Morphological and Genetic Diversity of Korean Native and Introduced Safflower Germplasm

  • Morphological and genetic diversity of thirty nine safflower germplasm were collected and evaluated by Principal Component Analysis (PCA) and Random Amplified Polymorphic DNA (RAPD) method. Stem length and seeding to flowering days of the safflower germplasm showed $26\~117cm\;and\;76\~179$ days of variation respectively. USA originated germplasm showed higher oil content as $39\%$, but that of Japanese showed lower as $26\%$. PCA made three different cluster groups according to some agronomic characteristics of safflower. Korea originated germplasm showed similar cluster group with that of collected from USA in the PCA of stem length. But in the seeding to flowering days, it showed similar cluster pattern with that of collected from Japan rather than USA. In the experiment of RAPD analysis, total five primers showed polymorphism at the several chromosomal loci. Korea, China Japan and South Central Asia originated germplasm were differently classified with USA and South West Asia originated germplasm with lower similarity coefficient value (0.47). Most of Korea originated germplasm were grouped with South Central Asia originated germplasm with higher similarity coefficient value (0.74) conferring similar genetic background between both of them. China and Japan originated germplasm were dendrogramed with Korea originated germplasm at the 0.65 and 0.50 similarity coefficient values respectively. Some common results were expected from both of PCA and RAPD analysis, but lower genetic heritability caused by relative higher portion of environmental variance and environment by genotype interaction at the expression of those of agronomic characteristics made constraint to find any reliable results.

Quantitative Changes in Phenolic Compounds of Safflower (Carthamus tinctorius L.) Seeds during Growth and Processing

  • Phenolic compounds in safflower seeds were recently found to stimulate bone formation and increase plasma HDL cholesterol levels in estrogen deficient rats, and to inhibit melanin synthesis. Nine phenolic compounds: $N-feruloylserotonin-5-O-{\beta}-D-glucoside,\;8'-hydroxyarctigenin-4'-O-{\beta}-D-glucoside,\;luteolin-7-O-{\beta}-D-glucoside$, N-(p-coumaroyl)serotonin, N-feruloylserotonin, 8'-hydroxy arctigenin (HAG), luteolin (LT), $acacetin-7-O-{\beta}-D-glucuronide$ (ATG) and acacetin (AT), were quantified by HPLC in safflower (Carthamus tinctorius L.) seeds during growth and processing. During growth, levels of the nine phenolic compounds in the seeds increased progressively with increasing growth stages, reached a maximum on July 30 (42nd day after flowering), and then remained relatively constant. During the roasting process, levels of phenolic compounds, except HAG, LT and AT, generally decreased with increased roasting temperature and time, whereas those of HAG, LT and AT increased progressively with increased roasting temperature and time. During the steaming process, levels of other phenolic compounds except HAG and AT generally tended to increase with increased steaming time, whereas those of HAG and AT were scarcely changed. During the microwave treatment, quantitative changes of phenolic compounds were similar to the roasting process, although there were some differences in levels of phenolic compounds between two heat treatments. These results suggest that the steamed safflower seeds after harvesting on late July may be useful as potential dietary supplement source of phenolic compounds for prevention of several pathological disorders, such as atherosclerosis and osteoporosis and aging.

Changes on the Quality of Safflower Seed Products Coated with Edible Films during Storage (가식성 필름 코팅에 따른 홍화씨 가공제품의 품질 변화)

  • This study analyzed the change of physical and chemical characteristics on the safflower seed produce coated edible film. L and a value of safflower seed product coated edible film were slowly increased during storage, while b value decreased. Moisture content were increased during storage. Acid valus welt less increased when edible film was ased. Peroxide values were also increased during storage, but peroxide value in products coated sodium caseinate or k-carraggenan was $50\%$ in the control after 30 days of storage. The value of sensual test was higher in products coated sodium caseinate or k-carraggenan than those of control. Coating with sodium caseinate or k-carraggenan was effective for acidification retardation and quality improvement of safflower seed products.

