추출조건에 따른 홍삼추출액의 사포닌 조성 및 이화학적 특성 (Saponin Composition and Physico-Chemical Properties of Korean Red Ginseng Extract as Affected by Extracting Conditions)

  • 홍삼은 건강을 유지하기 위한 약재로 오래전부터 우리나라에서 많이 애용되고 있는 한방소재로서 근래에 와서 홍삼의 유효 성분군이 사포닌으로 밝혀지면서 약리 효능적인 측면에서는 연구가 급진전되었으나, 홍삼의 유용 사포닌 성분을 안정성 높게 회수하기 위한 추출방법 및 홍삼을 추출하는데 있어 이화학적인 특성에 영향을 주는 조건에 대해서는 현재까지 연구된 바가 많지 않아 홍삼 추출조건에 대한 체계적인 연구가 필요한 실정이다. 이에 본 연구에서는 홍삼 추출기를 이용하여 추출조건에 따른 사포닌 성분 및 이화학적 변화에 대해 조사하여 비교분석하였다. 홍삼추출액의 총 사포닌 함량에 있어서는 추출온도와 시간이 증가함에 따라 총 사포닌 함량이 감소하는 경향을 나타내었으며, $75^{\circ}C$, 24시간 추출조건에서 0.774 mg/g으로 가장 높은 함량을 나타내었다. 개별 ginsenoside 함량에서도 대부분 추출온도와 시간이 증가될수록 함량이 감소하였지만, $Rg_3$의 경우에는 일정범위내에서의 추출조건에서는 함량이 오히려 증가하는 경향을 나타내었다. 이는 홍삼 추출 시 가열 과정에서 일부 ginsenoside가 $Rg_3$로 구조 전환되면서 그 함량이 증가한 것으로 추정된다. 또한 추출조건별 홍삼추출액의 이화학적 변화에 있어서는 사포닌의 경우와 반대로 추출온도와 시간이 증가될수록 $^{\circ}Brix$와 색차는 증가되는 반면, pH는 감소하는 등 전반적으로 품질 측면에서는 안정화되는 경향을 나타내었으며, 품질적인 특성만을 고려한 적정 추출조건은 $85^{\circ}C$ 내외에서 48시간 추출하는 것이 효율적이라 판단된다. 따라서 이러한 결과를 바탕으로 홍삼 추출액의 품질특성과 사포닌의 안정적인 추출을 위한 적정 추출조건은 ginsenosides의 손실이 최소화되는 $80^{\circ}C$ 내외에서 물리적 성질이 안정화되기 시작하는 48시간 이내로 추출하는 것이 적합한 것으로 사료된다. 이와 같은 결과는 홍삼 추출방법에 있어서 최적의 추출조건에 대한 정보를 주는 것이며, 더 나아가 고농도 사포닌성분이 함유된 기능성 홍삼제품 개발에 기초정보를 제공하는 결과라 사료된다.

Panax ginseng Improves Senile Testicular Function in Rats

  • We reported previously that the administration of Korean red ginseng water extract (KRG-WE) protected the guinea pig testis against damage induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (a potent endocrine disruptor). We also found that crude saponin from ginseng was the active ingredient responsible for this protection. Here, we examined the biological role of KRG-WE in an animal model of age-induced dysfunction of spermatogenesis. Twenty-four male Sprague-Dawley (six 2-month-old and eighteen 12-month-old) rats were used. The young and old control groups received only vehicle. The ginseng saponin (GS)- and KRG-WE-treated groups received GS (40 mg/kg body weight/day) and KRG-WE (200 mg/kg body weight/day), respectively, for 4 months. The number of cells, Sertoli cell index, Johnsen's score, and sex hormone levels decreased significantly with age. However, the administration of KRG-WE and GS markedly improved the number of germ cells, seminiferous tubular size, and Johnsen's score in the old rats. Ginseng produced a distinct testicular histological improvement in old rats. KRG-WE and GS elevated testosterone levels, while attenuating the aberrant increase in follicle stimulating hormone and luteinizing hormone levels. Sperm kinematics evaluated by a computer-assisted sperm analyzer demonstrated improvement in the percentage of motile sperm, progressive sperm motility, and curvilinear velocity associated with sperm quality, supporting the beneficial role of red ginseng in senile spermatogenesis. Overall, the total water extract had a more potent effect than the corresponding saponin fraction. In conclusion, Korean red ginseng rejuvenated age-induced testicular dysfunction. Additionally, the total water extract was more potent than the corresponding saponin fraction.

