Analysis of Polymeric Immunoglobulin Receptor Expression in Olive Flounder (Paralichthys olivaceus) against Viral Hemorrhagic Septicemia Virus

  • Polymeric immunoglobulin receptor (pIgR) mediates the transfer of polymeric immunoglobulin to protect organisms and is one of the most important mucosal effectors. In this study, the developmental stage- and tissue-specific expression of pIgR were observed before virus inoculation in olive flounder. pIgR was gradually expressed until the formation of immune tissue, exhibiting high expression in the late juvenile period; thereafter, pIgR expression gradually decreased and exhibited high expression in the spleen and skin. Moreover, pIgR expression after viral hemorrhagic septicemia virus infection was high in the kidney and spleen tissues at high density and low at low density. The results of this study can provide a basis for future studies on breeding density, virus expression, and immune system studies in fish.

  • We developed and subsequently characterized mouse antibodies (MAbs) against viral hemorrhagic septicemia virus (VHSV, genotype IVa), the causative agent of VHS. Five hybridoma clones secreting MAbs against VHSV were established. The MAbs recognized the glycoprotein (MAbs 2C10, 18H4, 23H6, and 30B7) and nucleocapsid protein (15E10) of VHSV by western blot analysis. All five MAbs reacted with VHSV-infected cells and tissue homogenates of VHSV-infected olive flounder (Paralichthys olivaceus) by western blot analysis. Whereas, no reactivity was observed in normal cells and tissue homogenates of normal olive flounder. Moreover, these MAbs reacted with VHSV, but did not react with other fish viruses (infectious hematopoietic necrosis virus, hirame rhabdovirus, spring viraemia of carp virus, infectious pancreatic necrosis virus, marine birnavirus, and nervous necrosis virus) by enzyme linked immunosorbent assay (ELISA). These results indicate that the MAbs are specific to VHSV and can be of value in VHSV detection.

Ecological Characteristics of Vibrio vulnificus in Estuary of Kum River (금강 하구에 있어서 Vibrio vulnificus의 생태학적 특성)

  • Vibrio unlnificus, a normal inhabitant in estuaries, is of great concern because it is a potent human pathogen causing septicemia, wound infection and gastrointestinal disease in Kum river, sampling was undertaken in five station from March, 26, 1993 to February, 22, 1994. Samples of 54 and 49 were collected from seawater and bottom deposit. The total detection rate of V. unlnificus was 11.7%. The detection rates of V. unlnificus in the seawater and the bottom deposit were 9.3% and 14.3% respectively. V. unlnificus was mainly detected in estuary water when temperature was above 23$^{\circ}C$ and salinity was below 15%. We suppose that water when temperature, salinity, pH and COD affect growth of V. unlnificus.

  • The viral hemorrhagic septicemia of rainbow trout, Salmo gairdneri was studied. The hematocrit values of diseased fish were very low than those of normal fish. And, the GOT and GPT values of serum of diseased fish were a little high than those of normal fish. Cytopathic effect of viral agents(serum and organs of diseased fish) was observed with inverted phase contrast microscopy. After 24hrs infection, the cells were showed the cytopathic effect.

  • birth-weight infants during the recovery phase of respiratory distress syndrome and has been associated with long-term pulmonary sequelae. The importance of surgical and medical abolition of left-to-right shunting in symptomatic neonates is established. Four preterm infants with birth weights under 1, 500gm with a PDA unresponsive to pharmacological closure underwent ligation. Two of preterm infants survived to be discharged and are developing normally. One infant has died due to respiratory distress syndrome, septicemia and necrotizing enterocolitis during hospital stay and other infant died due to septicemia after hospital discharge with follow-up for 6 months.

