어성초 메탄올 추출물로부터 항산화 효능을 가진 활성물질의 확인 (Identification of Antioxidant Compound Derived from Methanolic Extract of Houttuynia Cordata)

  • 본 연구는 어성초의 메탄올 추출물을 이용하여 항산화 효능이 가장 좋은 fraction을 찾고, 항산화 효능을 나타내는 성분을 분석하였다. 메탄올 추출에 의한 유기 용매 별 분획에서 항산화 효과가 가장 좋은 ethyl acetate 분획물을 칼럼 크로마토그래피하여 12가지의 fraction 중 가장 높은 항산화 효과를 보인 Fr. 10을 이용하여 DPPH 라디칼의 소거활성, 환원력, 지질과산화, 세포독성, DNA 산화 및 DCFH-DA를 이용한 세포내 과산화수소 제거효과를 조사하였다. DPPH radical scavenging activity, reducing power, TBARS, cell viability, DNA oxidation and DCF fluorescence 12가지의 fraction들 중 항산화 효능이 가장 좋은 Fr. 10의 DPPH radical 소거 활성 결과로 $64{\mu}g/ml$ 농도에서 양성대조군에 근접하는 60%의 억제능을 보였다. Reducing power 결과로 $32{\mu}g/ml$의 농도에서 140%로 양성대조군과 비슷한 결과 값을 보였다. TBARS 결과로 $2{\mu}g/ml$에서 양성대조군과 같은 값의 활성산소 억제능이 나타난다. 또한 cell viability 결과로 $32{\mu}g/ml$ 이상의 농도에서 세포 독성에 의해 생존율이 감소하였고, DCF fluorescence 결과로 농도의존적으로 $H_2O_2$에 의한 산화적 손상을 억제하는 것으로 나타났다. DNA oxidation 실험결과 $1{\mu}g/ml$ 이상의 농도에서 DNA의 손상을 감소시킴을 확인할 수 있었다. IR 및 LC-MS를 이용하여 Fr. 10의 유효성분을 조사한 결과 rutin (분자량, 610)으로 확인 되었다. 결론적으로 어성초의 메탄올 추출물로부터 분리한 Fr. 10은 농도 의존적으로 항산화 효능이 우수하게 나타났고, 어성초는 화장품 및 기능성 식품의 소재로 활용될 수 있음을 시사한다.

Structural Determination of Oxidation Products of Flavonoids in Alcoholic Aqueous Solution with Reactive Oxygen Species

  • Recently, much attention has been paid to the physiological functions of flavonoids associated with their antioxidant properties. However, there was a lack of information on the molecular mechanism at which flavonoids play the antioxidative role. We have already studied on the oxidation of quercetin with hydrogen peroxide and sodium hypochlorite in alcoholic aqueous solution and determined the oxidation products. Through the structural analysis of the oxidation products, it was clarified that the hydroxyl group at C-3 in the C ring plays the important role in the antioxidative action of quercetin. Successively, rutin and (+)-catechin were oxidized with sodium hypochlorite and their mono- and di-chlorinated derivatives were obtained. These facts indicate that these flavonoids can directly scavenge hypochlorous acid and the active site in this scavenging reaction is not the hydroxyl group at C-3.

항염작용을 갖는 신규 생약복합제 SK1306X의 분리 및 항염작용 (Preparation of Antiinflammatory Herbal Drug, SKI306X.)

  • Antiinflammatory activities of the solvent fractionates of several herbal medicines were investigated and SKI306X was prepared from the active principles of three herbal medicines, Prunella vulgaris, Trichosantlies kirilowii and Clematis mandshurica. SK1306X was shown to have strong inhibitory effects on acetic acid-induced pain, carrageenan-induced paw edema and adjuvant induced arthritis. LD50 of SKI306X was more than 5 g/kg in rat, so generally nontoxic. Chemical analysis revealed that oleanolic acid and rutin, which are known to have various antiinflammatory activities, were contained in it. These results suggest SK1306-X may become a useful drug for the treatment of inflammatory diseases such as rheumatoid arthritis.

