Effects of Potassium-Cyanoethylstarch (K-CES) and Polyacrylamide (PAM) on Growth of Spinach (Spinacia oleracea L.) (토양내 K-CES와 PAM처리가 시금치의 생육에 미치는 영향)

  • The objective of this study was to observe the effects of potassium-cyanoethylstarch (K-CES), which is a natural polymer derivate,. and polyacrylamide (PAM), which is a synthetic polymer, on the growth of spinach under environment-controlled biotron. K-CES was made from tapioca starch, and introductions of acrylonitrile was confirmed by FT-IR. Water absorbing capacities (WAC) of K-CES and PAM were 37.6 and 47.8 times, respectively. WAC of PAM was slightly higher than K-CES, WAC of PAM m saline solutions were more reduced than those of K-CES. It was introduced that K-CES treatment was more effective than PAM in higher salinity of water and soil. Dry super absorptive polymers were put into solution, and the solution was applied to soil as an soil conditioner. Both amendments, K-CES and PAM, increased the vegetative growth of the spinach when applied at 0.1% of soil weight. In general, the most favorable results for early vegetative growth of the spinach came from the PAM treatment, followed by the K-CES treatment.

Effects of Filtration on the Characteristics of Reused Waste Brine in Kimchi Manufacturing (배추 절임 중 반복사용 폐염수의 여과처리 효과)

  • The waste brine gained from successively reused brine during kimchi manufacturing can cause serious water pollution. We investigated the filtration effects on the physicochemical characteristics and microbial counts of the waste brine. Chinese cabbage was salted for five times successively, and the waste brines were filtered through sand and active carbon column. While original values of salinity and soluble solid contents of waste brine were 15.4% and $18.0^{\circ}$Brix$, respectively, we observed decrease of them to 0.1% and $0.0^{\circ}$Brix$, respectively, after filtration of the waste brine through sand followed by active carbon column. The filtration also recover pH value of the waste brine to its original value, which was decreased by successive salting from 8.3 to 6.0. We also observed that COD of waste brine increased to 63.2 ppm after five times of salting but decreased to 5.1 ppm after active carbon filtration. Total viable count was also increased with successive 5 steps of salting and was not detected after active carbon-filtration.

Effects of Salinity Stress on the Composition of Free Amino Acids of the Pacific abalone Haliotis discus discus (염분 스트레스가 둥근전복 Haliotis discus discus의 유리아미노산 조성에 미치는 영향)

  • Changes in 13 free amino acids (FAA) of Haliotis discus discus exposed to various salinities were investigated using a high performance liquid chromatography (HPLC). Taurine, glycine and alanine are three major contributors to the total FAA in the gill tissues of H. discus discus. Concentration of taurine was 114 imol/g dry tissue weights accounting for 76.64% of total FAA in H. discus discus. Levels of most FAAs in H. discus discus exposed to low and high salinities for 24 h decreased dramatically. Taurine concentration was slightly increased in the samples exposed to 20 psu and 25 psu for 48 h, and greatly increased after 120 h exposure. After 48 hrs exposure to 20 psu, 25 psu, 30 psu and 40 psu, methionine in H. discus discus was not detected; no methionine was detected in the sample exposed to 20 psu, 25 psu, 30 psu and 40 psu after 120 h of exposure. Taurine:glycine ratio increased depending upon hyper-hypoosmotic condition as well as period of osmotic stress. These data indicate that taurine, glycine and methionine play important role in regulating osmotic stress in H. discus discus. This study suggesting that FAA analysis is a useful tool to diagnose osmotic stress to H. discus discus.

Effect of Environment Factors on Growth and Mortality of Cupped Oyster, Crassostrea gigas (양식장 환경이 갯벌양식 굴 (Crassostrea gigas)의 성장과 폐사에 미치는 영향)

  • In order to evaluate how the effects of aquacultural environment, such as temperature, salinity, DO, SS, Chlorophyll-a, COD and nutritive salts on Cupped oysters, an investigational study was conducted between September to December of 2011. During the study, different intermediate cage farms on the surface of the sea were used to culture and compare the growth and survival rate of the Cupped oysters in different fishing environments. The different intermediate cage farms used were Wonsando-ri, Chang-ri, and Pado-ri. In January of 2012, these oysters were transplanted to a horizontal net farm in a mudflat until July of that year. The adaptation rates of the Cupped oysters were tested at differential exposure times at varying intertidal periods. Wonsando-ri showed the highest water temperature and Chlorophyll-a levels, while the salinity was found to be within range of stable conditions among all three intermediate cage farms. Once the Cupped oysters were at the horizontal net farm, the growth was measured at distinct tidal exposure time of 1, 3, and 5 hours, whereby the growth rate was highest at 3, 1, and 5 hours, respectively. In addition, the oysters cultivated in intermediate cage farms had longer shell lengths compared to shell heights, while oysters cultured in the horizontal net farm had larger shell heights than shell lengths.

