Effect of Phytagel on the Root Growth of Plantlets Redifferentiated from Ginseng(Panax ginseng) Embryogenic Cells (인삼 배발생세포에서 재분화된 유식물체의 뿌리 발육에 미치는 Phytagel의 영향)

  • This study was carried out to find out the optimum hardening condition for ginseng plantlets redifferentiated by tissue culture method. While a lot of root hair were observed on the root of seedling grown on the soil, few root hair were observed on the root of plantlet redifferentiated in vitro. On the medium solidified with $0.1{\%}$ phytagel, root hair was not observed and root weight, root width and laternal root development were also very poor. While raising the phytagel concentration on the media, root hair began to increase and root weight, root width and latemal root development were improved. Vascular tissue of plantlet grown on the medium with $0.1{\%}$ phytagel was very poor, but that of plantlet grown on the medium with $0.8{\%}$ phytagel was very good.

Modified Drum Priming and Exogenous Application of 24-Epibrassinolide (24-EBL) for Enhancing Germination under High Temperature Condition in Lettuce Seeds

  • This study was conducted to investigate the effects of modified drum priming and 24-Epibrassinolide (24-EBL) treatment to improve the seed quality for export. 40, 50 and 60% seed moisture content (SMC) of hydrated seeds were incubated for 16 and 24 h in a container with a relative humidity of 99% at 26 rpm for a modified drum priming treatment. The treated seeds were sown at $20^{\circ}C$ and $30^{\circ}C$ (12/12h, light/dark) with four replications of 25 seeds on pleated paper. The seeds were hydrated with water or 24-EBL solutions of $10^{-7}$, $10^{-8}$ and $10^{-9}M$, respectively. The germination of the modified drum primed seeds (24 h incubation after 60% SMC hydration) improved to 1.6 days mean germination time (MGT) and $46%{\cdot}day^{-1}$ germination rate (GR), while the untreated seeds showed 2.1 days MGT and $28%{\cdot}day^{-1}$ GR. The modified drum priming (60% SMC and 24 h incubation with $10^{-9}M$ 24-EBL) showed improved results in MGT (1.8 days) and GR (55%) at $20^{\circ}C$, whereas untreated seeds showed 2.3 days MGT and 44% GR. Under $30^{\circ}C$, germination of modified drum primed seeds was significantly improved in GP (80%), GR ($31%{\cdot}day^{-1}$), HS (55%) and MGT (3.3 days), however, untreated seeds showed decreased GP (27%), GR ($22%{\cdot}day^{-1}$), HS (55%) and MGT (4.8 days). This study showed that the germination of lettuce seeds is enhanced by 24 h drum incubation with 24-EBL and this method can be used effectively to achieve the benefits of early germination and uniform seedling development. In addition, these treatments circumvent thermo-dormancy of lettuce seed and have a possibility of high-quality and environment-friendly seed processing.

Differentiation of Flower Bud of Angelica gigas NAKAI (참당귀의 화아분화(花芽分化))

  • The most important subject in cultivation of A. gigas NAKAI would be how to decrease the bolting rate. Belcause the decrease and poor of the yields and quality is imevitable in such field where the bolting rate is high. This study was carried out to investigate the time of flower-bud differentiation as a part of improvements of cultivation method. Shoot apex was fixed in Farmer's solution(ETOH 3:Acetic acid 1), thin paraffin ribbon was made, and the tissue was stained by Safranin solution. Differentiation time of flower-bud in A. gigas differed greatly from individuals by the seedling size, generally begun from middle of May to late of June, it was time that two leaves spread. In this results, Photoperiocid types of A. gigas NAKAI were regarded as LI type or LS type of dull-photosensitivity.

  • This paper reports a simple rearing system for Cnaphalocrocis medinalis based on corn seedlings diet. C. medinalis population under this system has been maintained by four stages (egg, young larva-1st to 3rd instar, old larva-4th to prepupa, 3nd adult) at $25{\pm}2^{\circ}C,\;40{\pm}10%$ RH, 3nd 16L:8D photoperiod in a laboratory. We have elaborated a new egg collection method using a polystyrene container $(top\;{\phi}11.3\;{\times}\;bottom\;{\phi}\;{\times}\;H8cm)$ which has made a great contribution in easy collection of eggs and storage of them for long period. Under this system, pupation and adult emergence rate, and pupal weight of C. medinalis population were close to the other reports by com seedling diet, and superior to those by artificial diets. Therefore, this rearing system would be useful in obtaining C. medinalsis population suitable to each specific needs.

  • In the beginning of the germination of Castanea creata seeds, seedlings with identical genetic characteristics from only one seed were produced by longitudinally splitting the radicle and epicotyl in half with a razor blade. The results are summarized as follows: 1. 2-8 genetically identical Castanca crenata seedlings were produced by longitudinally splitting the radicles and epicotyles from one to four in number in half with a razor blade 1-3 times about 7 days apart, while the seeds were germinating. 2. The proportion of success of splitting seedlings was very high (90-100%). When some of the spliting seedlings were withered a sprout appeared again and it was possible the seedling to revive. 3. The production rate, growth and T/R rate of seedlings which were split only once were lower that those of the seedlings which were split several times. 4. The growth and T/R rate of split seedlings which were one of no more than four seedlings produced from one seed showed no difference in comparison with normal seedlings, but in seedlings which were one of more than four, the growth and T/R rate were considerably decreased. 5. With the progress of growth, the external shapes of splitting seedlings completely returned to the original state, and no problems in growth, were found.

