대전지역에서 호밀과 보리의 파종비율이 건물수량 및 사료가치에 미치는 영향 (Effect of Seed Blending Rates Between Rye and Barley on Forage Production and Quality in Daejon Area)

  • 본 시험은 호밀과 보리의 혼파에 의해 수량성과 품질이 높은 조사료 생산방법을 강구하고자 2004년 9월부터 2007년 6월까지 충남대학교 생명과학대학 내 부속 초지시험포장에서 수행하였다. 시험 재료는 청예용 호밀인 Koolgrazer와 대면보리였으며, 시험구 배치는 호밀 100%, 호밀 60+보리 40%, 호밀 50+보리 50% 및 호밀 40+보리 60% 등 호밀과 보리의 혼파비율을 4수준으로 하는 난괴법 3반복으로 시험하였으며 얻어진 결과는 다음과 같다. 3년 평균 ha당 건물수량은 호밀 100%구가 9,282 kg으로 가장 높았고 보리의 파종비율이 높아질수록 건물수량은 반대로 감소하는 경향이 뚜렷하였다(p<0.05). 식생비율은 보리의 파종비율이 각각 40%, 50% 및 60%로 높아짐에 따라 보리의 식생비율은 각각 30%, 41% 및 47%로 높아졌으나 파종비율 대비 보리의 식생비율은 그리 높지 못한 결과를 가져왔다. 조단백질 함량과 건물소화율은 호밀 100%구에 비하여 보리의 파종비율이 높아질수록 높아졌던 반면에 NDF, ADF, cellulose 및 liglin 함량은 낮아지는 결과를 보였다(p<0.05). ha당 조단백질 수량과 가소화건물 수량은 호밀 100%구에 비하여 보리를 혼파한 모든 처리 구에서 낮은 결과를 나타내었다(p<0.05). 따라서 대전지역에서 호밀과 보리를 혼파하여 이용할 경우, 청예용 보리의 다수확품종 보급이 선행되지 않고는 건물수량과 CPDM 및 DDM수량을 높이는 데는 어려움이 클 것으로 사료된다.

  • 본시험은 Canola 전지종실과 Canola 박 및 Canola 유의 유효에너지가(AM 및 TME)와 아미노산 이용을(TAAA)를 측정하기 위하여 실시하였다. 본시험에는 단관백색레그혼종(S.C.W.C.) 성계수탉이 실시되었으며, AME시험에서는 측정사료인Canola 전지증실, Canola 박 Canola 박+유(60:40)을 30% 그리고 Canola 유를 10% 배합한 4종의 시험사료와 기초사료를 각각 자유채식시켰고 예비시험기간은 3일 채분기간은 4일이었다. TME와 TAAA시험에서는 Canola 전지종실과 Canola 박은 단-사료로, Canola 박+유와 Canola 유는 각각 기초사료에 30%와 10%의 비율로 혼합하여 각각 1일 30g을 24시간의 절식기간 후에 강제투여(force feeding)하였고 48시간동안 전분을 채취하였다. 모든 에너지가는 질소정정 에너지가로 환산하였는데, Canola 전지종실, Canola 박, Canola 유 및 Canola +유의 AMEn가는 각각 4,485, 1,984. 8,275 및 5,655 kcal/kg이었고 TMEn가는 각각 4,577, 2,103, 8,487 및 5,630kcal/kg으로 측정되어 Canola 박+유는 Canola 전지종실보다 높은 에너지가를 나타냈으며(P<0.01), 전반적으로 Canola전지종실은 Canola 박보다 높은 TAAA가를 나타냈다.

