가든 세이지(Salvia officinalis L.) 추출물의 생리활성 탐색 (Biological Activity of Extracts from Garden Sage (Salvia officinalis L.))

  • 가든 세이지 추출물의 추출 최적 조건을 알아보고자 각각 추출 용매별, 농도별, 시간별로 비교하였다. 가든 세이지는 60% ethane에서 24시간 추출하는 것이 최적 추출조건으로 가장 효율적이라 판단되었다. 한편 생리활성 효과는 추출물의 phenol 농도를 $200\;{\mu}g/ml$로 조절하여 실험하였다. DPPH에 대한 전자 공여능은 60% ethanol 추출물에서 64.4%로 나타났으며, ABTS radical decolorlization은 60% ethanol 추출물에서 96.9%, antioxidant protection factor는 60% ethanol 추출물에서 2.30PF로 나타났으며, PF와 같이 지용성 물질의 항산화력을 나타내는 TBARS값은 centrol의 $0.62{\times}10^2\;{\mu}M$에 비해 60% ethanol 추출물에서 $0.28{\times}10^2\;{\mu}M$의 낮은 TBARS값을 나타내어 항산화 효과가 우수한 것으로 나타났다. Xanthine oxidase 활성억제 효과는 100의 저해율을 나타냈고, ACE활성억제 효과는 75.5%의 저해율을 나타내었으며, Helicobacter pylori균에 대한 활성도 $200\;{\mu}g/ml$ 농도의 액체배지에서 63.4의 저해율을 나타내어 생리활성효과도 우수한 것으로 나타났다.

  • salvia의 생장억제(生長抑制)와 측지발달(側枝發達), 개화반응(開花反應) 등(等)에 관(關)한 dikegulac과 ancymidol의 효과(效果)를 비교(比較)하기 위하여 salvia 품종(品種) Bonfire를 공시(供試)하여 dikegulac 0.06%, 0.12%(엽면살포(葉面撒布))와 ancymidol 50 ppm, 100 ppm(엽면살포(葉面撒布)) 및 0.1 mg/pot, 0.5 mg/pot(토양주입(土壤注入))을 처리(處理)하였다. dikegulac 처리(處理)는 정아(頂芽)의 생장(生長)을 저해(沮害)하여 초장(草長)을 감소(減少)시키고 측지(側枝)의 발달(發達)을 유도(誘導)하여 개화경수(開花莖數)를 증가(增加)시켰으나 개화기(開花期)는 지연(遲延)시켰다. 또 잎의 엽록소(葉綠素) 함량(含量)을 증가(增加)시켜 관상가치(觀賞價値)를 증대(增大)시켰는데 특(特)히 chlorophyll a의 함량(含量)이 월등(越等)히 높아졌다. ancymidol 처리(處理)는 정아(頂芽)의 생장(生長)을 저해(沮害)하지 않고 초장(草長)을 감소(減少)시켜 개화기(開花期)에는 거의 영향(影響)하지 않았으나 개화경(開花莖) 증가(增加)에는 0.1 mg 토양주입구(土壤注入區)를 제(除)하고는 거의 효과(效果)가 없었다. 또 잎의 엽록소(葉綠素) 함량(含量)이 ancymidol 처리(處理)에 의해 현저(顯著)히 증가(增加)되었는데 dikegulac 처리구(處理區)와 마찬가지로 chlorophyll a의 함량(含量)이 현저(顯著)히 높아졌다. dikegulac은 salvia에서 매우 효과적(效果的)인 화학적(化學的) 적심제(摘心劑)로 사용(使用)될 수 있음을 알 수 있었다.

곰보배추 추출물이 항산화 및 피부미백효과에 미치는 영향 (Effects of Salvia plebeia Herb Extracts on Anti-oxidant Activity and Whitening action)

