  • Saururus chinensis Baill (Saururaceae) is a perennial plant that has been used in the treatment of edema, jaundice and gonorrhea in Korean folk medicine. This study was carried out to investigate the inhibitive effects of Saururus chinensis Baill (SCB) on lipid metabolism in Sprague-DaweIy rat(SD-rat) accutely exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD). After 7 days from TCDD(1$\mu$g/kg) injection, SCB(200mg/kg) was administered into rats intraperitoneally for 4 weeks. (omitted)

삼백초와 약모밀의 내부형태 비교 연구 (A Comparative Anatomical Study of Saururus chinensis (Lour.) Baill. and Houttuynia cordata Thunb. (Saururaceae))

  • Objectives : Saururus chinensis and Houttuynia cordata (Saururaceae) are perennial herbs using for medicinal purposes in Korea. The objectives of this study are to compare anatomical key characters between two medicinal plants and to provide fundamental information for the identification of two herbal medicines by using anatomical features. Methods : Cross-sections of root, rhizome, stem, petiole, and leaf for each species were observed in this study. Materials were analyzed through dehydration, paraffin embedding and micro-sectioning, and double staining with Safranin O and Fast-Green FCF. Observations of permanent preparation were conducted using light microscope. Results : S. chinensis and H. cordata were distinguished with anatomical differentiations; Idioblasts with essential oil were scattered in the parenchyma cell of cortex, pith, and phloem of S. chinensis, on the other hand, in H. cordata, idioblasts were distributed ring-shaped in the cortex of the root. S. chinensis had two cycles of vascular bundles in the stem while H. cordata had one cycle. Hypodermis layer was conspicuous in a stem of H. cordata, crystals were observed the only parenchyma in a stem of S. chinensis, and epidermal oil cells were developed in the epidermis of H. cordata. S. chinensis had air cavity at the cortex and pith of the stem. The shape of cross-section was polygonal in the stem of S. chinensis and was circular in the stem of H. cordata. Conclusions : We investigated anatomical study of Korean S. chinensis and H. cordata. To identify two herbal medicines, we considered main anatomical features and provided identification key here.

  • 1. The cell viability was determined by MTT method. Their cytotoxic activities against three cancer cell lines such as A549, MDA-MB-231 and SNU-C4 cell line were tested. Among them, The methanol extract of Saururus chinensis Baill. showed the strongest cytotoxic effect against SNU-C4 cells. These results suggest that the methanol extract of Saururus Chinensis Baill. possessed a potential antitumorous agent. 2. In vitro the antitoxic activity of ethanol extract of Saururus Chinensis Bail on NIH 3T3 fibroblasts was evaluate by the MTT (3-(4,5-dimethyl- thiazol-2-yl)-2,5-diphenyl-2H-tetra캐lium bromide) and SRB (sulforhodamine B protein) assays. The number of NIH 3T3 fibroblasts were increased and tend to regenerate. These results suggest that Saururus Chinensis Bail extract retains a potential antitoxic activity. 3. Complexation of Cd (II) ion with ligands such as quercetin has been determined by UV-vis spectrophotometric method in tris buffer solution at various pH. It was found that only 1 : 1 Cd-complex is formed by the interaction between catechol moiety in ring B and cadmium [Cd(II)] in aqueous solution. The spectral parameters for Cd-complex were determined by Beer's law at various pH. It has shown that 1 : 1 Cd-complex has a maximum absorbance and red shift by the alkaline pH.

Renoprotective and antioxidant effects of Saururus chinensis Baill in rats fed a high-fructose diet

  • This study investigated the preventive effect of Saururus chinensis Baill against renal damage induced by a high-fructose diet in rats. The rats (n = 30) were fed either a cornstarch-based (65%), high-fructose (65%), or high-fructose (64.5%) diet with 0.5% S. chinensis Baill extract for 10 weeks. Twenty-four hour urine collections were obtained and the animals were sacrificed after an overnight fast. Serum urea and creatinine and urine albumin were measured using colorimetric methods, and creatinine clearance was determined. In addition, thiobarbituric acid reactive substances (TBARS), reduced glutathione (GSH), and the activity of superoxide dismutase (SOD) in the kidney were determined. Kidney samples were also examined histologically. The fructose-fed rats showed renal dysfunction, indicated by decreased creatinine clearance, increased albumin in the urine, and increased urea and creatinine in the serum. These renal function parameters were comparable to control levels in rats that consumed S. chinensis Baill. Fructose consumption increased renal TBARS and reduced GSH and SOD activity, whereas these levels were near-normal in the rats consuming S. chinensis Baill The kidneys of fructose-fed rats showed glomerular basement membrane thickening, mesangial matrix expansion, and tubule dilation. These pathological changes were not seen in the rats that consumed S. chinensis Baill. Therefore, S. chinensis Baill effectively alleviated fructose-induced renal damage in these rats, at least partially due to antioxidant activity.

