Development of New Strains of Wolfiporia cocos for Sclerotium Formation by 2-Way Cross-Breeding (이원교배에 의해 균핵 형성하는 복령 균주 개발)

  • Wolfiporia cocos is an important medicinal fungus that has been used in regions of Northeast Asia including Korea, Japan, and China. W. cocos is classified in Korea into two types (red bokryeong and white bokryeong) based on the internal colors (yellow orange-pale pink and white) of the sclerotium. Generally, the W. cocos type cultivated on farms produces white sclerotium. In this study, we endeavored to select strains that form sclerotium in sawdust medium using 2-way cross-breeding among two cultivated strains and three wild strains. Monospores were isolated from the fruiting bodies of cultivated and wild strains on potato dextrose agar. Thirty-nine strains of 338 hybrid strains isolated formed sclerotia with white or yellow colors upon culture for 3 months in Pinus densiflora sawdust medium. Selection for sclerotium forming strains using sawdust culture follows a very simple and easy procedure that is presented for the first time in this paper. We plan to test selected strains in the field to aid in developing new varieties for the future.

Mycelial Growth of Monokaryotic and Dikaryotic Strains of Lentinula edodes Cultivars for Sawdust Cultivation on the Agar and Sawdust Culture media (톱밥재배용 표고 품종의 단핵균사체와 2핵균사체의 한천과 톱밥배지에서의 균사생장)

  • In order to develop the culture material for breeding of Lentinula edodes, dikaryotic and monokaryotic mycelia were cultured in agar and sawdust medium. The cultivars were used Sanjo No. 701, Sanjo No. 705, Chamaram and Chuje No. 2 in this study. The mycelial growth of the cultivars were highest in PDA except for Sanjo No. 705, but Sanjo No. 705 showed the best mycelial growth in MCM. Regardless of the cultivars, the mycelial growth showed the most stable in MEA. There was no significant difference in mycelial growth among the cultivars in sawdust medium, but it was about 10% faster than that of Sanjo No. 701 and 705. Monokaryotic strains of Sanjo No. 701 and Chamaram showed less mycelial growth than dikaryotic mycelium. Dikaryotic mycelium of Chamaram showed better mycelial growth than that of Sanjo No. 701, but monokaryotic mycelium of Chamaram showed lower mycelial growth than Sanjo No. 701. The selected monokaryotic mycelium has a wide varience of mycelial growth, and the morphologies of the colonies are very diverse, so those are presumed that wide variences of monokaryotic are selected in genetically, and these monokaryotic mycelium are expected to be a good breeding materials.

Cultivation of Cauliflower Mushroom (Sparassis crispa) Using Coniferous Sawdust-based Media with Barley Flours (보릿가루가 첨가된 침엽수 톱밥을 이용한 꽃송이버섯 재배)

  • Cauliflower mushroom (Sparassis crispa) is an edible mushroom that shows remarkably high contents of $1,3-{\beta}-D-glucan$ compared to other edible mushrooms. The mushroom is known to give high antitumor and immunologic activities, thus the mushroom is recently cultivated in Japan and Korea. However, cultivation methods are being kept in secret or under patents by some companies with complicated procedures. This study was conducted to solve the problem by providing a simple method for the cultivation of cauliflower mushroom. We could produce the mushroom using sawdust-based media of Larix leptolepis, Pinus densiflora and P. koraiensis with addition of barley flours and sugar. The ratio of the sawdust : barley flours : sugar used for this study was 80:20:3 in dry-weight base, and the water content was adjusted as 65%. The productivity was somewhat different among the species of sawdust media and the strains of Sparassis crispa for the cultivation, and the mushroom production reached up to 177g from 650g base(productivity : 27%). Thus, this medium is strongly recommended as a practical cultivation method for cauliflower mushroom. By the way, further study for the determination of adequate concentration of barley flour and the selection of proper strain for each sawdust species is needed.

