Evaluation of Anti-Colitic Effect of Chung-Jang-Hwan (C-mix) in Mice

  • The inhibitory effect of Chung-Jang-Hwan (C-mix) consisted of Geranium nepalense subsp. thunbergii, Saururus chinensis, and Rubus coreanus were investigated in dextran sulfate sodium (DSS)-induced colitic mice by microarray analysis. Treatment with Cmix improved colitic symptoms, including colon shortening and myeloperoxidase activity. Treatment with DSS alone upregulated the expression levels of inflammation-related genes, including IL-$1\beta$, IL-6, CCL2, CCL4, CCL5, CCL7, CCL8, CCL24, CXCL1, CXCL2, CXCL5, CXCL9 and CXCL10, and other colitis-related genes such as COX-2, PAP, MMP family, S100a8, S100a9 and DEFA1 in mice. However, treatment with C-mix inhibited the expression levels of inflammation-associated genes induced by DSS. The increased expression levels of COX-2 and IL-$1\beta$, representative inflammatory genes, were confirmed by a quantitative realtime polymerase chain reaction analysis. These results indicate that C-mix may ameliorate colitis by the inhibitory regulation of inflammation-associated genes.

In Vitro Anti-bacterial and Anti-inflammatory Effects of Six Types of Herb Aqueous Extracts (일부 살충해독유(殺蟲解毒類) 한약의 Staphylococcus aureus에 대한 시험관 내 항균 및 항염 효과)

  • Objectives: The object of this study was to observe the in vitro anti-bacterial and anti-inflammatory effects of six single aqueous herbal extracts-Quisqualis Fructus (QuF), Meliae Cortex (MeC), Arecae Semen (ArS), Crassirhizomae Rhizoma (CrR), Ulmi Pasta Semen(UlS), Torreyae Semen(ToS)- against Staphylococcus aureus (S. aureus) and Lipopolysaccharide(LPS)-activated Raw 264.7 cells. Methods: Anti-bacterial activities against S. aureus of aqueous extracts of QuF, MeC, ArS, CrR, UlS and ToS were detected using standard agar microdilution methods. In addition, the effects on the cell viability, prostaglandin $E_2$ ($PGE_2$), nitric oxide (NO), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin (IL)-$1{\beta}$ and IL-6 productions of LPS activated Raw 264.7 cells were detected. The anti-bacterial and anti-inflammatory effects were respectively compared with lincomycin and piroxicam. Results: Minimal Inhibition Concentration (MIC) of aqueous extracts of QuF, MeC, ArS, CrR, UlS and ToS against S. aureus was respectively detected $5.625{\pm}4.075$ (3.125~12.500), $0.332{\pm}0.273$ (0.098~0.782), $1.094{\pm}0.428$ (0.782~1.563), $2.969{\pm}2.096$ (0.782~6.250), $9.375{\pm}4.419$ (3.125~12.500)>25 mg/ml. MIC of lincomycin was detected as $0.469{\pm}0.297$ (0.195~0.782) ${\mu}g/ml$ at same conditions. In addition, $ED_{50}$ against LPS-induced cell viabilities and cytokine releases of QuF, MeC, ArS, CrR, UlS and ToS was as follows - Cell viability: 66.370, 2.908, 1.747, 259.553, 18.150 and 34.160 mg/ml; NO production: 389.486, 0.294, 0.138, 523.060, 45.363 and 49.327 mg/ml; $PGE_2$ production: 114.271, 0.223, 0.046, 243.078, 8.829 and 28.947 mg/ml; TNF-${\alpha}$ production: 406.288, 0.343, 0.123, 9404.227, 125.406 and 140.775 mg/ml; IL-$1{\beta}$ production: 117.178, 0.135, 0.019, 237.451, 7.923 and 19.418 mg/ml; IL-6 production: 31.261, 0.105, 0.055, 128.434, 2.290 and 3.745 mg/ml. ED50 of piroxicam against LPS-induced cell viabilities, NO, $PGE_2$, TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 were detected as 35.179, 6.552, 1.162, 7.273, 7.101 and $5.044{\mu}g/ml$, respectively at same conditions. Conclusions: All six single aqueous herbal extracts showed anti-bacterial effects against S. aureus, in the order of MeC, ArS, CrR, QuF and UlS aqueous extracts except for ToS; they did not showed any anti-bacterial effects (MIC>25 mg/ml). They also showed anti-inflammatory effects against LPS-activated Raw 264.7 cells in the order of ArS, MeC, UlS, ToS, QuF and CrR aqueous extracts. It means that the ArS and MeC will be showed favorable potent anti-bacterial and related anti-inflammatory effects.

