ANALYSIS ON THE PREDICTOR OF DISEASE PROGRESSION IN REFRACTORY PERIODONTITIS (난치성 치주염의 질환진행 예견 인자에 관한 분석)

  • Refractory periodontitis manifest progressive attachment loss in a rapid and unrelenting manner regardless of the type or frequency of therapy applied. The purpose of this study was ta evaluate the relation between the level of cytokines in GCF and periodontopathic microflora with disease activity of refractory periodontitis. Selection of patients with refractory periodontitis (7 males, 3 females) were made by long term clinical observation including conventional clinical history and parameters. Teeth that showed pocket depth greater than 6mm were selected as sample teeth. Subjects were examined at baseline and after 3 months. Prior to baseline test, individual acrylic stent was fabricated. Reference grooves were made on each sample tooth site. Pocket depth and attachment loss were measured by Florida Probe. Gingival index was measured at 4 sites each sample teeth. Disease activity was defined as attachment loss of ${\ge}$ 2.1mm, as determined by sequential probing and tolerance method. The pattern and amount of alveolar bone resorption was observed with quantitative digital subtraction image processing radiography. Morphological analysis of subgingival bacteria was taken by phase contrast microscopy. Predominant cultivable bacterial distribution and frequency were compared between disease-active and disease-inactive site using immunofluorescence microscopy and selective microbial culturing. Levels of $interleukin-l{\beta}$, 2, 4, 6 and $TNF-{\alpha}$ in GCF and blood serum sample were quantified by ELISA. In active sites, P. intermedia was significantly increased to compare with inactive site. $IL-1{\beta}$, IL-2, IL-6 and $TNF-{\alpha}$ in GCF were increased in active sites and IL-2 in serum was increased in active patients significantly. Alveolar bone loss in active site was correlated with $IL-1{\beta}$, IL-2 in GCF. And loss of attachment in active site was correlated with IL-2 in GCF. These results demonstrate that IL-2 in serum, $IL-1{\beta}$, IL-2, IL-6 and $TNF-{\alpha}$ in GCF, P, intermedia might be used as possible predictors of disease activity in refractory periodontitis before it is clinically expressed as attachment loss and quantitative alveolar bone change.

  • To produce cupped oyster sprat consistently, we investigated the efficiency of nine settlement substrates using selective breeding of Pacific oyster Crassostrea gigas, with the goal of popularizing off-bottom culture on the west coast of Korea. We also compared the growth and survival of selectively bred sprat (diploid) with triploid sprat from an off-bottom culture system. Considering, the attachment rate and detachment efficiency, producing cupped oyster in a polypropylene gunny bag proved to be the most effective method. There were no differences in shell growth or total weight between the diploids from selective breeding and triploids from off-bottom culture for 5 months. However, the survival rate was 8 times higher in the diploids than the triploids. Transplanting sprat from selective breeding is one way to restore oyster farms and nurture off-bottom culture along the west coast.

  • Over the years, nanostructures have been developed to enable to support enzyme usability to obtain highly selective and efficient biocatalysts for catalyzing processes under various conditions. This review summarizes recent developments in the nanostructures for enzyme supporters, typically those formed with various inorganic materials. To improve enzyme attachment, the surface of nanomaterials is properly modified to express specific functional groups. Various materials and nanostructures can be applied to improve both enzyme activity and stability. The merits of the incorporation of enzymes in inorganic nanomaterials and unprecedented opportunities for enhanced enzyme properties are discussed. Finally, the limitations encountered with nanomaterial-based enzyme immobilization are discussed together with the future prospects of such systems.

  • Labeling of a protein with a specific dye or tag at defined positions is a critical step in tracing the subtle behavior of the protein and assessing its cellular function. Over the last decade, many strategies have been developed to achieve selective labeling of proteins in living cells. In particular, the site-specific unnatural amino acid (UAA) incorporation technique has gained increasing attention since it enables attachment of various organic probes to a specific position of a protein in a more precise way. In this review, we describe how the UAA incorporation technique has expanded our ability to achieve site-specific labeling and visualization of target proteins for functional analyses in live cells.

