백삼과 홍삼추출물의 학습수행과 Acetylcholinesterase 억제에 미치는 효과 (Effects of white ginseng and red ginseng extract on learning performance and acetylcholinesterase activity inhibition)

  • 백삼과 홍삼추출물 투여가 scopolamine으로 기억을 손상시킨 실험동물의 학습수행 개선과 뇌조직의 AChE 억제 및 ACh 함량을 평가하여 다음과 같은 결론을 얻었다. 1. 백삼과 홍삼추출물(200 mg/kg, p.o.) 투여는 scopolamine (2 mg/kg, i.p.)으로 유발된 기억손상 동물모델의 Morris water maze 시험에서 tacrine 투여군과 대조군처럼 인지훈련이 반복될수록 escape latency 시간이 현저히 감소하였다. probe test에서도 scopolamine 투여군보다 유의적으로 감소하여 인삼추출물이 장기기억(Long-term memory)과 working memory의 개선에 효과가 있음을 확인하였다. 2. Y-maze test를 통한 실험동물의 자발적 변동행동력은 실험군간 차이가 없었으며, 총 출입회수도 차이가 없어 인삼추출물은 실험동물의 일반적 운동성에 영향을 주지 않았다. 3. 해마조직의 AChE 활성은 인삼추출물 군에서 감소하는 경향을 보였으나 유의적 차이는 없었다. 대뇌피질과 혈청 AChE 활성은 홍삼추출물 투여군에서 유의적으로 감소하였다. 4. 인삼추출물 투여군의 뇌조직 ACh 함량은 대뇌피질과 해마조직에서 scopolamine 투여군보다 증가하는 경향을 보였으나 그룹간의 유의적인 차이는 없었다. 5. 이를 통하여 볼 때 인삼추출물 특히 홍삼추출물은 장기기억 활성촉진, AChE 억제, ACh 활성촉진으로 콜린성 신경계를 자극하여 기억 및 학습증진에 매우 효과적으로 작용하는 항치매 물질로 사료된다.

몰약 당귀 오미자 혼합제제 HPM-1의 Scopolamine에 의해 유도된 기억력 감퇴에 대한 개선 효과 (Ameliorating Effects of HPM-1 on Scopolamine-induced Memory Impairments in Mice)

  • Myrrh, Angelicae Radix, and Schisandrae Fructus have been used to treat diverse diseases including neurological disorder in the traditional Korean medicine. In the present study, we investigated the ameliorating effects of HPM-1, a combined extract of Myrrh, Angelicae Radix and Schisandrae Fructus, on scopolamine-induced memory impairments in mice. First, we assessed HPM-1, HPM-2 and HPM-3, in which Myrrh was extracted with water, 30% ethanol, and 30% ethanol/3% vinegar, respectively. The oral administration of HPM-1, HPM-2, or HPM-3 significantly reversed scopolamine-induced reduction of spontaneous alternation in the Y-maze task. In the passive avoidance task, HPM-1 or HPM-3 restored the decreased latency time of the retention trial by scopolamine treatment, but in terms of efficacy, HPM-1 showed more beneficial effects than HPM-3. In addition, HPM-1 administration reversed scopolamine-induced reduction of spontaneous alternation in the Y-maze task and scopolamine-decreased latency time in the passive avoidance in a dose-dependent manner. These results suggest that HPM-1 has the therapeutic potential in memory impairments.

Scopolamine 처리에 의한 인지 및 기억력 손상 마우스에서 박하의 효과 (Mentha arvensis Attenuates Cognitive and Memory Impairment in Scopolamine-treated Mice)

  • Mentha arvensis is used traditional medicine to treat various disorders. In the present study, M. arvensis were extracted by the solid-phase microextraction (SPME) method and analyzed by gas chromatograph-mass spectrometry (GC-MS). We investigated the protective effects and mechanisms of a M. arvensis extract on scopolamine-induced cognitive and memory impairment. Mice were orally pretreated with a M. arvensis extract or normal saline, and then behavior tests were conducted 30 min after scopolamine injection. The antioxidant capacities were analyzed by free radical scavenging (DPPH and ABTS). Acetylcholinesterase (AChE) activity were also measured using Ellman's method ex vivo test. In behavior tests, percent of spontaneous alteration, escape latency and swimming time in target quadrant were improved by the administration of the M. arvensis extract, which suggests that the M. arvensis extract improves memory function in the scopolamine-treated mice model. In addition, M. arvensis extract showed inhibition of the free radical and AChE activity. The results of the present study suggest that the M. arvensis extract ameliorates scopolamine-induced cognitive and memory deficits through the inhibition of free radicals and AChE activity. Therefore, M. arvensis may be a promising neuroprotective agent for management of learning and memory improvements in human dementia patients.

