Variation of Anthocyanin and Protein Contents in Glycine max L. (Merr) (Soybean) Germplasms from Korea

  • Soybean (Glycine max L. (Merr) is commonly consumed and found in major foods including soymilk, soy sauce, tofu, and soy sprout in Korea and east Asia. In addition, it is common to cook the whole seeds with rice. Soybean is known to have ranges of health benefits including antiaging, anticancer, neuroprotective and antidiabetic taken either as supplement or dietary food. Anthocyanins and flavonoids in G. max are found to be the main contributors to such wide arrays of health benefits. Due to increasing economic values of soybean, development of specialty soybean cultivars is becoming an area of interest worldwide. In this study, 746 black soybean accessions from National Agrobiodiversity Center were characterized as part of an attempt to identify important germplasms of G. max. Seed coats of each accession were analyzed for their total anthocyanin, cyanidin 3-O-Glucoside (C-3-O-G), delphinidin 3-O-glucoside (D-3-O-G), petunidin-3-O-glucoside (Pt-3-O-G), and their whole seeds for crude protein contents. HPLC was used to determine and quantify the anthocyanin compositions while crude protein was determined using Kjeldahl method by Kjeltec auto-analyzer (Kjeltec 8400, Foss, Sweden). Accessions were grouped according to their anthocyanins and protein contents; the mean content of which were correlated to agronomic traits including maturity date, one hundred seed weight, cotyledon color and seed lust color. The results indicated that the total anthocyanin content (TAC) ranged from 273.77 to 6250.52 mg/100 g, with mean value of 1853.03 mg/100 g while the crude protein content (CPC) being between 33.43 and 47.51%, with mean value of 40.81%. The highest number of accessions (45.97%) showed TAC between 1000~1900 mg/100 g while 30.96% of accessions showed CPC between 41~43%. Among the 746 accessions considered, 11 (IT142935, 175818, 175855, 177191, 177209, 177211, 177214, 177216, 177218, 177220, 177274) of them showed TAC above 4000 mg/100 g. C-3-O-G was found to be the major contributor to TAC showing strong correlation. Accessions with green cotyledon color showed high mean TAC compared to those having yellow cotyledon color, and accessions with dull seed lust color showed high mean TAC than those having shiny seed lust color. One hundred seeds weight and maturity date showed positive correlation with all anthocyanin contents, except for Pt-3-O-G in the latter case. The overall result of the present study could be used as background for developing new black soybean cultivars and breeds with high anthocyanin and protein contents. The result depicted that many of the accessions could be used as potential parental lines.

Accumulation of triple recessive alleles for three antinutritional proteins in soybean with black seed coat and green cotyledon

  • The black seed coat of soybeans contain anthocyanins which promote health. However, mature soybean seeds contain anti-nutritional factors like lipoxygenase, lectin and Kunitz Trypsin Inhibitor (KTI) proteins. Furthermore, these seeds can be used only after the genetic elimination of these proteins. Therefore, the objective of this study was to develop novel soybean genotypes with black seed coat and triple recessive alleles (lx1lx1lx2lx2lx3lx3, titilele) for lipoxygenase, lectin, and KTI proteins. From a cross of parent1 (lx1lx2lx3/lx1lx2lx3, ti/ti, Le/Le) and parent2 (lx1lx2lx3/lx1lx2lx3, Ti/Ti, le/le), 132 F2 seeds were obtained. A 3:1 segregation ratio was observed during F2 seed generation for the inheritance of lectin and KTI proteins. Between a cross of the Le and Ti genes, the observed independent inheritance ratio in the F2 seed generation was 9: 3 : 3 : 1 (69 Le_Ti_: 32 leleTi_: 22 Le_titi: 9 leletiti) (χ2=2.87, P=0.5 - 0.1). From nine F2 seeds with triple recessive alleles (lx1lx1lx2lx2lx3lx3, titilele genotype), one novel strain posessing black seed coat, and free of lipoxygenase, lectin and KTI proteins, was selected. The seed coat color of the new strain was black and the cotyledon color of the mature seed was green. The weight of 100 seeds belonging to the new strain was 35.4 g. This black soybean strain with lx1lx1lx2lx2lx3lx3, titilele genotype is a novel strain free of lipoxygenase, lectin, and KTI proteins.

시설재배지 토양중 잔류농약 경감을 위한 잎들깨 종자코팅제 선발 (Selection of coating materials to leafy perilla seed for reducing endosulfan residue in greenhouse soil)

  • 잎들깨 종자의 파종 밀도 조절을 위해 종자코팅 용도로 사용된 엔도설판 분제에 의한 시설재배지 토양중 endosulfan의 잔류량을 경감시키기 위하여 이를 대체할 수 있는 종자코팅재료의 선발 및 이들이 미치는 발아영향을 조사하였다. 코팅재료는 talc, kaoline, 패화석등 석회류가 가능하였다. 코팅재료의 종자부착을 위한 접착제로는 식용유, 전착제, 알코올, 증류수 순이었으나 종자 발아영향은 반대경향을 나타냈다. 코팅재료별 종자부착성은 패화석, 생석회, 석회, talc, kaoline이 양호하였으나 토양과의 색깔 구별성은 패화석등 석회류에 비해 talc, kaoline이 좋았다. 선발 코팅제를 종자에 부착시 잎들깨의 발아율은 talc, kaoline이 양호하였고 토양과의 색깔 구별성도 양호하였다.

