An Analysis of the Job Sequences of the Working Poor (근로빈곤층의 직업력 분석 -비빈곤층과의 비교를 중심으로-)

  • After the economic crisis of Korea, the character of Korean Poverty has changed. Most notably, many people are working but poor. Therefore it is important to understand the characteristics of the working poor, especially the unstable work experiences of the working poor since one of the causes of poverty is that. Prior research about the working poor has not fully proven this issue. This study is to examine the job sequences of the working poor. Thus I utilized the KLIPS(Korea Labor and Income Study), and analyzed it by event sequence analysis and optimal matching methods. The job sequences are divided as follows: total years of working in the labor market, the number of gaps and the length of gaps in their careers, and the characteristics of experienced jobs since they have entered the labor market from age 15. As a result, there are no statistically significant in the total years of working in the labor market. And the number of gaps and the length of gaps in their careers, and the characteristics of experienced jobs show that working poor have been experiencing more unstable than non-poor. Thus, almost all of the male working poor has unstable jobs their whole lives, and the female working poor's job sequences show distinct features according to women's life course. These results can give political implications to the anti-poverty policy in Korea.

Improvement of DS System using Transversal Filter for Advanced Duty Rate on Meteor Burst Channel (유성 버스트 채널 도통율 향상을 위해 Transversal Filter를 적용한 DS시스템의 개선)

  • In spite of many advantages of Meteor Burst Communications(MBC) on its transmission channel, the fact that its duty rate is less than 10 percents is a considerable deficiency of MBC. To overcome with this deficiency without paying large cost, we use a direct sequence(DS) simple reception system. This method doesn't need to add the components for acquisition or tracking the signal so that it keeps the cost efficiency of the MBC. But it has a disadvantage that its duty rate shows a large decline by noise. For this problem we adopt the proposed system applying a transversal filter circuit. The improvement by this method is more significant when the received signal is weak Simulation results show as t1 is 100ms, then the burst length would be increased by 35% when the initial value of the received signal power increases by 3 dB, t2 is 135ms. If the power increases by 6dB the burst length would be 170ms thus increased by 70%, which shows a great enhancement. Suppose now that t1 is 50ms and the power is increased by 3dB, then t2 would be increased by 70% to 85ms, and increasing the power by 6dB it would be increased by 130% to 115ms.

Performance of PN Code Based Time Hopping Sequences in M-ary Ultra Wide Band Multiple Access Systems Using Equicorrelated Signal Sets (동일 상관 신호군을 이용하는 M-ary UWB 다원 접속 시스템에서 PN 부호 기반 시간 도약 시퀀스의 성능)

  • In this paper, we evaluate the performance of PN (Pseudo Noise) code based time hopping sequences for M-ary UWB (Ultra Wide Band) multiple access systems using the equicorrelated signal sets. In particular, we consider two different types of M-ary UWB systems in UWB indoor wireless multipath channels: The first type of the systems (System #1) has identical symbol transmission rate regardless of the number of symbols M since the length of signal pulse train is fixed while M increases, and the second type of the systems (System #2) has the same bit transmission rate regardless of M since the length of signal pulse train is extended according to the increase of M. We compare the proposed systems with those using the ideal random time hopping sequence in terms of the symbol error rate performance. Simulation results show that the PN code based time hopping sequence achieves quite good performance which is favorably comparable to that of the ideal random sequence. Moreover, as M increases, we observe that System #2 shows better robustness against multiple access interference than System # 1.

New Records of Two Arcuospathidium Subspecies (Ciliophora: Haptoria: Arcuospathidiidae) from Korea