Superoxide Quenching Activity of Phenolic Compounds from the Whole Plant of Galium verum var. asiaticum

  • During the process of screening for antioxidative effects of natural plants in Korea, by measuring the superoxide quenching activity, methanol extract of the whole plant, Galium verum var. asiaticum (Rubiaceae), was found to show potent antioxidant activity. Subsequent activity-guided fractionation of methanol extract of Galium verum var. asiaticum led to the isolation of five phenolic compounds. Using spectroscopic techniques, the chemical structures were elucidated as: caffeic acid (1), narcissin (2), rutin (3), luteolin-7-O-${\alpha}$-L-rhamnopyranosyl (1 ${\rightarrow}$ 2)-${\beta}$-D-glucopyranoside (4), and luteolin-7-O-${\beta}$-D-glucopyranoside (5). These compounds were isolated for the first time from this plant. Among them, compound 1 showed the most significant riboflavin-originated superoxide and xanthine-originated superoxide quenching activities. Compounds 3 and 4 exhibited mild superoxide quenching effects compared with vitamine C.

  • Using the isolated perfused rat heart this study investigated 1) the cardiac uptake of idarubicin (IDA), 2) the role of P-glycoprotein (P-gp) in the uptake process, 3) the formation of IDOL from IDA in the heart, and 4) the effect of P-gp inhibitors (verapamil, amiodarone, PSC 833), doxorubicin, hypothermia, xanthine derivatives (caffeine, theophylline) and metabolism inhibitors (rutin, phenobarbital) on the pharmacokinetics and pharmacodynamics of IDA using a mathematical modeling approach. A minimal model was constructed; the differential equations were numerically solved and fitted to the data using the ADAPT II-software package using maximum likelihood estimation assuming that the measurement error has a standard deviation which is a linear function of the measured quantity[1].   • The relationships of inhibitory activities of inflammation and wound healing of flavonoids were studied in vitro and in vivo. Generally flavonoids have not only significantly anti-inflammatory activity in carrageenin-induced paw edema and Freund's complete adjuvant-induced arthritis, but also inhibitory activity of wound healing. The more inhibitory activities of wound healing flavonoids have, the more they have the anti-inflammatory activities; apigenin > guercetin > flovone > rutin > hesperidin > naringin. Their inhibitory mechanism seems to be inhibition of the inflammatory cell infiltration and fibroblast proliferation, and so they decreased the granulomatous activity and tensile strength.

한국산 재배대황엽의 약효성분 -엽의 후라보노이드- (Pharmaco-Constituents of Korean Cultivated Rhubarb Leaves -The Flavonoids from Leaves-)

  • As the continued studies for Korean cultivated rhubarb, MeOH extract of the leaves was fractionated with ether, ethylacetate, and n-butanol. From the ethyl acetate fraction of MeOH extract, one flavone glycoside, apigenin-8-${\beta}$-D-glucopyranoside(vitexin, $C_{21}H_{20}O_{10}$) and from the n-BuOH fraction of MeOH extract, two flavonol glycosids, kaempferol-3-O-(2,6-di-O-rhamnopyranosyl)-${\beta}$-D-galactopyranoside$(C_{33}H_{40}O_{19})$and quercetin-3-O-rutinoside(rutin, $C_{27}H_{30}O_{16}$) were isolated and identified through the physico-chemical properties and spectroscopic evidences(UV, IR, NMR, Mass) respectively.

은행잎 플라보놀배당체에 대한 셀루라제류의 영향 (The Effect of Cellulases on Flavonolglycosides of Ginkgo Leaf)

  • The extractability and stability of ginkgoflavonolglycosides under presence of several cellulose preparations were investigated. The enzymes used were macerosin, cellulose C and cellulase NC. The content variation of the glycosides was measured with HPLC method, using caffeic acid as an internal standard. The methanol extract of ginkgo leaf, containing the total flavonolglycosides of 4.46%, was used for the content comparison. By extraction with the enzymes, each or mixed, the peak levels of all the glycosides began to decrease after 1 or 2 hours. After 24 hour extraction, most of the glycosides were degraded to minor components. The flavonolglycosides in ginkgo leaf were also hydrolysed simply by the water extraction. After 24 hour extraction with water at $40^{\circ}C$, the peak levels of major glycosides were distinctly decreased. Rutin was hydrolysed by enzyme treatment or by ginkgo leaf itself. As a result, it was concluded that the commercially available cellulases and the ginkgo leaf itself contain the activities of $\beta$-glycosidase and $\alpha$-rhamnosidase. Kaempferol-3-O-(6'"-O-p-coumaroylglucosyl)-rhamnoside and four other ginkgo flavonolglycosides were not hydrolysed under the same condition.tion.
