Changes of hematological parameters in olive flounder Paralichthys olivaceus exposed to pentachlorophenol (Pentachlorophenol 노출에 따른 넙치, Paralichthys olivaceus의 혈액성분의 변화)

  • Experiments were carried out to determine the hematological changes in olive flounder, Paralichthys olivaceus, after sub-chronic dietary pentachlorophenol (PCP) concentrations (0, 0.1, 0.5 and 1.0 mg/kg) exposure 6 weeks. The RBC count, Ht and Hb were significantly decreased in PCP 0.5 mg/kg exposed flounder compared to control group. The flounder exposed to PCP concentration 0.5 mg/kg started to increase significantly in serum magnesium concentrations after 4 weeks. The low concentration of serum calcium concentration was observed at 6 weeks of the experiment in PCP concentration 0.5 mg/kg. Although serum total protein concentrations was significantly reduced compared to the control group at PCP concentration 0.5 mg/kg, serum glucose concentration was found to be significantly increased over the control group. A significant increment of GOT, GPT and LDH activities in the flounder serum was noticed after 4 weeks at PCP concentrations over 0.5 mg/kg without typical changes of Al-P. These results indicate that flounder can be affected by PCP in terms of inorganic matter, organic matter and enzyme activity in serum when they were exposed to the PCP concentrations 0.5 mg/kg or higher.

Antioxidant and Neuronal Cell Protective Effect of Purple Sweet Potato Extract (자색고구마 추출물의 항산화 효과 및 신경세포 보호효과)

  • The antioxidant and neuronal cell protective effects of water extract from purple sweet potato were investigated. The total phenolics and monomeric anthocyanin contents of purple sweet potato extract were 44.25 mg/g and 2,394 mg/L, respectively. The antioxidant activities of purple sweet potato extract were evaluated using various antioxidant tests, including 1,1-diphenyl- 2-picrylhydrazyl (DPPH), 2,2'-azino- bis-(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging activities, ferric reducing/antioxidant power (FRAP) and reducing power. In these assays, the extract of purple sweet potato presented significant radical scavenging activities, FRAP, and reducing power in a dose-dependent manner. MTT {3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyl- tetrazoliumbromide} reduction assay showed significantly increase in cell viability when PC12 cells were pretreated with purple sweet potato extract. Because oxidative stress is also known to increase neuronal cell membrane breakdown, we further investigated by lactate dehydrogenase (LDH) and neutral red uptake assay. Purple sweet potato extract inhibited oxidative stress-induced membrane damage in neuronal cells. Therefore, these data results demonstrated that water extract of purple sweet potato have antioxidant activity and neuronal cell protective effect thus it has great potential as a natural source for human health.

Ginsenoside F1 attenuates pirarubicin-induced cardiotoxicity by modulating Nrf2 and AKT/Bcl-2 signaling pathways

  • Background: Pirarubicin (THP) is an anthracycline antibiotic used to treat various malignancies in humans. The clinical usefulness of THP is unfortunately limited by its dose-related cardiotoxicity. Ginsenoside F1 (GF1) is a metabolite formed when the ginsenosides Re and Rg1 are hydrolyzed. However, the protective effects and underlying mechanisms of GF1 on THP-induced cardiotoxicity remain unclear. Methods: We investigated the anti-apoptotic and anti-oxidative stress effects of GF1 on an in vitro model, using H9c2 cells stimulated by THP, plus trigonelline or AKT inhibitor imidazoquinoxaline (IMQ), as well as an in vivo model using THP-induced cardiotoxicity in rats. Using an enzyme-linked immunosorbent test, the levels of malondialdehyde (MDA), brain natriuretic peptide (BNP), creatine kinase (CK-MB), cardiac troponin (c-TnT), lactate dehydrogenase (LDH), superoxide dismutase (SOD) and glutathione (GSH) were determined. Nuclear factor (erythroid-derived2)-like 2 (Nrf2) and the expression of Nrf2 target genes, including heme oxygenase-1 (HO-1), glutathione-S-transferase (Gst), glutamate-cysteine ligase modifier subunit (GCLM), and expression levels of AKT/Bcl-2 signaling pathway proteins were detected using Western blot analysis. Results: THP-induced myocardial histopathological damage, electrocardiogram (ECG) abnormalities, and cardiac dysfunction were reduced in vivo by GF1. GF1 also decreased MDA, BNP, CK-MB, c-TnT, and LDH levels in the serum, while raising SOD and GSH levels. GF1 boosted Nrf2 nuclear translocation and Nrf2 target gene expression, including HO-1, Gst, and GCLM. Furthermore, GF1 regulated apoptosis by activating AKT/Bcl-2 signaling pathways. Employing Nrf2 inhibitor trigonelline and AKT inhibitor IMQ revealed that GF1 lacked antioxidant and anti-apoptotic effects. Conclusion: In conclusion, GF1 was found to alleviate THP-induced cardiotoxicity via modulating Nrf2 and AKT/Bcl-2 signaling pathways, ultimately alleviating myocardial oxidative stress and apoptosis.

