MICROSCOPIC OBSERVATIONS OF FAT TRANSLOCATION IN THE TISSUE OF YELLOW CORVENIA DURING SALTING AND DRYING ("굴비" 제조과정중의 지방의 이동에 대한 조직학적 관찰)

  • Salted and dried yellow corvenia(Pseudosciaena manchurica), so called 'Gul-bi', is one of nation-widely consuming fish foo::ls. It is suitable for a long term preservation and its pro-duce is also a great deal on sea food processing in this country. The texture of 'Gul-bi', however, have often appeared to be a delicate factor for the quality of the product. The loss or dislocation of fat in the tissue of the fish resulted by salting and drying is believed to profoundly relate to the texture of product. In this paper, the tissue of yellow corvenia and movement of fat were microscopically observed before salting, immediately after salting, and after drying and the results observed in the tissues dry salted, brine salted, and brine salted with the addition of BHA were compared. The cross section of yellow corvenia muscle showed that a distinctive border by connective tissue between white and red muscle could not be seen in general, and red muscle was surrounded by hypodermic fatty tissues. In the tissue of fresh yellow corvenia, the fat was mainly distributed in hypodermic fat layer which located under the corium while rarely distributed in white muscle. It was found that some parts of the fat in the tissue were permeated into intermuscular tissue passing through the connective tissues during salting. The result Was the same in both dry-salting and brine-salting tissue. However, the fat translocated into intermuscular tissues disappeared during drying process in the salted without BHA tissues whereas in BHA added tissue. This result suggested that BHA may take a role of multiple effect in translocation of fat in tissues as well as in retarding oxidation. In an advanced stage of salted and dehydration, the muscle fibers were ajoined together and then limits between muscle fibers already became indistinguishable. And the migrated fat into intermuscular tissue aggregated around the connective tissue and are apt to gradually to flow out from the muscular system through these tissues.

Bioactive Component Analysis and Antioxidant Activity in Fresh Sprout of New Kalopanax septemlobus Cultivar (음나무 신품종 새순의 유용물질 함량분석과 항산화활성)

  • This study was carried out to analyze the nutritional composition, bioactive compound and antioxidant activity in fresh sprout of 3 new Kalopanax septemlobus cultivars. Cheong-San, Cheong-Song and Cheong Sun 1 had largest carbohydrate (52.90, 53.12, 55.28%), crude protein (34.23, 31.33, 33.58%) in the proximate composition and had largest K (2.29, 2.22, 2.23%) followed by Ca (0.44, 0.34, 0.37%), Mg (0.24, 0.19, 0.19%) in the mineral contents. Three new cultivars contained a total of 26, 25 and 28 different kinds of amino acids, respectively and the number of essential amino acid is 6, 6 and 7 species, respectively. Also, Cheong-San had higher total polyphenol contents and Cheong-Song had higher total flavonoid contents. DPPH free radical scavenging activity of 70% ethanol, water extracts of Cheong-San and Cheong-Song is higher than others at 100 ppm. In the ABTS radical cation decolorization activity, 70% ethanol extract of Cheong Sun 1, water extract of Cheong-San and Cheong-Song are higher activity than others and all samples have more than 85% ABTS racial cation decolorization activity at 500 ppm. Our results suggest that new K. septemlobus cultivars possess good antioxidant capacities with a high nutritional value and might have potential applications in the food and medical industries.

STUDIES ON THE EXTRACTION OF SEA WEED PROTEINS 3. Extraction of NaOH Soluble Proteins (해조단백질 추출에 관한 연구 3. NaOH 가용성 단백질의 추출)

  • In present study, the effect of various factors including the solvent concentration, extraction time, extraction temperature and the ratio of sample vs extraction solvent(w/v) upon the extractability of the NaOH soluble proteins of marine algae were investigated. Seven species of sun-dried algae, the major ones in consumption as food, namely Porphyra suborbiculata, Undaria pinnatifida(natural and cultivated), Sargassum fulvellum, Sargassum kjellmanianum, Ulva pertusa, Enteromorpha linza and Codium coarctatum were used for the extraction of the NaOH soluble protein. The frozen and masceratd samples were prepared by the same mettled described in previous paper(Lee, 1977). In case of the TCA insoluble protein, all samples reached maxima at 0.025M NaOH solution while the 0.05M for extractable total nitrogen. Variation of the ratio of sample vs solvent gave slight effect upon the extractability, 100 ml solvent added to 1 g dried sample was effective. The effect of extraction time on the extractability differed from species. The extractabilty of Enteromorpha linza, Ulva pertusa and Codium coarctatum reached maxima within 1 hour extraction and 2 hours for the cultivated Undaria pinnatifida while 3 flours for the natural Undaria pinnatifida, Sargassum fulvellum, Sargassum kjellmanianum and Porphyra suborbiculata. The most effective extraction temperature was $60^{\circ}C$ for all samples.

