Growth Stimulation of Pines by Artificial Inoculation with Mycorrhizal Fungus, Pisolithus tinctorius (균근균(菌根菌)의 인공접종(人工接種)에 의(依)한 소나무류(類)의 생장촉진(生長促進))

  • Two ectomycorrhizal fungi, Pisolithus tinctorius and Thelephora terrestris, were introduced form U.S.A. and inoculated to five pine species in Korea to evaluate the reported growth stimulation of host plants after inoculation. These fungi were grown as mycelial inoculum in large quantity and ioculated to the fumigated nursery soil just before seed sowing. At the end of the first growing season. Pisolithus stimulated the height growth of Pinus densiflora. P. thunbergii. P. rigida, and P. rigida x teada by55, 36, 69, and 37%, respectively, compared with control seedlings with no fumigation and no inoculation. When the growth stimulation was expressed with dry weight, Pisolithus increased dry weight of P. densiflora and P. rigida x taeda by 143% and 128%, respectively, over control seedling. Thelephora failed to stimulate growth of inoculated plants. Pinus koraiensis did not respond to the inoculation during the first growing season. It is concluded that artificial inoculation of nursery pine trees with selected mycorrhizal fungi should be seriously considered to improve the quality of planting stocks and to stimulate early plant growth. The potential for use of Pisolithus in reforestation on adverse sited is also discussed.

Plant Regeneration and Effect of Auxin and Cytokinin on Adventitious Shoot Formation from Seedling Explant of Taraxacum platycarpum (민들레 [Taraxacum platycarpum]유식물 절편으로부터 부정아 발생에 미치는 auxin과 cytokinin의 영향 및 식물체 재생)

  • Taraxacum platycarpum has been used as a medicinal plant. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on medium with various kinds of growth regulators. Adventitious shoot formation was achieved when cytokinin was used alone. Shoot formation was higher on MS medium containing 2 mg/L BAP compared to that with 2 mg/L kinetin and 2 mg/L 2-ip. Among root, hypocotyl and cotyledon, roots were the best explant for the adventitious shoot induction. Adventitious shoot formation from roots declined markedly by the combination of both 0.1 mg/L NAA and 2 mg/L BAP, while shoot formation from cotyledons was stimulated by the same combination. Root formation from the regenerated shoots was achieved on 1/3MS medium containing 0.2 mg/L NAA. Regenerated plantlets was acclimatized and transplanted to the soil, showing 100% survival.

Study on Nitrogen·Phosporus Absorption and Growth of Seedling of Prunus serrulata var. serrulata f. spontanea(E.H. Wilson) C. S. Chang by Treatment with Dried Swine Excrement (고형돈분 처리에 따른 벚나무의 유묘생장 및 질소·인산 흡수에 관한 연구)

  • Prunus serrulata var. serrulata f. spontanea (E. H. Wilson) C. S. Chang was treated with dried swine excrement at various concentration levels, and their growth reactions and the contents of total kjeldahl nitrogen and total phosphoric acid were analyzed. The result is as follows; 1. When the plants were treated with 0.25% of dried swine excrement, the seed germination rate of Prunus serrulata var. serrulata f. spontanea was higher than that of the control. However, it showed a tendency to decrease when the treatment concentration got higher than 0.25%. 2. The growth rate of species was highest when they were treated with 0.25% of dried swine excrement. It tended to decrease with higher treatment concentration than 0.25%. Significant difference was clear between the control and the experimental plants of Prunus serrulata var. serrulata f. spontanea. 3. The contents of nitrogen and phosphoric acid in Prunus serrulata var. serrulata f. spontanea also got higher as the concentration of dried swine excrement increased. However, the content of total kjeldahl nitrogen was highest in the leaves, and followed by the roots and stems. In contrast, the content of total phosphoric acid were highest in the roots, and followed by the leaves and stems. Therefore, stems had the lowest contents of both nitrogen and phosphoric acid. 4. The contents of total kjeldahl nitrogen and total phosphoric acid in the soil were measured before and after the growth experiment of Prunus serrulata var. serrulata f. spontanea treated with dried swine excrement. The contents of nitrogen and phosphoric acid significantly decreased after the experiment. In conclusion, the contents of total kjeldahl nitrogen and total phosphoric acid accumulated in the plants increased as the concentration level of dried swine excrement got higher. The seed germination and plant growth rates were best at 0.25% treatment of dried swine excrement. The treatment of dried swine excrement may bring high effects on increasing the plant growth rate but could damage the plants with higher concentration than they need. Different optimal concentration levels of dried swine excrement for different plants should be found before it is used as fertilizer.

