스코폴라민으로 유도한 기억력 장애 동물모델에서 명자나무(Chaenomeles speciose Nakai) 과실 추출물의 효과 (Effects of Chaenomeles speciose Nakai on Scopolamine Induced Memory Impaired Mouse Model)

  • Chaenomeles speciose Nakai (CSP) or Chaenomeles sinensis Koehne (CSS) (Rosaceae) has been used, traditionally, to treat muscle problems and gastric dampness in eastern Asia countries. Therefore, many studies have focused on investigating its active compounds and effects on muscle pain, arthritis and gastro-intestinal diseases. Recently, several studies reported that CSS extract degrade amyloid plaques and enhance synaptic acetylcholine level in vivo and in vitro. Although these two Chaenomeles species are used without differences, CSP is reported to contains more phenolic compounds which are known to enhance memory. Therefore, in this study, we investigated the memory ameliorating effects of CSP by employing the passive avoidance test, Y-maze task and novel object recognition test. CSP (30 or 100 mg/kg) ameliorated the declined memory induced by scopolamine injection and enhanced the brain-derived neurotrophic factor (BDNF) levels along with post synaptic density protein 95 (PSD 95) levels at the hippocampus of the scopolamine-injected mouse brain. These results suggested that CSP alleviates the cognition declines caused by cholinergic blockade via enhancing BDNF levels and PSD 95, and that it would enhance memory formation and be useful for treating memory declines.

증숙 더덕 추출물의 인지능력 개선 효과 (Effect of Steamed Codonopsis lanceolata on Spatial Learning and Memory in Mice)

  • Alzheimer's disease is progressive neurodegenerative disorder by the loss of memory and learning abilities. Codonopsis lanceolata (C. lanceolata) is traditional medicinal plant used for the treatment of inflammatory diseases. The aim of study was to evaluate the effect of steamed C. lanceolata on scopolamine-induced memory impairment in the Morris water maze test and passive avoidance test. In addition, this study investigated the neuroprotective effects of steamed C. lanceolata on glutamate-induced cell death in HT22 cells using MTT assay. The results showed that steamed C. lanceolata (500 mg/kg body weight, p.o.) reversed spatial memory impairment by scopolamine in Morris water maze test and passive avoidance test. Steamed C. lanceolata attenuated memory impairment by scopolamine compared with common C. lanceolata. In addition, administration of steamed C. lanceolata significantly also reduced cell death. We suggest that steaming process more improve cognitive enhancing and neuroprotective effect of C. lanceolata than common C. lanceolata.

Dehydroevodiamine·HCl enhances cognitive function in memory-impaired rat models

  • Progressive memory impairment such as that associated with depression, stroke, and Alzheimer's disease (AD) can interfere with daily life. In particular, AD, which is a progressive neurodegenerative disorder, prominently features a memory and learning impairment that is related to changes in acetylcholine and abnormal ${\beta}$-amyloid ($A{\beta}$) deposition in the brain. In the present study, we investigated the effects of dehydroevodiamine HCl (DHED) on cognitive improvement and the related mechanism in memory-impaired rat models, namely, a scopolamine-induced amnesia model and a $A{\beta}_{1-42}$-infused model. The cognitive effects of DHED were measured using a water maze test and a passive avoidance test in the memory-impaired rat models. The results demonstrate that DHED (10 mg/kg, p.o.) and Donepezil (1 mg/kg, p.o.) ameliorated the spatial memory impairment in the scopolamine-induced amnestic rats. Moreover, DHED significantly improved learning and memory in the $A{\beta}_{1-42}$-infused rat model. Furthermore, the mechanism of these behavioral effects of DHED was investigated using a cell viability assay, reactive oxygen species (ROS) measurement, and intracellular calcium measurement in primary cortical neurons. DHED reduced neurotoxicity and the production of $A{\beta}$-induced ROS in primary cortical neurons. In addition, similar to the effect of MK801, DHED decreased intracellular calcium levels in primary cortical neurons. Our results suggest that DHED has strong protective effects against cognitive impairments through its antioxidant activity and inhibition of neurotoxicity and intracellular calcium. Thus, DHED may be an important therapeutic agent for memory-impaired symptoms.

미치광이풀 모상근 배양에서 Tropane Alkaloids 생산성 증진을 위한 최적 생물학적 엘리시터 선발 (Selection of Optimal Biotic Elicitor on Tropane Alkaloid Production of Hairy Roots in Scopolia parviflora Nakai)

  • 미치광이풀의 모상근 뱌양에 의해 의학적으로 중요한 물질인 scopolamine과 hyoscyamine 생산을 증진시키기 위해 3종의 곰팡이와 1종의 효모 유래의 생물학적 elicitor를 supernatant와 homogenate의 형태로 처리하였다. Elicitor 중 Candida albicans는 supernatant와 homogenate 모두 scopolamine 생산성을 증가시켰다. Fusarium solani의 homogenate와 C. albicans의 supernatant 처리로 hyoscyamine 함량이 증가되었다. 그러나 나머지 fungus strain도 대조구에 비해 tropane alkaloids의 생산을 다소 증가시켰다. 본 연구에서는 C. albicans가 tropane alkaloids 생산에 적합한 elicitor임으로 구명되었다. 본 연구 결과는 미치광이풀 모상근 대량배양에 의한 tropane alklaoids 대량생산에 기여할 것이다.