The Effects of Dichroa febrifuga(DIF) Extract on the Alzheimer's Disease Model (상산(常山)이 Alzheimer's Disease 병태(病態) 모델에 미치는 영향(影響))

  • This experiment was designed to investigate the effect of Dichroa febrifuga(DIF) on the Alzheimer’s disease. The effects of DIF extract on $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ mRNA of THP-1 cell treated by $A{\beta}$ plus LPS and amyloid precursor proteins(APP), acetylcholinesterase(AChE), glial fibrillary acidic protein(GFAP) mRNA of PC-12 cell treated by $A{\beta}$ plus $rIL-1{\beta}$ and AChE activity of PC-12 cell lysate treated by $A{\beta}$ plus $rIL-1{\beta}$ and behavior of memory deficit mice induced by scopolamine and mice glucose, uric acid, AChE activity of memory deficit rats induced by scopolamine were investigated, respectively. The results were summarized as follows ; 1. DIF extract suppressed APP, AChE, GFAP mRNA in PC-12 cell treated by $A{\beta}$. 2. DIF extract suppressed $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ mRNA in THP-1 cell treated by LPS. 3. DIF extract suppressed AChE activity in cell lysate of PC-12 cell treated by $A{\beta}$. 4. DIF extract increased glucose, decreased uric acid and AChE significantly in the serum of the memory deficit rats induced by scopolamine. 5. DIF extract group showed significantly inhibitory effect on the memory deficit of mice induced by scopolamine in the experiment of Morris water maze. According to the above results, it is suggested that DIF extract might be usefully applied for prevention and treatment of Alzheimer’s disease and memory deficit.