  • Cognitive impairment is considered to be key research topics in the field of neurodegenerative diseases and in understanding of learning and memory. In the present study, we investigated neuroprotective effects of Schisandra chinensis (SC) and Ribes fasciculatum (RF) extracts in hydrogen peroxide-induced neuronal cell death in vitro and scopolamine-induced cognitive impairment in Sprague Dawley® (SD) rat in vivo. Apoptotic cell death in neuroblastic PC12 cell line was induced by hydrogen peroxide for 1 hour at 100 μM. However, mixture of SC and RF treatment prevented peroxide induced PC12 cell death with no neurotoxic effects. For in vivo experiment, the effect of SC and RF extracts on scopolamine-induced cognitive impairment in SD rat was evaluated by spontaneous alternation behavior in Y-Maze test. After 30 min scopolamine injection, the scopolamine-induced rats presented significantly decreased % spontaneous alteration and acetylcholine level, compared to non-induced group. However, treatment of SC+RF extracts rescued the reduced % spontaneous alteration with acetylcholine concentration from hippocampus in scopolamine-induced rats. These results suggested that mixture of SC and RF extract may be a potential natural therapeutic agent for the prevention of cognitive impairment.

Water-soluble ginseng oligosaccharides protect against scopolamine-induced cognitive impairment by functioning as an antineuroinflammatory agent

  • Background: Panax ginseng root is used in traditional oriental medicine for human health. Its main active components such as saponins and polysaccharides have been widely evaluated for treating diseases, but secondary active components such as oligosaccharides have been rarely studied. This study aimed to assess the impact of water-soluble ginseng oligosaccharides (WGOS), which were isolated from the warm-water extract of Panax ginseng root, on scopolamine-induced cognitive impairment in mice and its antineuroinflammatory mechanisms. Methods: We investigated the impact of WGOS on scopolamine-induced cognitive impairment in mice by using Morris water maze and novel object recognition task. We also analyzed the impact of WGOS on scopolamine-induced inflammatory response (e.g., the hyperexpression of proinflammatory cytokines IL-$1{\beta}$ and IL-6 and astrocyte activation) by quantitative real-time polymerase chain reaction and glial fibrillary acid protein (GFAP) immunohistochemical staining. Results: WGOS pretreatment protected against scopolamine-induced learning and memory deficits in the Morris water maze and in the novel object recognition task. Furthermore, WGOS pretreatment downregulated scopolamine-induced hyperexpression of proinflammatory cytokines interleukin (IL)-$1{\beta}$ and IL-6 mRNA and astrocyte activation in the hippocampus. These results indicate that WGOS can protect against scopolamine-induced alterations in learning and memory and inflammatory response. Conclusion: Our data suggest that WGOS may be beneficial as a medicine or functional food supplement to treat disorders with cognitive deficits and increased inflammation.

  • This study was conducted to investigate the cognitive improvement and memory recovery effects of Korean and Chinese Crataegus pinnatifida ethanolic extracts on scopolamine-induced memory impairment in mice. In vivo studies were carried out with mice treated with Korean Crataegus pinnatifida extracts (KCF) and Chinese Crataegus pinnatifida extracts (CCF) in doses of 5 and 50 mg/kg (p.o.) and scopolamine was injected 30 min before the behavioral testing. Antioxidant activity was assessed by 2,2-diphenyl-1-picryl hydrazyl (DPPH) assay and 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay, and acetylcholinesterase inhibition by Ellman's modified method. The chlorogenic acid and hyperoside as marker compounds of KCF and CCF was quantified by ultra-performance liquid chromatography analysis (UPLC). Results showed that KCF was more contained high content of chlorogenic acid and hyperoside than CCF. In addition, KCF was more exerted free radical (DPPH and ABTS) scavenging activity and blocked AChE activity than CCF. In vivo studies also showed that KCF administration has a further improved the memory of scopolamine-treated mice than CCF in Y-maze test, passive avoidance test and Morris water maze test. These results revealed that KCF more prevents scopolamine-induced memory impairments through antioxidant and acethylcholinesterase inhibition effect compared CCF.

