Morphological and Genetic Diversity of Korean Native and Introduced Safflower Germplasm

  • Morphological and genetic diversity of thirty nine safflower germplasm were collected and evaluated by Principal Component Analysis (PCA) and Random Amplified Polymorphic DNA (RAPD) method. Stem length and seeding to flowering days of the safflower germplasm showed $26\~117cm\;and\;76\~179$ days of variation respectively. USA originated germplasm showed higher oil content as $39\%$, but that of Japanese showed lower as $26\%$. PCA made three different cluster groups according to some agronomic characteristics of safflower. Korea originated germplasm showed similar cluster group with that of collected from USA in the PCA of stem length. But in the seeding to flowering days, it showed similar cluster pattern with that of collected from Japan rather than USA. In the experiment of RAPD analysis, total five primers showed polymorphism at the several chromosomal loci. Korea, China Japan and South Central Asia originated germplasm were differently classified with USA and South West Asia originated germplasm with lower similarity coefficient value (0.47). Most of Korea originated germplasm were grouped with South Central Asia originated germplasm with higher similarity coefficient value (0.74) conferring similar genetic background between both of them. China and Japan originated germplasm were dendrogramed with Korea originated germplasm at the 0.65 and 0.50 similarity coefficient values respectively. Some common results were expected from both of PCA and RAPD analysis, but lower genetic heritability caused by relative higher portion of environmental variance and environment by genotype interaction at the expression of those of agronomic characteristics made constraint to find any reliable results.

Quantitative Changes in Phenolic Compounds of Safflower (Carthamus tinctorius L.) Seeds during Growth and Processing

  • Phenolic compounds in safflower seeds were recently found to stimulate bone formation and increase plasma HDL cholesterol levels in estrogen deficient rats, and to inhibit melanin synthesis. Nine phenolic compounds: $N-feruloylserotonin-5-O-{\beta}-D-glucoside,\;8'-hydroxyarctigenin-4'-O-{\beta}-D-glucoside,\;luteolin-7-O-{\beta}-D-glucoside$, N-(p-coumaroyl)serotonin, N-feruloylserotonin, 8'-hydroxy arctigenin (HAG), luteolin (LT), $acacetin-7-O-{\beta}-D-glucuronide$ (ATG) and acacetin (AT), were quantified by HPLC in safflower (Carthamus tinctorius L.) seeds during growth and processing. During growth, levels of the nine phenolic compounds in the seeds increased progressively with increasing growth stages, reached a maximum on July 30 (42nd day after flowering), and then remained relatively constant. During the roasting process, levels of phenolic compounds, except HAG, LT and AT, generally decreased with increased roasting temperature and time, whereas those of HAG, LT and AT increased progressively with increased roasting temperature and time. During the steaming process, levels of other phenolic compounds except HAG and AT generally tended to increase with increased steaming time, whereas those of HAG and AT were scarcely changed. During the microwave treatment, quantitative changes of phenolic compounds were similar to the roasting process, although there were some differences in levels of phenolic compounds between two heat treatments. These results suggest that the steamed safflower seeds after harvesting on late July may be useful as potential dietary supplement source of phenolic compounds for prevention of several pathological disorders, such as atherosclerosis and osteoporosis and aging.

가식성 필름 코팅에 따른 홍화씨 가공제품의 품질 변화 (Changes on the Quality of Safflower Seed Products Coated with Edible Films during Storage)

  • 본 연구는 홍화씨 가공제품의 산패방지 및 품질 향상을 위하여 가식성 필름을 코팅하여 저장 중의 물리화학적 변화를 조사하였다. 가식성 필름 용제로 코팅한 홍화정의 색도 변화는 저장기간이 길어질수록 명도 L 값과 적색도 a 값은 조금씩 증가하였으며 황색도 b 값은 감소하였다. 코팅한 홍화정의 수분함량의 변화는 저장기간이 길어질수록 약간의 증가보였으나 큰 변화는 없었다. 산가는 저장온도에 관계없이 저장기간이 길어질수록 가식성 필름 용제로 코팅한 홍화정의 산가가 대조구에 비해 낮게 나타났다. 과산화물가는 저장기간이 길어질수록 점점 증가를 보였으나 저장 30일째에 $25^{\circ}C$$50^{\circ}C$의 sodium caseinate와 k-carraggenan으로 코팅한 것이 대조구에 비해 $ 50\%$낮았다. 저장기간에 따른 냄새에 대한 관능검사는 두가지의 가식성 필름 용제로 코팅한 홍화정이 대조구에 비해 모두 좋은 결과를 나타내었으며, 그 중에서도 k-carrageenan으로 코팅한 홍화정이 sodium caseinate로 코팅한 것 보다 좋은 결과를 보였다. 관능검사와 유지 산패 측정의 주요 인자인 산가와 과산화물가를 측정한 결과를 종합하여 볼 때, 홍화정의 산패취를 방지하고 품질의 보존을 위한 방법으로 가식성 코팅제인 caseinate와 k-carrageenan 용제로 코팅하는 것이 매우 효과적이었다.

