Effects of Types of Water Media and Concentration of Additives on Rooting and Survival of Ternstroemia japonica after Transplanting Cuttings (물배지의 종류 및 첨가물의 농도가 후피향나무 삽목묘의 발근 및 이식 후 활착에 미치는 영향)

  • Cuttings based on water media are relatively low-cost and easy to manage compared to those based on soil media. Nevertheless, species that root out in water media are very few. Water-medium rooted cuttings are not widely used. In water media, the survival rate of cuttings is high, but in many cases, they only form callus while not rooting. If cuttings with callus formed in a water medium were to root and survive after being transplanted in soil, water-medium cuttings could be used as an important means of propagating landscape tree species. In this study, cuttings of Ternstroemia japonica having various reactions to water media were soaked in different types of water media. Their callus formation and rooting conditions were analyzed. The water-medium cutting swere then transplanted in soil, and conditions for their survival were examined. The study concluded: 1. Cuttings that only formed callus in water media were able to root and survive after being transplanted. 2. Cuttings with satisfactory callus formation showed more satisfactory survival conditions than those with unsatisfactory callus formation. 3. Cuttings rooted in water media showed a high survival rate of 96.6% after being transplanted. 4. Water-medium cutting of Ternstroemia japonica proved to be more effective in Late July or August than in late June. 5. Cuttings pre-treated in 50ppm-IBA for 24 hours showed better survival conditions than those that were not pre-treated inIBA. 6. Cuttings pre-treated in 50ppm-IBA for 24 hours and soaked in water media with 0.02% atonic, 0.02% polyoxin, and 0.01% polyoxin showed a rooting and survival rate of 80 to 85%. Regardless their low rooting rate, water-medium cuttings can show a high survival rate when these factors are considered: proper cutting time, plant hormone treatment, and types of water media. Accordingly, water-medium cuttings will emerge as major means of propagating landscape tree species that require small volumes of multi-species.

  • Outdoor cuttings studies on two coniferous species, pitch pine and needle fir, have been made at Suwon, Korea extending from March to early May, 1956. The purpose of these studies was to find the effects of ages of parent trees, topophysis, dilute hormone solution treatments and the time at which the cuttings were collected on the rooting response of the cuttings. On September 2, rooting results were examined. Pitch Pine (Pinus rigida) The total of 1335 cuttings were collected from 3- and 10-year old trees. The cuttings treated were planted on March 13 and April 20. 1. In the cuttings collected from 3-year-old seedlings, the best result (34 percent rooted) was obtained by treating ends of cuttings with a mixture of 50 ppm IBA and 50 ppm NAA. 2. No rooting was observed in lammas shoots. 3. The cuttings from 3-year-old seedlings gave better rooting than those from 10-year-old trees. 4. The root origins were developed from the lower part of the cutting and the cut surface. Needle fir (Abies holophylla) One hundred twenty-five cuttings were collected from a 30-year-old tree. The cuttings treated with any one of four mixed hormone solutions were planted on March 23. 1. Fifteen cuttings out of 75 cuttings from the lower half of the crown were rooted. The cuttings obtained from the upper half of the crown did not root at all. 2. The cuttings treated by the hormone solution made up of 20 ppm IAA, 20 ppm IBA, 20 ppm NAA, 20 ppm thiamine and 5 percent sucrose gave the best rooting (22 percent). 3. All roots originated from callus tissues and irregularly arranged parenchymatous tissues near the cut base.

Growth and Rooting Rate of 'Maehyang' Strawberry as Affected by Irrigation Method on Cutting Propagation in Summer Season (하절기 '매향' 딸기의 삽목 번식 시 관수방법에 따른 생육 및 발근율)

  • This study was conducted to investigate the optimum irrigation method for propagation of cutting strawberry ($Fragaria{\times}ananassa$ Duch. cv. Maehyang) plantlet in summer season. The cutting strawberry plantlets were planted in 24-cell tray ($60{\times}34{\times}10cm$) filled with commercial mixed medium (Tosilee) and placed in semi closed-type small plastic tunnel. Each semi closed-type small plastic tunnel was treated as follows; control (non-treatment), over head irrigation (twice a day), capillary mats irrigation (twice a day), or fog irrigation (30 minutes turn on and 10 minutes off from 8:00 to 18:00). The strawberry plantlets were rooted during 8 days in the semi closed-type small plastic tunnel, and then plastic film was removed. Growth parameters, such as plant height, root length, number of primary roots, petiole length, leaf length, leaf width, crown diameter, SPAD, leaf area, fresh and dry weights of the shoot and root, were measured at 61 days after cutting. Relative humidity in tunnel was high in the order by fog irrigation, over head irrigation, capillary mats, and the control as 72.5, 56.3, 45.8, and 29% on average, respectively. However, the air temperature was similar in all treatments. On the 4 and 8 days, the rooting rate of strawberry plantlet was significantly higher in the over head irrigation and fog irrigation treatments. Plant height, petiole length, crown diameter, and leaf area were highest in the over head irrigation and fog irrigation treatments. In addition, fresh and dry weights of shoot were greater in over head irrigation and fog irrigation treatments than the others. Dry weight of root was differed significantly heaviest in the fog irrigation treatment. However, root length, no. of primary roots, SPAD value, and fresh weight of root were not significantly different in all treatments. These results indicated that growth and rooting for propagation of cutting strawberry plantlet 'Maehyang' were best achievement in the over head irrigation and fog irrigation treatments.