Il a $consacr{\acute{e}}$, dans ses $carri{\grave{e}}res$ de la mise en $sc{\grave{e}}ne$, toute son ${\acute{e}}nergie$ au $th{\acute{e}}{\hat{a}}tre$ sans jamais $fl{\acute{e}}chir$ contre le pouvoir dictatorial. Un critique a dit que son $th{\acute{e}}{\hat{a}}tre$ est semblable au $th{\acute{e}}{\hat{a}}tre$ de la $cruaut{\acute{e}}$. Pourtant, dans la mesure $o{\grave{u}}$ il respecte le texte dramatique, et que son $th{\acute{e}}{\hat{a}}tre$ a des tendances de la politique et qu'il oriente l'engagement du spectateur, il est certain que son $th{\acute{e}}{\hat{a}}tre$ est loin du $th{\acute{e}}{\hat{a}}tre$ de la $cruaut{\acute{e}}$. Ses $proc{\acute{e}}d{\acute{e}}s$ de la mise en $sc{\grave{e}}ne$ sont $tr{\grave{e}}s$ divers et il n'y a pas de $coh{\acute{e}}rences$ au niveau des styles dramatiques. $Malgr{\acute{e}}$ cela, nous pouvons trouver son $originalit{\acute{e}}$ de la mise en $sc{\grave{e}}ne$ dans la rencontre avec Lee hyun-wha. Le metteur en $sc{\grave{e}}ne$ ${\acute{e}}tablit$ son $esth{\acute{e}}tique$ $th{\acute{e}}{\hat{a}}trale$ en montant sur $sc{\grave{e}}ne$ de cinq $pi{\grave{e}}ces$ de Lee hyun-wha. Alors, en effet qu'est-ce que $l^{\prime}esth{\acute{e}}tique$ de la mise en $sc{\grave{e}}ne$ de Chae yoon-il produite par les ${\oe}uvres$ $th{\acute{e}}{\hat{a}}trales$ de Lee hyun-wha? Dans les $pi{\grave{e}}ces$ de Lee hyun-wha, il y a beaucoup de codes symboliques de la violence sexuelle comme le sadisme et le masochisme. L'implusion sexuelle $cach{\acute{e}}e$ dans le subconscient des personnages s'entrecroise avec le $m{\acute{e}}canisme$ du pouvoir politique. Alors, les abus sexuels et les mauvais traitements sur le corps deviennent une sorte de torture. L'expression cruelle des langages $sc{\acute{e}}niques$ n'est autre que la $r{\acute{e}}volte$ contre la $r{\acute{e}}alit{\acute{e}}$ actuelle de la politique. Son $th{\acute{e}}{\hat{a}}tre$ de torture est donc une sorte de $m{\acute{e}}canisme$ de la catharsis pour dissiper le cauchemar et l'inconscient noir. La fantasie sexuelle construite par le monde inconscient de l'auteur dramatique $s^{\prime}{\acute{e}}panouit$ dans le $th{\acute{e}}{\hat{a}}tre$ de Chae yoon-il qui $r{\hat{e}}ve$ toujours l'esprit de la $libert{\acute{e}}$. Une des convictions dramatiques du metteur en $sc{\grave{e}}ne$, c'est que le $th{\acute{e}}{\hat{a}}tre$ tout d'abord doit ${\hat{e}}tre$ choquant. Il dit : "l'art doit ${\hat{e}}tre$ ${\grave{a}}$ la fois ${\acute{e}}mouvant$ et bouleversant. Cependant, dans la mesure $o{\grave{u}}$ le film ou la $t{\acute{e}}l{\acute{e}}vivion$ fait aujourd'hui une vive impression aux spectateurs, la part $donn{\acute{e}}e$ au $th{\acute{e}}{\hat{a}}tre$ est le choc." Pour donner le choc aux spectateurs, le style de la torture est $tr{\grave{e}}s$ utile. $L^{\prime}esth{\acute{e}}tique$ $th{\acute{e}}{\hat{a}}tral$ du metteur en $sc{\grave{e}}ne$ respecte essentiellement l'intention de l'auteur. $N{\acute{e}}anmois$, le respect sur l'original ne se $d{\acute{e}}range$ pas $l^{\prime}interpr{\acute{e}}tation$ de son propre $id{\acute{e}}e$. Dans l'insistance de la torture, l'analyse rituelle sous la base du choc, il projecte le symbole $arch{\acute{e}}typal$ sur $sc{\grave{e}}ne$. Bien $s{\hat{u}}r$, il renforce le $caract{\grave{e}}re$ de la politique dans le $th{\acute{e}}{\hat{a}}tre$ de torture pour inviter des spectateurs de l'engagement. D'ailleurs, il y a l'aspect d'absurde dans le $th{\acute{e}}{\hat{a}}tre$ de torture qui insiste des violences ${\grave{a}}$ travers les langages $sc{\acute{e}}niques$ comme le $m{\acute{e}}tal$ froid, parce que le metteur en $sc{\grave{e}}ne$ $consid{\grave{e}}re$ que $l^{\prime}{\hat{e}}tre$ humain vit dans l'absurde.

Separation of 20(R&S) Prosapogenin Isomers of Ginsenoside-$Rg_2$ and -$Rg_3$ from Ginseng Saponins by Reversed-Phase High Performance Liquid Chromatography (인삼 사포닌에서 Ginsenoside-$Rg_2$와 -$Rg_3$의 이성질체인 20(R&S) Prosapogenin들의 역상 고속 액체 크로마토그래피에 의한 분리)

  • Using a reversed-phase high performance liquid chromatography, the separation of 20(S)-, 20(R)-prosapogenin stereo-isomers of ginsenoside-$Rg_2$ and of ginsenoside-$Rg_3$ in ginseng saponins has been carried out with binary solvent system. The optimum conditions for the isomer separation are as following: Nova-$Pak^{(R)}C_{18}$ (Waters, $3.9{\times}150mm$) column, $CH_3CN/CH_3CN$ (100:8, v/v) binary solvent system and the flow rate was 1.7 mL/min. The stereoisomers were separated with change of the mixture ratio of the solvent system, the solvent elution by gradient program, and then detected at 203 nm of UV detector. The simultaneous separation of mixture that were the $Rg_2$, $Rg_3$ isomers was easily performed in nonpolar solvent for $Rg_2$, polar solvent for $Rg_3$ at the same optimum conditions.

  • Dose response for the nuscheduled DNA synthesis induced by various concentration of MMS was dose dependent and directly proportional to dose increased. Time dependence of unscheduled DNA synthesis was continued up to 4 hours, with the peak appearing 2$\\sim$3 hours after treatment with MMS and $^3 H$-thymidine labeling. Single treatment with BUdR or IUdR does not induce unscheduled DNA synthesis. BUdR and IUdR greatly enhanced MMS-induced unscheduled DNA synthesis, but the dose responses were different from that of single treatment with MMS. IUdR was found to be more effective sensitizer on MMS-induced unscheduled DNA synthesis.