Anticancer Activity of the Safflower Seeds (Carthamus tinctorius L.) through Inducing Cyclin D1 Proteasomal Degradation in Human Colorectal Cancer Cells

  • The seed of safflower (Carthamus tinctorius L) has been reported to suppress human cancer cell proliferation. However, the mechanisms by which safflower seed inhibits cancer cell proliferation have remained nuclear. In this study, the inhibitory effect of the safflower seed (SS) on the proliferation of human colorectal cancer cells and the potential mechanism of action were examined. SS inhibited markedly the proliferation of human colorectal cancer cells (HCT116, SW480, LoVo and HT-29). In addition, SS suppressed the proliferation of human breast cancer cells (MDA-MB-231 and MCF-7). SS treatment decreased cyclin D1 protein level in human colorectal cancer cells and breast cancer cells. But, SS-mediated downregulated mRNA level of cyclin D1 was not observed. Inhibition of proteasomal degradation by MG132 attenuated cyclin D1 downregulation by SS and the half-life of cyclin D1 was decreased in SS-treated cells. In addition, SS increased cyclin D1 phosphorylation at threonine-286 and a point mutation of threonine-286 to alanine attenuated SS-mediated cyclin D1 degradation. Inhibition of ERK1/2 by PD98059 suppressed cyclin D1 phosphorylation and downregulation of cyclin D1 by SS. In conclusion, SS has anti-proliferative activity by inducing cyclin D1 proteasomal degradation through ERK1/2-dependent threonine-286 phosphorylation of cyclin D1. These findings suggest that possibly its extract could be used for treating colorectal cancer.

Safflower Bud Dietary Prevents Ovariectomy-induced Osteoporosis in Rats

  • Safflower (Carthamus tinctorius L.) seeds have long been clinically used in Korea to promote bone formation and prevent osteoporosis. In addition, the safflower buds (SB) were found to have more useful functional ingredients than safflower seed. Thus, we investigated the preventive effects of SB diet in ovariectomized (OVX) rats. The rats were divided into five groups; sham operated group, OVX alone group, OVX plus 17β-estradiol (E2 10 μg/kg, i.p.) and OVX plus SB diet feeding group (0.3% or 1%). Feeding of SB diet (0.3% or 3%) to OVX rats markedly increased trabecular formation in femur compared to OVX rats. Feeding of SB diet (0.3% or 3%) to OVX rats also decreased TRAP activity compared to OVX rats. These results suggest that SB diets have bone sparing effects by the decrease of osteoclast activity. We also observed that OVX rats fed with SB diet (0.3% or 3%) exhibited the decrease of calcium and phosphorus in serum compared to OVX-induced rats. Therefore, SB may be beneficial for the patients of osteoporosis, especially in postmenopausal women.

유지의 알칼리 이성질화에 의한 Conjugated Linoleic Acid 농축물의 제조 (Preparation of Conjugated Linoleic Acid Concentrate from Vegetable Oils by Alkali Isomerization)

  • 항산화 및 항암 작용이 있는 것으로 알려진 conjugated linoleic acid(CLA)를 식물성 유지로부터 알칼리 이성질화에 의해 고 농도의 CLA 농축물을 얻기 위한 최적 조건을 연구하였다. 지방산 조성이 상이한 식물성 유지를 알칼리 이성질화 하였을 때 CLA 생성량은 유지중 linoleic acid 함량이 많은 것이 가장 높았다. 홍화유를 알칼리 이성질화할 때 KOH 농도는 $8{\sim}11%$, 가열온도는 $180{\sim]185^{\circ}C$에서 cis-9, trans-11 CLA와 총 CLA 생성량이 가장 높았고, 가열시간은 CLA 생성량에 큰 영향을 미치지는 않았으나 $20{\sim}40$분이 가장 적당하였다. 홍화유를 silicic acid column chromatograph)에 의해 중성, 당 및 인지방질의 lipid class를 분획하여 알칼리 이성질화를 시켰을 때 중성지방질의 획분에 CLA가 가장 많이 함유되었다. 알칼리 이성질화시킨 홍화유를 요소처리와 HPLC 분획에 의해 95.4%의 CLA 농축물을 얻을 수 있었다.