Successional Changes in Seed Banks in Abandoned Rice Fields in Gwangneung, Central Korea

  • In order to understand the role of seed banks for restoration, seed banks in abandoned rice fields in the Gwangneung National Arboretum, central Korea were investigated using the seedling emergence method. The study sites represented three stages: an initial stage dominated by forbs such as Persicaria thunbergii and Juncus effuses var. decipiens, a middle stage dominated by Salix, and a late stage dominated by Quercus aliena and Prunus padus (in nearby riparian forest chosen as a reference stand). DCA ordination arranged the stands according to the number of years since abandonment. CCA ordination identified the dominant environmental variables correlated most closely with Axes 1 and 2 as $Mg^{2+}$ (intraset correlation was 0.827) and $K^+$ (intraset correlation was -0.677), respectively. Species richness and diversity decreased from the initial stage (H'=2.61) to the middle (H'=1.79) and late (H'=0.75) stages. A total of 49 species $(/m^2)$ and 18,620 seedlings $(/m^2)$ emerged out of the seed bank samples. The DCA ordination and similarity analysis detected a large discrepancy between the composition of the actual vegetation and the seed bank. We conclude that the contribution of seed bank to restoration is low. However, seed bank may help the recovery of forbs after disturbance. Some of our results are consistent with the tolerance model of succession whereas others follow the trajectory of the facilitation model. More research on succession will be required to understand the underlying mechanisms.

Control of Crisphead Lettuce Damping-off and Bottom Rot by Seed Coating with Alginate and Pseudomonas aeruginosa LY-11

  • Seedling damping-off and bottom rot caused by Rhizoctonia solani are yield limiting diseases of crisphead lettuce. To provide biocontrol measure in the management of the diseases, biocontrol strain Pseudomonas aeruginosa LY-11 was isolated from lettuce rhizosphere and introduced into crisphead lettuce rhizosphere by the seed coating delivery method. Alginate was used as a coating material to generate beads containing $10^6-10^{6.5}$ colony-forming units (CFUs) of viable bacterial cells of LY-11. When seeds germinated from the alginate beads containing the strain LY-11, the bacteria established mostly in plant rhizosphere to maintain at least $10^4$ CFU per gram of plant tissues. Crisphead lettuce seedlings germinated from the entrapped seeds were less affected from damping-off and bottom rot with disease control values of 70.4% and 85.4% respectively. Although P. aeruginosa LY-11 colonized plant rhizosphere and not phyllosphere, the result indicated that bottom rot caused by the foliar inoculation of R. solani was effectively reduced by the rhizobacteria. All data suggested that immobilized rhizobacterial application in seeds by alginate coating could control damping-off and induce induced systemic resistance of crisphead lettuce to reduce bottom rot.

Physiological responses of selected Philippine upland rice genotypes evaluated using drought and salinity stress

  • Screening for drought and salinity tolerance was undertaken for selected Philippine upland rice landraces during germinative and seedling stages to identify varieties which can potentially be grown in marginally dry and saline soils. While increasing PEG and NaCl concentrations caused obvious signs of injury to all rice genotypes, considerable varietal differences were noted in the nature of responses providing evidence that these genotypes possess broad intraspecific genetic variations for drought and salt tolerance. Inconsistent responses of these varieties during both growth stages highlight complexities involved in stress responses and underscore the futility of utilizing a single stage in the rice plant's life cycle for physiological screening. Notwithstanding these perplexing responses, G_Katiil and Ml-Pilit Tapul were observed to thrive relatively well despite increased salt and drought stress during early growth stages and may therefore possess genes needed in crop improvement efforts for drought and salinity tolerance. While these results do not reflect the entire spectrum of adaptive expression to drought and salinity stress during the life cycle of the upland rice plant, they nonetheless provide an easy, reliable and reproducible method for preliminary identification of drought and salt tolerant rice varieties.

Estimates of Early Selection Efficiency for Height Growth Using Age-Age Correlation in Pinus koraiensis (수령간 상관을 이용한 잣나무 수고 생장의 조기선발 효율 추정)

  • Korean white pine(Pinus koraiensis S.et Z.) is one of the important silvicultural species due to its quality timber in addition to edible pine - nut production. Selective breeding method is under progress to improve growth and nut production for this species. This study was carried out to obtain genetic information necessary for establishing advanced generation seed orchard, such as genetic parameters and efficiency of early selection. Open - pollinated cones were collected from 50 plus trees. In the spring of 1986, test plantations were established at 3 different sites with 4 - year - old seedlings. Seedling height was measured from age 5 to 10 in plantations. Genetic and phenotypic correlations based on the log of the ratio of two ages (LAK) were expressed as simple regression with statistical significance($r^2=0.7862$ and 0.8256, respectively). If 20 years is presumed to mature, the most efficient age for early selection is 3 years after planting with efficiency of 1.96.

Seed-borne Infection of Anthracnose Fungi Isolated from Diseased Red Pepper (병든 고추 종자에서 분리된 탄저병균의 종자전염)

  • Colletotrichum dematium, C. gloeosporioides and Glomerella cingulata were detected in seed samples collected from diseased red pepper (Capsicum annuum) using blotter method. C. gloeosporioides was the predominant species in seed samples tested and followed by C. dematium and G. cingulata. When the seed components were plated C. dematium, C. gloeosporioides and G. cingulata were detected from seed coat, endosperm and cotyledon. The three anthracnose fungi were recorded more frequently from seed coat than that of observed in the endosperm and cotyledon. Seed infection with C. dematium, C. gloeosporioides and G. cingulata caused seed rotting, damping off and seedling blight of red pepper plants. According to the inoculation experiments, it was shown that C. gloeosporioides was the most virulent among three species. C. dematium showed weak virulence when the plants were wounded, and G. cingulata was wound parasite or weakly virulent on red fruits. Benlate T (benomyl+thiram) and Homai (thiophnate-methyl+thiram) were effective to anthracnose fungi when treated to infected seeds.