  • 본 논문은 재배가 용이하고 민간요법에서 다양하게 이용되고 있는 곰보배추의 에탄올 추출물이 화장품 소재로서 가능성이 있는지를 알아보기 위해 항산화, 미백효능에 대하여 관찰하였다. B16F10 세포에서 곰보배추 에탄올 추출물 25, 50, $100{\mu}g/mL$ 농도에서 독성이 나타나지 않았다. DPPH radical 소거능을 관찰한 결과 모든 농도에서 소거능을 보여주었고, $50{\mu}g/mL$의 농도에서 65.2%, $100{\mu}g/mL$에서 77.6%의 강한 항산화 효능을 나타냈다. Raw 264.7 세포 내에서 ROS 생성 저해능을 관찰한 결과 농도 의존적으로 유의하게 나타냈고, $100{\mu}g/mL$ 농도에서는 39.1% 억제하는 것으로 나타났다. NO생성 억제를 관찰하기 위해 Raw 264.7 세포에 곰보배추 에탄올 추출물을 25, 50, $100{\mu}g/mL$ 농도별로 첨가하여 관찰한 결과 농도 의존적으로 NO생성을 억제하였다. 시험관 내에서 L-DOPA와 L-tyrosine을 이용하여 곰보배추 에탄올 추출물이 tyrosinase activity를 농도 의존적으로 억제하는 것을 나타냈다. 세포 내에서 MSH를 가한 B16F10 세포에 곰보배추 에탄올 추출물을 25, 50, $100{\mu}g/mL$의 melanin 함량을 관찰한 결과 농도 의존적으로 억제하는 것을 보여주며 $100{\mu}g/mL$에서 30.7%로 억제하였다. 따라서 곰보배추 에탄올 추출물이 항산화 기능이 있는 미백 기능성 화장품의 소재로서 개발 가능성이 충분히 있는 것으로 사료된다.

A Case of Hepatotoxicity by Salvia Plebeia R. Br.

  • Objective: To investigate the possibility of hepatotoxicity by supplemental foods or folk herbs such as Salvia Plebeia R. Br. Methods: A hospitalized male patient with alcoholic liver disease and electrolyte imbalance had recovered, and then followed by rapid hepatic serum enzymes after taking S. Plebeia. This study monitored the clinical outcome and biochemical parameters. Result: A 58-year male had drunk frequently, which led to alcoholic steatohepatitis and hospitalization. Two weeks after his discharge from hospital, he felt nausea, dizziness, and mild difficulties in speech and walking, resulting in re-hospitalization at the Korean Medical Hospital. The symptoms disappeared on correction of the electrolyte imbalance suspected to have been caused by severe sweating while working in the outdoors, and the patient was discharged. During treatment and monitoring of his health as an outpatient, the serum hepatic enzyme rapidly elevated approximately 10-fold in hepatic enzymes; the enzyme levels fluctuated according to whether or not he was taking the boiled water of S. Plebeia. The RUCAM score was 12, which met the criteria for toxic hepatitis by S. Plebeia. His general condition and abnormal hepatic enzymes recovered with cessation of S. Plebeia and administration of Chungganplus syrup (CGX). Conclusion: This study reports the hepatotoxic risk of Salvia Plebeia, which is commonly used as a folk remedy in Korea.

배암차즈기 추출물의 기능성원료 표준화를 위한 지표성분으로서 Hispidulin의 분석법 평가 (Analytical Method for the Validation of Hispidulin as a Marker Compound for the Standardization of Salvia plebeia R. Br. Extracts as a Functional Ingredient)

  • Background: In the present study, we established an HPLC (high performance liquid chromatography)-analysis method for the determination of marker compounds as a part of the material standardization for the development of health-functional foods from Salvia plebeia R. Br. extract. Methods and Results: The quantitative determination method of hispidulin as a marker compound was optimized by HPLC analysis using a YMC hydrosphere C18 column with a gradient elution system. This method was validated using specificity, linearity, accuracy, and precision tests. It showed a high linearity in the calibration curve with a coefficient of correlation ($r^2$) of 0.999995. The method was fully validated, and was sensitive, with the limit of detection (LOD) at $0.09{\mu}g{\cdot}m{\ell}^{-1}$ and limit of quantification (LOQ) at $0.27{\mu}g{\cdot}m{\ell}^{-1}$. The relative standard deviation (RSD) values of the data from intra- and inter-day precision were 0.05 - 0.22% and 0.32 - 0.42%, respectively, and the intra- and inter-day accuracy of hispidulin were 99.5 - 102.3% and 98.8 - 101.5%, respectively. The average content of hispidulin in Salvia plebeia R. Br. extract was $3.945mg{\cdot}g^{-1}$ (0.39%). Conclusions: These results suggest that the developed HPLC method is very efficient, and that it could contribute to the quality control of Salvia plebeia R. Br. extracts as a functional ingredient in health functional foods.

