Degradation of Phytate Pentamagnesium Salt by Bacillus sp. T4 Phytase as a Potential Eco-friendly Feed Additive

  • A bacterial isolate derived from soil samples near a cattle farm was found to display extracellular phytase activity. Based on 16S rRNA sequence analysis, the strain was named Bacillus sp. T4. The optimum temperature for the phytase activity toward magnesium phytate (Mg-$InsP_6$) was $40^{\circ}C$ without 5 mM $Ca^{2+}$ and $50^{\circ}C$ with 5 mM $Ca^{2+}$. T4 phytase had a characteristic bi-hump two pH optima of 6.0 to 6.5 and 7.4 for Mg-$InsP_6$. The enzyme showed higher specificity for Mg-$InsP_6$ than sodium phytate (Na-$InsP_6$). Its activity was fairly inhibited by EDTA, $Cu^{2+}$, $Mn^{2+}$, $Co^{2+}$, $Ba^{2+}$ and $Zn^{2+}$. T4 phytase may have great potential for use as an eco-friendly feed additive to enhance the nutritive quality of phytate and reduce phosphorus pollution.

  • The last decade of this centry is now the accepted birth date of that sub-discipline of pharmacy that is now called 'biopharmceutics'. Wagner defines biopharmaceutics 'as the study of the influence of fomulation on the therapeutic activity of a drug product.' More specifically, he states that biopharmaceutics encompasses the study of the relationship between the nature and intensity of the biological effects observed in animals or man and the following factors: 1. The nature of the form of the drug (ester, salt, complex, etc). 2. The physical state, particle size, and surface area. 3. Presence or absence of adjuvants with the drug. 4. The type of dosage form in which the drug is administered. 5. The pharmaceutical process (es) used to make the dosage form. The philosophy inherent in this definition has revolutionized our thinking with respect to product development, quality control, and to the practice of pharmacy itself. Althoughthe the emphasis herein will be on quality control, the interrelationship between this and the other areas of pharmacy will be evident. The principles of quality control dictate that a wide variety of techniques be used to evaluate the quality of a dosage form. Since quality must be built into a dosage form, the pharmaceutical scientist begins the process at the research stage, continues it during the production stage, and ends it by applying the tests and procedures established by parmacopeial commissions. These stages are usually separate and distinct and, because of this, product quality has become synonymous with compliance with pharmacopeial specifications.

Factors affecting the occurrence of wilt of strawberry caused by Fusarium oxysporum f. sp. fragariae in Korea

  • The occurrence of Fusarium wilt in strawberry fields in Korea was assessed from 2001 to 2003. Fusarium wilt was found from June to August in nursery beds, from September to October after planting in production beds, and from January to March during harvest. The symptoms seen were root rots, discolored vascular tissue in the crown and deformation and yellowing of central leaflets. The disease occurred in up to 30% of plants in 37 of 214 fields surveyed. Fusarium of sporum Schlecht. ex Fr. f. sp. fragariae was frequently isolated from cvs. Dochiodome, Maehyang, Redpearl, Samaberry and Akihime. Factors affecting the occurrence of Fusarium wilt were investigated; infested soils had high salt concentrations, low pH, OM, average P2O5 and exchangeable. Fusarium wilt was more frequent following conventional basal fertilization than after non-nitrogen basal fertilization and more frequent following the use of NH4-N than after NO3-N.

  • A two year's field experiment was conducted on newly reclaimed saline tidal lands to measure the effects of sand mulching on salinity of the soils and their relationship to crop performance. Hybrid sorghum cv. Pioneer 931 was grown under different mulching treatments using of medium sand and red earth (Fine loamy, Typic Hapludults). Salinity in the root zone was decreased markedly under soil mulching using of medium sand, especially during the dry season, and it caused a great increase in the root growth and R/T ratio. Seasonal values of electrical conductivity at sand mulching were 6.6 in April and 1.6 mmhos in August, but it was still high with a concentration of 12.7 (April) and 3.8 mmhos (August) in untreated check plot. Sand mulching increased plant growth and the rate of dry matter accumulation. However, treatment of red earth additionally over sand mulching produced lower dry matter yield than those of soil mulching using of medium sand only. Under salt stress sorghum plant showed a decrease in the leaf weight ratio (LWR) and it resulted in a low concentration of crude protein of the plant. Sand mulching enhanced leaf weight ratio and rate of protein synthesis.