Studies on Physicochemical and Biological Properties of Depolymerized Alginate from Sea Tangle, Laminaria japonicus by Heating Hydrolysis -4. Inhibition Effects of Acute Gastritis Induced by $HCl{\cdot}ethanol$ Solution and Gastric Ulcer Induced- (다시마 (Laminaria japonicus) Alginate의 가열가수분해에 따른 물리${\cdot}$화학적 및 생물학적 특성에 관한 연구 -4. $HCl{\cdot}ethanol$용액과 침수스트레스에 의해 유발되는 랫드 위궤양의 억제효과-)

  • Anti-ulcer effects of depolymerized alginate (HAG-10, average molecular weight 10,000; HAG-50, average molecular weight 50,000; HAG-100, average molecular weight 100,000) obtained by hydrolysis of alginate by heating at $121^{\circ}C$, against $HCl{\cdot}ethanol$ and water-immersion stress in rats were investigated. The acute gastritis, induced by $HCl{\cdot}ethanol$, and the gastric ulcer, induced by water-immersion stress, were inhibited dose-dependently by administration of HAG-50, HAG-100 and alginate. Histopathological lesions of the gastritis and gastric ulcer in rats treated with HAG-50, HAG-100 and alginate were significantly lower than those in rats fed with HAG-10. The inhibition rates (${\%}$) on acute gastritis induced by $HCl{\cdot}ethanol$ and gastritis ulcer induced by water-immersion stress in rats, were $13.00{\%}\;and\;15.74{\%}$of HAG-10, $41.15{\%}\;and\;35.72{\%}$ of HAG-50, $41.58{\%}\;and\;35.37{\%}$ of HAG-100, and $45.17{\%}\;and\;41.11{\%}$ of alginate, respectively. These results suggested that HAG-50, HAG-100 and slginate had a protective effect against the gastritis and gastric ulcer. The effect was not as visible when using HAG-10 in rats. From the present results, it was suggested that HAG-50 was an effective anti-ulcer agent against $HCl{\cdot}ethanol$and water-immersion stress in rats.

  • In order to find the optimum culture condition for marine free-living nematoda in the laboratory, various agar media were developed and experiments were carried on nematodes. Nematodes have collected from the bottom of the sandy sediments' surface layer (about 5cm sediment) of Taean Mallipo beach's intertidal zone. Especially, with regard to agar medium, Killian medium was transformed slightly, density of agar had made a difference and this agar medium(height 2.0 mm on diameter 60 mm Petri dish) was divided. It was mixed with 5 different species of microorganism as nematodes' live food and added each culture medium. Five individuals of Enoplolaimus sp. on each culture medium were grown in a culture medium which was set on $20^{\circ}C$ and light blocked. Moreover, as a result of the optimum culture condition, 1.0% density of agar showed the highest survival rates (the average time of survival is 246.8 hours). On the other hand, the 0.4% density of agar adding Killian medium, bacto peptone and beef extract showed the lowest survival rates, which indicates the average time of survival is 27.6 hours. About Bathylaimus sp., on Killian medium's 1.0% density of agar(no feeding amount) showed the highest survival rates, which connects that the hatching rate 94.7% after 99.5 hours and it laid 7 eggs averagely on the spawning amount and the hatching rate experiment.

Physicochemical Properties of Endosperm Starch and Breadmaking Quality of Rice Cultivars (쌀 품종의 배유 이화학적 특성에 따른 제빵 적성 비교)

  • The physicochemical properties of rice endosperm from five rice varieties and effects of milling on baking bread properties of rice flour were investigated. Five rice varieties exhibited different level of amylose content. The ratio of longer amylopectin chain length to the distribution of endosperm starches was the highest in Goami3. According to the RVA measurement of rice flours, the pasting temperatures of Seolgaengbyeo and Goamibyeo were lower than those of the other rice varieties. There were differences in the changes of swelling power of rice flour under increasing temperature. Each rice flour sample for bread-making was processed into two different particle size by using an air-classification mill, and significant differences were observed among cultivars in the average particle size and damaged starch content of rice flours. Rice flour of Seolgaengbyeo with fine particle size showed the highest value in specific volume after baking.

A Trial for Utilizing Flounder Skin Gelatin as an Emulsifier through Enzymatic Modification (가자미피 젤라틴의 효소적 수식에 의한 유화제의 시제)