Anticancer Activity of the Safflower Seeds (Carthamus tinctorius L.) through Inducing Cyclin D1 Proteasomal Degradation in Human Colorectal Cancer Cells

  • The seed of safflower (Carthamus tinctorius L) has been reported to suppress human cancer cell proliferation. However, the mechanisms by which safflower seed inhibits cancer cell proliferation have remained nuclear. In this study, the inhibitory effect of the safflower seed (SS) on the proliferation of human colorectal cancer cells and the potential mechanism of action were examined. SS inhibited markedly the proliferation of human colorectal cancer cells (HCT116, SW480, LoVo and HT-29). In addition, SS suppressed the proliferation of human breast cancer cells (MDA-MB-231 and MCF-7). SS treatment decreased cyclin D1 protein level in human colorectal cancer cells and breast cancer cells. But, SS-mediated downregulated mRNA level of cyclin D1 was not observed. Inhibition of proteasomal degradation by MG132 attenuated cyclin D1 downregulation by SS and the half-life of cyclin D1 was decreased in SS-treated cells. In addition, SS increased cyclin D1 phosphorylation at threonine-286 and a point mutation of threonine-286 to alanine attenuated SS-mediated cyclin D1 degradation. Inhibition of ERK1/2 by PD98059 suppressed cyclin D1 phosphorylation and downregulation of cyclin D1 by SS. In conclusion, SS has anti-proliferative activity by inducing cyclin D1 proteasomal degradation through ERK1/2-dependent threonine-286 phosphorylation of cyclin D1. These findings suggest that possibly its extract could be used for treating colorectal cancer.

Safflower Bud Dietary Prevents Ovariectomy-induced Osteoporosis in Rats

  • Safflower (Carthamus tinctorius L.) seeds have long been clinically used in Korea to promote bone formation and prevent osteoporosis. In addition, the safflower buds (SB) were found to have more useful functional ingredients than safflower seed. Thus, we investigated the preventive effects of SB diet in ovariectomized (OVX) rats. The rats were divided into five groups; sham operated group, OVX alone group, OVX plus 17β-estradiol (E2 10 μg/kg, i.p.) and OVX plus SB diet feeding group (0.3% or 1%). Feeding of SB diet (0.3% or 3%) to OVX rats markedly increased trabecular formation in femur compared to OVX rats. Feeding of SB diet (0.3% or 3%) to OVX rats also decreased TRAP activity compared to OVX rats. These results suggest that SB diets have bone sparing effects by the decrease of osteoclast activity. We also observed that OVX rats fed with SB diet (0.3% or 3%) exhibited the decrease of calcium and phosphorus in serum compared to OVX-induced rats. Therefore, SB may be beneficial for the patients of osteoporosis, especially in postmenopausal women.

Preparation of Conjugated Linoleic Acid Concentrate from Vegetable Oils by Alkali Isomerization (유지의 알칼리 이성질화에 의한 Conjugated Linoleic Acid 농축물의 제조)

  • The optimal conditions of alkali isomerization to obtain conjugated linoleic acid (CLA) concentrate from vegetable oils which have antioxidant and anticarcinogenic properties were studied. The result of alkali isomerization of various vegetable oils indicated that CLA content of safflower oil which contains more linoleic acid than any other vegetable oils was the highest of all experimental vegetable oils. During alkali isomerization, the amount of cis-9, trans-11 CLA and total CLA content in safflower oil was the highest at $8{\sim}11%$ KOH concentration and $180{\sim}185^{\circ}C$. But heating time had no effect on CLA formation after $20{\sim}40$ minutes. As a result of alkali isomerization of neutral lipid, glycolipid and phospholipid in safflower oil, CLA content of neutral lipid class was higher than any other lipid classes. By urea treatment and HPLC fractionation, total CLA content in alkali-isomerized safflower oil increased to 95.4% from 78.9%.