  • Reed-solomon(R-S) code is very effective to coerrect both random and burst errors over a noise communication channel. However, the required hardware is very complex if the B/M algorithm was employed. Moreover, when the error correction system consists of two R-S decoder and de-interleave, the I/O data bns lines becomes 9bits because of an erasure flag bit. Thus, it increases the complexity of hardware. This paper describes the R-S decoder which consisits of a error correction section that uses a direct decoding algorithm and erasure generation section and a erasure generation section which does not use the erasure flag bit. It can be shown that the proposed R-S dicoder is very effective in reducing the size of required hardware for error correction.

Antioxidant and Immunomodulatory Effects of Ulva pertusa kjellman on Broiler Chickens (육계에서 구멍갈파래의 항산화 및 면역조절 효과)

  • This study was conducted to test the efficacy of Ulva pertusa kjellman as immunomodulators under lipopolysaccharide (LPS) challenge in broilers by investigating their effects on serum superoxide dismutase (SOD) like ability, immunoglobulin concentration, and splenic cytokine mRNA expression. A total of ninety six1-d-old male broiler chicks (Ross 308) were divided into 4 treatment groups with 8 replicates of 3 birds in each group. NC (negative control, no immune substances), PC (positive control, ${\beta}$-glucan 25 ppm), Ulva P (Ulva pertusa kjellman Powder 3%), and Ulva E (Extract form Ulva pertusa kjellman 0.3%) were added in feed with respective substance. Heparinized venous blood and spleens were collected from five birds per dietary treatment at 5 wk of age. The SOD like activities in Ulva P and Ulva E were significantly increased in comparison with other groups (P<0.05). The immunoglobulin M content was lower in the Ulva E than other groups (P<0.05). Expression patterns of splenic cytokine mRNA in the IL-$1{\beeta}$, IL-2 and IL-6 were similar. Expression rate of IL-$1{\beta}$, IL-2 and IL-6 in Ulva pertusa kjellman (Ulva P, Ulva E) were decreased (P<0.05) in comparison to other groups. Expression rate of IL-$1{\beta}$, IL-2 and IL-6 were significantly lower in groups treated with Ulva E than Ulva P. In conclusion, dietary supplementation of Ulva pertusa kjellman improved SOD like activity and affect immune system by inhibiting inflammatory response in broiler chicks. In addition, it is more effective to use Ulva pertusa kjellman extract than powder for immunomodulatory function. These results suggest the possibility that Ulva pertusa kjellman could be used as the immunomodulator for inflammatory response in broiler chicks.

Immunological Features of Macrophages Induced by Various Morphological Structures of Candida albicans

  • Candida albicans is a dimorphic fungus that commensally colonizes human mucosal surfaces. The aim of this study was to assess the role of different C. albicans morphologies in inducing pattern recognition receptors (PRRs) and cytokines in macrophages. Macrophages may respond to pathogen-associated molecular patterns via TLR2 and TLR4 by expressing cytokines. The hyphal transition of C. albicans was induced by 20% serum (S), RPMI-1640 (R), or $39^{\circ}C$ culture (H). Macrophages were then challenged with either yeast (Y) or different hyphae cultures of C. albicans, followed by RT-PCR and FACS analysis of PRRs expression. In addition, macrophages were stimulated with either yeast or different hyphae cultures of C. albicans used by RT-PCR and Bio-Plex analysis of cytokines production. Macrophages expressed high levels of TLR4 and dectin-1 after stimulation with Y cells. In contrast, stimulation with H or R cells strongly increased the expression of TLR2 and dectin-2. Stimulation with Y cells significantly enhanced the expression of IL-$1{\beta}$ and weakly increased the expression of IL-6 and IL-12. Stimulation with hyphal cells (S, R, and H) strongly increased IL-10 expression, but weakly reduced IL-$1{\beta}$ expression. The phagocytosis activity and NO production of macrophages were decreased upon treatment with hyphal cells compared with yeast, and depended on the length of hyphae. In summary, the yeast and hyphae forms of C. albicans resulted in an induction of different PRRs, with accompanying differences in immune cell cytokine profiles.
