Effects of Column Length and Particle Diameter on Phospholipid Analysis by Nanoflow Liquid Chromatography-Electrospray Ionization-Mass Spectrometry

  • The effects of column length and particle size on the efficiency of separation and characterization of phospholipids (PLs) are investigated using nanoflow liquid chromatography-electrospray ionization-tandem mass spectrometry (nLC-ESI-MS-MS). Since PLs are associated with cell proliferation, apoptosis, and signal transduction, it is of increasing interests in lipidomics to establish reliable analytical methods for the qualitative and quantitative profiling of PLs related to biomarker development in adult diseases. Due to the complexity of PLs, the preliminary separation of PLs is necessary prior to MS analysis. In this study, length of capillary column and the particle size of reversed phase ($C_{18}$) packing materials are varied to find a reliable condition for the high speed and high resolution separation using 8 PL standard mixtures. From experiments, it was found that a capillary column of nLC-ESI-MS-MS analysis for PL mixtures can be minimized to a 5 cm long pulled tip column packed with 3 ${\mu}m$ $C_{18}$ particles without losing resolution.

  • Flow separation of recirculation zone by increasing of flow and change of its direction at confluence results in backwater due to conveyance reduction. The hydraulic characteristics of flow separation are analysed by experimental results of flow ratios of tributary and main streams and approaching angles. The boundary of flow separation by dimensionless length and width is defined by the streamline of zero and this definition agrees well to the existing investigation. Because flow separation doesn't appear in small flow ratio and approaching angle of $30^{\circ}$, the equation of flow separation with flow ratio and approaching angle is provided. In flow separation consideration and comparing with previous results, the existing equations of dimensionless length and width ratios by function of approaching angle, flow ratio, and downstream Froude number are modified and also contraction coefficient and shape factor are analysed. Dimensionless length and width ratios are proportional to the flow ratio and approaching angle. In analysis of water surface profiles, the backwater effects are proportional to the flow ratio and approaching angle and the magnitude at outside wall is greater than that of inside wall of main stream. The length, $X_l$ from the beginning of confluence to downstream of uniform flow, where the depth is equal to uniform depth, is characterized by width of stream, flow ratio, approaching angle, and contraction coefficient. The ratios between maximum water depth by backwater and minimum depth at separation are analysed.

An Experimental Study on Tip Velocity and Downstream Recirculation Zone of Single Groyne Conditions (단일 횡수제 조건에 따른 선단부 유속과 재순환 영역에 대한 실험 연구)

  • Recently, the concern of groynes that provide us with various ecological environment and improvement of scenary near river as well as hydraulic improvement has greatly increased for naturalized river and ecological river restoration. However there is no adequate design guidelines in Korea. Furthermore there is lack of research for installation of groynes in Korea. Thus, in this study, we have conducted hydraulic modeling test for kinds of groyne and changes of permeability and made a study of the analysis for installation factor of groyne. Experimental conditions were the fixed bed model for flow pattern of groyne tip and separation length about changes of groyne length and permeability. The Experiments were conducted to measure velocity and drift angle at groyne tip. The main study kas measured the groyne tip velocity for a factor of scour and drift angle for velocity increase at center of channel. We have suggested the equation about groyne tip velocity rate(tip velocity/ approach velocity) and area rate(groyne area/ flow area). And In recirculation zone of groyne downstream, To study the basic data of groyne about installation interval has analyzed the change of separation length and incidence angle. We have suggested the equation about separation length rate(separation length/ groyne length), changes of permeability and installation angle.

A study of backward-facing step flow in a rectangular duct (후향계단이 있는 사각덕트 내부의 유동특성 연구)

  • This study is to analyze turbulent flow over a backward-facing step in a rectangular duct. The side wall effects on the internal flow were determined by varying the aspect ratio(defined as the step span-to-height ratio) from 1 to 20. In the flow behind a backward-facing step, separation, recirculation and redeveloping is occurred frequently. These phenomena appear in a particular variation by varying the aspect ratio. The results show that the aspect ratio has an influence on the velocity and reattachment length. When the AR is increased, the reattachment length is increased. For 6 over aspect ration, the rate of increase is decreased. The length of recirculation in the upper corner is increased, as the increase of aspect ration. It's width is not changed in the variation of aspect ration. The transverse, streamwise and spanwise velocities were decreased along the flow down stream of the step.