  • Human Erythropoietin (EPO) gene is cloned in quail fibrosarcoma cell, QT35. Because molecular weight of EPO is similar to that of serum albumin, cell culture with serum containing medium makes purification of EPO very difficult. Using fractional factorial study, we have developed serum free medium for the recombinant QT35 cell lines, QT N4D4 and QT SY-IMP, which have cloned EPO with glutamine synthetase (GS) gene amplification system and with puromycin selective marker, respectively. Among the seven frequently used medium components, fibronectin, BSA, and EGF were the most important for EPO production. However, sufficient fibronectin supplement to the medium did not make any good attachment of QT35 to culture plate over 3 days. Therefore, to maximize EPO production, we attempted a medium-shift at confluence from serum containing medium to serum free medium(QT SFM6). Using the medium-shift protocol with QT SFM6, nearly the same productivity of EPO was achieved comparing with that without medium-shift. This result was true in both QT35 cell lines in three types of culture, i.e. T flask, microcarrier and roller bottle cultures.

Idol Fan's Acceptance Process of Idol Image -Focusing on BTS- (아이돌 팬의 아이돌 이미지 수용 과정 -BTS를 중심으로-)

  • The purpose of study was to analyze the process of accepting by BTS fans. The methodology used in this study was qualitative research using grounded theory and the results of the study follow. As a result of open coding, 47 concepts, 23 subcategories, and 14 categories were derived. In the axis coding stage, the casual conditions were "Recognition of BTS image," "Fascinated by BTS image," "Simple consumption of BTS image," and "Arising and expansion of curiosity about BTS image." The contextual conditions were "Accessibility of BTS image" and "Abundance of BTS image searching paths." The central phenomenon appeared to be "Immersing and studying BTS image." The arbitration conditions were "Capabilities required to reproduce BTS image" and "Motivation for contributing BTS image." The actions/interactions were "Presenting BTS image," "Contribution to the spread of BTS image," and "Involved in forming new BTS image." The result was "Emergence and expansion of new BTS image meaning" and "Strengthening attachment to BTS." Through process analysis, it was found that acceptance of BTS images consisted of five stages: "Recognition of BTS image," "Becoming curious about BTS image," "Searching BTS image," "Intervention of BTS image." and "Reproduction of BTS image." As a result of deriving the core categories through selective coding, the core category was "Forming a bond while participating in the BTS image."

Proteomic Changes by Acupuncture Stimulation at HT7 in the Hippocampus of Rat Pups (신문혈 자침이 어린 백서 해마의 단백질 발현에 미치는 영향)

  • Objectives : Hippocampus, a region of temporal lobe, plays an important role in the pathogenic mechanisms of brain diseases such as Alzheimer's disease, depression and temporal lobe epilepsy. This research is designed to investigate hippocampal changes after acupuncture stimulation at Shinmun(HT7) using 2-dimensional gel electrophoresis(2-DE). Methods : On postnatal-day 15, rat pups were randomly devided into Normal(NOR) or HT7 group. All of Pups kept with their mothers for 7 days, but pups in HT7 group received acupuncture stimulation at HT7 daily. On postnatal-day 21, hippocampus of each rat pup was dissceted 30 minutes after last acupuncture stimulation and the protein expressions were investigated using 2-DE. Results : After acupuncture stimulation at HT7, expression of 20 proteins were significantly increased. Succinate semialdehyde dehydrogenase, glyceraldehyde-3-phosphate dehydrogenase-like, transketolase, aconitate hydratase and phosphoglucomutase-1 were related to glucose methabolism. Eukaryotic initiation factor(eIF) 4A-II, eIF 4A-III, mitochondrial Tu translation elongation factor and chain A of crystal structure of the 70-Kda heat shock cognate protein involve in the protein synthesis in ribosome. Tubulin ${\beta}$-4 chain, tubulin T ${\beta}$-15 and tubulin ${\alpha}$-1B chain comprise cytoskeleton. Glutathione S-transferase(GST) ${\omega}$-1, GST P and GST Yb-3 can reduce oxidative stress. ${\beta}$-soluble N-ethylmaleimide-sensitive fusion protein attachment protein is required for vesicular transport between the endoplasmic reticulum and the Golgi apparatus, glycerol-3-phosphate dehydrogenase plays a major role in lipid biosynthesis, creatine kinase U-type catalyses the conversion of creatine and consumes adenosine triphosphate to create phosphocreatine and adenosine diphosphate. Platelet-activating factor acetylhydrolase IB subunit alpha and voltage depedent anion-selective channel protein 2 were also increased. Conclusions : The results suggest that acupuncture stimulation at HT7 may enhance glucose and lipid metabolism, protein synthesis, cytoskeletal substance and anti-oxidative stress in hippocampus.