Effect of Nitrogen and Age of Alfalfa ( Medicago sativa L. ) Seeding on Growth and Nodulation When Growth under a Cool Environment (저온하에서 질소시비가 근류균을 접종한 Alfalfa의 생육부위에 미치는 영향에 관하여)

  • There are differences in recommendations in the USA as to whether nitrogen fertilizer should be applied when establishing alfalfa (Medicago sativa L). The reason for not applying nitrogen is because some researchers found the addition of nitrogen reduced nodulation of alfalfa plants. A replicated experiment was conducted under controlled environmental conditions at the University of Nevada-Reno, Reno, Nevada, USA, to determine the effects of nitrogen application on seedling growth and nodulation of alfalfa when grown in a cool environment. A sterile sand was used in the growing media to which a complete nutrient solution minus nitrogen was applied volumetrically to each pot daily. Half of the pots received $NH_4NO_3$, at the rate of 11.2 kg/ha, at seeding and two and four weeks after planting, giving a total nitrogen application rate of 33.6 kg/ha. Rhizobia inoculant (R-12) consisted of a mixture of strains 171-15a. 1682c and 80 PI 265 of Rhizobium meliloti. Inoculant was applied to the seeds prior to planting and to the sand media at two and four weeks after seeding. Twenty seeds were planted in pots 14.0 cm in diameter and 11.5 cm deep. Seedlings were thinned after emergence to ten plants per pot. They were grown in a controlled environment chamber with a 16-hour light period. Soil temperatures at 6 cm depth ranged from $5.7^{\circ}C\;to\;21.5^{\circ}C$ and had a daily mean of $16.2^{\circ}C$ Plants were harvested at weekly intervals for seven weeks at which time root, shoot and total length, dry weight, volume and number of nodules per plant were determined. Root, shoot and total length were not affected by nitrogen fertilizer. However, application of nitrogen increased the size of the seedlings as determined by dry weight and volume when compared to plants which were not fertilized. This indicates that rhizobia did not fix enough atmospheric nitrogen to promote good growth. Nitrogen application resulted in significantly more nodules per plant. The effect of nitrogen fertilizer became more apparent as the plant became older. Results of this experiment show there are benefits from applying nitrogen at a low rate when establishing alfalfa under a cool environment.

Effects of Paper Mill Sludge-Fertilizer Application on the Growth Performances of Tree Seedlings (제지(製紙)슬러지 가공비료(加工肥料)의 시용(施用)이 수묘(樹苗)의 생육(生育)에 미치는 영향(影響))

  • Effects of paper mill sludge-fertilizers were investigated on the growth performances of tree seedlings under field conditions. Two types of sludge-fertilizers applied were an organic sludge compost and a processed sludge-pellet fertilizer strengthened with several components of organic and inorganic nutrients. Three species of tree seedlings studied were ibota privet(Ligustrum obtusifolium) and yellow poplar(Liriodendron tulipifera) of 1-0 year seedlings and also eastern white pine(Pinus strobus) of 2-2 year seedlings. Nitrogen and phosphorus contents of plot soil treated with sludge compost+sludge-pellet fertilizer+sludge-pellet fertilizer plot were increased to 8 times and 2.6 times as those of control plot, respectively. The growth performances of root collar diameter and seedling height were repeatedly measured up to five times through the growing season and the results were analyzed statistically by analysis of variance of randomized block design and Duncan's multiple range test. The growth performances of dry weight were measured after last fifth measurements.

