  • 1. The experiment was conducted to investigate the physiological characteristics on ten isolates of bacterial leaf blight pathogen of rice, Xanthomenas eryzae Dowson. Seven out of tin isolates were isolated from infected leaves of various rice varieties including IR strains in Korea. Isolates S-20 and S-103 were originated from IRRI in Philippines, and isolate H-5809 was allocated by National Institute of Agricultural Science in Japan. 2. All isolates Produced hydrogen sulfide and ammonia 9as from peptone sol. media, and reduced methylen blue, Gelatin liquefaction occurred by all isolates tested, although each isolate showed different degree of liquefaction No coagulation of the casein in milk by the isolates was observed, though some amount of acid production occurred in litmus milk by ail isolates tested. 3. All isolates utilized glucose and galactose, and slight utilization of esculin, mannitol, raffinose, salicin and saccharose was observed. Lactose, starch and dextrin, however, were not utilized at all by all isolates tested in the study.

Studies on the Production of Gibberellic acid (지베렐린 생산에 관한 연구)

  • By the treatment of Gibberella fujkuroi I-892 with mutagen such as UV light and N-methyl-N'-nitro-N-nitrosoguanidine, a mutant G. fujkuroi G-471 was selected as the highest producer of gibberellic acid among 800 mutant strains. It showed 30% increase of production yield compared with that of the parent strain. At optimum medium composition (saccharose 1.0%, ammonium tartarate 50mM, malt extract 1.0% KH$_2$PO$_4$ 0.5%, MgSO$_4$0.5%, FeSO$_4$0.0002%, trace element sol.0.002% (v/v), the yield of submerged culture increased by 30% after 7 days culture at 24$^{\circ}C$ (253mg/$\ell$). In submerged culture, the initial pH showed much effects on the increase of gibberellic acid production. The highest yield of the production was attained with pH adjustment to 4.0 at the initial stage of fermention.

  • For the purpose of utilizing cellulose resources by cellulolytic enzymes of Roseofomes subflexibilis, it's cultural conditions for the production of cellulolytic enzymes in synthetic media were investigated. The results obtained were summarized as follows : The optimum temperature and pH for the enzyme production were $30^{\circ}C$ and pH 4.0, respectively. Among the carbon sources, glucose was good for the production of cellulase. The optimum concentration of saccharose as 1.2%. As a organic nitrogen source, yeast extract was good for the mycelial growth. The optimum concentration of yeast extract as 1.5%. As a inorganic nitrogen source, $NH_{4}H_{2}PO_{4}$ was good for the mycelial growth. The optimum concentration of $NH_{4}H_{2}PO_{4}$ were 1.1%. The mineral salt of $Al_{2}(SO_{4})_{3}$ was effective and the optimum concentration was 0.1 M.

Fabrication and Characterization of Spherical Carbon-Coated Li3V2(PO4)3 Cathode Material by Hydrothermal Method with Reducing Agent

  • Spherical $Li_3V_2(PO_4)_3$ (LVP) and carbon-coated LVP with a monoclinic phase for the cathode materials are synthesized by a hydrothermal method using $N_2H_4$ as the reducing agent and saccharose as the carbon source. The results show that single phase monoclinic LVP without impurity phases such as $LiV(P_2O_7)$, $Li(VO)(PO_4)$ and $Li_3(PO_4)$ can be obtained after calcination at $800^{\circ}C$ for 4 h. SEM and TEM images show that the particle sizes are $0.5{\sim}2{\mu}m$ and the thickness of the amorphous carbon layer is approximately 3~4 nm. CV curves for the test cell are recorded in the potential ranges of 3.0~4.3 V and 3.0~4.8 V at a scan rate of $0.01mV\;s^{-1}$ and at room temperature. At potentials between 3.0 and 4.8 V, the third $Li^+$ ions from the carbon-coated LVP can be completely extracted, at voltages close to 4.51 V. The carbon-coated LVP exhibits an initial specific discharge capacity of $118mAh\;g^{-1}$ in the voltage region of 3.0 to 4.3 V at a current rate of 0.2 C. The results indicate that the reducing agent and carbon source can affect the crystal structure and electrochemical properties of the cathode materials.

