Molecular Cloning and Characterization of the Yew Gene Encoding Squalene Synthase from Taxus cuspidata

  • The enzyme squalene synthase (EC 2.5.1.21) catalyzes a reductive dimerization of two farnesyl diphosphate (FPP) molecules into squalene, a key precursor for the sterol and triterpene biosynthesis. A full-length cDNA encoding squalene synthase (designated as TcSqS) was isolated from Taxus cuspidata, a kind of important medicinal plants producing potent anti-cancer drug, taxol. The full-length cDNA of TcSqS was 1765 bp and contained a 1230 bp open reading frame (ORF) encoding a polypeptide of 409 amino acids. Bioinformatic analysis revealed that the deduced TcSqS protein had high similarity with other plant squalene synthases and a predicted crystal structure similar to other class I isoprenoid biosynthetic enzymes. Southern blot analysis revealed that there was one copy of TcSqS gene in the genome of T. cuspidata. Semi-quantitative RT-PCR analysis and northern blotting analysis showed that TcSqS expressed constitutively in all tested tissues, with the highest expression in roots. The promoter region of TcSqS was also isolated by genomic walking and analysis showed that several cis-acting elements were present in the promoter region. The results of treatment experiments by different signaling components including methyl-jasmonate, salicylic acid and gibberellin revealed that the TcSqS expression level of treated cells had a prominent diversity to that of control, which was consistent with the prediction results of TcSqS promoter region in the PlantCARE database.

Keratin 17 identified by proteomic analysis may be involved in tumor angiogenesis

  • Angiogenesis is crucial for solid tumor growth. By secreting angiogenic factors, tumor cells induce angiogenesis. However, targeting these angiogenic factors for cancer therapy is not always successful, suggesting that other factors may be involved in tumor angiogenesis. This work shows that 25 protein spots were differentially expressed by two-dimensional gel electrophoretic analysis when HepG2 cells induced endothelial cell differentiation to tube in vitro, and most of them were upregulated. Twenty-one proteins were identified with MALDITOF-MS, and the other four were identified by LTQ-MS/MS. Keratins were identified as one class of these upregulated proteins. Further study indicated that the expression of keratin 17 in cultured endothelial cells is likely microenvironment regulated, because its expression can be induced by HepG2 cells and bFGF as well as serum in culture media. Increased expression of keratins in endothelial cells, such as keratin 17, may contribute to the angiogenesis induced by HepG2 cells.

Mediating Effect of Self-identity on the Relationship between Aggression and School Life Adaptation of Early Adolescent in Single Parent Families (한부모가정 초기 청소년의 공격성이 자아정체성을 매개로 학교생활적응에 미치는 영향)

  • Objectives: This study was conducted to analyze the mediating effect of self-identity on the relationship between aggression and school life adaptation of early adolescents. Methods: This study used the 5th-year data of the Korean Children & Youth Panel Survey (KCYPS) including 159 middle school 2nd students from single-parent families. The data was analyzed with open source statistics program R 3.5.0 to determine whether self-identity had a mediating effect on school life adaptation as an independent variable. Results: A moderate correlation was found among aggression, self-identity, and school life adaptation. As a result of the Sobel test, self-identity of early adolescents demonstrated a significant mediating effect on the relationship between aggression and school life adaptation. Conclusions: Given the significant mediating factor of self-identity, the strategies for early adolescents to boost their overall school life adaptation should be considered through intervention programs that help to enhance their self-identity.

The Effect of Forest Experience on School Life Satisfaction of Korean Boarding Middle School Students

  • We surveyed to assess the impact of the mood states and sentiments resulting from forest experience on the school life satisfaction of Korean boarding middle school students (n=90). The average mood state score was 2.32 on a scale of 1-5, with the subscale of vigor scoring the highest (3.19). The average sentiment score was 3.08, with the subscale of subjective happiness scoring the highest (3.08). The average school life satisfaction score was 3.66, with the subscale of interpersonal relationships scoring the highest (3.91). The mood states resulting from forest experience explained 39.6% of the variance in school life satisfaction, and the regression model showed a significant fit (F=9.077, p<0.001). Similarly, sentiments resulting from forest experience explained 47.4% of school life satisfaction, with the regression model showing a significant fit (F=10.569, p<0.001). It can be concluded that forest experience positively impacts the school life satisfaction of Korean boarding middle school students.