중부산간 초지에서 초종 및 혼파조합이 목초 수량, 식생 구성 및 사료가치에 미치는 영향 (Effect of Species and Seed Mixture on Productivity, Botanical Composition and Forage Quality in Middle Mountainous Pasture)

  • 본 시험은 중부 산간지(충북 괴산, 해발 520 m)에서 초종 및 혼파조합에 따른 신규조성 초지의 생산성 변화를 평가하기 위하여 수행하였다. 총 7개의 처리(T1 : 오차드그라스, T2 : 티머시, T3 : 톨페스큐, T4 : 페레니얼라이그라스, T5 : 켄터키블루그라스, T6 : 티머시, 12 + 오차드그라스, 10 + 톨페스큐, 10 + 알팔파, 5, 및 T7 : 티머시, 15 + 오차드그라스, 12 + 켄터키블루그라스, 5 + 화이트클로버, 5)를 두고 2014년 가을에 조성하여 2015년 1년동안 수량 및 생육특성을 조사하였다. 초장은 오차드그라스와 페레니얼라이그라스에서 컸으며 건물함량은 켄터키블루그라스에서 높게 나타났다. 식생은 1차 수확시 잡관목의 비율이 높았으나 2차 및 3차에서는 목초의 비율이 높았다. 또한 페레니얼라이그라스는 2차 및 3차 수확시 잡초의 비율이 가장 높게 나타났다. 생초 및 건물수량은 오차드그라스와 페레니얼라이그라스 단파 초지에서 높게 나타났으며(p<0.05) 켄터키블루그라스에서 가장 낮았다. 혼파초지는 톨페스큐 위주 초지가 켄터키블루그라스 위주 혼파초지보다 더 높았으나 통계적 유의성은 없었다(p<0.05). 조단백질 함량은 2차 및 3차 수확시는 16.91 및 14.79%로 높았으며, 2차 수확시 티머시, 톨페스큐, 페레니얼라이그라스는 17.46~18.81%의 높은 조단백질 함량을 보여주었다. 3차 수확시 켄터키블루그라스는 17.57%로 가장 높았다. In vitro 건물 소화율은 1차에서는 낮았으나 2차 및 3차에서는 비슷하게 높은 수준을 유지하였고 켄터키블루그라스는 매 수확시기에서 가장 낮은 IVDMD 함량을 보여주었다. TDN 함량은 3차 수확시 가장 높았으며 1차 수확에서 낮게 나타났다. 전체적으로 사료가치에 있어서 3차 수확시 가장 우수한 것으로 나타났으며 1차 수확시 가장 낮았다. 이상의 결과를 종합하여 볼 때 생산성 면에서는 오차드그라스 및 페레니얼라이그라스 단파 초지 그리고 T6 혼파조합이 우수하였고 사료가치적 측면에서는 티머시 및 페레니얼라이그라스 단파 그리고 T7 혼파조합이 우수한 것으로 나타났다.

  • Currently, there are researches about environment-friendly road construction plans led by the Ministry of Construction and Transportation in progress. Therefore, in order to create the revegetation techniques of thin layer-soil combination media hydroseeding measures by actively using native herbs and native woody plants instead of using imported foreign grasses as a concept of sustainable environment-friendly land development, this thesis is going to identify problems that can appear when applying the thin- layer-soil-media hydroseeding measures by the suggested in the "Slope revegetation design and guidelines" proposed by the Ministry of Construction and Transportation, and to propose improvement plans for the problems.To this aim, a seeding mixture selection test was conducted by the goal of slope restoration, and a test group for artificial slope was created. As for a test for June sowing, it was intended to identify appropriate combination quantity by conducting a test that differentiates the combination quantity, and as for a test for September sowing, an artificial slope test was conducted by creating an artificial bank for earth and soil and applying 1~2cm and 3~4cm thickness after differentiating the seed combination volume and slope aspects.

