찹쌀가루를 첨가한 솔설기의 재료배합비에 따른 관능적ㆍ텍스쳐 특성 (Sensory and textural characteristics of Solsulgi using varied levels of pine leaves powder and different types of sweeteners)

  • The objectives of this study was to investigate the sensory and quality characteristics of Solsulgi made from rice flour and glutinous rice flour containing 1, 2, or 3% of pine leaves powder. The results of sensory evaluation showed that Solsulgi containing 1% pine leaves powder had high overall acceptability, chewiness and sweetness preference. In the results of textural analysis, the hardness was decreased by adding pine leaves powder. Hunter color L-value of Solsulgi decreased by increasing the level of pine leaves powder. The more pine leaves powder was added, the a-value and b-value of Solsulei were decreased. The moisture content was higher in Solsulgi with sugar than honey and oligo-saccharide.

Purification and Characterization of Two Isolectins with Arginase Activity from the Lichen Xanthoria parietina

  • Two glycoproteins were purified and biochemically characterized from the lichen X. parietina. Both behaved as enzymes with arginase activity and haemaglutinins. Secreted arginase (SA) contained galactose and glucose in the saccharide moiety and an isoelectric point of 4.54. The algal binding-protein (ABP) had N-acetyl-glucosamine and glucose as glycosidic residues and an isoelectric point of 3.53. Both proteins had the same molecular mass (58.6 kDa) and the same qualitative amino acidic composition. The results allowed us to consider these glycoproteins as isolectins, which have significant physiological roles in the relationship between photobiont and mycobiont of symbiotic association.

  • The purpose of this study was to find the effect of honey on the sugar metabolism In Sprague-Dawley rats. All experimental rats were fed ad libitum, for seven weeks, 68% saccharide diet anti 10% or 20% honey from acacia, sumac and miscellaneous flower honey, respectively, and sucrose. The amount of glucose in whole blood was increased in all groups fed with honey except the group fed with 10% sumac honey solution. The amount of fructosamine in serum of rat taken diet with honey solution was increased in comparison with normal group, control group, sucrose group. The amount of fructosamine in serum of rat taken diet with honey solution was high in order of sumac honey > miscellaneous flower honey > acacia honey. The amount of lactate and pyruvate in whole blood of rat taken diet with sucrose group or honey group were decreased in comparison with the normal group and control group. The amount of $\beta$-hydroxybutyrate in serum of rat taken diet with sucrose or honey was increased in comparison with the normal group, control group.

  • The rheological properties of seven mixed sugar solutions of glucose, fructose, maltose, sucrose, and two kinds of honey, the chestnut and the acacia, were discussed. The fluidity of the above samples were studied by using the applied pressure horizontal viscometer with which glycerine was confirmed to be a Newtonian flow at $25^{\circ}C$. Time required to pass the constant volume of the sample fluids through the capillary was measured at various pressure differences, ${\Delta}p$, and the ${\Delta}p$ vs ${\Delta}p{\cdot}t$ in the Poiseuille equation was plotted. Shear rate against shearing stress, on the other hand, was plotted for the sample fluids and it was found that all the sample fluids have shown a Newtonian behavior. It was interesting to note that the shear viscosity increased as the concentration of sugar solutions increased, and as the concentration of sucrose became higher than that of maltose, although the total concentration of sugar solution is constant.

  • Penicillium sp.-L4, a causative fungus of rot in citrus fruits, was isolated and its mode of hydrolytic enzyme production was investigated. Carboxymethylcellulase (CMCase), polygalacturonase(PGase), extra- & intra-cellular $\beta$-glucosidase and cellobiase were produced drastically by addition of substrates in minimal media. Production of the hydrolytic enzymes were induced efficiently by cellobiose and cellooligosaccharides which were the products of cellulose hydrolysis, but repressed by addition of mono-saccharide such as glucose, raffinose, galacturonic acid. The relative activity of p-nitrophenyl-$\beta$-D-glucopyranoside(PNPG) hydrolysis was higher than that of cellobiose hydrolysis in extracellular enzymes, and reverse is true in intracellular enzymes. Intact enzyme production of P. sp.-L4 on lemon peel lesion was sequential. $\beta$-Glucosidase and CMCase were produced first and followed by PGase. The enzyme productivities and pH in lesions were coincident with optimal pH of each enzyme activities.