  • A study was carried out to investigate the effects of safflower seed powder on the improvement of lipid metabolism in high fat and high cholesterol fed rats. Male Sprague Dawley rats of 10 weeks old, weighing 325$\pm$5g, were divided into two groups; the control group(C group, high fat(10% lard) and high cholesterol(1% cholesterol)) and safflower seed group(S group, 10% safflower seed powder), they were fed experimental diets for 6 weeks. Food intake, body weight gains and organ weight had little differences between the groups. Concentration of lipoprotein in serum was remarkably lower in S group than in C group. Serum cholesterol levels were significantly lower in S group(72.94$\pm$4.08 mg/dl) than in C group(89.41$\pm$4.19mg/dl). The level of serum HDL cholesterol was higher in S group than in C group. The level of serum LDL C was significantly lower in S group than in C group. The ratio of HDL cholesterol to total cholesterol were higher in the S group than in the C group, too. The value of atherogenic index(AI) was determined to be low in S group. The content of liver triglyceride and cholesterol in the S group was lower than that of C group. ACAT activities which involves in cholesterol esterification in liver, was not significantly different between two groups.

Effect of Seed Size on Seed Germination and Growth Characteristics in Safflower (Carthamus tinctorius L.) (홍화의 종자 크기별 발아 및 생장특성 분석)

  • In order to evaluate the effect of seed weight on different aspect of safflower (Carthamus tinctorius L.) seed germination and growth characteristics. Quantity of sinapine leaked from seed was greater as the viability of seeds was dropped by the time elapsed of seed aging model and long storage condition in safflower (Carthamus tinctorius L.). The cultivar of safflower was Jin-Sun and the seeds that are separated to three different weights of small, middle, and large were used in this experiment. Large seeds revealed the highest germination percent, coleoptiles fresh weight, coleoptiles dry weight, radicle fresh weight and 1000 seed weights than other seed weight. Seed weight had little effect on yield while seed number exerted a positive influence. Interestingly, yield per plant and its major components, number of capsules and capsule weights, revealed a negligible relationship with oil content.

Major Morphological Characteristics of Safflower(Carthamus tinctorius L.) Accessions (홍화 수집종의 주요 형태적 특성)

  • Morphological characteristics of 16 collected safflower (Carthamus tinctorius L.)accessions were studied at Gyongbuk A.T.A. Uisong Medicinal Plant Experiment Station in 1998. As for plant height, 5 lines were tall(more than 131cm), which were mainly foreign safflower. Middle and small were 5 and 6 accessions, respectively. Number of branch per plant was 11∼13 in 2 accessions of domestic safflowers while foreign safflowers have more than 50 branches per plant. Leaf shapes were oblong(3 lines), lanceolate(10 lines) and broadly linear(3 lines). There were three types of leaf margin shape which were repand(7 lines), serrate(4 lines) and parted serrate(5 lines). Flower color was white(1 line), yellow(13 lines) and red orange(2 lines). As for flower head shape, 14 lines were conical and 2 lines were flattened. There were thorns on bract of flower head in 11 lines but in 5 lines there were not. Domestic safflower lines had seeds of conical shape. Contrastively, seed shapes of foreign saffower lines were oval.

Effects of antler, red ginseng, safflower, ipriflavone and estrogen on hormones, Ca, P and ALP levels in ovariectomized rats (난소적출 rat에 녹용, 홍삼, 홍화, ipriflavone 및 estrogen을 투여 했을 때 호르몬, Ca, P 및 ALP수준에 미치는 영향에 관한 연구)