Scopolamine 유발 기억력손상 흰 쥐에서 영계감조탕가용골모려(笭桂甘棗湯加龍骨牡蠣)의 인지기능개선과 항산화 효과 (Anti-amnesic and Antioxidant Effect of Yeongkyekamjotanggayonggolmoryo Aqueous Extracts on Scopolamine-induced Memory Impairment in Mice)

  • Objectives: The purpose of this in vivo study is to observe anti-amnesic effects of Yeongkyekamjotanggayonggolmoryo (YGYM), a novel mixed herbal prescription, Ossis Mastodi and Ostreae Testa added Yeongkyekamjo-tang, on scopolamine induced amnesia in C57BL/6 mice through acetylcholine (ACh) and acetylcholinesterase (AChE) activity, Choline acetyltransferase (ChAT) mRNA expression, and antioxidant effects. Methods: Six groups, total 20 intact or 100 Sco treated mice were used in this study after one week of acclimatization period. Half the animals were used for passive avoidance task tests and hippocampus ACh content, AChE activity, and ChAT mRNA expression were measured. The other half was subjected to an underwater maze test and then the cerebral cortex antioxidant defense system was measured. Results: In the passive avoidance experiment, there was significant decrease in residence time in the bright room and in the underwater maze test, escape latency to escape from the esophagus significantly increased compared with the normal control group. At the final sacrifice, ACh content and ChAT mRNA expression decreased, AChE activity increased, and cerebral cortical MDA increased GSH content, SOD and CAT activity in Sco control mice, as compared to intact vehicle control mice. However, these Sco treatment-related memory loss through AChE activation destroyed the cerebral cortex antioxidant defense system, and was inhibited dose-dependently by 28 days consecutive oral pretreatments of YGYM extracts 500, 250, 125 mg/kg. Conclusions: In the above results, YGYM extract that oral administration of YGYM extracts alleviates the antioxidant defense system, through preservation of ACh mediated by upregulation of ChAT mRNA expression, and increase of AChE inhibition and brain antioxidant defense systems.

  • The ameliorating effect of the saponin rich fraction (SRF) prepared from the roots extract of Platycodon grandiflorum (Campanulaceae) on the scopolamine-induced cognitive dysfunction in mice were investigated by way of Moris water maze task. The mice with repeated oral administration of the SRF for four weeks showed markedly reduced the swimming time and swimming distance to reach the platform as well as the increase of number of crossing the platform on the Moris water maze task performed after scopolamine intoxication. The results suggest that SRF obtained from the roots extract of P. grandiflorum exert a cognitive enhancing effect on scopolamine-induced memory impairment in mice.

Alzheimer's Disease 병태모델에서 녹용대보탕(鹿茸大補湯)의 신경세포 보호효과 (The neuroprotective effects of Nokyongdaebo-tang(Lurongdabutang) treatment in pathological Alzheimer's disease model of neural tissues)

  • Objectives : Alzheimer's disease(AD) is the most common form of dementia, which is characterized by progressive deterioration of memory and higher cortical functions that ultimately results in total degradation of intellectual and mental activities. Nokyongdaebo-tang(Lurongdabutang) has been usually used for the treatment for the deficiency syndrome dementia and amnesia. This experiment was designed to investigate the effect of the Nokyongdaebo-tang(Lurongdabutang) hot water extract on pathological AD model. Methods : The effects of the Nokyongdaebo-tang(Lurongdabutang) hot water extract on cultured spinal cord cells induced by ${\beta}$-amyloid were investigated. The effects of the Nokyongdaebo-tan(Lurongdabutang) hot water extract on the memory deficit mice induced by scopolamine were investigated. Results : 1. ${\beta}$-amyloid treatment on cultured spinal cord cells increased both GFAP-staining intensity of astrocytes and caspase 3 immunoreactivity on cultured cells. Then, Nokyongdaebo-tang(Lurongdabutang) treatment reduced the labeling intensity for both GFAP and caspase 3 proteins in culture cells. 2. Scopolamine treatment into mice increased levels of GFAP-positive astrocytes and caspase 3-labeled cells of the hippocampal subfields dentate hilar region, CA3 and CA1 area. In vivo administration of Nokyongdaebo-tang(Lurongdabutang) attenuated labeling intensity for those two proteins in the same hippocampal areas. Similar effects were observed by the treatment of galanthamine, an inhibitor of acetylcholinesterase. Conclusions : This experiment shows that the Nokyongdaebo-tang(Lurongdabutang) may play a protective role in damaged neural tissues. Since neuronal damage seen in degenerative brains such as AD are largely unknown, the current data may provide possible insight into therapeutic strategies for AD treatments. Nokyongdaebo-tang(Lurongdabutang) might be effective for the prevention and treatment of AD.