Isolation and Characterization of Phosphate Solubilizing Bacteria Pantoea Species as a Plant Growth Promoting Rhizobacteria (식물 생장 촉진 활성을 가진 인산분해 미생물 Pantoea 종의 분리 및 특성 규명)

  • Plant growth-promoting rhizobacteria (PGPR) have gained worldwide importance and acceptance due to their agricultural benefits. These microorganisms are potential tools for sustainable agriculture, with effects on plant growth, biofertilization, induced systemic resistance, and biocontrol of plant pathogens. In this study, four different Pantoea species were isolated from field soil, and their plant growth-promoting characteristics were studied. Based on 16S rDNA gene sequencing analyses, the se were grouped into Pantoea ananatis, Pantoea citrea, Pantoea dispersa, Pantoea vagans and named as Pa1, Pc1, Pd1, Pv1, respectively. All of these strains have their ability for solubilization of insoluble phosphate depending on pH decrease at the range around pH 5 at 1days after inoculation and production of plant hormone indole acetic acid (IAA) with 85.3±16.3 μg/ml of Pa1, 183.9±16.8 μg/ml of Pc1, 28.8±17.3 μg/ml of Pd1 and 114.1±16.5 μg/ml of Pv1, respectively. Pa1, Pc1 and Pd1 also have high activity for production of gibberellin (GA3) hormone with 331.1±19.2 μg/ml of Pa1, 288.5±16.8 μg/ml of Pc1, 309.2±18.2 μg/ml of Pd1, but Pv1 does not. Furthermore, all these species have significantly promoted the growth of the lettuce seedling plants at the range around 32~37% for fresh weight and 10~15% for shoot length enhancement, so that these microbe could be used as a potential bio-fertilizer agents.

Effect of Seed Density, Number of Seeds Sown Per Hole and Thinning Treatment on Growth Characteristics and Disease Occurrence in Greenhouse-Cultivated Ginseng (인삼 하우스 직파재배시 파종밀도, 혈당 파종립수, 솎음처리가 생육과 병 발생에 미치는 영향)

  • This study was conducted to investigate the effects of sowing density, number of seeds sown per hole, and thinning treatment on growth characteristics and disease occurrence in Panax ginseng under direct sowing cultivation in a blue plastic greenhouse. Seedling were grown from 2 or 3 seeds sown, and the healthiest was only retained, while the rest were thinned out at the foliation stage. $NO_3$-N, $P_2O_5$, and organic matter content differed significantly between growth conditions in the plastic greenhouse and in conventional shade in the soil. Disease also tended to be higher in the conventional shade than in the plastic greenhouse. Plant height and stem length showed an increasing trend with increasing sowing density and number of seeds sown per hole. However, these measures noticeably decreased when thinning treatment was conducted. Growth of the subterranean part of ginseng was not markedly influenced by sowing density, the number of seeds sown per hole, or thinning treatment. Root weight, which is an important factor in yield, was significantly affected by the number of seeds sown and thinning treatment. Interestingly, root weight tended to be higher in the thinning treatment plot than the untreated control plot. Damping-off and root rot increased noticeably as the number of seeds sown increased. Disease also tended to be substantially higher in the thinning treatment plot than the untreated control. However, physiological disorder of the plants did not vary with sowing density, the number of seeds sown, or thinning treatment.