Optimization of Culture Media for Solid-state Culture of Pleurotus ferulae

  • In order to elucidate the possibility of artificial production of p. ferulae by solid-state culture, the optimization of culture conditions was carried out. When $NH_4H_2PO_4$ and $CaCO_3$ were used in the cultures using test tube with 30 g of Populus sawdust at $25^{\circ}C{\pm}1$ in the dark, the favored mycelial growth was observed with $1\%$ of $NH_4H_2PO_4$ and the production of polysaccharide was 7.85 mg/100 mg of mycelium with $1\%$ of $CaCO_3$. The mixtures of $80\%$ of Populus Sawdust and $20\%$ of rice bran at $60\%$ of water content were determined to be optimal for the production of fruiting bodies in the sawdust culture. When three treatments containing various ratios of garlic powder were conducted, yields of fruiting bodies were drasti[ally higher than those of Synthetic mixture without garlic powder The highest yield (143 g/bag) was obtained with $7\%$ garlic powder. The yield of synthetic mixture containing $7\%$ of garlic powder was $83\%$ higher than that of Sawdust culture. The reason why garlic powder did support growth was not clear but it is possible that garlic powder might contain effective components for the formation of fruiting body. The optimal synthetic mixture composition consisted of cotton seed $77\%$, lime $6.4\%,\;K_2HPO_4\;0.2\%,\;KH_2PO_4\;0.2\%,\;CaHPO_4\;0.2\%$, corn flour $4\%$, wheat flour $5\%$, and garlic pow-der $7\%$.

Object Detection Based on Deep Learning Model for Two Stage Tracking with Pest Behavior Patterns in Soybean (Glycine max (L.) Merr.)

  • Soybean (Glycine max (L.) Merr.) is a representative food resource. To preserve the integrity of soybean, it is necessary to protect soybean yield and seed quality from threats of various pests and diseases. Riptortus pedestris is a well-known insect pest that causes the greatest loss of soybean yield in South Korea. This pest not only directly reduces yields but also causes disorders and diseases in plant growth. Unfortunately, no resistant soybean resources have been reported. Therefore, it is necessary to identify the distribution and movement of Riptortus pedestris at an early stage to reduce the damage caused by insect pests. Conventionally, the human eye has performed the diagnosis of agronomic traits related to pest outbreaks. However, due to human vision's subjectivity and impermanence, it is time-consuming, requires the assistance of specialists, and is labor-intensive. Therefore, the responses and behavior patterns of Riptortus pedestris to the scent of mixture R were visualized with a 3D model through the perspective of artificial intelligence. The movement patterns of Riptortus pedestris was analyzed by using time-series image data. In addition, classification was performed through visual analysis based on a deep learning model. In the object tracking, implemented using the YOLO series model, the path of the movement of pests shows a negative reaction to a mixture Rina video scene. As a result of 3D modeling using the x, y, and z-axis of the tracked objects, 80% of the subjects showed behavioral patterns consistent with the treatment of mixture R. In addition, these studies are being conducted in the soybean field and it will be possible to preserve the yield of soybeans through the application of a pest control platform to the early stage of soybeans.

