm probabilities. As aresult of performance analysis, it is shown that mean acquisition time increases as fading rate becomes faster andthe numbre of users increases, and it decreases as a matched-filter length becomes logner. The variation of parameter values hs much influence on acquisition performance as SNR/chip becomes smaller.

  • In this paper, we analyze the compression of optical soliton which is obtained by proceeding the optical pulse in FSDD(Fiber with Slowly Decreasing Dispersion) using both NSE(Nonlinear Schrodinger Equation) and GNSE(General Nonlinear Schrodinger Equation) and compare the results. We replace the FSDD with a sequence of fibers having different dispersion values and pompre the results with those obtained in FSDD. It is found that the same results in peak value and FWHM(Full width Half Maximum) can be obtained by replacing FSDD with a sequence of fibers having proper length. We vary the shape of initial pulse which is the input of FSDD and suggest the condition to obtain higher compression rate.

  • In this study, we examine the effect of spreading sequence matched filtering on the power parameters of ultrawideband (UWB) multiuser interference (MUI) under different multiple access (MA) scenarios. More specifically, we investigate the manner in which the length of the sequence MF affects the average power, peak power, and the peak-to-average ratio (PAR) of the matched filtered version of an MUI signal. The results of the analysis performed for a simplified scenario are supported by the simulation results obtained for a realistic multipath environment.

  • In this paper, we discuss recovery schemes for errors occurring when image data encoded with variable length coding (VLC) is transmitted through additive white Gaussian noise (AWGN) and multiple-access interference in direct sequence code division multiple access (DS/CDMA) systems. VLC such as JPEG is so sensitive to channel errors that severe degradation in decoded images occurs even if only one or two bits have errors. This is due to the loss of synchronization at the image decoder. We propose a resynchronization scheme using a power allocation method in wireless DS/CDMA transmission. Through simulation, we know that the proposed method has a more robust resynchronization capability and higher objective and subjective quality than the conventional method.

Molecular Phylogeny of Silk Producing Insects Based on Internal Transcribed Spacer DNA1

  • Silk moths are the best studied silk secreting insects and belong to the families Bombycidae and Saturniidae. The phylogenetic relationship between eleven silk producing insects was analyzed using the complete DNA sequence of the internal transcribed spacer DNA 1 locus. The PCR amplification and sequence analysis showed variation in length ranging from 138 bp (Antheraea polyphemus) to 911 bp (Hyalopora cecropia). Microsatellite sequences were found and was be used to distinguish Saturniidae and Bombycidae members. The nucleotide sequences were aligned manually and used for construction of phylogenetic trees based on Maximum parsimony and Maximum likelihood methods. The topology in both the approaches yielded a similar tree that supports the ancestral position of the Antheraea assama.

Identification of Four Cyst Nematodes using PCR-RFLP in Korea (PCR-RFLP를 이용한 국내 분포 씨스트선충 4종의 동정)

  • To identify four cyst nematodes (Heterodera schachtii, H. trifolii, H. glycines, H. sojae) that are economically important plant-parasitic nematodes in Korea, restriction fragment length polymorphism (RFLP) by 8 endonucleases (PstI, VspI, AlwI, RsaI, MvaI, EcoRI, Eco72I, Hinf I) was performed based on sequence difference of mitochondrial DNA cytochrome c oxidase subunit I (COI) gene. As a result, species-specific DNA band patterns by RsaI endonuclease were observed in H. schachtii. The specific patterns was in H. trifolii by 3 endonucleases (VspI, AlwI, Hinf I), and was in H. glycines by Hinf I. While, H. sojae was not digested by 4 endonuclease (VspI, AlwI, RsaI, Hinf I). This study showed that four cyst nematodes could be distinguished using RFLP by 4 endonucleases (RsaI, VspI, AlwI, Hinf I) based on the sequence difference of COI gene.

Efficient Processing of Subsequence Searching in Sequence Databases (시퀀스 데이터베이스를 위한 서브시퀀스 탐색의 효율적인 처리)

  • This paper deals with the subsequence searching problem under time-warping. Our work is motivated by the observation that subsequence searches slow down quadratically as the average length of data sequences increases. To resolve this problem, the Segment-Based Approach for Subsequence Searches (SBASS) is proposed. The SBASS divides data and query sequences into a series of segments, and retrieves all data subsequences. Our segmentation scheme allows segments to have different lengths; thus we employ the time warping distance as a similarity measure for each segment pair. For efficient retrieval of similar subsequences, we extract feature vectors from all data segments exploiting their monotonically changing properties, and build a spatial index using feature vectors. The effectiveness of our approach is verified through extensive experiments.

Identification, Growth and Pathogenicity of Colletotrichum boninense Causing Leaf Anthracnose on Japanese Spindle Tree

  • Leaf anthracnose was observed on leaves of Japanese spindle tree in Seoul, Korea from autumn 2003 to spring 2004. The causal fungus was purely isolated from he leaf spot lesions and cultured on PDA. The colony on PDA was cream to orange but blackish in the center n old cultures. Conidia were formed in blackish orange asses and were cylindrical in shape, measured 13-17${\times}$5-7 ${\mu}$m in size. Blackish brown setae were often observed on PDA and ranged up to 100 ${\mu}$m in length. Based on morphological and ITS region sequence analyses, the fungal strain was identified as Colletotrichum boninense. Koch’s postulates were fulfilled by inoculating tree leaves with 1 ${\times}$ $106^6$ conidia per ml in a moist chamber. This is the first study on the pathogenicity, growth and phylogenetic characteristics of C. boninense causing leaf anthracnose on Japanese spindle tree in Korea.

Study of Electrophoretic Karyotypes of Fusarium Section Liseola (Fusarium 균의 section Liseola에 대한 핵형 연구)

  • CHEF-PFGE(Contour-Clarnped Homogeneous Electric field- Pulsed Field Gel Electrophoresis) was used to identify electrophoretic karyotype for eight strains belonging to the Fzisoriuni section Liseolo. Chromosome numbers were nine to thirteen bands, ranging in size Cram 0.75 to 6.45 Mb. The total genome size was eslimated to range from 38.19 Mb to 43.12 Mb and numerous chromosome-length polymorphisms (CLPs) were observed. For the chromosome localizalion of the gene, 1GS sequence(2.6 Kb) of rDNA from F: moniliforme, chs-2 gene(2.8 Kb) and 4 - 3 gene(3.8 Kb) from Neuuospora cmssa were wed as probes.