  • Fro the purpose of producing glouse from cellobiose or oligo saccharide and obtaining genetic information of beta-1,4-glucosidase gene, alpha beta-1,4-glucosidase gene of Pseudomonas sp. LBC505, potent cellulase complex and xylanase producing strain, was cloned in Esherichia coli and Bacillus subtilis into pUC19 and pBD64, respectively. Recombinant plasmid pGL1 contained 1.2kb EcoRI fragment was isolated from transformants forming blue color around colony on LB agar plate containing 20 ng/ml of 5-bromo-4-chloro-3-indolyl-beta-D-glucopyranoside(X-glu) and ampicillin.

  • Recently, sea algae cultivation as carbon sink and carbon dioxide fixation have been considered. Also, various researches on bioenergy derived from sea algae and the utilization of fibers, saccharide, and lipid of sea algae have been performing. Till now, algae fibers has been used for manufacturing of paper and reinforcing of polymer composites and the extracts of sea algae are used for cosmetics, pharmaceutical materials and food such as agar. Especially, algae fiber has so similar properties to cellulose in terms of crystallinity and functional groups that it can be utilized as reinforcements of biocomposites. Biocomposites as alternatives of glass fiber reinforced polymer composites are environmentally friendly polymer composites reinforced with natural fibers and are actively applying to the automobiles and construction industries. In this paper, characteristics of algae fiber and biocomposites reinforced with algae fiber as environmentally friendly energy materials have been introduced.

  • The objective of this study was to invesitigate the sensory and quality characteristics of Chicksulgi containing different ratios of ingredients such as arrowroot starch (10%, 20%, and 30%), sugar, honey, oligo-saccharide and water, by sensory evaluation and mechanical examination. The results of sensory evaluation showed that Chicksulgi containing 10% arrowroot starch had higher scores in overall acceptability, color and flavor preference. In the textural analysis of Chicksulgi, the springiness, cohesiveness, adhesiveness were the highest in the samples with 10% arrowroot starch, and the chewiness, gumminess, and hardness were the highest in the 30%-added samples. The hunter's color L value of Chicksulgi was decreased by the increase of arrowroot starch. The more arrowroot starch was added, the redness of Chicksulgi were increased, and yellowness of Chicksulgi were decreased. The moisture content of Chicksulgi was higher in the samples with 10% arrowroot starch than those with 30%.

  • The effects of inflammation on the changes in some blood metabolites and ovarian response to super-ovulation treatment in sheep were investigated. Inflammation was induced artificially with LPS(Lipopoly-saccharide E. coli; 0.2mg/kg). Changes in the levels of NEFA, GOT, ${\gamma}$-GTP and body temperature after LPS injection were observed. Superovulation was carried out about 2 months after LPS injection. The body temperature, the levels of NEFA and GOT increased, and the level of total cholesterol decreased transiently. However, the level of ${\gamma}$-GTP did not change significantly. Superovulation results were not different between LPS injection and control groups. It was indicated that superovulation results did not decrease in the sheep which showed regular estrous cycle even after the induction of inflammation.

Determination of Mono- and Oligosaccharides Derivatized with p-Aminobenzoic Ethyl Ester by Reverse Phase HPLC

  • Mono- and oligosaccharides are derivatized with p-aminobenzoic ethyl ester (ABEE), strongly absorbs UV light at 254 nm, in the presence of sodium cyanoborohydride. C18-bonded silica column is used for the separation of sugar-ABEE derivatives in an isocratic mode. RP-HPLC conditions are optimized by using ternary mixture as mobile phase and $45^{\circ}C$ as a column temperature. Sugar-ABEE derivatives are separated well within a short run time (ca. 25 min) by reverse-phase partition chromatographic mode. The ($1{\rightarrow}6$) linkage type of dihexose-ABEE derivatives has shorter retention time than ($1{\rightarrow}4$)-linkage type. After acid hydrolysis of glycoproteins with 2M trifluoroacetic acid, monosaccharide composition and contents are determined. This procedure is very useful for the simultaneous analysis of neutral and amino sugars in a single chromatographic step using RP-HPLC without reacetylation of deacetylated amino sugars, which are produced by acid hydrolysis.