Study on School Life Adaptation of Middle School Students who are Beneficiary of Educational Welfare Service (교육복지서비스 이용 중학생의 학교생활적응에 관한 연구)

  • The purpose of this study is to examine if there is any difference due to the general characteristics of middle school students who are beneficiaries of educational welfare services and a special quality of use about educational welfare services. Additionally, the examination has been performed to understand the relationship that exists between the satisfaction about educational welfare services, and adaptation to school life and self-respect of middle school students who are beneficiaries of educational welfare services. The results of the study showed meaningful higher scores by male students than female students only in the environmental adaptation area among lower areas in school life adaptation due to sex. Due to school year, third-year students showed meaningful high scores in school life adaptation. The satisfaction about programs in each area of educational welfare services of middle school students who are beneficiaries of educational welfare services showed a meaningful static relationship with school life adaptation. The inspection of the relationship between satisfaction about educational welfare services and 5 lower areas of school life adaptation showed statistically meaningful results in the order of adaptation to the teacher, adaptation to the environment and adaptation to the class, which are lower areas. The examination on the relationship between self-respect and adaptation to school life showed that self-respect has a static relationship with all lower areas of school life and overall school life adaptation. However, no meaningful relationship was shown between the satisfaction about educational welfare services and self-respect. Also, it was found that the factors of grade, self-respect, the number of service utilization have an effect on school life adaptation. This study has some limitations. But in this study, which is different from other studies which dealt with students who had used partial areas of educational welfare services, students who were using all 4 areas of educational welfare services were selected as subjects. In addition, this study is significant in that basic data has been offered for establishment of a policy on educational welfare services in middle schools in the future.

Comparative Genomics Uncovers the Genetic Diversity and Synthetic Biology of Secondary Metabolite Production of Trametes

  • The carbohydrate-active enzyme (CAZyme) genes of Trametes contribute to polysaccharide degradation. However, the comprehensive analysis of the composition of CAZymes and the biosynthetic gene clusters (BGCs) of Trametes remain unclear. Here, we conducted comparative analysis, detected the CAZyme genes, and predicted the BGCs for nine Trametes strains. Among the 82,053 homologous clusters obtained for Trametes, we identified 8518 core genes, 60,441 accessory genes, and 13,094 specific genes. A large proportion of CAZyme genes were cataloged into glycoside hydrolases, glycosyltransferases, and carbohydrate esterases. The predicted BGCs of Trametes were divided into six strategies, and the nine Trametes strains harbored 47.78 BGCs on average. Our study revealed that Trametes exhibits an open pan-genome structure. These findings provide insights into the genetic diversity and explored the synthetic biology of secondary metabolite production for Trametes.

New Recorded Species in Three Genera of the Sordariomycetes in Korea

  • In an ongoing survey of Korean indigenous fungi, three fungal strains belonging to the Sordariomycetes were isolated from soil samples. These strains were designated KNU16-001, KNU16-002, and KNU16-009, and identified as Ambrosiella grosmanniae, Acremonium sclerotigenum, and Trichocladium asperum, respectively, based on morphological characterization and phylogenetic analysis using internal transcribed spacer region sequences of ribosomal DNA. This is the first report of these species in Korea.

  • Purpose: The purpose of this study was to explore and understand life and meaning in life of elementary school students. Methods: Data from 1,790 higher grade elementary school students were analyzed for content of meaning in life. Results: The students described the following: 1) Meanings of living existence included, 'living things', 'importance', 'human life', 'family', 'event', 'eternity', 'origin of life', 'disease', and, 'protection'. 2) Four categories were found for reasons why human life is important, 'solitary', 'limitation', 'valuables', and 'living'. 3) Things that one can do to protect the value of human life were, 'doing safely', 'keep things well', 'altruistic behavior', 'doing sound activity', 'preservation & development', and 'devotion of oneself'. 4) Meaning of life included 'to accomplish plans', and 'to do things' 5) Cases when students felt life had meaning were 'perform meaningful behavior', 'comfortable emotionally', and 'receiving help from others'. 6) The methods by which to live a meaningful life were 'working diligently', 'serving', and 'doing rightly'. Conclusion: These results suggest that students recognize the meaning of life in a most positive way and with a relatively wide scope, but as a few students had a negative orientation, age-appropriate and more systematic education programs are needed.

Effects of Logotherapy on Life Respect, Meaning of Life, and Depression of Older School-age Children (의미요법을 적용한 생명존중 교육 프로그램이 초등학생의 삶의 의미, 생명존중 인식, 우울에 미치는 효과)

  • Purpose: This study was done to identify effects of a life-esteemed education applied logotherapy on life respect, meaning of life, and depression on older elementary school students. Methods: A nonequivalent control group and non-synchronized design was conducted with a convenience sample of 142 students. The program named 'My Precious Life'. consisted of one session per week for five weeks. Students were assigned to the experimental group (n=70) or the control group (72). Data were analyzed using descriptive statistics, Chi-square, Fisher's exact test, t-test, and repeated measured ANOVA with the SPSS/PC 18.0 program. Results: Meaning of life and life respect increased significantly and depression decreased significantly for participants in the experimental group. Conclusion: The results of the study indicate that life-esteemed education applied logotherapy is effective in improving meaning of life and life respect and in decreasing depression in elementary school students. It can also be used to prevent existential distress and to intervene as a motif for having hope in life.