  • 황색포도상구균이 Capsular polysaccharide (CPS)는 백혈구의 탐식에 저항하기 때문에 젖소의 유방염 발생의 중요한 병원상 인자로 오랫동안 연구자들의 관심이 집중되어 왔다. 따라서 CPS는 젖소의 유방염을 최소화시키기 위한 유방염 백신개발에 있어 잠재적인 백신 항원물질로 알려져 왔다. 본 연구의 복적은 황색포도상구균의 CPS 항원물질을 좀더 효과적으로 발현시키고 향후 백신항원으로 사용시 좀 더 간편하게 정제하는 방법을 찾아보고자 본 연구를 수행하였다. CPS는 락토스와 덱스트로스를 첨가한 brain heart infusion media 에서도 부분적으로 발현되었지만 CPS가 가장 잘 발현되는 배지 조건은 젖소 유래의 유청을 10% 첨가한 조건에서 가장 발현율이 높았다. 또한 CPS발현은 salt agglutination test, india ink법 및 투과전자현미경을 이용하여 확인하였으며 균체표면의 hydrophobicity에서는 락토스, 덱스트로스 및 젖소 유래의 유청을 첨가한 배지조건에서 다른 조건에 비해 높았다. CPS의 가장 손쉬운 검사법은 균체를 절편하지 않고 직접 투과전자현미경상에서 염색없이 관찰하는 것이 가장 간편한 방법이었다. CPS정제는 상층액을 농축시킨후 이온교환크로마토그라피와 젤 여과법을 이용하여 정제하였으며 CPS의 분자량은 대부분 97kDa이상이었다. CPS 회수율은 배지와 균체 1리터당 1.5mg이었다. 결론적으로 CPS를 백신항원으로 포함하고자 할 경우, CPS의 최적 발현조건은 젖소 유래의 유청을 10% 첨가한 Brain heart infusion 배지에서 배양하여 사용하는 것이 가장 좋은 방법이라고 생각된다.

  • The development of automobile, vessel, rail road, and machine industry leads an increase of foundry production used as their components, which cause a by-product, waste foundry sand (WFS). The amount of the WFS produced in Korea is over 700,000 tons a year, but most WFS has been buried itself and only $5{\~}6\%$ WFS is recycled as construction materials. Therefore, it is necessary for most WFS to research other ways which can be used in a higher value added product. The study on recycling it as a fine aggregate for concrete or green sand has been in progress in America and Japan since 1970s and 1980s respaectively. In this study, two types of WFS were used as a fine aggregate for concrete. Nine types of concrete aimed at the specified strength of 30 MPa were mixed with washed seashore coarse sand in which salt was removed, and WFS and then appropriate mixture proportion of concrete was determined. Moreover, basic properties such as air contents, setting time, bleeding, workability and slump loss of the fresh concrete with WFS were tested and compared with those of the concrete mixed without WFS. In addition, both compressive strength of hardened concrete at each ages and tensile strength of it at the age of 28 days were measured and discussed.

Influence of an Anaerobic Fungal Culture (Orpinomyces sp.) Administration on Growth Rate, Ruminal Fermentation and Nutrient Digestion in Calves

  • The study was to see the effect of administration of ruminal fungi on feed intake, growth rate, rumen fermentation and nutrient digestion of calves (Tharparkar$\times$Holstein-Friesian, average age: 10 months, average body weight: 130 kg). The 6 calves in first group were fed a mixture consisted of 50% wheat straw and 50% concentrate (Maize 62%, Groundnut cake 35%, Mineral mix. 2% and Common salt 1%) along with 1 kg green oats $animal^{-1}$ $day^{-1}$ while second group calves were fed the above-mentioned diet in addition to a dose of 160 ml ($10^{6}$ CFU/ml) fungal culture $calf^{-1}$ $week^{-1}$. The average dry matter intake per day was slightly lowered in fungal fed calves yet feed conversion ratio was higher. The average daily weight gain was significantly higher (15.37%) in fungal administered group as compared to control. The nutrient digestibility was increased for crude fibre, NDF and ADF with fungal administration. Digestible energy value of straw-based diet in terms of percent TDN also increased. The pH and $NH_{3}$-N were lower whereas TVFA, total-N, TCA-N and number of zoospores were higher in rumen liquor in fungal administered group.

