Apoptotic Effects of psiRNA-STAT3 on 4T1 Breast Cancer Cells in Vitro

  • Background: The aim of this study was to investigate the effect of a Lipofectamine2000 (Life2000) Transfection Reagent transfected psiRNA-STAT3 plasmid on 4T1 breast cancer cells. Materials and Methods: MTT was used to detect the cell proliferation of breast cancer 4T1 cells at different periods (0h, 6h, 8h, 10h); the cell cycle was assessed by flow cytometry; variation of apoptosis and mitochondrial membrane potential was observed under a fluorescence microscope; immunohistochemical staining was used to determine the expression of caspase-3 and cyclin-D1 protein. Results: An obvious effect of inhibition to 4T1 cancer cells could be observed at 8h after the psiRNA-STAT3 was transfected. Typical alterations of apoptotic morphological features were visible in the psiRNA-STAT3 treatment group. Mitochondrial membrane potential decreased significantly, the number of cells was increased in G0/G1 phase, and the number of cells was decreased in S phase, and the data were statistically significant (p<0.05), compared with the Scramble and Mock groups. Expression of caspase-3 protein was increased significantly, while that of cyclin D1 was significantly decreased. Conclusions: Life2000 transfected psiRNA-STAT3 plasmid can inhibit 4T1 tumor cell proliferation and promote apoptosis of 4T1 tumor cells, which process depends on the regulation of expression of cyclin D1 and caspase-3 protein.

The Structure of Reversible DTCNN (Discrete-Time Celluar Neural Networks) for Digital Image Copyright Labeling (디지털영상의 저작권보호 라벨링을 위한 Reversible DTCNN(Discrete-Time Cellular Neural Network) 구조)

  • In this paper, we proposed structure of a reversible discrete-time cellular neural network (DTCNN) for labeling digital images to protect copylight. First, we present the concept and the structure of reversible DTCNN, which can be used to generate 2D binary pseudo-random images sequences. We presented some, output examples of different kinds of reversible DTCNNs to show their complex behaviors. Then both the original image and the copyright label, which is often another binary image, are used to generate a binary random key image. The key image is then used to scramble the original image. Since the reversibility of a reversible DTCNN, the same reversible DTCNN can recover the copyright label from a labeled image. Due to the high speed of a DTCNN chip, our method can be used to label image sequences, e.g., video sequences, in real time. Computer simulation results are presented.

  • The amount of digitalized contents has been rapidly increased, but the main distribution channel of them is Internet which is easily accessible. Therefore 'security' necessarily arises as one of the most important issues and the method of protecting contents becomes a major research topic as much as data coding techniques. In recent years, many developers have studied on techniques that allow only authorized person to access contents. Among them the scrambling method is one of well-known security techniques. In this paper, we propose a simple and effective digital video scrambling method which utilizes the intra block properties of a recent video coding technique, H.264. Since intra prediction modes are adopted in H.264 standard, it is easy to scramble a video sequence with modification of the intra prediction modes. In addition to its simplicity, the proposed method does not increase bit rate after scrambling. The inter blocks are also distorted by scrambling intra blocks only. This paper introduces a new digital video scrambling method and verifies its effectiveness through simulation.

  • IPTV(Internet Protocol Television) is an emerging technology in which telecommunication and broadcasting technologies are converged. IPTV service providers usually multicast scrambled contents. And only legitimate subscribers who pass the access control can de-scramble and use the contents. In order to ensure revenues, providers oftentimes employ password-based authentication protocols that ensure secure authentication processes between set-top box and smart card. In this paper, we propose a fingerprint-based 1-out-of-n authentication protocol which provides convenient and more secure authentication process in some organizational environments. The proposed scheme shows that only those who provide legitimate fingerprint information can be authenticated even in a situation that both password and smart card are compromised.