  • Arcuospathidium is a haptorian ciliate genus composed of 18 species, and only one species has been reported in Korea. Here, we identify two unrecorded Arcuospathidium subspecies by morphological observation of both living and protargol-impregnated specimens with the small subunit ribosomal RNA (18S rRNA) gene sequence. These subspecies, Arcuospathidium cultriforme cultriforme (Penard, 1922) Foissner, 1984 and A. cultriforme scalpriforme (Kahl, 1930) Foissner, 2003, were isolated from various terrestrial habitats in July and August 2013, respectivley. Arcuospathidium cultriforme cultriforme is similar to A. cultriforme scalpriforme by a knife-shaped body, a twisted-shaped macronucleus, number of dorsal brushes, position of dorsal brushes, and shape of macronucleus but former mainly differs from the body length to oral bulge length ratio (27-38% vs. 41-53%), extrusome (one types vs. three types), cyst shape (roughly faceted wall vs. smooth surface and thin wall) and number of somatic kinety rows(18-30 vs. 30-44). Additionally, we analyzed the 18S rRNA gene sequences of two A. cultriforme subspecies and compared them with the sequences from GenBank to confirm their identification at the molecular level. As the results of genetic analysis, the 18S rRNA gene sequence of the Korean A. cultriforme cultriforme population is most similar to that of Austrian population. Also, the sequence of the Korean A. cultriforme scalpriforme population is most similar to that of another population with some nucleotide differences.

Rapid and Efficient Molecular Cloning of Rat Liver Full-length LDH A-cDNA (효율높은 cloning system을 통한 Rat Liver 전장 낙산탈수소효소 A-cDNA의 제조 및 분리동정)

  • It is still difficult and time consuming to obtain cDNA sequences that contain the entire nucleotide sequence of the corresponding mRNA. A rapid and high efficient cloning method to obtain full-length cDNA segments is thus developed. The cloning procedure described here consists of the construction of oligo(dT)-tailed vector primer using pWR34 plasmid, polyadenylation of mRNA-cDNA heteroduplex using terminal deoxytransferase, and replacement of MRNA strand with DNA by RNase H and DNA polymerase I. The restriction endonuclease analysis shows that the size of inserted-cDNA is in the range of 1.5~4.0 kb long suggesting that most of cloned cDNA are full-length or nearly full-length cDNA. The plasmid-DNA recombinants obtained were 4$\times$$10^5$~$10^{6}$ per $\mu\textrm{g}$ of rat liver poly (A$^+$)mRNA, which is 4 to 10 fold higher cloning efficiency in comparison to the presently used methods for full-length cDNA cloning. The results indicate that the described cloning system is much simpler, less time consuming, and very efficient cloning method to construct a cDNA library.

Generation of Full-Length Infectious cDNA Clones of Middle East Respiratory Syndrome Coronavirus

  • Middle East respiratory syndrome coronavirus (MERS-CoV) was first identified in Saudi Arabia in 2012 and related infection cases have been reported in over 20 countries. Roughly 10,000 human cases have so far been reported in total with fatality rates at up to 40%. The majority of cases have occurred in Saudi Arabia with mostly sporadic outbreaks outside the country except for the one in South Korea in 2015. The Korean MERS-CoV strain was isolated from the second Korean patient and its genome was fully sequenced and deposited. To develop virus-specific protective and therapeutic agents against the Korean isolate and to investigate molecular determinants of virus-host interactions, it is of paramount importance to generate its full-length cDNA. Here we report that two full-length cDNAs from a Korean patient-isolated MERS-CoV strain were generated by a combination of conventional cloning techniques and efficient Gibson assembly reactions. The full-length cDNAs were validated by restriction analysis and their sequence was verified by Sanger method. The resulting cDNA was efficiently transcribed in vitro and the T7 promoter-driven expression was robust. The resulting reverse genetic system will add to the published list of MERS-CoV cDNAs and facilitate the development of Korean isolate-specific antiviral measures.

Translational Enhancement by the 5' Leader of Tobacco Mosaic Virus and Soybean Glycinin Gene in Transgenic Tobacco Plants (담배 모자이크 바이러스와 대두 Glycinin 유전자의 5' Leader Sequence를 이용한 외래 유전자의 전이효율 증진)

  • To increase the expression of a foreign protein in transgenic plant, the benefits of 5'-untranslated leader sequences of tobacco mosaic virus (TMV) RNA or soybean glycinin gene, Gy2, fused to a protein coding sequence were exploited. pGA643-derived plasmid contains 355 promoter of cauliflower mosaic virus, protein coding sequence of maize 10 kDa zein (10kZ) and Gy2 terminator. The leader from Gy2 or TMV RNA was inserted between the promoter and the coding sequence in each construct. The recombinant DNAs were introduced into tobacco plants by Agrobacterium mediated leaf disc transformation method. Although the transgene without the leader had more transcripts than the others, mRNAs containing the leader were translated more efficiently. It might be due to difference in the length of 5'-untranslated sequence and context surrounding the AUG codon, but could be sequence specific rather. These results suggest that the leader sequences of Gy2 and TMV play important roles as an enhancer in translational control of foreign gene in transgenic tobacco plant.