Induction of Glutathione S-transferase Activity by the Extracts of Glycyrrhiza uralensis Fischer (감초 추출물이 glutathione S-transferase의 유도 활성에 미치는 영향)

  • In the present study, we investigated the glutathione S-transferase (GST) induction of medicinal plants using a cultured PC12 cells. The methanol extracts of Dendrobium nobile Lindley, Schizonepeta tenuifolia Briquet, Glycyrrhiza uralensis Fischer, Paenoia lactiflora Pall were tested. As the result, exposure of PC12 cells to G. uralensis resulted in the significant induction of GST. On the continuous experiment, G. uralensis was extracted with methanol, ethanol, and acetone. Among these extracts, methanol extracts showed the highest GST induction. The methanol extracts were further fractionated with hexane, diethyl ether, ethyl acetate, and water layer according to the degree of polarity. The diethyl ether layer showed the highest exhibited GST induction on PC12 cells. Based on these results suggest that the extracts of G. uralensis can be applicable for the development of a new antioxidant agent.

Effects on Alzheimer's disease by Jimitang in CT105-overexpressed SK-N-SH cell lines (CT105로 유도된 신경모세포종 세포주에서 지미탕(指迷湯)의 항치매효과)

  • Alzheimer's disease(AD) is a geriatric dementia that is widespread in old age. In the near future AD may be the biggest problem in public health service. Although a variety of oriental therapies in the study of Jimitang have been traditionally utilized for the treatment of AD, their pharmacological effects and active mechanisms have not been fully elucidated. This study in an investigation of effects of Jimitang on apoptotic cell death induced by CT105 overexpression in SK-N-SH neuroblastoma cell lines. DNA fragmentation, neurite outgrowth assay and LDH activity assay were examined. The regeneratory and inhibitory effects on Alzheimer's disease in pCT105-induced neuroblastoma cell lines by Jimitang water extract were examined. Findings from these experiments have shown that Jimitang inhibits the synthesis or activities of CT105, which has neurotoxicities and apoptotic activities in cell lines. In addition, pretreatment of $Jimitang(>50\;{\mu}g/mL\;for\;12\;hours)$ partially prevented CT(105)-induced cytotoxicity in SK-N-SH cell lines, and were inhibited by pretreatment. $Jimitang(>50\;{\mu}g/mL\;for\;12\;hours)$ repaired CT(105)-induced neurite outgrowth when SK-N-SH cell lines were transfected with CT(105). Results of this study show that. in the Jimitang group, the apoptosis in the nervous system in inhibited, the repair against the degerneration of neuroblastoma cells by CT105 expression is promoted. In addition, Jimitang was found to inhibit DNA fragmentation induced by CT105 overexpression, and promote neurite outgrowth. These findings suggest that Jimitang is beneficial for the treatment of AD.

Effects of Callophyllis japonica Extracts on the Glucide and Lipid Metabolism in Hypercholesterolemic Rats (고콜레스테롤혈증 흰쥐의 당질 및 지질 대사에 볏 붉은잎(Callophyllis japonica)추출액이 미치는 영향)

  • This study was designed to observe the effects of the feeding Callophyllis japonica extracts on the improvement of the blood glucose, lipid in the serum of dietary hypercholesterolemic rats(S.D. strain, ?) fed the experimental diets for 4 weeks. Concentrations of total cholesterol and LDL-cholesterol in serum were significantly lower in the Callophylis japonica extracts group than in the choloesterol supplemented diet group. Concentrations of HDL-cholesterol in serum were higher in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. Concentrations of free cholesterol, triglyceride, phospholipid and blood glucose in serum were significantly lower in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. The activities of aspatate aminotransferae(AST), alanine aminotransferase(ALT), lactate dehydrogenase(LDH) and alkaline phosphatase(ALP) in serum were lower in the CAllophyllis japonica extracts supplemented group than in the cholesterol diet group. From the above research, the physiological activity substances in Callophylis japonica extracts were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hypercholesterolemic rats.