  • Obesity increases the risk of many adult diseases, such as atherosclerosis, diabetes, hypertension, ischemic heart disease and breast cancer. Inhibition of adipogenesis is an effective way in the anti-obesity management. Because of main components of Saururus chinensis is flavonoid, it has been showed some improvement by its antioxidant effects on the atherosclerosis, heart disease and diabetic hyperlipidemia. But mechanism of anti-obesity effect of S. chinensis is not clear. We investigated the effects of ethanol extracts of S. chinensis on adipogenesis in the 3T3-L1 pre-adipocyte. The 3T3-L1 cell line is commonly used to study adipogenesis in vitro. In this study, ethanol extracts of S. chinensis significantly decrease the lipid accumulation in the 3T3-L1 cells proved by measuring triglyceride contents and Oil red O staining. The proposed mechanism of inhibition of adipogenesis in the 3T3-L1 cells with ethanol extracts of S. chinensis is down-regulation of transcriptional factors and adipocyte-specific genes such CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$) and Peroxisome proliferator activated receptor ${\gamma}$ ($PPAR{\gamma}$) in concentration dependent pattern. These results suggest that ethanol extracts of S. chinensis inhibits adipognesis in the 3T3-L1 cells and can be used as a safe and efficient natural substance to manage anti-obesity.

  • It has been well established that S. mutans is the major etiological agent in dental caries, one of the most common oral diseases worldwide. The present study was designed to investigate the effect of Saururus chinensis (S. chinensis) ethanol extracts on the growth, acid production, biofilm formation, adhesion, and insoluble glucan synthesis of S. mutans. The ethanol extracts of S. chinensis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.025, 0.05, 0.1, 0.2 and 0.4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 80% at the concentration of 0.05 mg/ml and complete inhibition was observed at the concentration of 0.4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan form sucrose, ethanol extract of S. chinensis showed more than 10% inhibition over the concentration of 0.025 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.025 ${\sim}$ 0.4 mg/ml of the ethanol extract of S. chinensis. Our research strongly suggested S. chinensis was a promising natural product for the prevention of dental caries.

삼백초 Ethyl acetate 분획물의 예쁜꼬마선충 내의 항산화 효과 (Anti-oxidative Effects of Ethyl acetate Fraction of Saururus chinensis in Caenorhabditis elegans)

  • Saururus chinensis (Lour.) Baill. (Saururaceae) has been used as a traditional medicine for the treatment of dysuria, leukorrhea, eczema, jaundice and ascites in Korea, China and Japan. Ethanol extract of S. chinensis was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions. Among those fractions the ethyl acetate soluble fraction showed the most potent DPPH radical scavenging and superoxide quenching activities. To verify antioxidant activities of ethyl acetate fraction, we checked the activities of superoxide dismutase (SOD) and catalase, and intracellular ROS level and oxidative stress tolerance in Caenorhabditis elegans. Furthermore, to see if increased stress tolerance of worms by treating of ethyl acetate fraction was due to regulation of stress-response gene, we quantified SOD-3 expression using transgenic strain. Consequently, ethyl acetate fraction elevated SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Moreover, ethyl acetate fraction-treated CF1553 worms exhibited significantly higher SOD-3::GFP intensity.

In Vitro and Cellular Antioxidant Activity of a Water Extract of Saururus chinensis

  • The water extract of Saururus chinensis was investigated for oxygen radical absorbance capacity (ORAC), reducing capacity, metal chelating activity, and intracellular antioxidant activity using HepG2 cell. When 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH) was used for the generation of peroxyl radicals in vitro, S. chinensis extract (SC-E) showed the strong and concentration-dependent scavenging activity through donating protons which could be explained by its reducing property. When hydroxyl radicals were generated in vitro through the addition of $Cu^{2+}$ and $H_2O_2$, SC-E demonstrated the antioxidant activity depending on its concentration. In HepG2 cell model, most of intracellular oxidative stress generated by AAPH was efficiently removed by SC-E. However, when $Cu^{2+}$ without $H_2O_2$ was used as an oxidant in the intracellular assay, SC-E partially reduced the oxidative stress caused by $Cu^{2+}$ in cellular antioxidant activity assay system. These results indicate that SC-E could be utilized for the development of functional foods as antioxidant resource in the near future.