  • Two cases of sudden death without clinical signs in a four-month-old female Hanwoo (Bos taurus coreanae) and sixteen-month-old female Holstein (Bos taurus taurus) were investigated. Severe pneumonia and septicemia were found on postmortem examination. Petechial and ecchymotic hemorrhages were present in the subcutaneous tissues of the neck region. The lungs remained expanded without collapsing, and the mediastinal lymph nodes were enlarged. A significant number of hemorrhagic bubbles were present in the tracheal lumen. On histopathology, the alveolar blood vessels were moderately congested, and inflammatory cells and multinucleated syncytial cells were found in the alveolar space. The pathogen isolated from the lesion was identified as Bibersteinia trehalosi using MALDI-TOF and 16S rRNA gene and sodA gene sequence analyses. To the best of our knowledge, this is the first report of B. trehalosi infection in cattle in Korea.

Vibrio vulnificus infection: a persistent threat to public health

  • Vibrio vulnificus is a gram-negative bacterium that can cause serious, potentially fatal infections. V. vulnificus causes three distinct syndromes: an overwhelming primary septicemia caused by consuming contaminated seafood, wound infections acquired when an open wound is exposed to contaminated warm seawater, and gastrointestinal tract-limited infections. Case-fatality rates are higher than 50% for primary septicemia, and death typically occurs within 72 hours of hospitalization. Risk factors for V. vulnificus infection include chronic liver disease, alcoholism, and hematological disorders. When V. vulnificus infection is suspected, appropriate antibiotic treatment and surgical interventions should be performed immediately. Third-generation cephalosporin with doxycycline, or quinolone with or without third-generation cephalosporin, may be potential treatment options for patients with V. vulnificus infection.

Neonatal invasive Streptococcus gallolyticus subsp. pasteurianus infection with delayed central nervous system complications

  • Group D streptococci are known to cause newborn septicemia and meningitis, but the Streptococcus bovis group strains rarely cause serious neonatal infections in Korea. Central nervous system (CNS) complications of neonatal S. bovis group infection have rarely been reported. In adults, S. bovis group strains cause bacteremia and endocarditis, and are associated with gastrointestinal malignancy. However, only a few studies have reported meningitis and septicemia in infants. Here, we describe a case of bacteremia and meningitis due to Streptococcus gallolyticus subsp. pasteurianus with a delayed CNS complication in an infant. A 28-day-old male infant was admitted to the hospital with a 1-day history of fever. Cultures of blood, cerebrospinal fluid, and urine showed the presence of S. bovis group strain-S. gallolyticus subsp. pasteurianus. He was discharged after 21 days of intravenous ampicillin and cefotaxime administration. Two weeks later, he was readmitted with a fever and short episodes of tonic-clonic movements. Brain magnetic resonance imaging showed marked bilateral frontal subdural effusion. He was discharged after 31 days of antibiotic therapy, and no neurological sequelae were observed at the 9-month follow-up. In conclusion, we present a rare case of neonatal S. gallolyticus subsp. pasteurianus infection causing urinary tract infection, septicemia, meningitis, and delayed CNS complications. This case emphasizes the need for physicians to be aware of S. bovis infection in infants.

Analysis of antigenicity of viral hemorrhagic septicemia virus (VHSV) glycoprotein from cultured olive flounder Paralichthys olivaceus (양식 넙치, Paralichthys olivaceus에서 분리한 viral hemorrhagic septicemia virus (VHSV)의 항원성 분석)

  • The amino acid sequence of glycoprotein of Korean VHSV isolate (KR'01-1) was analyzed using the DNAStar Protean system. Based on the flexibility, hydrophilicity, antigenic index and surface probability, three regions (Gp1, Gp2 and Gp3) were selected as potential antigenic determinants. Three oligopeptides containing the amino acid sequences of the three regions were synthesized and polyclonal antibodies were raised against them. The activities of the antibodies were analyzed by Western blotting and virus neutralization test. The results showed that antibodies raised against oligopeptides Gp1 and Gp2 neutralized the infectivity of VHSV, suggesting that they can be possible candidates for subunit vaccines against VHS diseases in olive flounder.