  • Sixteen vegetable food extracts were screened for the presence of saponin with the methods of foaming test, hemolytic test and Liebermann-Burchard reaction. Six samples were found to contain saponin. The ethanal extract of six positive samples and its hydrolysates, sapogenin parts and sugar parts were applied t.l.c to detect saponin moiety.

  • As a part of studies on the quality control of crude drug preparation drinks, ginseng saponins were identified by HPLC. Ginsenoside-Rb1 was determined quantitatively by HPLC. Ginsenoside MeOH/H2O(65:35:10, v/v) on Si-gel plate. Ginsenoside-Rb1 content determined by HPLC on Lichrosorbtract drinks was 57.5-70.4% compared to the content in the red ginseng extract.

  • Two triterpenoid saponins were isolated from the methanol extract of Caltha minor leave(Ranunculaceae). The structure of these saponin were elucidated as hederagenin-3-O-$\alpha$-rhamnopyranosyl(1$\rightarrow$2)-$\alpha$-L-arabinopyrano side and 3-O-$\alpha$-L-rhamnopyranosyl(1$\rightarrow$2)-$\alpha$-L-arabinopyranosyl hederagenin 28-O-$\alpha$-L-rhamnopyranosyl(1$\rightarrow$4)-$\beta$-D-glucopyranosyl(1$\rightarrow$6)-$\beta$-D-glucopyranosyl ester.

홍삼의 가열추출 과정중 유기산 중화에 의한 사포닌의 가수분해 억제 (Controls of the Hydrolysis of Ginseng Saponins by Neutralization of Organic Acids in Red Ginseng Extract Preparations)

  • Glucosidic bonds at the C20 position of the sapogenins were hydrolyzed easily in the lower pH, higher temperatures and longer times to give prosapogenins and sugars. The glucosidic bond of saponin at the C3 of ginsenoside-Rb1, which is secondary carbon, was relatively stable due to the low electron density of -0.2. But the bond of saponin at the C20 position, which is tertiary carbon with the relatively high electron density of -0.3, was liable to be hydrolyzed even in weakly acidic solution by the increase of heating time. On the other hand, red ginseng contained 13.34 mg/g of citric acid, 8.78 mg/g of malonic acid, 3.70 mg/g of oxalic acid, 2.13 mg/g of malic acid and 0.44 mg/g of succinct acid. Ginseng saponins were very stable in ginseng extract neutralized with sodium carbonate or sodium bicarbonate corresponding to the equivalent amount of the total organic acid in the red ginseng.

인삼 조사포닌의 조제 방법 개선 (Improved Method for the Preparation of Crude Ginseng Saponin)

  • This stuffy was carried to establish a new efficient method for the preparation of edible crude ginseng saponin. The conventional butanol extraction and resin adsorption methods were compared for the contents of total crude ginseng saponin and major ginsenosides. Seventy- percent methanol extract was applied to Diaion HP-20 column and the resin was washed with Hn and eluted with absolute methanol. The methanol elute was dried in vivo and analyzed for its ginsenosides. Use of ethanol instead of methanol to make edible crude ginseng saponin gave a similar result. Butanol extraction was performed by the conventional method. The final aqueous layer from butanol extraction was passed through Diaion HP-20 column followed by elution with methanol and Diaion HP-20 passed fraction was extracted with butanol to recover remaining components, respectively, in order to determine saponin loss. TLC and HPLC qualitatively and quantitatively monitored Ginsenosides, respectively. Loss of ginsenosides was higher in butanol extraction method than in Diction HP-20 adsorption method. In addition, saponin fractions prepared by Diction HP-20 adsorption method showed higher content of each ginsenoside, showing 8.2% higher purity than that of butanol extracted fraction. From these results, we propose the resin adsorption method as a new efficient measure for the preparation of crude ginseng saponin, which is edible by using spirit instead of methanol.

