Characterization of Sclerotinia sclerotiorum Isolated from Paprika

  • A fungal isolate collected from infected paprika (Capsicum annuum var. grossum) was characterized as Sclerotinia sclerotiorum based on its ability of sclerotium formation, physiological and molecular properties. When the isolate was grown on potato dextrose agar, oatmeal agar, and malt extract agar, it grew most well on PDA. Optimal temperature and pH for its growth were $25^{\circ}C$ and pH 7, respectively. The fungal isolate produced sclerotia on PDA within 10 days, and the color and shape of the sclerotia were similar to those of S. sclerotiorum. The ITS rDNA regions including ITS1 and ITS2 and 5.8S sequences were amplified using ITS1F and ITS4 primers from the genomic DNAs of the paprika isolate and other known pathogenic S. sclerotiorum isolated from different crops in Korea, and their nucleotide sequences were determined. Sequence comparison analysis showed the ITS rDNA of the paprika isolate shares 100% sequence identity with those of S. sclerotiorum isolated from red pepper, lettuce and a S. sclerotiorum isolate registered in GenBank DNA database. Neighbor joining analysis based on the ITS rDNA sequence revealed the paprika isolate has very close phylogenetic relationships with known Sclerotinia sclerotiorum isolates. This is the first report that S. sclerotiorum has been found associated with paprika rot in paprika growing countries.

Biological control of Lettuce Sclerotinia rot using Bacillus mojanvinensis Pro-EB 15 strain.

  • This studies were investigated the occurrence of Sclerotinia rot by Sclerotinia Sclerotiorum at the lettuce field in Uiryeong-Gun, Gyeongsangnam-Do and were isolated the most effective microorganism for the biological control to the pathogen, S. sclerotiorum YR-1 strain from diseased soil and lettuce leaves. For the pathogenicity test, the most suitable inoculmn density of YR-1 strain was selected as the mycelial suspension of 40m1 showing disease incidence of 80%, and the symptom showed as same as at the fields, the leaves and stem had rotten and developed white downy mycelial at the diseased lesion on the leaves and stems, and produced black and irregular sclerotinia. On the PDA dual test, about 300 isolates were examined the antifungal activity to the pathogen, YR-1 strain, and among them, A-2, A-7, and RH-4 strain were selected most effective antagonistic bacteria. At pots test, the control value of A-7 strain showed the highest value as 85% which was more effective than that of others in a growth chamber. For the promotion of control effect, the selected 3 isolates were spayed on the lettuce leaves as a sole and/or mixed treatments in a growth chamber, the mixed treatment of A-7 and RH-4 strain showing the control value of 90% was most effective than that of sole treatment with A-7 or RH-4 strain showing the control value of 80%, respectively and mixed treatment with A-2 and A-7 strain and A-2, A-7 and RH-4 strain. In addition, 3 bacteria re-isolated from diseased soils, and all of the selected 6 isolates investigated the control effect at pots in a growth chamber, According to the results, A-7 and Pro-EB 15 strain showed the control value of 91.0% and 90.1% respectively, and they were selected most effectual antagonistic bacteria to control lettuce sclerotinia rot and identified as the Bacillus mojanuinensis by 16s RNA analysis. This is the first report on the biological control using by B. mojanvinensis to the lettuce Sclerotinia rot.

Newly Recorded Problematic Plant Diseases in Korea and Their Causal Pathogens

  • Since 1993, a total of 50 problematic plant diseases unrecorded in Korea were surveyed in Gyeongnam province. Totally 34 new host plants to corresponding pathogens investigated in this study were 5 fruit trees, 9 vegetables, 12 ornamental plants, 3 industrial crops, and 5 medicinal plants. Among the newly recorded fruit tree diseases, fruit rot of pomegranate caused by Coniella granati and Rhizopus soft rot of peach caused by Rhizopus nigricans damaged severely showing 65.5% and 82.4% infection rate. Among the vegetable diseases, corynespora leaf spot of pepper caused by Corynespora cassiicola and the crown gall of pepper caused by Agrobacterium tumefaciens, powdery mildew of tomato caused by Oidiopsis taurica were the most severe revealing 47.6%, 84.7%, and 54.5% infection rate in heavily infected fields, respectively. In ornamental plants, collar rot of lily caused by Sclerotium rolfsii, gray mold of primula caused by Botrytis cinerea, soot leaf blight of dendrobium caused by Pseudocercospora dendrobium, sclerotinia rot of obedient plant caused by Sclerotinia sclerotiorum showed 32.7 to 64.8% disease incidence. On three industrial plants such as sword bean, broad bean, and cowpea, eight diseases were firstly found in this study. Among the diseases occurring on broad bean, rust caused by Uromyces viciae-fabae and red spot caused by Botrytis fabae were the major limiting factor for the cultivation of the plant showing over 64% infection rate in fields. In medicinal plants, anthracnose of safflower caused by Collectotrichum acutatum was considered the most severe disease on the plant and followed by collar rot caused by Sclerotium rolfsii.(중략)