Production of Tropane Alkaloids by Optimal Culture Conditions in Adventitious Root of Hyoscyamus niger L.

  • Scopolamine and hyoscyamine are important anticholinergic compounds obtained from Hyoscyamus niger. Adventitious roots induced from rhizome of H. niger and roots were cultured in SH medium supplemented with 3% (w/v) sucrose and 0.5 mg/L IBA. Roots were grown rapidly after 10 days of cultures. Scopolamine production was increased 7 times and hyoscyamine production was increased 12 times after 10 day of cultures. SH medium was best in root growth. But, highest scopolamine productivity was observed in WPM medium, followed White medium and best hyoscyamine productivity was resulted in MS medium. Sucrose was increased scopolamine and hyoscyamine production were increased the medium supplemented by sucrose comparing to than those by other carbon sources.

  • The effects of gastric acidity on the bioavailability of scopolamine hydrobromide and ${\ell}$-hyoscya-mine were studied in rabbits. Each drug was administered orally at dose of 0.2mg/kg to gastric acidity controlled rabbits. The plasma concentration of scopolamine and ${\ell}$-hyoscyamine were determined by selected ion monitoring in GC/MSCScopolamine; m/Z= 138, ${\ell}$-Hyoscyamine; m/z= 124). Rabbits with hypoacidity showed significantly higher Cp and AVC than rabbits with hyperacidity after oral administration of each drug. From the results of this experiment, it is desirable that dosage regimen of dose of scopolamine and hyoscyamine should be adjusted when the drugs should be administered with antacids in clinical practice.

The effects of various biotic elicitors on the accumulation of scopolamine and hyoscyamine by adventitious hairy root cultures of Scopolia parviflora

  • Hyoscyamine and scopolamine are the pharmaceutically valuable anticholinergic drugs. The aim of our study is to increase the contents of both hyoscyamine and scopolamine by means of elicitation. Various biotic elicitors derived from 3 fungi and 1 yeast were inoculated to the hairy root cultures. When homogenate and supernatant of elicitors treated with hairy root cultures, that of yeast elicitor was only increased scopolamine production. However, all of the fungal elicitors did not influenced the productions of scopolamine and hyoscyamine. Our results will contribute to mass production of tropane alkaloids by hairy root cultures of Scopolia parviflora.

Cognitive-Enhancing Effect of Dianthus superbus var. Longicalycinus on Scopolamine-Induced Memory Impairment in Mice

  • Dianthus superbus (D. superbus) is a traditional crude drug used for the treatment of urethritis, carbuncles and carcinomas. The objective of this study was to confirm the cognitive enhancing effect of D. superbus in memory impairment induced mice and to elucidate the possible potential mechanism. Effect of D. superbus on scopolamine induced memory impairment on mice was evaluated using the Morris water maze and passive avoidance tests. We also investigated acetylcholinesterase (AChE) activity and brain-derived neurotropic factor (BDNF) expression in scopolamine-induced mice. HPLC-DAD analysis was performed to identify active compounds in D. superbus. The results revealed that D. superbus attenuated the learning and memory impairment induced by scopolamine. D. superbus also inhibited AChE levels in the hippocampi of the scopolamine-injected mice. Moreover, D. superbus increased BDNF expression in the hippocampus. Eight compounds were identified using HPLC-DAD analysis. The content of 4-hydroxyphenyl acetic acid was higher than contents of other compounds. These results indicated that D. superbus improved memory functioning accompanied by inhibition of AChE and upregulation of BDNF, suggesting that D. superbus may be a useful therapeutic agent for the prevention or treatment of Alzheimer's disease.