홍화종실분말 식이가 고지방-고콜레스테롤섭취 흰쥐의 지질대사에 미치는 영향 (effects of Safflower Seed(Carthamus timctorious L. Powder on Lipid Metabolism in High Fat and High Cholesterol-Fed Rats)

  • A study was carried out to investigate the effects of safflower seed powder on the improvement of lipid metabolism in high fat and high cholesterol fed rats. Male Sprague Dawley rats of 10 weeks old, weighing 325$\pm$5g, were divided into two groups; the control group(C group, high fat(10% lard) and high cholesterol(1% cholesterol)) and safflower seed group(S group, 10% safflower seed powder), they were fed experimental diets for 6 weeks. Food intake, body weight gains and organ weight had little differences between the groups. Concentration of lipoprotein in serum was remarkably lower in S group than in C group. Serum cholesterol levels were significantly lower in S group(72.94$\pm$4.08 mg/dl) than in C group(89.41$\pm$4.19mg/dl). The level of serum HDL cholesterol was higher in S group than in C group. The level of serum LDL C was significantly lower in S group than in C group. The ratio of HDL cholesterol to total cholesterol were higher in the S group than in the C group, too. The value of atherogenic index(AI) was determined to be low in S group. The content of liver triglyceride and cholesterol in the S group was lower than that of C group. ACAT activities which involves in cholesterol esterification in liver, was not significantly different between two groups.

홍화의 종자 크기별 발아 및 생장특성 분석 (Effect of Seed Size on Seed Germination and Growth Characteristics in Safflower (Carthamus tinctorius L.))

  • In order to evaluate the effect of seed weight on different aspect of safflower (Carthamus tinctorius L.) seed germination and growth characteristics. Quantity of sinapine leaked from seed was greater as the viability of seeds was dropped by the time elapsed of seed aging model and long storage condition in safflower (Carthamus tinctorius L.). The cultivar of safflower was Jin-Sun and the seeds that are separated to three different weights of small, middle, and large were used in this experiment. Large seeds revealed the highest germination percent, coleoptiles fresh weight, coleoptiles dry weight, radicle fresh weight and 1000 seed weights than other seed weight. Seed weight had little effect on yield while seed number exerted a positive influence. Interestingly, yield per plant and its major components, number of capsules and capsule weights, revealed a negligible relationship with oil content.

홍화 수집종의 주요 형태적 특성 (Major Morphological Characteristics of Safflower(Carthamus tinctorius L.) Accessions)

  • 홍화의 새로운 품종육성을 위하여 국내외에서 수집된 홍화의 외관상 나타나는 형태적인 특성을 조사한 결과를 요약하면 다음과 같다. 1. 수집된 홍화의 16계통중 단간(90∼110cm)인것이 6계통, 중간(111∼130cm)인 것은 5계통, 장간(131cm이상)인것은 5계통이었으며 장간은 국외 도입계통이었다. 2. 국내 수집 종 2계통은 주당 분지수가 11∼13개의 소분지형이었으며 도입종들은 50개이상의 다분 지형이었다. 3. 잎의 형태는 장타원형이 3계통, 피침형이 10계통, 넓은 선상형은 3계통으로 피침형이 가장 많았으며 잎의 가장자리 거치형태는 파도 물결 모양의 둔거치가 7계통으로 가장 많았고 4계통은 거치가 있었으며 깊게 찢어진 심열거치는 5계통이었다. 4.꽃의 색깔은 백색 1계통, 황색 13계통, 주황색 2계통이었으며 두장화의 형태는 원뿔형이 대부분이며(14계통)편형이 2계통이었고 화두 포엽의 끝은 가시형이 11계통이었고 무가시형은 5계통이었다. 5. 종자의 형태는 원뿔형이 11계통, 타원형은 5계통으로 국내 수집계통은 모두 원뿔형이었으며 도입계통은 타원형으로 대립이었다.