Leucaena Seeds as Protein Supplement in the Rations of Growing Sheep

  • The study was carried out to evaluate leucaena seeds as a protein replacement of mustard seed cake (MSC) in the concentrate mixture of growing lambs. Fifteen owing male lambs (Local${\times}$Corridale) with an average body weight of 16.3 kg were allocated into three dietary treatments (T1, T2, and T3) with five animals in each group. Animals were offered dry mixed grass, berseem hay and concentrate mixture to meet their nutrient requirements. In concentrate mixture of T1, (Control) MSC was used as protein source, while in T2 and T3 groups, 25 and 50% of MSC was replaced by leucaena leucocephala seeds. On completion of three months (90 days) of feeding, a digestion cum-metabolism trial was conducted to determine DMI, nutrient utilization, and nitrogen balance. Changes in body weight were recorded at 15 day internals and eating patterns were recorded for 3 consecutive days at the end of the feeding trial. MSC had higher CP contents than leucaena seeds (27.0%). Mimosine contents in leucaena seeds were 1.1 compared to 0.2 and 0.4% in concentrate mixture of T2 and T3 group, respectively. Dry matter intake varied non-significantly ($79.3{\pm}1.2$ to $83.4{\pm}1.3g/kg$ $w^{0.75}$) across the dietary treatments. Digestibility of DM and cell wall polysaccharides (NDF, ADF. Cellulose and hemicellulose) were comparable, however CP digestibility was relatively lower in leucaena luecocephala seeds based groups (T2 $45.5{\pm}1.7$ and T3 $46.7{\pm}3.5$) compared to MSC supplemented group (T1 $47.7{\pm}0.9%$). The growth rate of lambs was non-significantly higher in T1 ($79.2{\pm}5.4$) compared to T2 ($73.8{\pm}8.8$) and T3 ($73.9{\pm}7.0$), respectively. The animals were in positive nitrogen balance and N-balance varied from 1.8 to 2.9 g/d across treatment groups. The eating rate (% of total offered) of concentrate up-to 15 min was relatively higher in T1 (82.4) than T2 (74.2) and T3 (77.8%). However no effect of leucaena seeds was recorded on total DMI of animals. The results of the study revealed that the inclusion of up to 50% leucaena seeds, as protein source in concentrate mixture of lambs had no adverse effect on DMI, nutrient utilization, eating patterns, nitrogen balance and growth performance of lambs.

EFFECT OF LOW DEGRADABLE DIETARY PROTEINS ON HEPATIC METABOLISM OF EARLY LACTATING BUFFALOES

  • Role of low degradable protein in milk production of early lactating Murrah buffaloes has been studied in relation to energy status of test animals. Replacement of conventional concentrate mixture with low degradable cotton seed cake resulted in appreciable changes in circulatory transaminases and phosphatase levels. The enzymes viz. glutamate oxaloacetate and glutamate pyruvate transaminase and alkaline phosphatases increased with feeding of said cake indicating stress on hepatic tissue. Animals seemed to overcome stress by feeding enhanced levels of same protein along with improved feed intake, body weight and milk production.

  • $eta$-Sialon synthesis was investigated via the simultaneous reduction and nitridation of Hadong Pink Kaolin using the graphite as a reducing agent at 135$0^{\circ}C$ under 80% $N_2-20%H_2$ atmosphere. When Hadong Pink Kaolin-graphite-silicon nitride seed(molar ratio ; $SiO_2:C:Si_3N_4$=1;3.5:0.05) mixture was heated at 135$0^{\circ}C$ for as long as 20h in 80%$N_2-20%H_2$ atmosphere a homogeneous $eta$-Sialon$(Si_{3.5}Al_{2.5}O_{2.5}N_{2.5})$ was mainly formed together with a small amount of $\alpha$-$Si_3N_4$.

물오리나무(Alnus hirsuta)의 칼루스 유도 및 자엽유래 칼루스로부터의 원형질체 배양 (Induction of Callus and Culture of Protoplasts from Cotyledon-Originated Callus in Alnus hirsuta)

  • Callus-inducing ability of Alnus hirsuta was examined by culturing various tissues (leaf, hypocotyl, cotyledon and seed) on NT (Nagata & Takebe) medium, supplemented with 2.5$\mu$M 2,4-D. Leaf-originated callus was cultured on media varying in auxin (IBA and NAA) and cytokinin (BAP) concentrations to examine the effects of auxin and cytokinin on callus growth. Maximum growth was obtained at 10 $\mu$M IBA+10$\mu$M BAP and 10$\mu$M NAA without cytokinin. Cell suspensions established from cotyledon-originated callus yielded viable protoplasts after incubation for 16-18 hours in an enzyme mixture (1% (w/v) Onozuka R-10 0.5% (w/v) Macerozyme, CPW salts and 13% (w/v) mannitol, pH 5.8). Protoplasts were cultured on NT medium, supplemented with glucose, hormones and coconut milk. After 6 weeks of culture, protoplasts sustained cell divisions to form microcallus, which showed various colors from red to white.