Variations of Flow Thalweg Alignment and Separation Region around a Groyne (단일수제 설치에 따른 흐름중심선과 흐름분리영역의 변화)

  • In this study, hydraulic characteristics of thalweg line occurred near the single groyne and recirculation zone around downstream of the structure were discussed from the experiments which conducted in fixed flat-bed channel flume. The thalweg line is the stream line where the maximum velocity occurs and flow separation appeared at the tip of groyne changes the thalweg alignment. In this study, the variations in flow fields which were caused by different length and permeability of groyne were measured by LSPIV(Large Scale Particle Image Velocimetry), and also the characteristics of thalweg line and separation area were analyzed. From these results, it is found that length to thalweg line from the channel wall $T_{CL}$ and height of separation area $S_h$ vary the channel width up to 75%, 50% in the change of length and permeability of groyne, however the Froude number does not affect on $T_{CL}$ and $S_h$. Velocity along the thalweg $U_{CL}$ Increased by twice the mean velocity, and the maximum velocity occurred in downstream where the distance $5{\sim}6$ times of groyne length away from the groyne, which does not get affected by length and permeability of groyne and Froude number.

  • An experimental study on the flow over the axisymmetric backward-facing step was carried out. The purpose of the present study is to investigate the effect of the boundary layer thickness at the separation point on the reattachment length and to understand the structure of the recirculating flows. Local mean and fluctuating velocity components were measured in the separating and reattaching axisymmetric turbulent boundary layer over the wall of convex cylinder placed in a water tunnel by using 2-color 4-beam fiber optics laser Doppler velocimetry. The study demonstrated that the reattachment length increases with increasing boundary layer thickness. It was also observed that the reverse flow velocity and turbulent kinetic energy decrease with an increase in the momentum thickness at the separation point. The measured velocity field suggests that the boundary layer thickness at the separation can affect definitely on the formation of corner eddy.

  • Laminar separation bubble and transitional flow over an airfoil are investigated at a moderate range of Reynolds numbers. In this research, a Reynolds-Averaged Navier-Stokes code is coupled with an empirical transition model that can predict transition onset points and the length of transition region. Without solving the boundary layer equations, approximated e-N method is directly applied to the RANS code and iteratively solved together. The computational results are compared with the experimental data for NACA0012 airfoil. Results of transition onset point and length are compared well with experimental and XFOIL prediction. In high angle of attack the present RANS results show better agreement than XFOIL results using the boundary layer equations.

Capillary Electrophoresis of Single-stranded DNA

  • We have studied the migration behavior of single-stranded DNA using capillary gel electrophoresis under various conditions. It was found that optimum electric fields should be less than 150 V/cm for the good tradeoff between the separation time and the resolution. It seems that the gel matrix with the combination of different polymer average molecular weights is important to extend the maximum readable DNA bases. The total gel concentration less than 3.1% in the mixed gel system showed good separation efficiency up to 600 bases. The best result was obtained with the poy(ethylene)oxide (PEO) gel concentration of 1.2% of Mr 8,000,000 and 1.8% of Mr 600,000. We observed that the capillary length between 50 cm to 100 cm (effective length) should be employed for the optimization between the total DNA migration time and the maximum readable length. A trizma base-boric acid-ethlyenediaminetetraacetic acid (EDTA) (TBE) buffer was commonly used for DNA sequencing, but we found that 3-[tris(hydroxymethyl)methyl amino]-1-propane sulfonic acid (TAPS) buffer worked as well for the single-stranded DNA separation. Especially, TAPS buffer showed a good resolution for very short DNA bases (1 to 30 bases).

  • A gas-liquid centrifugal separator is widely used in industry because of its simple geometry and little maintenance. Also, these separators have considerable advantages over filters, scrubbers or precipitators in term of compact design, low pressure drop and higher capacity. A gas-liquid centrifugal separator is a device that utilizes centrifugal force and low pressure to separate liquid from gas by density difference. Design parameters such as length of separation space, swirl vane exit angle, inlet to outlet pipe diameter ratio, models for separation efficiency and low pressure drop as a function of physical dimension are not available in literature. In present study, length of separation space (from vane to gas exit opening) has been studied using CFD. The 3-D Navier-Stokes equations are numerically solved using a fully implicit finite volume scheme. Based upon the obtained solutions, tangential velocities, centrifugal forces, vortices and total pressure losses are analyzed to find the best design parameters.

  • A method to measure the crack length in rubbery materials is described. Through dimensional analysis and experiments, an equation is derived to give the crack length as a function of the change of strain energy density in a region remote from the crack. The function is provided in a form of separated terms of loading and material, the validity of which is experimentally proved using separation parameters.