Effects of Foliar Application of Chitosan and Wood Extraction on Rooting and Tuber Formation of Plug Seedlings in Potatoes (Chitosan과 목초액 엽면살포에 의한 감자 플러그 삽목표의 발근 및 괴경형성효율)

  • To do mass multiplication of plug seedlings in potatoes, apical stem cuttings originated from virus-free microtubers were cut to one-two internodes and transplanted into the plug-tray. After 10days, we applied Chitosan and Wood Extraction on rooting and tuber formation of plug seedlings. To improve field adaptability of plug seedlings, rooted cuttings with a height of 20cm after 20days of cutting were transplanted ito the fields, We applied 500~2000ppm Chitosan on growth characteristics and tuber formation of that. The above and underground growths, such as plant height and number of leaves were significantly more vigorous after treatment with 500~1,000ppm Chitosan and 2,000ppm Wood Extraction, the spray treatment was carried out five times at intervals of four days after ten days of transplanting. T-N, K, P, Mg and Na, were higher as the concentrations of chitosan and Wood Extraction were higher. The growth and tuber yield in plug seedlings planting plot and seed potatoes planting plot were effectively highter as foliar application of Chitosan(500~2,000ppm) was done after planting the plot. T-N content in leaves and tuber was higher as the concentration of Chitosan was high. A similar tendency was shown in K, P and Mg. In the small tuber(under 30g), the number of tubers and tuber yield were relatively increased in the seed potatoes planting plot, but the large tubers(over 80g) yield was higher in the plug seedlings planting plot, and in order to increase tuber yield in plug seedlings it was necessary to add plant density to the field.

Characteristics of flowering, fruit setting and propagation of Callistemon citrinus (Curt.) Skeels (병솔꽃나무(Callistemon citrinus (Curt.) Skeels)의 개화결실 및 증식특성)

  • This study was performed to obtain the information on fruit set, propagation and flowering habits of the exotic ornamental bottle brush tree (Callistemon citrinus). Trees have fruit capsules for four years since the fruit set and they have enlarged for three years. While 2 and 3-year-old capsules have similar seeds in number and size, 1-year-old ones have fewer and smaller seeds. Most dried capsules open when they have 75% relative water contents. Two year old seeds which they were dried at 30% germinated better than others. Removal of capsule bearing twigs resulted in greatly increased subsequent flower numbers (an average of 14 flowers per tree) compared with twigs where seed capsule were allowed to remain. In cutting for propagation, semi-hard wood branches dipped in 100mgι$^{-1}$ IBA for 24 hours rooted at 86.4 % and had an average of 12.6 primary roots.

Push-out bond strength of a self-adhesive resin cement used as endodontic sealer

  • Objectives: The aim of the present study was to investigate the bond strength of RelyX Unicem (3M) to root canal dentin when used as an endodontic sealer. Materials and Methods: Samples of 24 single-rooted teeth were prepared with Gates Glidden drills and K3 files. After that, the roots were randomly assigned to three experimental groups (n = 8) according to the filling material, (1) AH Plus (Dentsply De Trey GmbH)/Gutta-Percha cone; (2) Epiphany SE (Pentron)/Resilon cone; (3) RelyX Unicem/Gutta-Percha cone. All roots were filled using a single cone technique associated to vertical condensation. After the filling procedures, each tooth was prepared for a push-out bond strenght test by cutting 1 mm-thick root slices. Loading was performed on a universal testing machine at a speed of 0.5 mm/min. One-way analysis of variance and Tukey test for multiple comparisons were used to compare the results among the experimental groups. Results: Epiphany SE/Resilon showed significantly lower push-out bond strength than both AH Plus/Gutta-Percha and RelyX Unicem/Gutta-Percha (p < 0.05). There was no significant difference in bond strength between AH Plus/Gutta-Percha and RelyX Unicem/Gutta-Percha (p > 0.05). Conclusions: Under the present in vitro conditions, bond strength to root dentin promoted by RelyX Unicem was similar to AH Plus. Epiphany SE/Resilon resulted in lower bond strength values when compared to both materials.