홍화 재배유형 및 수확시기가 종실 수량 및 품질에 미치는 영향 (Effect of Cultivation Methods and Harvesting Time on Yield and Quality of Safflower(Carthamus tinctorius L.) Seeds)

  • 홍화의 노지및 하우스재배의 생육과 수량 및 수확시기별 종실의 품질에 대한 시험결과를 요약하면 다음과 같다. 하우스비가림재배가 노지재배에 비해 출현기는 3일, 개화기는 8일이 빨랐으며, 생육도 양호하여 10g당 종실수량도 노지재배 보다 $25\%$ 증수되어 노지재배 보다 비가림재배가 유리하였다. 조지방함량은 비가림하우스재배가 노지재배보다 높았고 종자의 색택도 비가림하우스재배에서 변색없이 흰색이었으나 노지재배에서는 변색되어 품질이 나빴다. 노지재배의 조지방 함량은 수확기까지 높아졌으며 개화성기 20일 이후 수확에서는 차이가 없었으나, 종실의 색깔은 개화성기 30일 이후 점차 변하여 개화성기 후 40일 수확은 흑갈색으로 종실품질이 크게 떨어져 종실의 수확시기는 개화성기 후 20일에서 30일 사이가 가장 좋았다.

쓴메밀(Fagopyrum tataricum) 유전자원의 루틴 함량 비교 (Comparison in Rutin Content of Tartary Buckwheat(Fagopyrum tataricum))

  • 아시아 지역에서 수집한 야생 쓴메밀의 종자 및 식물체 내의 루틴함량을 조사하였다. 쓴메밀은 종자 및 식물 전체부위에서 숙근메밀이나 단메밀보다 높은 루틴 함량을 나타냈다. 특히, 꽃은 숙근메밀의 2.2배, 단메밀의 9.5배, 줄기는 각각 3.1배와 24.9배, 종자는 3.2와 65배나 높았다. 3종에 대한 식물체 부위별 루틴함량은 모두 꽃>잎>종자>줄기>뿌리의 순으로 높았다. 종피색에 따른 루틴함량은 암회색>검은색>갈색, 회갈색 순의 경향을 나타냈으며, 종자의 형태는 Slender>Notched>Round 순으로 루틴함량이 높았다. 재배종은 야생종에 비해 루틴함량이 잎은 1.3배, 줄기가 1.4배, 종자가 1.2배의 높은 경향을 보였다. 부탄지역 수집품종은 잎, 줄기, 종자의 루틴함량이 다른 지역보다 높은 경향을 보였고, 슬로베니아 및 파키스탄지역 수집품종은 잎, 줄기에서, 인도 및 네팔지역 수집품종은 종자에서 가장 낮은 루틴함량의 경향을 보였다.

정수장 원생동물 분석방법의 국내 실험실간 교차시험 연구 (Interlaboratory Study for Cryptosporidium and Giardia Test Methods in Water)

  • This interlaboratory study was designed to evaluate protozoan test methods in water and to predict the major causes of deviation of the methods. Each of four laboratories with previous experience of protozoa analysis in water participated, and met the initial performance criteria of EPA 1623 method provided. The protozoan analysis procedure consists of filtrations, concentration, immunomagnetic separation, dyeing (staining) and counting with observation. We tested three different filtration equipments: capsule filter for 10 L of surface water, and high volume (HV) capsule filter and membrane filter for 100 L of finished water. When the recovery of each step of the procedure was evaluated with EasySeed, the commercial stock of each 100 Cryptosporidium and Giardia, immunomagnetic separation and filtration step were the most crucial steps affecting the stability of the recovery, especially for Cryptosporidium. There was no significant difference of recovery among the filtration methods. Recovery of protozoa from source water were evaluated with spiked EasySeed as matrix tests. The recoveries of Giardia increased significantly in the matrix tests compared those in the deionized water. We also applied red stained mixture stocks of Cryptosporidium and Giardia called ColorSeed as internal standards of water sample tests. The recoveries of both EasySeed and ColorSeed in samples tested were within the range of the criteria, however, the Giardia recoveries using ColorSeed decreased significantly. Further optimization study with ColorSeed will be necessary, considering the convenience of using the internal standard without additional sample analysis. The significant factors of the recovery variation were identified as the differences of laboratories as well as water quality and type of the stock for spiking. The results of this study emphasize the importance of the quality assurance program for protozoan analysis lab in water.