Safety investigation of the moisturizing medium prepared using the Chinese oak mushroom (Lentinula edodes) based on the presence of residual pesticides, heavy metals, and radioactive materials (중국산 표고(Lentinula edodes) 보습배지의 잔류농약, 중금속 및 방사능 안전성 분석)

  • In order to provide basic data for preparing management standards and to verify the safety of the Chinese oak mushroom-derived moisturizing medium-which is synthesized and imported in large quantities-the presence of 321 residual pesticides, 7 heavy metals, and 3 radioactive materials was analyzed in the moisturizer samples. Examination of residual pesticides in seven moisturizing medium samples prepared using the Chinese oak mushroom and three domestic sawdust samples used for mushroom culture revealed the presence of cypermethrin and iprodione in three moisturizer samples, but the contents of these pesticides were below the standard limits. Zn was detected in ten samples, Cu was detected in nine samples, and Ni was detected in four samples, but their contents were below the standard limits. Pb, Cd, Cr, and Hg were not detected in any sample. No radioactive materials were detected in the samples. In addition, fruiting bodies of the oak mushroom were observed in each medium. Examination did not reveal the presence of any residual pesticides or harmful compounds. In this study, the use of the moisturizing medium prepared using the Chinese oak mushroom was found to be safe. As residual pesticides, heavy metals, and radioactivity-even in trace amounts-remain concentrated in the human body, continuous verification of the safety of hazardous substances and pollutants during the systematic cultivation and management of these mushrooms is required.

Cultural Characteristics of Veiled Lady Mushroom, Dictyophora spp.

  • This study was carried out to obtain the basic data for artificial culture of veiled lady mushroom (Dictyophora spp). The optimal conditions for the mycelial growth were $25^{\circ}C$ and pH 5.0 for all isolates except the optimal temperature of $30^{\circ}C$ for D. echinovolvata ASI 32002 and Phallus rugulosus. The optimal medium for Dictyophora spp. was PBA (potato bamboo sawdust extract agar) medium. The strain ASI 32002, D. echinovolvata, grew faster than. D. indusiata ASI 32003 and Phallus rugulosus ASI 25007 on the medium. Carbon sources such as glucose, maltose and inuline were favorable for stimulating a mycelial growth of the two strains of ASI 32002 and ASI 32003. Asparagine and glutamine appeared to be favorable to the strain ASI 32002 and ASI 32003, where as alanine, one of nitrogen source also favorable to the strain ASI 32002. The optimum C/N ratio of the two isolates of ASI 32002 and ASI 32003 was about 25 : 1 when 2% glucose as carbon source was mixed with the basal medium. While, in the case of 4% as carbon source, the optimum C/N ratio was about 30 : 1.

Performance Characteristics of Agitated Bed Manure Composting and Ammonia Removal from Composting Using Sawdust Biofiltration System (교반식 축분 퇴비화 및 톱밥 탈취처리 시스템의 퇴비화 암모니아 제거 성능)

  • Sawdust biofiltration is an emerging bio-technology for control of ammonia emissions including compost odors from composting of biological wastes. Although sawdust is widely used as a medium for bulking agent in composting system and for microbial attachment in biofiltration systems, the performance of agitated bed composting and sawdust biofiltration are not well established. A pilot-scale composting of hog manure amended with sawdust and sawdust biofiltration systems for practical operation were investigated using aerated and agitated rectangular reactor with compost turner and sawdust biofilter operated under controlled conditions, each with a working capacity of approximately $40m^3\;and\;4.5m^3$ respectively. These were used to investigate the effect of compost temperature, seed germination rate and the C/N ratio of the compost on ammonia emissions, compost maturity and sawdust biofiltration performance. Temperature profiles showed that the material in three runs had been reached to temperature of 55 to $65^{\circ}C$ and above. The ammonia concentration in the exhaust gas of the sawdust biofilter media was below the maximum average value as 45 ppm. Seed germination rate levels of final compost was maintained from 70 to 93% and EC values of the finished compost varied between 2.8 and 4.8 ds/m, providing adequate conditions for plant growth.

