김스낵 제조시 도라지 첨가량에 따른 반죽액의 품질 특성 (Quality Characteristics of Dough Liquid according to the Addition Ratio of Doraji in Seaweed Snack Manufacturing)

  • This study was examined the quality characteristics of dough liquid according to the additional ratio of Doraji in seaweed snack manufacturing. Firstly, the results of Doraji type (dry & powder) were as follows: Carbohydrates 77.57~79.29, crude protein 9.10-9.25, crude fat 0.96~1.33 and calories 355~366 kcal, pH 5.42~5.45, sugar $3.53{\sim}3.96^{\circ}brix$, color 33.82~44.25 (L), 2.27~3.52 (B) and total free amino acids 2,200~2,699 mg/100 g. Total polyphenol contents had dry extracts 1,931.18 mg% and powder extract 1,382.43 mg%, DPPH and ABTs radical scavenging activities tended to increase with higher treatment concentration. Next, the results showed that dough liquid for seaweed snack manufacture which was added Doraji were as follows: Color became deep poppy red with increased addition of Doraji. The texture of adhesiveness, cohesiveness, chewiness, and brittleness tended to decrease with addition of Doraji. The springness showed the opposite tendency. Accordingly, these results suggest that 20% of dry Doraji extract is a proper proportion so that it can be added to the rice dough liquid to produce form Doraji (dry and powder) containing seaweed snacks.

Growth Enhancement of the Microalga Tetraselmis suecica by an Extract of the Green Alga Monostroma nitidum

  • Cell growth of the marine microalga Tetraselmis suecica was regulated by the addition of seaweed extracts in its culture medium. Of 26 species of seaweed tested, water-soluble extracts from Monostroma nitidum and Pachymeniopsis elliptica and methanol-soluble extract from Sargassum confilsum enhanced the growth of T suecica cells. The water extract of M. nitidum was the most effective, producing up to a 2-fold increase in cell density with the addition of 1 mg/mL of extract to the culture medium. Cell size, gross biochemical composition, fatty acids, and digestion efficiency all differed marginally between cultures of T suecica grown with and without the M. nitidum extract.

Measurement of Antioxidant Activities and Phenolic and Flavonoid Contents of the Brown Seaweed Sargassum horneri: Comparison of Supercritical CO2 and Various Solvent Extractions

  • Seaweed Sargassum horneri extracts were obtained using supercritical carbon dioxide ($SC-CO_2$) and different solvents. $SC-CO_2$ was kept at a temperature of $45^{\circ}C$ and pressure of 250 bar. The flow rate of $CO_2$ (27 g/min) was constant during the entire 2-h extraction period, and ethanol was used as a cosolvent. Six different solvents [acetone, hexane, methanol, ethanol, acetone mix methanol (7:3), and hexane mix ethanol (9:1)] were used for extraction and agitated by magnetic stirring (250 rpm) in the dark at $25^{\circ}C$ for 20 h; the ratio of material to solvent was 1:10 (w/v). Antioxidant properties of S. horneri extracted using $SC-CO_2$ with ethanol and different solvents have shown good activity. The highest activity belongs to $SC-CO_2$ with ethanol extracted oil, showing DPPH, ABTS, total phenolic content, and total flavonoid levels of $68.38{\pm}1.21%$, $83.51{\pm}1.25%$, $0.64{\pm}0.02mg/g$, and $5.57{\pm}0.05mg/g$, respectively. The S. horneri extracts showed a significant correlation between the antioxidant activity and phenolic content. Based on these results, the $SC-CO_2$ extract (ethanol) of the seaweed extract from brown seaweed may be a valuable antioxidant source.

다양한 해조류를 첨가하여 제조한 밥의 in vitro 항산화 활성 (In vitro Antioxidant Activity of Cooked Rice Containing Various Seaweeds)

  • An emerging interest in healthy eating has led to an increase in the consumption of rice mixed with various types of grains. Cooked rice was prepared with five different seaweeds, Hizikia fusiformis, Sargassum fulvellum, Enteromorpha compressa, Undaria pinnatifida, and Gracilaria verrucosa, and the antioxidant activity was measured. In addition, the antioxidant activities of 80% ethanol and methanol extracts of the five seaweeds were compared. Total phenolic content (TPC), total reducing power (TRP), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) cation radical scavenging activities of the ethanol extracts were higher than those of the methanol extracts. The TPC of raw seaweed ethanol extracts was from 7.58 to 26.27 mg gallic acid equivalent (GAE)/g. The antioxidant activities of both extracts of Hizikia fusiformis were the highest among the five seaweeds, and the antioxidant activities of the cooked rice were lower than those of the raw seaweed extracts. The DPPH radical scavenging activities of cooked white rice, mixed grains, barley, and a mixture of white rice and barely added with Hizikia fusiformis, Sargassum fulvellum and Undaria pinnatifida were 3.17, 23.12, 31.11 and 10.66%, respectively. These results demonstrate the addition of seaweeds to cooked rice helps to improve the antioxidant activity compared to white rice alone.

