부위별 꽈리(Physalis alkekengi var. francheti) 추출물의 항산화효과 (Antioxidative Activities of Different Part Extracts of Physalis alkekengi var. francheti (Winter Cherry))

  • 본 실험에서는 꽈리를 열매, 꽃받침, 잎, 줄기, 뿌리 등 부위별로 구분하여 메탄올로 추출하고 추출 수율, 총페놀 함량, 총플라보노이드 함량, DPPH radical 소거능, 아질산염 소거능, superoxide anion radical 소거능, 환원력, 철 이온에 대한 킬레이트 효과 등을 측정하였다. 추출 수율은 열매가 가장 높았고 그 다음으로 꽃받침, 잎, 줄기, 뿌리 순이었다. 총페놀 및 총플라보노이드 함량은 잎 추출물이 가장 높았고 그 다음으로 꽃받침 추출물이었다. DPPH radical 소거능은 1 mg/mL 및 5 mg/mL의 농도에서 잎 추출물이 가장 높았고 꽃받침 추출물도 높은 활성을 나타내었다. 아질산염 소거능은 모든 부위에서 농도에 따라 유의적인 차이 없이 높게 나타났다. 꽈리 추출물의 superoxide anion radical 소거능은 꽃받침과 잎 추출물이 높은 활성을 나타내었다. 환원력은 잎 추출물에서 가장 높게 나타났고 철 이온에 대한 킬레이트 효과는 꽃받침, 줄기 및 뿌리 추출물이 높은 활성을 나타내었다. 이상의 결과를 종합하여 보면 꽈리의 잎 추출물에서 총 페놀 함량, 총플라보노이드 함량과 항산화 활성이 탁월하게 높게 나타났고 그 다음으로 꽃받침 추출물이 우수한 항산화 활성을 나타내어 향후 이를 이용한 천연 항산화제로 개발 가능성이 시사되었다.

  • Water and methanolic extracts of Ecklonia cava, a marine brown alga, were prepared by ultrasonic extraction (UE) and conventional extraction (CE) methods. The radical-scavenging activity and the inhibitory effects against hydrogen peroxide ($H_2O_2$)-induced DNA damage of the extracts were investigated. All extracts prepared by CE exhibited higher total polyphenolic content than that in the extracts prepared by UE. Extraction yield and total phenolic content increased as the UE time increased. The radical-scavenging activities increased as the UE time increased. All extracts prepared by CE exhibited higher 1,1-diphenyl-2-pricrylhydrazyl (DPPH) and hydroxyl radical-scavenging activities than did those prepared by UE. Extracts prepared by UE showed stronger scavenging activities on alkyl radical and $H_2O_2$ than those prepared by CE did. Methanolic extract with UE 12 h (100MEU-12h) and methanolic extract with CE 24 h (100MEC-24h) were selected and evaluated by comet assay for their inhibitory effect against $H_2O_2$-induced DNA damage. 100MEU-12h showed slightly greater protective effect against $H_2O_2$-induced DNA damage than 100MEC-24h. Thus, UE can be effectively used as a seaweed extraction technique, and there is potential for scale-up of the extraction process.

미숙감귤의 항산화 및 항노화 활성에 대한 평가 (Evaluation on antioxidative and antiaging effect of immature fruits from Citrus unshiu Markovich)

  • Objective : This study was designed to investigate effect of immature fruits from Citrus unshiu Markovich to mature them on antioxidative and antiaging-related enzyme activities in vitro. Methods : Fruits from Open field-cultivated C. unshiu Markovich (ripe and unripe fruits of Citri) made a purchase in Jeju island. We measured total polyphenol and flavonoid contents in mature C. unshiu Markovich (MC) and immature C. unshiu Markovich (IMC) 70% ethanol extract. DPPH free radical, ABTS radical, and hydroxyl radical scavenging activities in both extracts were determined. Also, reducing power, SOD-like activities, elastase inhibition, and collagenase inhibition activities of these extract were assayed at 5, 1, 0.5, and 0.1 mg/㎖. Results : The total polyphenol contents in the ethanol extract of IMC were higher than those of MC (26.99 ± 0.89 mg/TAEg). And total flavonoid contents in them were very similar to both extract (MC; 7.25 ± 0.4 mg/RUEg and IMC; 7.44 ± 0.18 mg/RUEg). DPPH free radical scavenging activities, ABTS radical scavenging capacity, reducing power, and SOD-like activities of IMC-treated group showed significant increase compared to those of MC group in all treated concentrations. Particularly, DPPH free radical scavenging and elastase inhibition activities in IMC-treated group at 5 mg/㎖ showed similar with positive control group. Besides, IMC extract at 5 mg/㎖ concentration surpassed positive control (BHA or Vit. C) in SOD-like activities and ABTS radical scavenging capacity. Conclusion : These results suggest that IMC ethanol extract may be used as a natural antioxidant and a antiaging material for development of functional foods and cosmeceuticals.