  • Three horticultural herbaceous plants and a natural herbaceous plant were tested to determine the growth responses, biomass and uptake of cadmium(cd), lead(pb) by application of cd and pb soil treatment in pot culture. The ecological effects on the growth of the plants were investigated to determine the tolerance for the heavy metal pollutants cd and pb. the marginal concentrain of cd treatment on the growth of the each plant was below the 1, 000 ppm treatment of cd. The marginal concentration of pb treatment was below the 1, 000ppm treatment of pb in cultivation of salvia splendens ker., celosia cristata l. and below the 3, 000ppm treatment of pb in cultivation of portulaca grandiflora hook., sedum saramentosum bunge. the resistance for cd of sedum saramentosum bunge, celosia criastata l., portulaca grandiflora hook. and salvia splendens ker. was in the listed order. The resistence for pb was in order of sedum saramentosum bunge, portulaca grandiflora hook. Salvia splendens ker.and celosia criastata l.stems. The flowering of portulaca grandiflora hook. was sustained in the pb 1, 000ppm treated group only. The higher the concentration of pb in the soil cultivated the plants was, the less the content of leaf chlorophy11 in each plant was. The number of stomata per unit leaf area was the highest in salvia splendens ker. and in order of celosia l., sedum saramentosum bunge., portulaca grandiflora hook., the higher the cd and pb concentration of cd and pb treatment was, the more the concent of cd and pb in the part of each plant increased. the content of cd and pb in the stems of salvia spiendens ker. was the highest in the 1, 000ppm-treated ground and in order of the roots, the leaves and the flowers.

곰보배추 추출물로 염색한 면직물의 자외선 차단율, 소취성 및 항균성 (Study of UV Protection, Deodorization and Antimicrobial Properties of Cotton Fabrics Dyed with the Liquids Extracted from Salvia Plebia R. Br.)

  • This study examined the stainability and functionality of cotton dyed with the extract of Salvia Plebia R. Br. as a research preceding the development of health functional materials and fashion products with a healing motif. The CIELAB measurements of the cotton fabric dyed with the extract of Salvia Plebia R. Br. showed 73.32 for $L^*$, 1.7 for $a^*$, and 37.78 for $b^*$, while the Munsell measurements exhibited 2.63Y in color, 7.18 of brightness, and 5.49 of chroma. The degree of staining resulting from color fastness to laundering, and that resulting from color fastness to perspiration (acid and alkaline), as well as the level of color fastness to crocking (wet and dry) all stayed between relatively higher levels of 4 and 5. The fabric dyed also exhibited SPF 50+, an outstanding sun blocking performance. The deodorization rate was also excellent, rising from 97% to over 99% when the time elapsed increased from 30 minutes to 120 minutes. The fabric dyed also showed a 99.6% antimicrobial activity against staphylococcus aureus ATCC 6538, and a 71.4% antimicrobial activity against Klebsiella pneumoniae ATCC 4352. The aforementioned findings indicate that fabrics dyed with the extract of Salvia Plebia R. Br. contain the potential to be developed for use as health-related materials and fashion products that promote healing.

단삼(丹蔘)이 손상된 뇌신경세포에 미치는 영향 (The Effect of the Salvia miltiorrhiza on Axon Regeneration Following Central Nervous System Injury)

  • Object: Reactive gliosis that is induced by central nervous system (CNS) injury is involved with up-regulation of CD81 and GFAP. The present study was to examine the effect of the Salvia miltiorrhiza on CD81 and GFAP regulation following brain injury. Methods: Immunoblot and ELISA methods were used to define the level of CD81 and GFAP in the astrocyte cultured from rat brain. Then immunohistochemistry was used to detect CD81 and GFAP in the injured rat brain. Results: The following results were obtained. 1. We did western blot and ELISA to detect the protein isolated from the whole cell and they showed that CD81 and GFAP decreased. 2. We injected Salvia miltiorrhiza extract intravenously to brain-injured rats for 7 days and 30 days, and the immunohistochemistry analyses showed that CD81 and GFAP decreased significantly. Conclusion: These results indicate that Salvia miltiorrhiza could suppress the reactive gliosis, which disturbs the neural regeneration following CNS injury, by controlling the expression of CD81 and GFAP.

  • 광합성 세균, 젖산균, 효모 등의 미생물을 토양 관주, 엽면 시비의 방법으로 식물(salvia)에 처리하고 식물의 생장에 미치는 효과를 검토하였다. 액체 배양한 광합성 세균을 100배 희석하여 토양 관주할 경우 다른 미생물군을 단독으로 처리한 경우에 비해 식물의 생장이 우수하여 대조구보다 약 160%의 생장 촉진 효과를 나타내었다. 미생물군을 혼합하여 처리한 경우에는 세 종류의 미생물을 혼합한 후 10배 희석하여 처리한 경우에 가장 우수한 효과를 나타내었으며 대조구보다 212% 식물 생장이 촉진되었다.