사리풀 (Hyoscyamus niger L.) 모상근의 Tropane Alkaloid 생성에 미치는 배지, 배양주기, Sucrose 및 Dextrose의 영향 (Effects of Media, Culture Peroids, Sucrose and Dextrose on Tropane Alkaloid Production in Hairy Root Cultures of Hyoscyamus niger L.)

  • Agrobacterium tumefaciens $A_4$T에 의해 형질전환된 사리풀 (Hyoscyamus niger L.)의 모상근으로부터 tropane alkaloids (scopolamine, hyoscyamine)의 생산성 향상을 위한 최적배지, 배양기간 및 탄수화물의 영향을 조사하였다. Tropane alkaloids의 생산성이 우수한 모상근 (HNl8, HN57)의 최적 배지는 SH배지이며, 모상근의 생장, tropane alkaloids 함량 및 생산성에서 조사한 6가지 배지보다 높게 나타났다. Tropane alkaloids생산을 위한 최적 배양기간은 HNl8 clone이 6주, HN57 clone은 5주로 나타났으며, 배양기간에 따라 scopolamine의 함량은 계속적으로 증가하는데 반하여 hyoscyamine의 함량은 일정 수준을 유지하는 것으로 나타났다. 두 세포주는 sucrose농도가 7%까지 증가할수록 생장률이 증가하는 반면, tropane alkaloids 함량은 감소하였으며, tropane alkaloids생산성은 3% sucrose처리구에서 가장 높게 나타났다. HN18 clone의 경우 최적 sucrose(3%)및 dextrose (2%)에서 유사한 tropane alkaloids 생산성을 나타내었다. 하지만 HN57 clone은 sucrose 처리구에 비하여 dextrose 처리구에서 매우 낮은 tropane alkaloids생산성을 나타내었다.

Effect of Black Ginseng on Memory Improvement in the Amnesic Mice Induced by Scopolamine

  • This study compared the effects of black, white, and red ginseng extracts (WGE, RGE, BGE, 200 mg/kg, p.o.) on learning and memory deficits associated with scopolamine treatment (SCOP, 2 mg/kg, i.p.). Tacrine (THA, 10 mg/kg, p.o.) was used as a positive control. Ginseng significantly reversed SCOP-induced memory impairment in the passiveavoidance test and also reduced escape latency in training trials of the Morris water maze test. The increased acetylcholinesterase (AChE) activity produced by SCOP was significantly inhibited by WGE and RGE (p<0.001). SCOP administration had no effect on choline acetyltransferase (ChAT) activity, but RGE and BGE significantly increased ChAT activity (p<0.05). SCOP administration increased oxidative damage in the brain. Treatment of amnesic mice with ginseng extracts decreased malondialdehyde (MDA) levels and restored superoxide dismutase (SOD) and catalase (CAT) activity to control levels. These results suggest that black ginseng enhances cognitive activity by regulation of cholinergic enzymes and antioxidant systems.

사이프러스 에센셜 오일의 흡입이 전임상 실험동물의 손상된 학습능력과 기억력에 미치는 영향 (Cypress Essential Oil Improves Scopolamine-induced Learning and Memory Deficit in C57BL/6 mice)

  • Objectives : Increasing evidence supports the biological and pharmacological activities of essential oils on the central nervous system such as pain, anxiety, attention, arousal, relaxation, sedation and learning and memory. The purpose of present work is to investigate the protective effect and molecular mechanism of cypress essential oil (CEO) against scopolamine (SCO)-induced cognitive impairments in C57BL/6 mice. Methods : A series of behavior tests such as Morris water maze, passive avoidance, and fear conditioning tests were conducted to monitor learning and memory functions. Immunoblotting and RT-PCR were also performed in the hippocampal tissue to determine the underlying mechanism of CEO. Results : SCO induced cognitive impairments as assessed by decreased step-through latency in passive avoidance test, relatively low freezing time in fear conditioning test, and increased time spent to find the hidden platform in Morris water maze test. Conversely, CEO inhalation significantly reversed the SCO-induced cognitive impairments in C57BL/6 mice comparable to control levels. To elucidate the molecular mechanisms of memory enhancing effect of CEO we have examined the expression of brain-derived neurotrophic factor (BDNF) in the hippocampus. CEO effectively elevated the protein as well as mRNA expression of BDNF via activation of cAMP response element binding protein (CREB). Conclusions : Our findings suggest that CEO inhalation effectively restored the SCO-impaired cognitive functions in C56BL/6 mice. This learning and memory enhancing effect of CEO was partly mediated by up-regulation of BDNF via activation of CREB.
