The effect of permanganate concentration and refluxing time on the test COD results were investigated, as a validation test of the revised procedure. The amended protocol involves the use of a more dilute(1/2.5) permanganate solution(0.010N) and a shorter(1/3) refluxing period(20 min). This revision resulted only 70% of the COD value determined by the old procedure with 8 somewhat improved precision(CV 4.6%${\rightarrow}$3.3%). The dominant factor that influences the decrease in the COD result is the oxidant concentration, being two thirds of the overall effect. The same results were obtained in the comparison test using seawater samples.
Global wanning induced by greenhouse gases such as carbon dioxide is a serious problem for mankind. Carbon dioxide ocean disposal is one of the promising options to reduce carbon dioxide concentration in the atmosphere because the ocean has vast capacity for carbon dioxide sequestration. However, the dissolution rate of liquid carbon dioxide in seawater must be known in advance in order to estimate the amount of carbon dioxide sequestration in the ocean. Therefore, the solubility, the surface concentration, the droplet size and other factors of liquid carbon dioxide at various depths are calculated. The results show that liquid carbon dioxide changes to carbon dioxide bubble around 500 m in depth, and the droplet is completely dissolved below 500 m in depth if carbon dioxide droplet is released both at 1000 m in depth with the initial droplet diameter of 0.011 m or less and at 1500 m in depth with the diameter of 0.015 m or less. In addition, the hydrate film acts as a resistant layer for the dissolution of liquid carbon dioxide. The surface concentration of carbon dioxide droplet with the hydrate film is about 50% at 1500 m in depth and about 60% at 1000 m in depth of the carbon dioxide solubility. Also, the ambient carbon dioxide concentration in the plume is an another crucial parameter for complete dissolution at the intermediate ocean depth, and the injection of liquid carbon dioxide from a moving ship is more effective than that from a fixed pipeline.
Edwardsiella tarda predominantly causes edwardsiellosis in fish at high temperature, but is rarely isolated from water when water temperature is low. However, E. tarda is viable but nonculturable (VBNC) in low water temperature, but it can be revived when water temperature rises and cause disease to fish. Therefore, in order to prevent disease, it is very important to identify pathogens that are in the VBNC state in environmental water. In this study, E. tarda cells in the VBNC state were detected by the ethidium monoazide (EMA)-PCR method using the low-temperature oligotrophic sea water microcosm obtained by inoculation of E. tarda at a concentration of $10^8CFU/ml$. In order to distinguish between live and dead bacteria in E. tarda, each sample was treated with EMA at different concentrations, photoactivated with a 500 W halogen lamp, and PCR was performed with E. tarda specific primer. At the concentration of $10^7CFU/ml$ bacterium, DNA amplification was observed only in the live cells when treated with $60{\mu}g/ml$ of EMA, and smaller amounts of live cells could be distinguished from dead cells by adjusting the EMA concentration. In addition, the VBNC cells of E. tarda in the oligotrophic low temperature seawater microcosm were estimated to be in the range of $10^4{\sim}10^5CFU/ml$ by EMA-PCR. Therefore, it is possible to detect VBNC cells that will act as potential pathogens in environmental water using EMA-PCR method, and quantitative confirmation using concentration change is also possible.
The comparison of the chemical properties of semen of black porgy Acanthopagrus schlegelii long-term acclimated reared in freshwater (BFW) and seawater (BSW) with sperm activity of salinity and ion composition. The chemical properties of seminal plasma on BFW of the factors that most there was not significant difference in the BSW. However, osmolality in seminal plasma of BFW and BSW was $307.0{\pm}4.6$ and $337.3{\pm}10.1$ mOsm/kg, respectively, where BFW showed significant lower concentration in contrast to BSW. Salinity effect on sperm motility of BFW and BSW in 0 psu solution, no sperm motility was observed, whereas in 10 psu solution, both BFW and BSW sperms showed low motility and short time post sperm activation. However, diluted in 20 and 32 psu solutions, highest motility and long time post sperm activation were observed in BFW and BSW sperm. SAI of BFW and BSW varied in depend on the osmolality regardless of ion kind and it showed the highest value in the similar osmolality of artificial seawater (956 mOsm/kg). Accordingly, even in sperm released from BFW, factors initiating sperm motility are determined by osmolality.
A simple procedure, readily available at low cost with a sensitivity sufficient to determine trace levels of iron in seawater is proposed, which utilizes adsorptive accumulation of the iron/catechol complex on the mercury drop electrode in a borate medium of pH 8.0. Optimal conditions include a solution concentration of 2 mM catechol, 2.5 mM borate and a pH of 8.0, an accumulation potential of - 0.25 V is applied for 1∼3 min, and the potential scan is in the differential pulse mode. The limit of detection is 1.5 nM Fe using a preconcentration time of 3 min. The interference from copper can be eliminated and baseline slope is greatly improved, because its peak is well separated from that of iron in the proposed medium.

Mass mortality of mariculture fish due to high summer temperatures is a major issue in the mariculture industry in many coastal waters of Korea, yet measures to mitigate the impact are generally limited. We injected a micro-bubble of liquefied oxygen into the bottom of rockfish cages (about 6-8 m deep) in order to maximize the dispersal of micro-bubbled seawater and reduce fish mortality. The injection of low-temperature oxygen in micro-bubbles lowered the water temperature at the injection area by as much as 1℃ and increased dissolved oxygen concentration by 0.5 ppm. In early August, following a week with persistent high water temperature (above 28.5℃), there was an increase in fish mortality despite the micro-bubble system, which resulted in approximately 7% death of the total introduced fish population. However, this mortality appeared to be much lower than mortality reported in a neighboring mariculture facility (approximately 50% mortality). We also estimated the volume that can be recirculated with pumped seawater using a micro-bubble system. We suggest that this approach of injecting liquefied oxygen through a micro-bubble system may reduce fish mortality during high temperature periods.