Antioxidant and Anti-Inflammatory Effect of Probiotic Lactobacillus plantarum KU15149 Derived from Korean Homemade Diced-Radish Kimchi

  • Lactobacillus plantarum KU15149 was demonstrated to have probiotic behavior and functions, including antioxidant and anti-inflammatory activity. L. plantarum KU15149 obtained from homemade diced-radish kimchi has a high survival rate under artificial gastric acid (pH 2.5, 0.3% pepsin) and bile salt (0.3% oxgall) conditions. However, L. plantarum KU15149 did not produce β-glucuronidase, which is known to be a carcinogenic enzyme with resistance to several antibiotics, such as gentamycin, kanamycin, streptomycin, tetracycline, and ciprofloxacin. L. plantarum KU15149 strongly adhered to HT-29 cells and had high antioxidant activity in terms of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging and β-carotene bleaching assays. L. plantarum KU15149 also exhibited a pronounced inhibition of nitric oxide (NO) production, along with expression of nitric oxide synthase (iNOS) and cyclooxygenase -2 (COX-2) as well as pro-inflammatory cytokines, such as TNF-α, IL-1β, and IL-6, when RAW 264.7 cells were stimulated with LPS. Therefore, L. plantarum KU15149 exhibited pharmaceutical functionality as a potential probiotic.

Influence of Arbuscular Mycorrhizal Fungus and Kinetin on the Response of Mungbean Plants to Irrigation by Seawater

  • An experiment was carried out to investigate the effects of pre-inoculation with the mycorrhizal fungus Glomus clarum and foliar application of kinetin on the growth of mungbean plant irrigated wht different dilution of seawater. Arbuscular-mycorrhizal(AM) infection significantly increased dry weight, height, chlorophyll, sugar and protein content, nitrogen and phosphorus-use efficiencies, leaf conductivity, transpiration rate, nitrogenase, acid and alkaline phosphates activities of all salinized mungbean plants in comparison with control and non-mycorrhizal plants irrespective of the presence or absence of kinetin. Mycorrrhizal plants showed higher concentrations of N, P, K, Ca and Mg and lower Na/N, Na/P, Na/K, Na/Ca and Na/Mg ratios than non-mycorrhizal plants when irrigated with certain dilution of seawater. Mungbean plants showed 597% and 770% dependency on AM fungus G. clarum in absence and presence of kinetin, respectively, for biomass production under a level of 30% of seawater. The average value of tolerance index for mycorrhizal plants accounted 267% and 364% in absence and presence kinetin respectively. This study provides evidence for the benefits of kinetin which are actually known for mycorrhizal than non-mycorrhzal plants. AM fungus and kinetin protected the host plants against the detrimental effects of salt. However, mycorrhizal infection was much more effective than kinetin applications. Thus management applications of this arbuscular mycorrhizal endophyte(G. clarum) with kinetin could be of importance in using seawater in certain dilution for irrigation in agriculture.

A Study of Sasang Constitution in Kimchi (김치에 대한 사상체질의학적(四象體質醫學的) 고찰(考察)(I))

  • Purposes This study is for the constitutional health in daily life by adding the Sasang Constitutional points to the kimchi's nutritional ones. It lays the foundation of the development of both new medical kimchis and production of various kinds of kimchis according to different constitution. Methods We have examined the Keemy(氣味) and effect(效能) of kimchi represented in the Korean medicine [Dongyibogam(東醫寶鑑), Hyangyaggibseongbang(鄕藥集成方), Choongyakdaesajeon(中葯大辭典)] and Leejema's literature [DongyiSoosebowon(東醫壽世保元), Dongmooyugo(東武遺稿), Chobon kwon(草本卷)]. Results 1) There are 18 kinds of materials in kimchi, 9 main materials and 9 additional ones. 2) There are mustard, red pepper, garlic, leek, ginger, salt, green onion, and dropwort as good materials of kimchi for Soeumin, which are mostly Shinon(辛溫) or Yeoll(熱). 3) There are eggplant, oyster, chinese cabbage, shrimp, cucumber, and sesame as good materials of kimchi for Soyangin, which are mostly Gam(甘) or Ham(寒) or Ryang(凉), Han(寒). 4) There are bracken, perilla, radish, and taro as good materials of kimchi for Taeumin. which are mostly ShinGam(辛甘) or OnPyeong(溫平), Ryang(凉) Han(寒).