Cultural Conditions for Mass Production of Antagonistic Bacillus subtills CAP134 (길항균 Bacillus subtillis CAP134의 대량생산을 위한 배양조건)

  • Cultural conditions for mass production of the antagonistic bacteria, Bacillus subtills CAP134 against pathogens causing major airborne diseases to apple tree, effect of temperature, pH, carbon and nitrogen source in the culture broth were investigated. The bacterial growth was most vigorous when the temperature and pH of the culture broth was 30~$35^{\circ}$C, and 7, respectively. As for carbon source, dextrose was best followed in order by dextrose(monosaccharides)>sucrose(disaccharides)$\geq$saccharose(di-saccharides)>starch (polysaccharides). Among different sugars, bacterial growth was favored by in the order of brown, black and white sugars, indicating that the bacterial growth might be promoted by the minor elements presented as impurities in the less purified sugars. As for nitrogen source, organic forms were better to bacterial growth than inorganic forms, that is polypeptone was best followed in order by soy sauce, soybeen milk and inoganic nitrogens. Differences in bacterial growth among different forms of inorganic nitrogen were negligible.

  • Isolates of Ophiocordyceps heteropoda (Kobayasi) collected from Mt. Halla on Jeju-do, Korea were tested for mycelial growth on different agar media and in the presence of different carbon and nitrogen sources. Similarly, isolates were also incubated at different temperatures as well as under continuous light and dark conditions. Growth was better on Hamada agar, basal medium, and malt-yeast agar, but poor on Czapek-Dox agar. Different carbon sources such as dextrin, saccharose, starch, lactose, maltose, fructose, and dextrose resulted in better growth. Complex organic nitrogen sources such as yeast extract and peptone revealed the most effective growth. Mycelial growth was best at $25^{\circ}C$. The growth rate was faster in the dark than the light, but mycelial density was less compact in the dark.

  • We have selected an valuable pmbiotic strain; Lactobacillus sakei BK19 which has wide antagonic spectrum against fish pathogens . Present study investigated cultural characterization of L. sakei BK19 including pH tolerance , susceptibility of antibacterial agents and growth pattern with different environment such as nutritions, temperature and salinity. L. sakei BK19 showed Significantly higher resistance at low pH(around pH 4) environment and relative high antibiotic tolerance . In the study of optimal culture condition, maltose and saccharose provided the optimal nutritional culture condition while lactose and mannitol were unable to supply its carbon source for the fermentation of L. sakei BK19. Moreover. L. sakei BK19 showed good growth at the temperature range of 15 to $45^{o}C$ und the NaCl concentration of 0 to 7%. Hence, this particular probiotic strain may be benificial both in seawater and fresh weter conditions.

  • The synthesis alcohol-oxidase[EC 1.1.3.13] was investigated in the yeasts, Candida boidinii CBS 8106 and C. boidinii CBS 2428, during growth on different carbon sources. Alcohol-oxidase was undetectable in all strains submitted to the test in the mineral salts medium containing 1.0% glucose, but its production was rapidly increased when the carbon source was changed glucose to 1.0% methanol after 24hrs of incubation. When cells were grown on the various carbon sources (glucose, xylose, lactose, glycerol, galactose, saccharose, sorbose, lactic acid or acetic acid), the alcohol-oxidase activity was undetected. These carbon sources together with methanol yielded far better synthesis of alcohol-oxidase than in the case of carbon sources alone. Alcohol-oxidase was active towards alcohol of shorter alkyl-chain length than C5 and unsaturated alcohols. Its affinity for these alcohols decreased with the increasing length of the alkyl-chain. The apparent Km values for the methanol of Candida boidinii CBS 8106 and C. boidinii CBS 2428 were 1.96 and 1.21, respestively.

  • Hot water extractives of sawdust/particle from domestic and foreign wood species, which were composed of pitch pine, Korean pine, larch, Italy poplar, acacia and oak as Korean wood species, Malaysian oil palm and German spruce were quantitatively analyzed with thin layer chromatography. Sugar components of saccharose, galactose, glucose, fructose and arabinose were contained in these wood species. It was assumed that arabinose and glucose are major inhibitory components against cement hardening in larch and oil palm, respectively, since both species contain a large amount of each sugar. In contrast, fructose might not influence so badly on a cement hardening, when considering that fructose was contained much in Italy poplar with a good cement hardening character. Galactose was a minor component.