RAW 264.7 대식세포에서 유산균으로 발효한 다시마와 톳의 항염증 효과 (Anti-inflammatory Effects of Fermented Laminaria japonica and Hizikia fusiforme Water Extracts with Probiotics in LPS-stimulated RAW264.7 Macrophage Cell Line)

  • This study was conducted to investigate alterations of seaweed composition upon Lactobacillus rhamnosus GG (LGG) fermentation as well as potential anti-inflammatory effects and mechanism (s) of water extracts and fermented water extracts of Laminaria japonica (LJ) and Hizikia fusiforme (HF) in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. Total polyphenol, total sugar, and reducing sugar contents were measured in LJ and HF water extracts before and after fermentation by LGG. Alterations of inflammatory cytokine levels in cell culture media were measured by ELISA, and levels of phosphorylation of c-jun NH2-terminalkinase (JNK) and extra cellular signal regulated kinase (ERK) were examined by Western blot analysis. LGG fermentation of LJ and HF altered total polyphenol and sugar contents in water extracts of LJ and HF. LPS-induced production of pro-inflammatory cytokines such as IL-6 and $TNF-{\alpha}$ was significantly reduced by HF-f compared to control in RAW264.7 cells. Consistent with reduction of anti-inflammatory cytokine, interleukin (IL)-6, and tumor necrosis factor $(TNF)-{\alpha}$ levels by HF-f, HF-f also significantly reduced phosphorylation of ERK and JNK in LPS-stimulated RAW264.7 cells. In addition, LJ-f and HF also significantly reduced phosphorylation of JNK and ERK induced by LPS in RAW264.7 cells. Overall, our result suggests that HF-f among the four tested seaweed extracts is the most potent anti-inflammatory agent, and its mechanism of action is partially mediated by reduction of JNK and ERK phosphorylation as well as IL-6 and $TNF-{\alpha}$ production in LPS-stimulated RAW264.7 cells.

해조류의 항산화 활성 및 아세틸콜린에스테라제 저해 활성 (Antioxidant Activities and Acetylcholinesterase Inhibitory Activities from Seaweed Extracts)

  • 본 연구에서는 퇴행성 질환인 치매의 예방 및 치료를 위한 물질 탐색으로써 여러 가지 해조류 추출물을 가지고 항산화 활성 및 AChE 저해활성을 살펴보았다. DPPH법으로 항산화 활성을 살펴본 결과, 최종농도 40 ${\mu}g$/mL에서 지충이(91.3%), 모로우붉은실(90.7%), 감태(89.9%), 큰비쑥(85.9%)이 80% 이상의 radical 소거활성을 나타내었다. AChE 저해활성에서는 최종농도 10 ${\mu}g$/mL에서 큰비쑥 추출물(30.2%)이 가장 높은 활성을 나타내었으며, 다음으로는 김 추출물(26.6%), 쇠미역(25.3%), 참홑파래(23.4%), 감태(21.7%), 구멍쇠미역(20.4%), 지충이(19.9%) 순으로 총 7종의 해조류가 약 20% 이상의 AChE 저해활성을 나타내었다. 7종의 해조류를 가지고 TLC bioassay를 통하여 살펴본 결과, 여러 compound들에 의해 AChE 저해 활성이 나타나는 것으로 생각된다. 따라서 본 연구 결과는 치매치료제 및 억제를 위한 신규의 생물 소재로 해조류가 가치가 있음을 시사하고 있다.

Screening of Korean Marine Plants Extracts for Inhibitory Activity on Protein Tyrosine Phosphatase 1B

  • Crude extracts of 69 marine organisms (27 salt marsh plants and 42 seaweeds) were screened for the inhibitory activity against the protein tyrosine phosphatase 1B (PTP1) in vitro. The most active extracts were methanol extracts from Derbesia marina (80.6% in inhibitory activity) and Symphycladia latiscula (85.6%) at the concentration of $15{\mu}g/mL$. Methanol extracts of Codium adhaerens and Hisikia fuziformis were moderately inhibitory with 71.2 and 69.1% inhibition, respectively. It was peculiar that only the extracts from seaweeds show inhibitory activity where those from salt marsh plants do not show any significant effect.

해조류 Pulp 분리방법 및 응용연구 (Application Study and Pulp Separation Method by see weeds)

  • 본 연구에서 개발 예정인 해조류 펄프 분리 가공은 해조류자체의 배당체 및 다당류의 복합체로서 개발상 많은 어려움이 있다. 그러나 정제 공정을 개발하고 응용기술을 도입하여 아직 미개척분야인 펄프 응용기술을 확립할 수 있도록 한다. 따라서 본 제품이 개발 대량 생산되어지면 수출향상에 기여함은 물론 생분해성 기능성소재 개발의 실용화로 어민 소득증대 향상에 기여할 수 있다. 따라서 연구의 최종 목표는 1,2차년도의 기술적 데이터를 종합하여 해조류 펄프를 적용하여 생분해성이 향상되고, 고분자의 특수성질을 갖는 새로운 기능성 생분해성 플라스틱, 포장재료 등의 원재료 제품 및 시제품개발을 목표로 한다.