  • This study was performed to elucidate the effects of antler, red ginseng, safflower seed, ipriflavone and estrogen on ovariectomized rats. The rats were fed with Ca and P deficient diet for five weeks to induce osteoporosis. After this period, these animals were fed with normal feed and treated every other day with antler(600 mg/kg, p.o), red ginseng(200 mg/kg, p.o), safflower(200 mg/kg, p.o), ipriflavone(80 mg/kg, p.o) and estrogene(400$\mu\textrm{g}$/kg, i.m) for 5 weeks. During the treatment, the rats were examined for serum concentrations of estradiol, calcitonin, Ca, p and alkaline phosphatase(ALP) activities. The results are summarized as follows 1. The levels of serum 17 $\beta$-estradiol after five weeks of treatment were showed 39.6$\pm$3.0 pg/ml by antler, 33.2$\pm$2.5pg/ml by red ginseng, 34.9$\pm$2.4pg/ml by safflower, 28.1$\pm$3.1 pg/ml by ipriflavone and 40.6$\pm$3.0pg/ml by estrogen-treated group. They were lower than 50.8$\pm$3.1pg/ml of normal control group which had not received ovariectomy. They, however, were significantly higher than 26.8$\pm$1.8pg/ml of ovariectomized non-treatment group(p<0.05). 2. The levels of serum calcitonin after five weeks of treatment were showed 0.60$\pm$0.02 ng/ml by antler, 0.55$\pm$0.04ng/ml by red ginseng, 0.59$\pm$0.02ng/ml by safflower, 0.56$\pm$0.04ng/ml by ipriflavone and 0.62$\pm$0.02ng/ml by estrogen-treated group. They were lower than 0.67$\pm$0.03pg/ml of normal control group. However, they were significantly higher than 0.45$\pm$0.05ng/ml of ovariectomized non-treatment group(p<0.05). 3. The levels of serum Ca of the rats after five weeks of treatment with antler, red ginseng, safflower, ipriflavone and estrogen were 23.51$\pm$2.19$\mu\textrm{g}$/$m\ell$, 25.22$\pm$3.44$\mu\textrm{g}$/$m\ell$, 23.20$\pm$0.02$\mu\textrm{g}$/$m\ell$, 24.76$\pm$3.57$\mu\textrm{g}$/$m\ell$, 23.07$\pm$3.66$\mu\textrm{g}$/$m\ell$, respectively. They were a bit higher than 21.43$\pm$2.22$\mu\textrm{g}$/$m\ell$ of normal control group. And non-treatment group showed 26.12$\pm$0.29$\mu\textrm{g}$/$m\ell$ which was significantly higher than that of control group(P<0.05). 4. The serum P concentraions after five weeks of treatment were showed 12.11$\pm$2.14$\mu\textrm{g}$/$m\ell$ by antler, 13.18$\pm$1.64u91m4 by red ginseng, 12.67 $\pm$2.31$\mu\textrm{g}$/$m\ell$ by safflower, 12.38$\pm$2.07$\mu\textrm{g}$/$m\ell$ by ipriflavone, 11.86$\pm$1.93$\mu\textrm{g}$/$m\ell$ by estrogen-treated group. They were a bit higher than 11.29$\pm$1.23$\mu\textrm{g}$/$m\ell$ of normal control group. And non-treatment group showed 13.42$\pm$1.87$\mu\textrm{g}$/$m\ell$ which was higher than that of control group but not significant. 5. The levels of serum ALP after five weeks of treatment were showed 164.8$\pm$3.8IU/ml by antler, 277.7$\pm$4.8IU/ml by red ginseng, 288.5$\pm$4.5IU/ml by safflower, 214.7$\pm$5.7IU/ml by ipriflavone and 159.4$\pm$5.4IU/ml by estrogen-treated group. They were significantly higher than 144.1$\pm$3.5IU/ml of normal control group(p<0.05). However, they were significantly lower than 336.9 $\pm$12.7IU/ml of ovariectomized non-treatment group(p<0.05). Antler and safflower elevated serum estradiol and calcitonin, and decreased serum ALP significantly. Therefore they were thought to have therapeutic effect on osteoposis by making inhibitory effect on osteoclasts rather than activating osteoblasts.