  • 미생물성 효소를 이용하여 인삼saponin중 조성비율이 가장 큰 ginsenoside-Rb$_1$을 약효면에서 보다 우수한 ginsenoside-Rd로 전환하고자 인삼부패균 중 Rhizopus 속의 한 균주를 선정하여 이 균주에서 얻은 효소를 ammonium sulfate 분별 침전법으로 조정제하여 사용하였다. 기질로 사용하기 위해 홍미삼 extract로부터 ginsenoside-Rb$_1$이 36.4%, ginsenoside-Rd 가 12.2%의 조성비율을 갖인 total saponin을 정제하였고 이어 ginsenoside-Rb$_1$의 함량을 증가시키기 위해 더욱 정제한 결과 ginsenoside-Rb$_1$이 54. 5%, ginsenoside-Rd가 1.1%인 ginsenoside Rb group saponin을 얻었다. 이들 기질 saponin에 본 효소를 작용시켜 본 결과 두 기질 모두 다른 ginsenoside pattern에는 변화없이 ginsenoside-Rb$_1$만이 선택적으로 감소하고 반면에 ginsenoside-Rd의 함량이 비례적으로 증가됨을 TLC 및 HPLC의 방법으로 조사하였으며 이로써 효소에 의한 인삼saponin의 선택적전환 가능성을 확인하였다.

흰쥐 심장의 수축력에 미치는 인삼성분의 효과 (Study on the Contractile Force of the Isolated Hearts from Ginseng Components Treated Rats)

  • The rate of deterioration of contractile force of isolated hearts from control and panax ginseng treated rats was determined and response of contractile force of the hearts from ginseng treated rats to several autonomic and other drugs was investigated. Rats weighing 150-250g were administrered orally with ginseng ethanol extract (100mg/kg) and total ginseng saponin (50mg/kg/day) for a week. Ginsenoside Rb$_{1}$ (5mg/kg/day) and ginsenoside Re (5mg/kg/day) were administered respectively for a week. The isolated hearts from rats were perfused with Krebs-Henseleit solution by using Langendorff perfusion apparatus. The control group was only able to maintain approximately 75.5% of their initial strength after 60 min of perfusion, whereas ginseng ethanol extract, total ginseng saponin treated hearts were able to sustain nearly their initial strength even after 60 min. Ginsenoside Rol treated hearts also sustained 93% of their initial strength, but there was no significant difference in the deterioration percentage of the contractile force of ginsenoside Re treated hearts. Experiments were conducted to study the response to perfusion of ginseng treated animal heart with epinephrine, isoproterenol, propranolol, and phenobarbital. The isolated hearts were perfused with Krebs-Henseleit solution containing epinephrine (10$^{-6}$ M), isoproterenol ($10^{-7}$M), propranolol ($10^{-6}$M) and phenobarbital (7{\times}10^{-3}M$) respectively. The maximum inotropic effect of epinephrine and isoproterenol was observed after 2~3 minutes of drug perfusion. Effect of epinephrine on ginseng ethanol extract and total ginseng saponin treated hearts was reduced compared with control. On the other hand, this phenomenon was not observed in ginsenoside Re treated rats but on ginsenoside $Rb_{1}$ treated rats. The positive inotropic effect of isoproterenol was reduced in the hearts from ginseng treated rats compared with control heart, Propranolol or phenobaribital decreased the contractile force in the control rats. The depressant effect of propranolol and phenobarbitat on ginseng treated rat hearts was less than those of control rat hearts. The result suggest that ginseng ethanol extract , ind total ginseng saponin and ginsenoside $Rb_{1}$ may protect the deterioration of contractile force of the heart and may attenuate the response to several drugs on hearts.

직파와 이식재배에 따른 인삼의 생육특성 및 엑스와 조사포닌 함량 (Comparison of Growth Characteristics, and Extract and Crude Saponin Contents in 4- Year-Old Ginseng Cultured by Direct Seeding and Transplanting Cultivation)

  • 금산 및 음성지역의 4년생 인삼재배 농가포장에서 직파재배 5개소와 이식재배 5개소를 임의로 선정하여 직파와 이식재배에 따른 생육특성 및 엑스와 조사포닌 함량을 비교한 결과는 다음과 같다. 직파재배는 4년근 생존율이 평균 48%로 이식재배의 86%보다 떨어지나 입모수가 평균 96주/3.3$m^2$순로 이식재배의 57주보다 많고 엽면적지수가 커 수량성이 높은 반면, 주당근중은 작았다. 직파재배는 이식재배에 비해 동체의 신장이 양호하나 지근의 발달이 불량하여 동체중의 비율이 높고 지근중의 비율이 낮았으며, 직파재배는 적변 발생율이 적으나 동체와 지근부위의 엑스와 조사포닌 함량이 낮았다.