난소적출 rat에 녹용, 홍삼, 홍화, ipriflavone 및 estrogen을 투여 했을 때 호르몬, Ca, P 및 ALP수준에 미치는 영향에 관한 연구 (Effects of antler, red ginseng, safflower, ipriflavone and estrogen on hormones, Ca, P and ALP levels in ovariectomized rats)

  • This study was performed to elucidate the effects of antler, red ginseng, safflower seed, ipriflavone and estrogen on ovariectomized rats. The rats were fed with Ca and P deficient diet for five weeks to induce osteoporosis. After this period, these animals were fed with normal feed and treated every other day with antler(600 mg/kg, p.o), red ginseng(200 mg/kg, p.o), safflower(200 mg/kg, p.o), ipriflavone(80 mg/kg, p.o) and estrogene(400$\mu\textrm{g}$/kg, i.m) for 5 weeks. During the treatment, the rats were examined for serum concentrations of estradiol, calcitonin, Ca, p and alkaline phosphatase(ALP) activities. The results are summarized as follows 1. The levels of serum 17 $\beta$-estradiol after five weeks of treatment were showed 39.6$\pm$3.0 pg/ml by antler, 33.2$\pm$2.5pg/ml by red ginseng, 34.9$\pm$2.4pg/ml by safflower, 28.1$\pm$3.1 pg/ml by ipriflavone and 40.6$\pm$3.0pg/ml by estrogen-treated group. They were lower than 50.8$\pm$3.1pg/ml of normal control group which had not received ovariectomy. They, however, were significantly higher than 26.8$\pm$1.8pg/ml of ovariectomized non-treatment group(p<0.05). 2. The levels of serum calcitonin after five weeks of treatment were showed 0.60$\pm$0.02 ng/ml by antler, 0.55$\pm$0.04ng/ml by red ginseng, 0.59$\pm$0.02ng/ml by safflower, 0.56$\pm$0.04ng/ml by ipriflavone and 0.62$\pm$0.02ng/ml by estrogen-treated group. They were lower than 0.67$\pm$0.03pg/ml of normal control group. However, they were significantly higher than 0.45$\pm$0.05ng/ml of ovariectomized non-treatment group(p<0.05). 3. The levels of serum Ca of the rats after five weeks of treatment with antler, red ginseng, safflower, ipriflavone and estrogen were 23.51$\pm$2.19$\mu\textrm{g}$/$m\ell$, 25.22$\pm$3.44$\mu\textrm{g}$/$m\ell$, 23.20$\pm$0.02$\mu\textrm{g}$/$m\ell$, 24.76$\pm$3.57$\mu\textrm{g}$/$m\ell$, 23.07$\pm$3.66$\mu\textrm{g}$/$m\ell$, respectively. They were a bit higher than 21.43$\pm$2.22$\mu\textrm{g}$/$m\ell$ of normal control group. And non-treatment group showed 26.12$\pm$0.29$\mu\textrm{g}$/$m\ell$ which was significantly higher than that of control group(P<0.05). 4. The serum P concentraions after five weeks of treatment were showed 12.11$\pm$2.14$\mu\textrm{g}$/$m\ell$ by antler, 13.18$\pm$1.64u91m4 by red ginseng, 12.67 $\pm$2.31$\mu\textrm{g}$/$m\ell$ by safflower, 12.38$\pm$2.07$\mu\textrm{g}$/$m\ell$ by ipriflavone, 11.86$\pm$1.93$\mu\textrm{g}$/$m\ell$ by estrogen-treated group. They were a bit higher than 11.29$\pm$1.23$\mu\textrm{g}$/$m\ell$ of normal control group. And non-treatment group showed 13.42$\pm$1.87$\mu\textrm{g}$/$m\ell$ which was higher than that of control group but not significant. 5. The levels of serum ALP after five weeks of treatment were showed 164.8$\pm$3.8IU/ml by antler, 277.7$\pm$4.8IU/ml by red ginseng, 288.5$\pm$4.5IU/ml by safflower, 214.7$\pm$5.7IU/ml by ipriflavone and 159.4$\pm$5.4IU/ml by estrogen-treated group. They were significantly higher than 144.1$\pm$3.5IU/ml of normal control group(p<0.05). However, they were significantly lower than 336.9 $\pm$12.7IU/ml of ovariectomized non-treatment group(p<0.05). Antler and safflower elevated serum estradiol and calcitonin, and decreased serum ALP significantly. Therefore they were thought to have therapeutic effect on osteoposis by making inhibitory effect on osteoclasts rather than activating osteoblasts.