  • CFRP(Carbon Fiber Reinforced Plastics) of the advanced composite materials as structural materials for vehicle, has a wide application in light-weigh structural materials of airplanes, ships and automobiles because of high strength and stiffness, However, there is a design variable to be considered in practical application of the laminate composite materials, these materials are vulnerable to transverse impact. This paper is to study the effects of stacking sequence and curvature on the penetration characteristics of composite laminate shell. They are stacked to $[0_3/90_3]S,\;[90_3/0_3]s\;and\;[0_2/90_3/0]s,\;[90_2/0_3/90]s$ and their interlaminar number two and four. They are manufactured to various curvature radius (R=100, 150, 200mm and $\infty$), When the specimen is subjected to transverse impact by a steel ball, the velocity of the steel ball was measured both before and after impact by determing the time for it to pass two ballistics-screen sensors located a known distance apart. The critical penetration energy of specimen A and B with less interfaces were a little higher than those of C and D. As the curvature increases, the critical penetration energy increases linearly because the resistance to the in-plane deformation as well as bending deformation increases, which need higher critical penetration energy. The specimen A and C have higher critical penetration energy than B and D because of different stacking sequences. We examined crack length through a penetration test. For the specimen A with 2interfaces, the longest circumferential direction crack length were observed on the first interface from the impact point. For the specimen B 4-interface, the longest circumferential direction crack length were observed on the second interface from the impact point.

Rapid and Unequivocal Identification Method for Event-specific Detection of Transgene Zygosity in Genetically Modified Chili Pepper

  • To identify unintended vertical gene-transfer rates from the developed transgenic plants, rapid and unequivocal techniques are needed to identify event-specific markers based on flanking sequences around the transgene and to distinguish zygosity such as homo- and hetero-zygosity. To facilitate evaluation of zygosity, a polymerase chain reaction technique was used to analyze a transgenic pepper line B20 (homozygote), P915 wild type (null zygote), and their F1 hybrids, which were used as transgene contaminated plants. First, we sequenced the 3'-flanking region of the T-DNA (1,277 bp) in the transgenic pepper event B20. Based on sequence information for the 3'- and 5'-flanking region of T-DNA provided in a previous study, a primer pair was designed to amplify full length T-DNA in B20. We successfully amplified the full length T-DNA containing 986 bp from the flanking regions of B20. In addition, a 1,040 bp PCR product, which was where the T-DNA was inserted, was amplified from P915. Finally, both full length T-DNA and the 1,040 bp fragment were simultaneously amplified in the F1 hybrids; P915 ${\times}$ B20, Pungchon ${\times}$ B20, Gumtap ${\times}$ B20. In the present study, we were able to identify zygosity among homozygous transgenic event B20, its wild type P915, and hemizygous F1 hybrids. Therefore, this novel zygosity identification technique, which is based on PCR, can be effectively used to examine gene flow for transgenic pepper event B20.

A Space Efficient Indexing Technique for DNA Sequences (공간 효율적인 DNA 시퀀스 인덱싱 방안)

  • Suffix trees are widely used in similar sequence matching for DNA. They have several problems such as time consuming, large space usages of disks and memories and data skew, since DNA sequences are very large and do not fit in the main memory. Thus, in the paper, we present a space efficient indexing method called SENoM, allowing us to build trees without merging phases for the partitioned sub trees. The proposed method is constructed in two phases. In the first phase, we partition the suffixes of the input string based on a common variable-length prefix till the number of suffixes is smaller than a threshold. In the second phase, we construct a sub tree based on the disk using the suffix sets, and then write it to the disk. The proposed method, SENoM eliminates complex merging phases. We show experimentally that proposed method is effective as bellows. SENoM reduces the disk usage less than 35% and reduces the memory usage less than 20% compared with TRELLIS algorithm. SENoM is available to query efficiently using the prefix tree even when the length of query sequence is large.