Uptake Patterns of N and P by Reeds (Phragmites australis) of Newly Constructed Shihwa Tidal Freshwater Marshes (시화지구 인공습지에서 갈대에 의한 질소 및 인 흡수)

  • This study was conducted to examine the seasonal pattern of N and P uptake by reeds (Phragmites australis) planted in newly constructed Shihwa tidal freshwater marshes. Reed and soil samples were collected from the wetland periodically from June 2000 to May 2002. Reed samples were analyzed for dry weight and content of N and P Soil organic matter content and salinity were also determined. Dry matter content of reed increased during the growing season but decreased in the fall and winter. However, this seasonal pattern was not so evident in the second year. In particular, throughout the measurement period, dry matter content of reed was lowest at a site showing high soil salinity. Regression analyses between dry matter content of reed and soil EC(1:5) suggested that dry matter content per unit square meter would decrease by 1.5 kg with every 1 dS m/sup -1/ increase in soil EC(1:5). The amount of N and P assimilated by reed significantly decreased from the fall and was lowest in the spring. Net decrease in N content from reed during the fall and next spring was calculated as 34.5 and 24.6 g m/sup -2/ in the first and second years, respectively, while the corresponding P loss was 4.0 and 1.8 g m/sup -2/. Soil organic mailer content increased in the fall and winter, but decreased in the spring and summer. The results of this study suggested that the removal of N and P by reed would be considerable during the growing season but the nutrients taken up by reeds would return as detritus to the marshes in the fall and winter. Based on the results of the study, therefore, the harvest of the reed at the latter part of the growth would be recommended to prevent further water quality degradation. However, the long-term effects of reed harvest needs further study.

Changes in Root Water Uptake and Chlorophyll Fluorescence of Rice (Oryza sativa L. cv. Dongjin) Seedling under NaCl Stress (NaCl 스트레스에 따른 벼 유식물의 뿌리 수분흡수와 엽록소형광의 변화)

  • The physiological and photochemical responses of rice seedling to NaCl stress were investigated through measuring leaf relative water content (RWC), root water uptake and chlorophyll fluorescence. When plants were exposed to increased salinity stress, the visual symptoms of injury were significant at ${\geq}$500 mM NaCl concentration for 4 and 5 day stress periods. The differences in Fv/Fm between control treatment and plants treated with 500 mM and 1,000 mM NaCl were evident after 5 day and 4 day, respectively, whereas in root water uptake its effect was observed at 500 mM and 1,000 mM NaCl at 2 day of salt-stressed periods. Leaf RWC in salt-stressed plants decreased gradually with increasing salinity in exogenous solution and duration of salt stress, and these decrease showed leaf RWC of 58-68% atduration over 2 day stress of 1,000 mM NaCl treatment and 88% at 1 day stress. NaCl stress led to a significant inhibition of the light-induced greening in etiolated rice plants, especially in 4 and 5 day salt-stressed plants, which linearly decreased with NaCl concentration ($R^2$=0.812 and 0.918, respectively). The effects of NaCl stress in rice seedlings indicate that water uptake in root is more sensitive to increasing NaCl concentration and stress duration than Fv /Fm in leaves compared with the same NaCl concentration.

Effects of Temperature, Salinity and Diet on the Productivity of the Cyclopoid Copepod, Apocyclops royi (수온, 염분 및 먹이에 따른 기수산 cyclopoid 요각류, Apocyclops royi의 생산력)

  • The productivity of cyclopoid copepod, Apocyclops royi fed by various diets (Isochrysis galbana, Tetraselmis suecica, Phaeodactylum tricornutum, concentrated freshwater Chlorella and baker's yeast) was investigated at tile different temperatures ($16-36^{\circ}C$) with different salinities (5-34 ppt). A. royi was cultured in 6 ml vessels (12 wells culture plate). Total production (188 inds.) and daily production (13.4 inds.) of nauplii by A. royi female at $32^{\circ}C$ were significantly higher than those of nauplii at the different temperatures (P<0.05). Development time from nauplii to copepodite and from nauplii to adult tended to increase with increasing water temperature up to 32. And total production (169 inds.) and daily production (9 inds.) of nauplii by A. royi female at 10 ppt were significantly higher than those of nauplii at the different salinities (P<0.05). The fastest development time from nauplii to copepodite and from nauplii to adult was observed at 10 ppt and 15 ppt, respectively (P<0.05). The highest total production of A. royi nauplii and fastest development time from nauplii to adult were obtained in females fed Isochrysis galbana (P<0.05). These results may indicate that the optimum culture temperature and salinity for A. royi are $32^{\circ}C$ and 10 ppt, respectively, and Isochrysis galbana is one of the suitable diets for this copepod.