  • In order to effectively utilize the by-products of sea-food, the utilization of enzyme-modified flounder(Limanda aspera) skin gelatin as an emulsifier was investigated. In the experiment, the gelatin was extracted from the flounder skin with the heat-treatment at $60^{\circ}C$ and in pH 5.0 for 3 hrs with four volumes of distilled water and emulsifiers were enzymatically modified L-leucine alkyl esters$(L-leucine-OC_n$ : n= 2, 4, 6, 8 and 10) to the gelatin$(EMFSG-C_2,\;EMFSG-C_4,\;EMFSG-C_6,\;EMFSG-C_8,\;EMFSG-C_{10})$ for improving the functional properties such as emulsifying activity, emulsifying viscosity, whippability, electric conductivity, critical micelle concentration and interface tension, etc. Also, the functional properties of the L-leucine alkyl ester modified gelatins were compared with those of Tween-60 as reference. Molecular weights of the enzymatically modified flounder skin gelatin(EMFSG) were 20.5kDa. in $EMFSG-C_2.\;19.5 kDa.\;in\;EMFSG-C_4\;and\;16.5kDa.\;in\;EMFSG-C_6,\;EMFSG-C_8$ and $EMFSG-C_{10}$. respectively. Emulsifying activity and emulsifying viscosity in the modified gelatins were risen with increase of carbon number of the introduced L-leucine alkyl esters. Among the modified gelatins, $EMFSG-C_6$ exhibited the highest emulsifying stability and foaming stability, whereas $EMFSG-C_8$ showed the highest whippability. The electric conductivities of the all $EMFSG-C_n$ were linearly risen to critical micelle concentration(CMC) , therefore $EMFSG-C_{10}$ exhibited the lowest CMC value and interface tension, and dense particles in the microscopic observation. In conclusion, the best quality in functional properties was assured on $EMFSG-C_{10}$.

  • According to facts revealed up until the present, there are a total of 68 known phyla on earth, including 55 phyla of bacteria and 13 phyla of archaea. The human large intestine has 9 phyla of microorganisms, which is a relatively lower diversity compared to the general environments of soil or sea. The diversity of intestinal microorganisms is affected by the characteristics of the host (genetic background, sex, age, immune system, and gut motility), the diet (non-digestible carbohydrates, fat, prebiotics, probiotics), and the intake of antibiotics, which in turn have an effect on energy storage processes, gene expressions, and even metabolic diseases like obesity. Probiotics are referred to as living microorganisms that improve the intestinal microbiota and contribute to the health of the host; in addition, probiotics usually comprise lactic acid bacteria. Recently, bacteriotherapy using probiotics has been utilized to treat sicknesses like diarrhea and irritable bowel syndrome. Prebiotics are a food ingredient which can selectively adjust intestinal microorganisms and which comprise inulin, fructooligosaccharides, galactooligosaccharides, and lactulose. In recent days, attention has been paid to the use of dietary cellulose in the large intestine and the production of short chain fatty acids (short-chain fatty acids) in relation to obesity and anticancer. More research into microorganisms in the large intestine is necessary to identify specific microorganism species, which are adjusted by diverse non-digestible carbohydrates, prebiotics, and probiotics in the large intestine and to understand the connection between sicknesses and metabolites like short chain fatty acids produced by these microorganism species.

Molecular and Physiological Aspects of Breeding Program for Development of Hybrids between Abalones Distributed in the Coast of Korea (한국산 전복을 이용한 교잡종 개발)

  • Abalones are gastropod mollusks belonging to the genus Haliotis. Fishery products are continuously declining worldwide while abalone production from aquatic farms has considerably increased. Although abalones are regarded as very important marine resources and considered to have big potential in sea-food farming industry around world, the slow growth rate of Pacific abalones is considered to be one of the most serious problems. For the genetic improvements in cultured abalone, advances in various breeding techniques for abalone have been reached through the introduction of selection, crossbreeding, hybridization, and polyploidy in several commercially important abalone species. Six species of abalone have been reported to be distributed along the coasts of Korea: Haliotis discus hannai, Haliotis discus discus, Haliotis madaka, Haliotis gigantea, Haliotis diversicolor diversicolor, and Haliotis diversicolor supertexta. The hybridization between these abalones may be one of the advanced technologies, and the preliminary experiments of interspecific hybrids between abalone species distributed in northern pacific areas including Korea, China, and Japan have been conducted. In this study, we reviewed the phylogenetic relationship of northern pacific abalone species which have the potential traits for aquaculture in Korea and their identifications. We also examined the development of molecular markers and some other aspects of the genetic approaches for successful development of hybrids.

Recovery of Protein Hydrolysate from Hoki (Johnius belengeri) Frame with Tuna Pyloric Caeca Crude Enzyme and Its Functionalities (참치 유래 조효소를 이용한 민태(Johnius belengeri) Frame으로부터 단백질 가수분해물의 회수 및 그 기능성)

  • Enzymatic hydrolysis with tuna pyloric caeca crude enzyme(TPCCE) was performed to recover a protein hydrolysate from hoki frame, fish processing by-product. Optimum hydrolytic conditions were pH 10.0, temperature $50^{\circ}C$, and incubation time 12 hrs, and then the degree of hydrolysis was about 60%. The yield of the hydrolysate from hoki frame by enzymatic hydrolysis was approximately 77% on a dry weight basis. The prepared protein hydrolysates were also fractionated through a series of 30, 10, 5 and 1 kDa molecular weight cut-off (MWCO) membranes in order to investigate the effect of their functionalities according to the difference of their molecular size. As the result of studying functionalities of the hydrolysates, 1 K hydrolysate showed the highest solubility over all pHs, and 30 and 10 K hydrolysate showed more excellent emulsifying property and whippability than the other hydrolysates.