Domestic review of Korean medical treatment on Carbon Tetrachloride-induced Hepatotoxicity - Focusing on Treatment of Korean Herbal Medicine (본초학적 접근을 통해 본 국내에서 연구된 한약재의 사염화탄소로 유발된 간손상에 대한 간보호 효과)

  • The purpose of this study is to inspect trends of the paper of Korean medical treatment on Carbon Tetrachloride-induced Hepatotoxicity and try to establish the future direction for development of Korean herbal medicine. We reviewed 79 papers which had been published from 1981 to 2010 in journals published in Korea. According to these studies, Carbon Tetrachloride-induced hepato-celluar degeneration and necrosis induced to increase in serum aspartate amintransferase (AST), alanine aminotransferase (ALT), Alkaline phosphatase (ALP), ${\gamma}$-Glutamyl transferase (${\gamma}$-GTP), lactate dehydrogenase (LDH), bilirubin, blood urea nitrogen (BUN) and albumin levels. In biochemical analyses, antioxidant enzymes such as superoxide dismutase (SOD), Glutathione S-transferase (GST) and catalase in hepatic tissue were remarkably incresed by Carbon Tetrachloride treatment. We found that some of the herbal extracts have a protective effect against Carbon Tetrachloride-induced Hepatotoxicity. More studies of oriental medicinal herbs are required for developing a treatment of hepatotoxicity.

Protective Effect of Pterocarpus santalinus on Galactosamine Induced Liver Damage

  • The present study was carried out to investigate the hepatoprotective effect of the extract of Pterocarpus santalinus Linn on acute hepatotoxicity induced in Wistar albino rats by a single dose of Galactosamine (400 mg/kg). Suspensions of methanolic extract of heartwood of P. santalinus (200 and 400 mg/kg) in 0.3% Carboxy Methyl Cellulose (CMC) were administered p.o. to experimental animals and hepatoprotective activity was monitored by estimating aspartate amino transferase (ASAT, GOT), alanine amino transferase (ALAT, GPT), alkaline phosphatase (ALP), total bilirubin (TB), lactate dehydrogenase (LDH), total cholesterol (TC), triglycerides (TGL), albumin, total protein (TP) levels. The methanolic extract significantly reduced the elevation of serum transaminases and alterations of biochemical parameters induced by hepatotoxin, and alleviated the degree of liver damage. The results were supported by histopathological studies of liver samples showing regeneration of hepatocytes in treated animals. Silymarin (25 mg/kg), a known hepatoprotective drug was used for comparison. Based on the results obtained, it can be concluded that P. santalinus exerts hepatoprotective activity and may serve as a useful adjuvant in several clinical conditions associated with liver damage.

The Clinical Study on Abnormal Liver Function Patients Caused by Obesity (비만증(肥滿症)과 간기능(肝機能) 이상(異常)을 동반(同伴)한 환자(患者) 11례(例)에 대한 임상적(臨床的) 고찰(考察))

  • Objectives: Obesity is regarded as the aggregation of needless risk factors, for instance, cardiovascular disease, joint disease, induce cancer. We studied on interrelation between abnormal liver function and obesity. Methods: We analyzed liver function, T.Cholesterol, Triglyceride before and after lose weight treatment. The collateral condition is over 6 weeks period on obesity treatment, no liver injury and no complicated another disease on personal past history and found out abnormal impression on biochemical liver function blood test. Results: The improvement rate of LFT, compare with before treatment is 10.6% in T.Bilirubin, 11.1% in ALP, 21% in AST, 38% in ALT, 37.3% in r-GTP, 9.2% in LDH and decreased 2.7% in T.Protein, increased 2.3% in Albumin. Hyperlipidemia is 19.4% in T.Cholesterol, 42.5% in Triglyceride. Conclusions: LFT and Hyperlipidemia of abnormal liver function patients, caused by obesity, is improved to normal limit in proportion to reduce patient's weight.

Induction of Apoptosis by Methanolic Extract of Rubia Cordifolia Linn in HEp-2 Cell Line is Mediated by Reactive Oxygen Species

  • Rubia cordifolia Linn, which belongs to the Rubiaceae family, is a well-known herb used in Ayurvedic medicine. In the present study, we investigated the influence of a methanolic extract (RC) on the induction of apoptosis in HEp-2 (human laryngeal carcinoma) cell line, as evidenced by cytotoxicity, morphological changes and modification in the levels of pro-oxidants. Inhibition of cell proliferation and lactate dehydrogenase (LDH) release increased in a time and dose-dependent manner. Further, reduced glutathione (GSH), glutathione transferase (GST) and protein levels decreased and lipid peroxidation increased significantly on RC treatment in a dose dependent manner when compared to controls. Based on the results we determined the optimal dose as 30mg/ ml and the apoptotic effect of RC extract (30 mg/ml) on HEp-2 cells was confirmed by fluorescent microscopy and transmission electron microscopy (TEM) based on morphological and ultrastructural changes. RC extract suppressed the proliferation of HEp-2 oral cancer cells inducing apoptotic cell death in vitro. These results point to potential of RC extract as an agent for the treatment of laryngeal squamous cell carcinoma.