Capillary Characteristics of Water and Cations in Multi-layered Reclaimed Soil with Macroporous Subsurface Layer Utilizing Coal Bottom Ash

  • Serious problems in reclaimed land agriculture are high soil salinity and poor vertical drainage, so desalinization in these soils is very difficult. Also, although desalinization is accomplished in reclaimed top soils, before long, soils are resalinized according to capillary rise of salts from the subsurface soils. To resolve these problems, multi-layered soil columns with subsurface layer of macroporous medium utilizing coal bottom ash (CBA) were constructed and the effects of blocked resalinization of these soils were investigated. In this experiment soil samples were collected from Munpo series (coarse-loamy, nonacid, mixed, mesic, typic Fluvaquents). The soil texture was silt loam and the EC was $33.9dS\;m^{-1}$. As for groundwater seawater was used and groundwater level of 1 cm from the bottom was maintained. The overall rate of capillary rise was $2.38cm\;hr^{-1}$ in soil 60 cm column, $0.25cm\;hr^{-1}$ in topsoil (30 cm) + CBA (5 cm) + subsurface soil (10 cm) column and $0.08cm\;hr^{-1}$ in topsoil (30 cm) + CBA (10 cm) + subsurface soil (10 cm) column. In multi-layered soil columns with CBA 20, 30 cm layer, wetting front due to capillary rise could not be seen in top soil layer. After 70 days capillary rise experiment water soluble Na+ accumulated in top soil of soil columns with CBA 20, 30 cm was diminished by 92.8, 96.5% respectively in comparison with Na+ accumulated in top soil of soil 60 cm column because CBA layer cut off capillary rise of salts from the subsurface soil. From these results we could conclude that the macroporous layer utilizing CBA placed at subsurface layer cut off capillary rise of solutes from subsurface soil, resulting in lowered level of salinity in top soil and this method can be more effective in newly reclaimed saline soil.

Increase of Salt and Low Temperature Tolerance by Overexpressing Glutathione S-Transferase (GST) Gene (염분과 저온에 대한 내성증진을 위한 GST 유전자의 과발현)

  • Cotton Glutathione S-Transferase (GST: EC 2.5.1.18) was cloned and overexpressed in tobacco (Nicotiana tabacum) plants. Northern blot analysis confirmed the successful transformation of cotton gst gene in tobacco plant. Type I and Type ll transcript patterns were identified in transgenic tobacco plants and only Type I transcripts were discussed in this paper, The activity of GST in the type II transgenic plants was about 1.5-fold higher than those of the wild type and non-expresser by using 1-chloro-2,4-dinitrobenzene (CDNB) and reduced glutathione as the substrate. The expression of cotton GST in tobacco plants proved that Gh-5 could be translated into functional protein. Type II transgenic plants produced functional GST in the cells. The effects of cotton GST in the seedlings was evaluated by growing the control and transgenic seedlings at $15^{\circ}C$ in the growth chamber in the light. Overexpressors were grown well compared to the control plants (non-expressors). lo test far tolerance to salinity, seeds of Gh-5 overexpressors and the wild type Xanthi seedlings were grown at 0, 50, 100, 150, and 200 mM NaCl solution. Gh-5 transgenic seedlings showed higher growth rate over control seedlings on 50 and 100 mM NaCl solution. There was no difference in growth rate at 150 and 200mM NaCl concentration.

Effects of Freshwater Flooding on the Properties of the Accelerated Thermally Aged CSPE (가속열화 된 CSPE의 특성에 미치는 담수침지의 영향)

  • The accelerated thermal aging of a CSPE were carried out for 0, 80.82, 161.63 days at $100^{\circ}C$, which are equal to 0, 40 and 80 years, respectively. The volume electrical resistivities of the non-accelerated thermally aged CSPE and the accelerated thermally aged CSPE for 40y and 80y were $9.620{\times}10^{12}{\sim}1.246{\times}10^{13}{\Omega}{\cdot}cm$, $5.066{\times}10^{12}{\sim}7.576{\times}10^{12}{\Omega}{\cdot}cm$ and $7.195{\times}10^{12}{\sim}9.208{\times}10^{12}{\Omega}{\cdot}cm$ at room temperature, respectively. The dielectric constant of the non-accelerated thermally aged CSPE and the accelerated thermally aged CSPE for 40y and 80y were 3.355~4.030, 2.996~3.963 and 3.020~4.776 at room temperature, respectively. After seawater and freshwater flooding, the volume electrical resistivity of the CSPE trend slightly upward according to drying day at room temperature. After seawater flooding, the dielectric constant of the accelerated thermally aged CSPE were not measured. After seawater flooding, bright open pores of the accelerated thermally aged CSPE were partly transferred to dark close pores due to salinity. After freshwater flooding, dark close pores of the accelerated thermally aged CSPE were partly transferred to bright open pores because salinity of them is decreased. An insulation property of a cable in NPPs was decreased because of the seawater flooding, and an insulation property of them was recovered through the freshwater flooding. As a result, it is considered that an insulation property of a contaminated cable through Tsunami can be recovered if it is cleaned quickly.