정향 및 목향과 백지를 혼합한 한약재 에탄올 추출물의 항산화 효능 (Anti-oxidant effect of ethanol extract from mixture including Caryophylli Flos, Aucklandiae Radix and Angelicae Dahuricae Radix)

  • Objectives : Herbal medicinal mixture (JMB) are consisted of Caryophylli Flos, Aucklandiae Radix and Angelicae Dahuricae Radix. Each of herbal medicines has studied on anti-oxidant effect. So this study was conducted to investigate efficacy and potency of JMB on anti-oxidation. Methods : The JMB was extracted at room temperature by 80% ethanol. And total polyphenol contents, total flavonoid contents in JMB ethanol extract were determined by colorimetric method. Also, DPPH, ABTS free radical scavenging capacity and reducing power of JMB ethanol extract were measured at 100, 500, 1000, $5000{\mu}g/m{\ell}$ concentrations by spectrometric assay. Positive control was used BHA (butylated hydroxyanisole). Results : The total polyphenol contents and total flavonoid contents of the extract were 55.38 mg/TAEg, 513.72 mg/RUEg, respectively. Also, DPPH free radical scavenging capacity and reducing power of JMB ethanol extract in treated concentrations (100, 500, 1000, $5000{\mu}g/m{\ell}$) increased dose dependently. In particular, DPPH free radical scavenging capacity of JMB ethanol extract at 500, 1000, $5000{\mu}g/m{\ell}$ was similar to positive control (BHA) at high concentration (50, $100{\mu}g/m{\ell}$). ABTS free radical scavenging capacity of JMB ethanol extract at 500, 1000, 5000 ug/ml was similar to BHA at high concentration (50, $100{\mu}g/m{\ell}$). Also, reducing power was showed that JMB ethanol extract at $5000{\mu}g/m{\ell}$ was similar to BHA at high concentration (50, $100{\mu}g/m{\ell}$). Conclusions : These results suggest that JMB ethanol extract has effects to scavenge free radicals. Therefore, JMB has potential and applicable benefits for development of materials and products to have anti-oxidation functions.

  • A strong antioxidant activity, which was measured by the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl(DPPH) radical, was detected in the methanol extract of Salvia miltiorrhiza Bunge (Labiatae). By activity-directed fractionation, compounds 1 and 2 were isolated as antioxidant principles of S. miltiorrhiza. Compounds 1 and 2 were identified as dimethyl lithospermate and 3-(3, 4-dihydroxyphenyl)lactamide, respectively, on the basis of spectral data. The radical scavenging effect of compounds 1 and 2 on DPPH radical exceeded that of L-ascorbic acid which is a well known antioxidant. These two compounds also showed prominent inhibitory activity against free radical generation in dichlorofluorescein (DCF) method and cytoprotective effect against t-BHP in cultured liver cell.

메밀 지상부의 DPPH 라디칼 소거작용과 활성 플라보노이드의 분리 (DPPH Radical Scavenging Effect of the Aerial Parts of Fagopyrum esculentum and Isolation of Bioactive Flavonoids)

  • The inhibitory effect of the aerial parts of Fagopyrum esculentum on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical was examined. The n-butanol fraction from the methanol extract of title plant showed stronger inhibitory effect than other fractions on DPPH radical. Two flavonoids were isolated from n-butanol fraction having the potent activity and elucidated as quercetin-3-O-${\alpha}$-L-rhamnoside and quercetin-3-O-rutinoside on the basis of spectral evidence. The $IC_{50}$ values of these compounds on DPPH radical were 6.56 ${\mu}M$ and 8.37 ${\mu}M$, respectively.