Studies on the Seed Transmission of Colletotrichum spp. in Red Pepper (Capsicum annuum) (고추 탄저병균(炭疽病菌)의 종자전염(種子傳染)에 관(關)한 연구(硏究))

  • Colletotrichum acutatum, C. coccodes, C. dematium and C. gloeosporioides were detected in seed samples of red pepper (Capsicum annuum). C. dematium and C. gloeosporioides were the predominant species, maximum seed infection of the species in some samples were 84% and 28%, respectively. C. acutatum and C. coccodes were recorded only in low percentages of 1-2. The blotter method proved more suitable for detecting Colletotricum spp. than the deep freezing blotter or agar plate methods. Plating of seed components showed that C. dematium and C. gloeosporioides were recovered more frequently from seed coat, and decreasing amounts of infection were observed in the endosperm and cotyledon. Seed-borne C. dematium and C. gloeosporioides caused seed rot, damping-off, seedling blight and brown discoloration of cotyledon and hypocotyl when infected seeds were sown in agar of test tube or in soil. Inoculation experiments showed that C. acutatum was pathogenic to red fruit of red pepper and C. coccodes was highly pathogenic to red fruit and weakly pathogenic to leaf of the plant. C. dematium was highly pathogenic to leaf and green fruit and C. gloeosporioides was pathogenic to not only leaf but also green and red fruits. Host range of the four seed-borne species of Colletotrichum was also investigated.

Suppressive Effects of Crude Extracts of Bacillus sp. CT16 and Neobacillus sp. JC05 against Egg Hatch of Meloidogyne incognita (근권세균 Bacillus sp. CT16과 Neobacillus sp. JC05의 배양액 추출물에 의한 뿌리혹 선충의 알 부화 억제 효과)

  • Root-knot disease caused by Meloidogyne incognita is major soil pathogen and cause severe economic damages to vegetable crops. In this study, we selected rhizobacteria for biocontrol of the root-knot nematode, M. incognita, and identified; performed bioassay of the bacterial extracts in cucumber seedlings. The crude extracts of strains CT16 and JC05 out of 180 strains inhibited egg hatching and increased juvenile mortality in vitro assay; based on 16S rRNA sequences analysis, the two strains were identified as Bacillus sp. CT16, and Neobacillus sp. JC05. After extracting the bacterial supernatants by using various organic solvents, n-butanol and n-hexane extracts of strain CT16 and n-butanol extract of strain JC05 showed inhibitory activity of egg hatching depending on concentrations. Subsequently, n-butanol extracts of two strains significantly suppressed formation of egg masses in cucumber seedling. Therefore, these results indicated that strains CT16 and JC05 could be used as potential biocontrol agents against M. incognita.

Monitoring of Particulate Matter Concentration for Forage Crop Cultivation during Winter Season in Saemangeum (새만금 내 동계 사료작물 재배에 따른 미세먼지 농도 변화 모니터링)

  • The Saemangeum has a dry surface characteristic with a low moisture content ratio due to the saline and silt soil, so the vegetation cover is low compared to other areas. In areas with low vegetation cover, wind erosion has a high probability of scattering dust. If the vegetation cover is increased by cultivating crops that can withstand the Saemangeum reclaimed environment, scattering dust can be reduced by reducing the flow rate at the bottom. Thus, the purpose of this study is to analyze the effect of suppressing the generation of fine dust and scattering dust by cultivating winter forage crops on the Saemangeum reclaimed land. While growing 0.5 ha of barley and 0.5 ha of triticale in Saemangeum reclaimed land, the concentration of fine dust was monitored according to agricultural work and growth stage. Changes in the concentrations of PM-10, PM-2.5, and PM-1.0 were monitored on the leeward, the windward and centering on the crop field. As a result of monitoring, PM-1.0 had little effect on crop cultivation. the concentration of PM-10 and PM-2.5 increased according to tillage and harvesting, and tillage had a higher increasing the concentration of PM-10 and PM-2.5 than that of harvesting. According to the growth stage of crops, the effect of suppressing scattering dust was shown, and the effect of suppressing scattering dust was higher in the heading stage than in the seedling stage. So, it was found that there was an effect of suppressing scattering dust other than the effect of land covering. Through this study, it was possible to know about the generation and suppression effect of scattering dust according to crop cultivation.