파드득나물 균핵병균(Sclerotinia sclerotiorum)의 균핵 헝성 및 생존에 미치는 온도의 영향 (Effect of Temperature on Sclerotia Formation and Viability of Sclerotinia sclerotiorum Causing Sclerotiorum Rot of Cryptotaenia japonica)

  • 남양주, 양주, 양평에서 파드득나물 균핵병의 발생생태를 조사한 결과, 균핵병은 늦가을부터 초봄까지 주로 동계기간에 발생했으며, 지역별로는 양평에서 발생이 심했다. 포장에서 분리한 파드득나물 균핵병균은 기보고된 Sclerotinia sclerotiorum의 균학적 특성과 일치하였고, 균사생장은 5~3$0^{\circ}C$에서 이루어졌으며, 15~3$0^{\circ}C$에서 활발하게 생장하였다 균핵형성은 20~$25^{\circ}C$에서 많이 형성되었으며, 낮은 온도에서 형성된 균핵의 크기가 큰 경향이었다. 균핵생존율은 균핵크기가 작을수록(~2 mm > 3~4 mm > 5 mm~) 높았으며, 치상온도에서의 정치시간이 길수록 높게 나타났다.

길항세균을 이용한 상추 균핵병의 생물학적 방제 (Biological Control of Sclerotinia sclerotiorum in Lettuce Using Antagonistic Bacteria)

  • 상추 균핵병균에 대하여 길항력을 나타내는 세균을 분리하기 위해 2007년부터 2008년에 걸쳐 초봄에 경기도 남양주시 관내 상추 시설재배지에서 균핵병이 발병한 포장의 토양을 수집하였다. 수집한 토양을 희석평판법을 이용하여 총 196개의 세균을 분리하였고, 분리한 세균을 실험실에서 균핵병균(Sclerotinia sclerotiorum)과의 대치배양을 통해 균사생장억제율이 80% 내외로 우수한 길항력을 나타내는 26개 균주를 선발하였다. 선발된 26개 균주를 16S rDNA염기서열분석으로 동정한 결과 Bacillus megaterium, B. cereus, B. muralis, B. velezensis, Arthrobacter nicotianae, A. oryzae, Pseudomonas fuscovaginae, P. flavescens, Stenotrophomonas maltophilia, Sphingobacterium faecium으로 동정되었다. 길항세균의 균사생장억제율이 높게 나타낸 10개 균주를 대상으로 여름철 포트실험을 통해 균핵 생존에 미치는 영향을 조사한 결과 B. cereus(C210)와 B. megaterium(DK6)이 각각 20%와 35%의 균핵 생존율을 나타냈다. 겨울철에 온실 내에서 포트실험을 통해 상추 균핵병에 대해 방제효과를 조사한 결과 대조구가 80%의 발병율을 나타냈으나, 26개 균주 중 9개 균주가 20% 정도의 발병율을 나타내 향후 상추 균핵병의 생물적 방제를 위한 길항세균으로서의 활용가치가 기대된다.

상추 균핵병 생물적방제를 위한 Brevibacillus brevis B23과 Bacillus stearothermophilus B42의 선발 (Selection of Brevibacillus brevis B23 and Bacillus stearothermophilus B42 as Biological Control Agents against Sclerotinia Rot of Lettuce)

  • 버섯폐배지로부터 잡균의 증식을 최소화하기 위해 버섯폐배지현탁액을 nutrient agar(NA) 배지에 직접 혼합하여 분주한 결과 배양기에 출현한 균체수가 희석평판법에 의한 것보다 훨씬 적었으며 형태적으로 Bacillus 속 세균이 분리될 확률이 높았다. 대장균(E. coli) $DH5{\alpha}$에 대하여 항균력을 보이는 1차 분리 균주를 신령버섯폐배지에서 12균주, 양송이폐배지에서 10균주, 느타리폐배지는 20균주 등 총 42균주를 분리하였다. 1차 선발한 균주를 최소 배지상에서 8종의 중요 식물병원균과 대치 우수한 길항력을 나타내는 17개의 균주를 2차 선발하였는데 이 들 중에서 배지상에서 생장이 우수하고 중요 식물병원균에 대하여 넓은 길항력을 가지는 B23과 B42균주를 최종 선발하였다. 이들은 각각 Brevibaciilus brevis와 Bacillus stearothermophilus로 동정되었다. 이들 길항균의 효과를 높이고 안전화를 위해서 유기농업에서 활용가능한 전분과 글리세린 및 계란노른자를 보조제로 선발하였다. 길항균과 이들 보조제를 혼합할 경우 대부분의 처리에서 세균의 밀도가 30일간 효과적으로 유지되거나 증가되었다. B23 단독 처리의 경우에는 상추균핵병의 발병율이 약 28% 정도였으나 전분(starch)과 글리세롤(glycerol) 및 계란노른자(Egg yolk)를 각각 0.2% 혼합 처리시는 발병율이 5% 이하로 나타나 길항세균 단독으로 처리하였을 경우보다 상추균핵병원의 방제 상승효과를 확인하였다.