Annual variation of water qualities in the Shihwa Lake were observed 18 times from June 1996 to October 2001. We studied at the station of the upper streams and near the water gate of lake. After the flow of the outer seawater through the water gate, the surface salinity in Shihwa Lake increased to the range of 25-30 psu in both stations after October 1998. Due to the declination of the salinity differences between the surface and the bottom water, the pycnocline in which had existed until 1997 has weakened, and made the water column mix vertically. This led to the improvement of anoxic/hypoxic environment at bottom waters after April 1998. However, despite the continuous flow of the outer seawater, the concentrations of chlorophyll-a at surface layer were varied from $2{\mu}g/l\;to\;60{\mu}g/l$, and these values indicated the eutrophication. The following organic matter load was greatly influencing the surface layer's COD concentration. During the rainy season, the salinity at the surface layer to the below 15 psu resulting in stratification between the surface and bottom layer. Organic matters that were provided from the surface layer to the bottom layer due to active primary production in the year exhausted dissolved oxygen at the bottom layer, and the bulks of organic matters at bottom gave rise to hypoxic or anoxic environment. It was observed that the enrichment of ammonia and phosphate were main factors to worsen the water quality of the Shihwa Lake. The results of examining the annual variations in Shiwha Lake through principal component analysis shown that water characteristics in the rainy season were similar with those before input of outer sea water.
According to the IPCC climate change scenario (A1B scenario), the surface seawater temperature of the South Sea of Korea by 2100 may be $2-3.5^{\circ}C$ higher than at present, and seawater pH may decrease from 8.1 to 7.8, due to the increase in atmospheric $CO_2$, which is predicted to increase in concentration from 380 to 750 ppm. These changes may not only intensify the strength of typhoons/storm surges but also affect the function and structure the marine ecosystem. In order to assess the impact of climate change on the marine ecosystem in Korean waters, the project named the 'Assessment of the impact of climate change on marine ecosystem in the South Sea of Korea' has been supported by the Ministry of Land, Transport and Maritime Affairs, from 2008. The goal of this project is to enhance our ability to adapt and prepare for the future environmental changes through the reliable predictions based on the knowledge obtained from projects like this. In this respect, this project is being conducted to investigate the effects of climate/marine environment changes (ocean warming and acidification), and to predict future changes of the structure and function of the ecosystem in the South Sea of Korea. This special issue contains 6 research articles, which are the highlights of the studies carried out through this project.
We investigated the biological activities of ethanol extract from the seawater algae, Chlorella elliposidea C020 such as antibacterial activity, anti-oxidant activity, tyrosinase inhibitory activity and hemolytic activity against human erythrocytes. Extract was obtained from various solvent, methanol, ethanol, acetone and ethanol + acetone (1:1, v/v%), 95% ethanol proved to be best extraction solvents. The contents of ethanol extract were higher in freeze-dried sample than that in frozen-thawing. Antibacterial activities of ethanol extract showed strong inhibitory effect against Bacillus subtilis PM125, Bacillus licheniformis and fish pathogenic bacteria, Vibrio parahaemolyticus KCTC2471 and Edward tarda NUF251. However, this extract didn't worked against antifungal activity against Candida albicans KCTC1940. And, ethanol extract was without hemolytic activity against human erythorocytes. The ethanol extract showed 75% of free radical scavanging effect on 2.0 mg/mL using DPPH method. In tyrosinase inhibition assay of ethanol extract, $IC_{50}$ (Inhibition Concentration) was measured as 10.87 mg/mL. Conclusionally, ethanol extract of Chlorella elliposidea C020 has good candidate for bioactive materials.

Aluminum alloys have poor corrosion resistance compared to the pure aluminum due to the additive elements. Thus, anodizing technology artificially generating thick oxide films are widely applied nowadays in order to improve corrosion resistance. Anodizing is one of the surface modification techniques, which is commercially applicable to a large surface at a low price. However, most studies up to now have focused on its commercialization with hardly any research on the assessment and improvement of the physical characteristics of the anodized films. Therefore, this study aims to select the optimum temperature of sulfuric electrolyte to perform excellent corrosion resistance in the harsh marine environment through electrochemical experiment in the seawater upon generating porous films by variating the temperatures of sulfuric electrolyte. To fabricate uniform porous film of 5083 aluminum alloy, we conducted electro-polishing under the 25 V at $5^{\circ}C$ condition for three minutes using mixed solution of ethanol (95 %) and perchloric (70 %) acid with volume ratio of 4:1. Afterward, the first step surface modification was performed using sulfuric acid as an electrolyte where the electrolyte concentration was maintained at 10 vol.% by using a jacketed beaker. For anode, 5083 aluminum alloy with thickness of 5 mm and size of $2cm{\times}2cm$ was used, while platinum electrode was used for cathode. The distance between the two was maintained at 3 cm. Anodic polarization test was performed at scan rate of 2 mV/s up to +3.0 V vs open circuit potential in natural seawater. Surface morphology was compared using 3D analysis microscope to observe the damage behavior. As a result, the case of surface modification showed a significantly lower corrosion current density than that without modification, indicating excellent corrosion resistance.