산초 추출물의 S. mutans 항균활성 및 항산화 효능 연구 (A Study on the Anti-microbial Effect on S. mutans and Anti-oxidant Effect of Zanthoxylum pericarpium Extract)

  • Objectives : The Zanthoxylum pericarpium has been used as oriental spicy seasoning and a medicinal plant from old times. This study was performed to determine the anti-oxidant efficacy of Zanthoxylum pericarpium extract and the anti-microbial effects. Methods : We got Zanthoxylum pericarpium extract using PSE (pressurized solvent extraction) method. The anti-microbial effect of Zanthoxylum pericarpium extract was assessed on Streptococcus mutans(S. mutans) and anti-oxidant effect of the extract was assessed by measuring DPPH radical scavenging activity and SOD like activity. Results : 1. Zanthoxylum pericarpium extract had high anti-microbial activity on S. mutans. 2. DPPH radical scavenging activity significantly increased in the Zanthoxylum pericarpium extract. 3. SOD like activity also significantly increased in the Zanthoxylum pericarpium extract. Conclusions : The PSE extract from Zanthoxylum pericarpium has good anti-microbial and anti-oxidant effects in a concentration-dependent manner.

대추잎 추출물의 생리활성 작용 (Physiological Activity of Zizyphus jujuba Leaf Extracts)

  • This study was designed to investigate the possible utilization of Zizyphus jujuba leaves as a source of functional ingredients. The physiological activity of different solvent fractions prepared from ethanol extract of Zizyphus jujuba leaves were analyzed. Xanthine oxidase inhibitory effect was very high in all fractions except chloroform fraction. The very high electron donating ability was observed in the ethylacetate fraction and the effect was similar to 0.1% tocopherol. Nitrite scavenging effect of all fractions was more than 40% even at low concentration of 1mg/ml and was increased with increasing concentration. Angiotensin I converting enzyme inhibitory activity was appeared in ethyl acetate and chloroform fractions only at high concentraton.

Protective Effect of Brazilin on Cisplatin Nephrotoxicity

  • Cisplatin is one of the most effective antitumor agents currently available for cancer therapy. However, its clinical use has been limited by its severe side effects, especially nephrotoxicity. Therefore, brazilin, which has a radical scavenging effect, was given intraperitoneally to evaluate the effect on cisplatin nephrotoxicity in rats. Remarkable protective effects against nephrotoxicity of cisplatin were observed when brazilin was administered to rats simultaneously with cisplatin. Hepatotoxicity induced by combination treatment of cisplatin and brazilin was evaluated by measuring serum glutamic pyruvate transaminase and serum glutamic oxalate transaminase. Combination treatment did not affect the levels of sGPT and SGOT, and any combination treatment did not induce metallothionein in kidney. Brazilin which has radical scavenging effect directly reduced nephrotoxicity of wisplatin in vivo. Thus, it seems that nephrotoxicity of cisplatin was caused by free radicals. The present results Indicate that brazilin, when it is given with cisplatin, may provide protection against cisplatin nephrotoxicity in rats.

황기추출물의 보습 및 항산화 효과 (Moisturizing and Anti-oxidation Effect of Astragalus membranaceus Root Extract)

  • 화장품 원료로서 황기추출물을 산지 및 추출방법에 따라서 추출한 후, 보습효과 및 항산화효과를 시험하였다. 황기는 산지별로서 제천산, 정선산, 영주산, 태백산 및 중국산 1년 근을 사용하였다. 황기추출물에 포함된 유효성분의 한 지표성분으로서 선정한 formononetin을 HPLC 분석법을 이용하여 분석하였다. 항산화능 평가로서 자유라디칼 및 활성산소 소거효과의 경우에 75% 에탄을 수용액을 이용한 황기추출물은 정제수를 이용한 황기추출물과 비교하여 더 우수한 것으로 나타났다. 75% 에탄을 추출물의 자유라디칼 소거효과는 50% 소거효율에 대한 농도($IC_{50}$)가 2.162 mg/mL이었으며, 활성산소 소거효과의 경우는 $IC_{50}$ 값이 2.981 mg/mL로 나타났다. 특히 에틸아세테이트 분획층에 포함되어 있는 이소플라보노이드는 항산화제로 알려진 제니스테인과 유사한 자유라디칼 소거효과를 나타냈다. 황기추출물의 보습효과는 제천산 황기추출물이 가장 양호하였으며, 초음파추출을 이용한 황기추출물이 상대적으로 양호하였다.