길항미생물 Bacillus subtilis GG95를 이용한 상추 균핵병의 생물학적 방제 (Biological Control of Lettuce Sclerotinia Rot by Bacillus subtilis GG95)

  • 상추재배시 문제가 되고 있는 균핵병과 시들음병에 대한 미생물 방제제 개발을 위하여 병해가 발생된 상추 포장에서 미생물을 분리하고, 길항미생물을 선발하여 균의 특성을 조사하고 균핵병에 대한 예방효과를 검정한 결과는 다음과 같다. 경기도 주요 상추 재배지에서 균핵병은 10월부터 발생하여 다음해 5월까지 발생하였으며, 12월에는 최대 40%까지 발생하여 피해를 주고 있었다. 균핵병과 시들음병에 모두 억제효과가 있는 균주를 대치배양을 통해 선발하였다. 선발됨 균주는 Vitek 2의 BCL카드를 이용한 생화학적반응과 형태적 확인, 16s rRNA 분석을 통해 동정한 결과 Bacillus subtilis로 동정되었고 B. subtilis GG95로 명명하였다. B. subtilis GG95의 배양특성을 조사한 결과 온도 $25{\sim}30^{\circ}C$, 산도 7의 배양조건에서 배양이 잘 되었으며, 배양에 적정한 영양원은 탄소원 전분, 질소원 tryptone과 yeast extract를 무기염류 $MgCl_2$를 사용하였을 때 가장 잘 되었다. 영양원에 따라 균핵병과 시들음병에 대한 길항효과가 좋은 탄소원은 fructose, maltose였고 질소원은 peptone였으며, 무기염류는 균핵병은 $CaCl_2$, $K_2HPO_4$, 시들음병은 $MgSO_47H_2O$로 병원균에 대한 길항효과가 차이가 있었다. 상추 균핵병에 대한 B. subtilis GG95의 포장검정 결과 $1{\times}10^8cfu/mL$ 농도의 길항미생물 배양액을 포기당 10 mL씩 3회 관주하고 균핵병원균을 접종한 후 균핵병의 발병정도를 조사한 결과 88% 방제효과가 나타났다.

Biological Control of Soil-borne Diseases with Antagonistic Bacteria

  • Biological control has many advantages as a disease control method, particularly when compared with pesticides. One of the most important benefits is that biological control is an environmental friendly method and does not introduce pollutants into the environment. Another great advantage of this method is its selectivity. Selectivity is the important factor regarding the balance of agricultural ecosystems because a great damage to non target species can lead to the restriction of natural enemies' populations. The objective of this research was to evaluate the effects of several different bacterial isolates on the efficacy of biological control of soil borne diseases. White rot caused by Sclerotium cepivorum was reported to be severe disease of garlic and chive. The antifungal bacteria Burkholderia pyrrocinia CAB08106-4 was tested in field bioassays for its ability to suppress white rot disease. In field tests, B. pyrrocinia CAB08106-4 isolates suppressed white rot in garlic and chive, with the average control efficacies of 69.6% and 58.9%, respectively. In addition, when a culture filtrate of B. pyrrocinia CAB08106-4 was sprayed onto wounded garlic bulbs after inoculation with a Penicillium hirstum spore suspension in a cold storage room ($-2^{\circ}C$), blue mold disease on garlic bulbs was suppressed, with a control efficacy of 79.2%. These results suggested that B. pyrrocinia CAB08106-4 isolates could be used as effective biological control agents against both soil-borne and post-harvest diseases of Liliaceae. Chinese cabbage clubroot caused by Plasmodiophora brassicae was found to be highly virulent in Chinese cabbage, turnips, and cabbage. In this study, the endophytic bacterium Flavobacterium hercynium EPB-C313, which was isolated from Chinese cabbage tissues, was investigated for its antimicrobial activity by inactivating resting spores and its control effects on clubroot disease using bioassays. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivated the resting spores of P. brassicae, with the control efficacies of 90.4%, 36.8%, and 26.0%, respectively. Complex treatments greatly enhanced the control efficacy by 63.7% in a field of 50% diseased plants by incorporating pellets containing organic matter and F. hercynium EPB-C313 in soil, drenching seedlings with a culture solution of F. hercynium EPB-C313, and drenching soil for 10 days after planting. Soft rot caused by Pectobacterium carotovorum subsp. carotovorum was reported to be severe disease to Chinese cabbage in spring seasons. The antifungal bacterium, Bacillus sp. CAB12243-2 suppresses the soft rot disease on Chinese cabbage with 73.0% control efficacy in greenhouse assay. This isolate will increase the utilization of rhizobacteria species as biocontrol agents against soft rot disease of vegetable crops. Sclerotinia rot caused by Sclerotinia sclerotiorum has been reported on lettuce during winter. An antifungal isolate of Pseudomonas corrugata CAB07024-3 was tested in field bioassays for its ability to suppress scleritinia rot. This antagonistic microorganism showed four-year average effects of 63.1% of the control in the same field. Furthermore, P. corrugata CAB07024-3 has a wide antifungal spectrum against plant pathogens, including Sclerotinia sclerotiorum, Sclerotium cepivorum, Botrytis cinerea, Colletotrichum gloeosporioides, Phytophotra capsici, and Pythium myriotylum.