Biomimetic sequestration of $CO_2$ and reformation to $CaCO_3$ using bovine carbonic anhydrase immobilized on SBA-15 (생체모방공학을 이용한 bovine carbonic anhydrase를 SBA-15에 고정화하여 이산화탄소분리와 재구성된 $CaCO_3$ 연구)

  • The biocatalytic capture of $CO_2$, and its precipitationas $CaCO_3$, over bovine carbonic anhydrase (BCA) immobilized on a pore-expanded SBA-15 support was investigated. SBA-15 was synthesized using TMB as a pore expander, and the resulting porous silica was characterized by XRD, BET, IR, and FE-SEM analysis. BCA was immobilized on SBA-15 through various approaches, including covalent attachment (BCA-CA), adsorption (BCA-ADS), and cross-linked enzyme aggregation (BCA-CLEA). The immobilization of BCA on SBA-15 was confirmed by the presence of zinc metal in the EDXS analysis. The effects of pH, temperature, storage stability, and reusability on the biocatalytic performance of BCA were characterized by examining para-nitrophenyl acetate (p-NPA) hydrolysis. The $K_{cat}/K_m$ values for p-NPA hydrolysis were 740.05, 660.62, and $680.11M^{-1}s^{-1}$, respectively, where as $K_{cat}/K_m$ for free BCA was $873.76M^{-1}s^{-1}$. The amount of $CaCO_3$ precipitate was measured quantitatively using anion-selective electrode and was found to be 12.41, 11.82, or 11.28 mg $CaCO_3$/mg for BCA-CLEA, BCA-ADS, or BCA-CA, respectively. The present results indicate that the immobilized BCA-CLEA, BCA-ADS, and BCA-CA are green materials, and are tunable, reusable, and promising biocatalysts for $CO_2$ sequestration.

Effects Of Nicotine And PDGF On The Cell Activity Of Human Gingival Fibroblasts And Periodontal Ligament Cells. (니코틴과 PDGF-AB가 배양인체 치은섬유모세포 및 치주인대세포의 활성에 미치는 영향)

  • The ability of fibroblasts attached to teeth is paramount important in reestablishing the lost connective tissue attachment after periodontal therapy. The migration and proliferation of periodontal ligament cells are desired goal of periodontal regeneration therapy. PDGF is well known to regulate the cell activity of mesenchymal origin cell. Tobacco contains a complex mixture of substance including nicotine, various nitrosamines, trace elements, and variety of poorly characterized substances. Human gingival fibroblasts and periodontal ligament cells were cultured from extracted tooth for non-periodontal reason. Cultured human gingival fibroblasts and periodontal ligament cells in vitro were treated with PDGF, nicotine in time dependent manner. Cellular activities were determined by MTT assay. The purpose of this study was to determine the effects of Nicotine and PDGF, respectively and the effect of PDGF presence of nicotine on human gingival fibroblasts and periodontal ligament cells. The results were as follows : 1. In the cell activities of human gingival fibroblasts and periodontal ligament cells were similar or decreased to control value at 1st day. At 2nd day, cellular activities of both group were increased to control value. At 3rd day, cellular activities of both group were returned to the control value. 2. In the cell activities of PDGF on human gingival fibroblasts and periodontal ligament cells, cell activities significantly increase from control group on periodontal ligament cells compared to gingival fibroblast group at 3rd day. 3. In the cell activities of PDGF and nicotine combined application on human gingival fibroblasts and periodontal ligament cells, it seems likely that the nicotinic effect of gingival fibroblasts were higher than periodontal ligament cells and the PDGF effect of periodontal ligament cells were higher than gingival fibroblasts. This results suggested that PDGF might stimulate the selective growth on periodontal ligament cells.

A Grounded Theory Approach on the Parting Experience of Korean Middle-aged Women with their Children (한국 중년여성의 성인 자녀와의 분리경험에 대한 근거이론적 접근)

  • Purpose: This research was conducted to explore the experience of middle-aged women parting with their children and to develop a grounded theory. Method: The participants were 11 women in their 50's who had experienced parting with their children due to schooling, military service and marriage. The grounded theory methodology based on symbolic interactionism was used. Results: 106 concepts, 30 subcategories and 14 categories were derived from the analysis through processes of open coding, axial coding and selective coding. The central phenomenon was 'happy but sad' and the core category was a process of 'independence that is sorrowful yet delightful'. Four types were identified; the independent-type; submissive -type; family-reliant-type ; and fragile-type; based on the attachment to the child, state of mind, satisfaction of the situation, family support system and self-achievement. Conclusion: This research identified that the modern Korean middle-aged women who are considered to be the 'sandwich generation' since they have experienced a turbulent history and the change from a confucian parent-child lifestyle to a couple-oriented one, moved away from an attitude dependent on children in parting with them and started preparing for an independent late life. Based on the results, the verification research is advised on the variables that affect the experience of parting with children.