Implementation of an Obfuscator for Visual C++ Source Code (비주얼 C++소스 코드를 위한 obfuscator 구현)

  • Automatic obfuscation is known to be the most viable method for preventing reverse engineering intentional1y making code more difficult to understand for security purposes. In this paper, we study and implement an obfuscation method for protecting MS Visual C++ programs against attack on the intellectual property in software like reverse engineering attack. That is, the paper describes the implementation of a code obfuscator, a tool which converts a Visual C++ source program into an equivalent one that is much harder to understand. We have used ANTLR parser generator for handling Visual C++ sources, and implemented some obfuscating transformations such as 'Remove comments', 'Scramble identifiers', 'Split variables', 'Fold array', 'Insert class', 'Extend loop condition', 'Add redundant operands', and 'Insert dead code'. We have also evaluated the performance and effectiveness of the obfuscator in terms of potency, resilience, and cost. When the obfuscated source code has been compared with the original source code, it has enough effectiveness for software protection though it incurs some run-time overheads.

  • Feed industry began to demand that beyond simple feeds to meet the needs of customers with a premium catering by economic development situation and, depending on the user's convenience and customer needs. But tableware introduced in the feed system, the introduction of RFID. High and low temperatures and salinity problems that RFID chips, the feed system malfunction. In this paper, infused with IT Convergence Technology was carried out for a smart lunch by putting RFID chips in the Food Court distribute food. Food court meal to ensure stability of the system was a smart lunch that Hot food ($125^{\circ}C$) and cold ($-40^{\circ}C$) and salinity reflecting foods in High temperature, low temperature experiments, through salt spray test. In addition, the existing straight distribute food, scramble distribute food, Marketplace distribute food, Food Court and the advantages of a comparative analysis of distribute food investigated. Through this paper distribute food will contribute to the Food Court. IT convergence technology (RFID, LCD, S/W, H/W) to apply the feed industry in the development of Smart Food Court.

Efficient H.264/AVC Video Scrambling Methods for Digital Rights Management (디지털 저작권 관리를 위한 효율적인 H.264/AVC 비디오 스크램블링 방법)

  • This paper describes efficient H.264/AVC video scrambling methods for digital rights management. The proposed scrambling methods are to scramble level and suffix in entropy encoding and MVD in motion estimation of the H.264 video compression process. Other scrambling methods have been proposed but they degrade the compression efficiency or make it difficult to achieve real-time processing due to the large amount of computational efforts. Since the proposed scrambling methods resolve the drawbacks of other approaches, they do not cause image distortion and the original compression efficiency is maintained. We verified our scrambling methods and evaluated the performance by conducting several experiments with H.264 reference program. Finally, we implemented video player system using USB dongle in order to apply the proposed scrambling/descrambling methods to H.264 video compression.

MicroRNA-126 Regulates the Expression of Stem Cell Transcription Factors (Sox2 and Lin28) in Various Ovarian Tumors (MicroRNA-126은 난소 종양세포의 줄기세포 전사인자 (Sox2와 Lin28) 발현을 조절한다)

  • Stem cell-like tumor cells are reported to be the main reason for tumor recurrence and metastasis. As one of the new approaches to overcome cancer, studies are emerging to inhibit the expressions of stem cell transcriptional factors (Oct4, Sox2, Klf-4, and Lin28) in cancer cells. MicroRNAs are master genetic regulators that can control development and differentiation of stem cells. In this study using various ovarian tumors (Skov3, Ovcar3, Tov112D, Tov21G, PA-1 and Hsc832(c)T), we examined the expressions of stem cell-related transcription factors, and the biological changes in cell survival and growth by miR-126 that targets stem cell transcriptional factors. We observed that treatment of miR-126 induced the morphological changes and cell suspension in most cells. In addition, miR-126 induced gradual regression of cell division except Skov3 cells, especially significant time-dependent reduction in Tov112D, Tov21G and PA-1. When we examined the expression of stem cell transcriptional factors, Sox2 was shown to be down-regulated after miR-126. Our results demonstrate that miR-126 treatment can provide the reversible environment to regulate cell division and to induce cell death of ovarian tumors, suggesting the molecular biological clues for clinical usage.