Change of Rooting Potential as Affected by Rooting Promoter Treatment in Cuttings of Corylopsis coreana (히어리(Corylopsis coreana)의 삽목번식시 발근촉진제의 처리에 따른 발근력 변화)

  • This study was conducted to determine the effects of several rooting promoter treatments on rooting of Corylopsis coreana cutttings for mass production. In first year experiment, no effect of rooting promoters was observed in hardwood cuttings taken on March 20. However, In softwood cuttings of June 20, rooting potential was greatly increased by IBA $100mg{\cdot}L^{-1}$ dipping treatment for 24 hours. Particularly, IBA $100mg{\cdot}L^{-1}$ 24 hour dipping treatment greatly shortened the days required for the rooting. In second year experiment, the result of first year was reconfirmed. Almost of all the cuttings (97.8% of rooting rate) were well rooted after only 30 days after cutting by IBA $200mg{\cdot}L^{-1}$ 24 hour dipping treatment. In this treatment, numbers and length of root were also prominent compared with any other treatment results.

Effects of Simulated Acid Rain on Chemical Properties of the Experimental Soil of pinus densiflora S. et Z. and Forsythia Koreana Nak. Seedlings (人工酸性雨가 소나무 및 개나리 盆植苗土壤의 化學的 性質에 미치는 影響)

  • For the purpose of examining the effects of simulated acid rain on the chemical properties of the experimental soil in Pinus densiflora seedlings and Forsythia koreana rooted cuttings, the experimental design of randomized block arrangement with three replications was implemented in the experimental field of Yesan National Agricultural Junior College. One-year-old Pinus densiflora seedlings and Forsythia koreana cuttings were planted in the pots filled the mixed soils (nursery soil: forest soil of siliceous sandy loam = 1 : V/V)in the early spring of 1985. The regime of artificil acid rain, in terms of spray frequency per monthly and spray amount at single treatment per plot, was simulated on the basis of climatological data averaged from 30 years records. Simulated acid rain (pH 2.0, pH4.0, and pH 5.5 as control) containing sulfuric and nitric acid in the ratio of 3:2 (chemical equivalant basis) diluted ground water, were treated on the experimental plants under condition of cutting off the natural precipitation with vinyl tunnel, during the growing season (May 1 to August 31) in 1985. THe results obtained in this study were as follows; 1. Soil acidity was dropped, and exchangeable aluminum contents in the soil was dramatically increased in both species, with decreasing pH levels of acid rain. 2. Exchangeable potassium, clacium, magnesium contents, and base saturation degree of the soil were highly drcreased in two species as the pH levels of acid rain decreased. 3. In two species, sulfate concentrations in the soil were decreased of pH 4.0 treatment, and remarkably increased at pH 2.0 treatment of acid rain in comparison with control. 4. Total nitrogen and available phosphate contents of the soil were not affected by acid rain treatment in the both species, and Fe contents at pH 2.0 treatment were highest among three acid rain treatments.

In vitro propagation of a rare and endangered species, Echinosophora koreensis Nakai, by axillary bud culture (희귀 및 멸종위기 수종 개느삼의 액아배양을 통한 기내번식)

  • An efficient micropropagation was established by using axillary bud explants from two-year-old tree(Echinosphorea koreensis Nakai), which has been known as a rare and endangered species. Among various basal media tested, DKW medium was shown to be the best for axillary shoot elongation. The addition of both BA and TDZ to the medium induced 6 to 10 shoots per explant during eight weeks of culture, without showing any abnormal morphology at the shoot proliferation stage. However, high concentration of TDZ(>0.05 mg/L) appeared to cause hyperhydration on either leaf or shoot at the later developmental stage. Approximately 20% of shoots produced roots by the addition of 1.0 mg/L NAA but not by IBA($0.2{\sim}1.0$ mg/L). Ex vitro micro-cuttings were better source for root induction; up to 58.6% of the micro-cuttings rooted when 100 mg/L IBA was applied to the soil(vermiculite). More than 90% of plantlets with roots were successfully acclimatized and grew normally in the field. Therefore, we suggest that this endangered tree species can be effectively micropropagated by axillary bud culture system developed in this study.

Cutting Propagation of Eleutherococcus senticosus MAXIM (가시오가피의 거목번식방법(揷木繁殖方法))

  • Eleutherococcus senticocus MAXIM containing compounds for a cordial and lumbago is a native plant in this country. However it is generally difficult to propagate them by seed. This study was carried out to improve propagation efficacy by cutting method using Eleutherococcus senticosus MAXIM collected Mt. Odae for three years$('90{\sim}'92)$. Rooting was delayed by 3 to 12 days but callus formation and rooting percentage was high by 75%, 30% respectively in the treatment of Rooton-F powder, a chemical for rooting promotion, compared with non-treatment. Rooting percentage and rooting characters were not differenced by treatment for remove rooting inhibition compounds and soil-media mixured by 1:1 of vermiculite and perlite was suitable for elevating rooting ratio. A. chiisanensis and A. sieboldianum were rooted well in any time to be cut, while rooting activity of E. senticosus was different by time to be cut. Rooting ratio of E. senticosus was 26%, 36% and 60% when cutten March 20, July 20 and september 20, respectively.