결명자 에탄올 추출물 및 식이섬유의 이화학적 특성 (Physicochemical Properties of Ethanol Extracts and Dietary Fiber from Cassia tora L. Seed)

  • This study was conducted to investigate the physicochemical properties of the ethanol extracts and soluble dietary fiber from Cassia tora L. seed. The proximate composition of Cassia tora, soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract, molecular mass distribution, sugar contents and viscosity of soluble fiber from Cassia tora were analyzed. Cassia tora contains 12.6% of moisture, 5.2% of ash, 13.4% of crude protein, 7.2% of crude fat, 8.8% of insoluble fiber and 48.3% of soluble fiber. The effects of extract condition on soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract were investigated. The soluble solid contents were higher in 70% or 50% ethanol extracts than those in 100% ethanol extracts and showed highest value in grind sample extracts. In Hunter's color value, 100% ethanol extracts and whole Cassia tora sample extracts were higher in L and b value, but on the contrary, were lower in a value, than those of the other. The highest emodin and rhein contents were observed in 70% and 50% ethanol extracts, respectively, and showed higher value in room temperature extracts than in heating extracts. The molecular mass of soluble fiber from Cassia tora seed was estimated by gel filtration chromatography. Most soluble fiber(80%) exhibited a molecular mass range of between 50~2000 kDa. The major sugars of soluble fiber from Cassia tora seed were identified as xylose, mannose and galactose. The apparent viscosity of 0.5% soluble fiber from Cassia tora seed was 33 mPas showing a higher value than pectin or xanthan gum.

들깨의 발아가 들깨지방질의 특성에 미치는 영향 (Effects of Seed Germination on Characteristics of Perilla Seed Lipids)

  • Color, lipid and fatty acid composition, and tocopherols and polyphenols contents of perilla seed lipids in response to seed germination were studied. Perilla seeds were germinated at $30^{\circ}C$ in the dark for 12, 36, or 48 h, after which total lipids were extracted by the Folch method using chloroform and methanol (2:1, v/v). Seed germination resulted in a decrease in yellowness and greenness in perilla seed lipids, but there were no significant changes in composition of the lipids including major neutral lipids (>90%). Contents of phosphatidylcholine and phosphatidylethanolamine in the perilla seed lipids significantly increased in response to germination. Linolenic acid (>63%) was the most abundant fatty acid. Seed germination tended to decrease the relative content of linolenic acid and increase the contents of oleic and stearic acids. Contents of antioxidants, especially ${\alpha}$-tocopherol and polyphenols, increased in response to seed germination. As the germination period was extended, the antioxidant content increased. Therefore, increases in useful components, phosphatidylcholine, phosphatidylethanolamine, ${\alpha}$-tocopherol, and polyphenols contents by seed germination can contribute to the improvement of perilla seed utilization in food industry.

고추씨박 급여가 육계의 발육 및 복강지방 축적에 미치는 영향 (Effects of Red Pepper Seed Oil Meal Supplementation on Performance and Abdominal Fat Accumulation in Broilers)

  • This experiment was conducted to determine the effect of dietary red pepper seed oil meal (RPSOM) on the performance and the fat pad content in broiler chicks. feeding trial was conducted with 360 birds broiler chicks for 8 weeks. The levels of dietary RPSOM were 0, 5, 10 and 15% and mixed in diet consisting of corn and soybean meal. Body weight, feed intake, feed conversion and mixed in diet consisting of corn and soybean meal. Body weight, feed intake, feed conversion and viability were not significantly different among treatments. The percentage of abdominal fat and skin color were not significantly differents among treatments. The results of the feeding trial show that RPSOM can be used within 10% in broiler diet.

종자 혼입율과 입도를 달리한 고추가루의 저장 중 색과 매운맛 성분의 변화 (Changes of Colors and Pungent Principles of Red Pepper Powder with Different Seed Contents and Particle Sizes During Storage)

  • Red pepper powders with variuos seed contents (0%, 10%, 20%) and particle sizes were stored at 0$^{\circ}C$, 20$^{\circ}C$, and 30$^{\circ}C$ for 90 days, and the changes in their characteristics were monitored. The initial moisture content was about 13%, and the powders were packed in 0.1 mm-thick polypropylene (PP) bags. Moisture content of each power remained constantly, so PP bags appeared to be effective to prevent moisture transmission. All the values of L*, a* and b* were higher in fine particles than in coarse particles. Storage at 0$^{\circ}C$ did not change any color values, however, overall color values devreased apparently when stored at 30$^{\circ}C$ for 90 days. Capsanthin contents decreased gradually at all storage temperatures and more rapidly at higher temperature. The contents of capsaicinoids in fine particles (49∼59 mg%) were twic as much as those in coarse particles (15∼36 mg%), and they did not change throughout the storage.