Sawdust Cultivation of Lentinula edodes Using a Detachable Plastic Bottle (분리형 용기를 이용한 표고버섯 톱밥재배 연구)

  • Currently, plastic bags are being used for sawdust cultivation of shiitake mushroom. However, due to serious environmental problems caused by the use of plastic bags, we studied the efficacy of bottle cultivation method to replace the sawdust bag method. Small detachable plastic bottles (400 g capacity) filled with Quercus spp. sawdust and wheat bran (4:1 w/w) media were incubated for 80 and 120 days. The weight loss (%) of the media was higher for the NIFoS 2464 strain at an approximate light intensity of 300 Lux than light intensity of 500 Lux; the light intensity was associated with the loss of sawdust medium-weight during the cultivation period. The highest yield was observed when the strain was cultivated for 80 days under dark conditions, 40 days under 500 Lux light, and air circulation fan speed of 30 rpm. When incubated for 120 days, mushroom yield in the bottle media was higher at 40 days of light exposure than 20 days of light exposure. In the bottle media incubated for 80 days under dark conditions, the mushrooms fruited due to repetitive water spraying on the top of the media and light stimulation during the fruiting period. The media could be separated from the bottles because the media shrank after the first harvest. These separated plastic bottles could be re-used for mushroom cultivation, thereby reducing the amount of plastic waste.

Growth characteristics and productivity of oyster mushroom according to adding of Rhus verniciflua sawdust (옻나무톱밥 첨가에 따른 느타리버섯의 생육 특성 및 수량성)

  • This study was carried out to investigated optimum mixing ratio of Korean natural Rhus verniciflua for production of functional oyster mushroom. Mycerial growth was some fast at addition of R. verniciflua sawdust, and not significant difference by increase of R. verniciflua substrate. Yields of fruiting body show the highest to 150.2 g/850 ml of medium which are addition 20% of R. verniciflua sawdust, and diameter of pileus and thick of stipes were the highest at addition 10% of R. verniciflua sawdust. The L value of stipes were the highest at addition 10% of R. verniciflua sawdust and the L value of pileus were decreased by increase of R. verniciflua substrate, but there was no significant difference in the a-value and the b-value.

Condition of mycelial culture and inoculum volume of spawn on cultivation of Agrocybe cylindracea (버들송이의 균사배양조건 및 최적 접종량 설정)

  • Studies were made to optimize the media composition and cultural condition for mycelial growth of Agrocybe cylindracea. Sawdust spawn of media composition for optimal growth was found to be pine sawdust combination of 30% wheat bran and poplar sawdust combination of 20% corn bran were the best of the optimal combination. The optimal concentration of white sugar was 1.0~1.5%. The nitrogen sources was found to be yeast extract and soybean powder. Also, optimal concentration were $0.7g/{\ell}$ and $0.1g/{\ell}$, respectively. The mineral sources of optimal medium compositions were $MgSO_4{\cdot}7H_2O\;0.3g/{\ell}$, $KH_2PO_4\;0.5g/{\ell}$ and $K_2HPO\;1.2g/{\ell}$. Optimal amount of inoculum for cultivation of A. cylindracea were $20{\sim}25g/850m{\ell}$ and $25m{\ell}/850m{\ell}$ in the sawdust spawn and liquid spawn, respectively.

Fruiting body productivity of Lentinula edodes 'Hwadam' as determined by nutrient supplements and storage period of sawdust spawn (배지영양원 및 톱밥종균 저장기간에 따른 표고 '화담'의 자실체 생산성)

  • This study was conducted to establish an appropriate period of use of sawdust spawn at low temperatures and a nutrient supplement medium for cultivation of Lentinula edodes 'Hwadam'. Of the nutrient supplements, the total yield of rice bran (5%) + corn flour (5%) treatments were 673.3 g, which was higher than rice bran (551.6 g) and wheat bran (546.7 g) treatments, respectively. As shown by the growth of Lentinula edodes 'hwadam' during to the sawdust spawn storage period (at 4℃), the period of spawn running, browning, fruiting body formation, and development was 27 d, 81 d, 5 d, and 11-13 d, respectively, regardless of the length of the storage period at 4 ℃. After 3 months of storage of sawdust spawn, the number of fruiting bodies and yield decreased as the storage period increased. Therefore, the period of use of sawdust spawn (at 4 ℃) for the stable production of fruiting bodies of Lentinula edodes 'Hwadam' was a maximum of 3 months.