Pharmacophore Identification for Peroxisome Proliferator-Activated Receptor Gamma Agonists

  • Peroxisome proliferator-activated receptors (PPARs) are members of nuclear receptors and their activation induces regulation of fatty acid storage and glucose metabolism. Therefore, the $PPAR\gamma$ is a major target for the treatment of type 2 diabetes mellitus. In order to generate pharmacophore model, 1080 known agonists database was constructed and a training set was selected. The Hypo7, selected from 10 hypotheses, contains four features: three hydrogen-bond acceptors (HBA) and one general hydrophobic (HY). This pharmacophore model was validated by using 862 test set compounds with a correlation coefficient of 0.903 between actual and estimated activity. Secondly, CatScramble method was used to verify the model. Hence, the validated Hypo7 was utilized for searching new lead compounds over 238,819 and 54,620 chemical structures in NCI and Maybridge database, respectively. Then the leads were selected by screening based on the pharmacophore model, predictive activity, and Lipinski's rules. Candidates were obtained and subsequently the binding affinities to $PPAR\gamma$ were investigated by the molecular docking simulations. Finally the best two compounds were presented and would be useful to treat type 2 diabetes.

Gastric Emptying in Patients with Diabetes: Gastric Emptying Time, Retention Rate and Effect of Cisapride (당뇨병 환자의 위배출능 : 위배출시간, 위저류율 및 Cisapride의 영향)

  • Gastic emptying scan in diabetic patients is widely used to assess the degree of motility disturbance and the symtoms such as nausea, vomiting, bloating, abdominal pain and early gastric fullness which we can't find anatomic lesion by fiberoscopic or barium study In order to determine the relationship among diabetic gastropathy, neropathy, retinopathy and disease duration, gastric emptying scan using $^{99m}Tc-tin$ colloid labeled scramble egg in hamburger was performed in 10 healty male controls and 50 diabetic patients which were subdivided to no neuropathy, peripheral neuropathy and autonomic neuropathy groups according to the degree of diabetic neuropathy and no retinopathy, background retinopathy and proliferative retinopathy groups according to the degree of diabetic retinopathy. After medication of cisapride for 2 weeks, we observed the presence of improvement of gastric motility in diabetics. The results were as following: 1) In controls, gastric emptying time (GET1/2) was $75{\pm}13.6$ min and 2 hour gastric retension rate (GRR2) was $32{\pm}11.1%$. 2) In diabetics, GET/2 was prolonged more than 2 hours and GRR2 was $58{\pm}23.1%$. According to degree of neuropathy, GET1/2 was prolonged more than 2 hours in all three groups and GRR2 was $54{\pm}24.1%$ in no neuropathy group, $57{\pm}24.3%$ in peripheral neuropathy group and $69{\pm}24.6%$ in autonomic neuropathy group. According to degree of retinopathy, GET1/2 was $110{\pm}23.4$ min in no retinopathy group and prolonged more than 2 hours in other two groups and GRR2 was $45{\pm}21.6%$ in no retinopathy group, $71{\pm}19.7%$ in background retinopathy group and $73{\pm}21.5%$ in proliferative retinopathy group. 3) After cisapride medication for 2 weeks, GET1/2 and GRR2 were improved as $90{\pm}14.6$ min and $40{\pm}13.8%$ (initial GET1/2 and GRR2 were above 2 hours and $61{\pm}15.4%$). We can conclude from above findings that gastropathy in diabetic neuropathy suggesting main underlying factor in motility disorder The degree of retinopathy and disease duration were correlated with severity of gastropathy in diabetics. From the results of gastric emptying scan, we can conclude that cisapride was useful drug for improving diabetic gastropathy and gastric emptying scan was valuable for assessing severity of diabetic gastropathy as non-invasive method.
