Effect of Biosolids on Heavy Metal Bioavailability and Organic Acid Production in Rhizosphere of Zea mays L. (Biosolid 처리가 옥수수 근권의 유기산 생성과 중금속 유효도에 미치는 영향)

  • A greenhouse experiment was conducted to evaluate the composition of organic acids on the bioavailability of heavy metals in the rhizosphere of corn (Zea mays L.) over 12 weeks after treatment of biosolids. After planting the plants were sampled at 12th week and analyzed for heavy metals in shoot and root and organic acids in the rhizosphere. Results showed that biomass yields of Zea mays L. grown on biosolids-treated media were significantly higher than those grown on standard media. The effect of biosolids on biomass yields of Zea mays L. was in order of Riverside ${\approx}$ Los Angeles ${\approx}$ MWRDGC > Milorganite > Nu-earth > standard media. Metal uptake by Zea mays L. was closely related with the contents contained on biosolids treated. In the plot treated with Nu-earth the uptake of Cd and Zn by shoot was significantly higher than those at the plots treated with other biosolids. The uptake patterns of Cd and Zn by root were similar to those of shoot. The uptake of Cr and Ni was significantly higher with application of Nu-earth over other biosolids. In all cases, the major organic acids in the rhizoshpere were lactic, acetic, propionic, butyric, and oxalic acids. Other organic acids were glutaric and succinic and occasionally, pyruvic and tartaric acids were also found. And the organic acid compositions did not vary significantly with the treatments of biosolids. Butyric and acetic acids were distinctively dominant both in the standard and the biosolids treatments.

Nursery Growing Media Practice: Impact on Seed Germination and Initial Seedling Development of Hymenodictyon orixensis (Roxb.) Mabberley - A Vulnerable Native Tree Species

  • Hymenodictyon orixensis (Roxb.) Mabberley (locally known as Bhutum in Bangladesh) is both an ecologically and economically valuable multipurpose tree species for afforestation and reforestation programs in Bangladesh. Seed germination and seedling development study of H. orixense were conducted to find out the response to different growing medium, e.g., polybag (15×10 cm (T0) and 20×15 cm (T1)), sand medium in propagator house (T2), conventional nursery bed (T3), and root trainer (T4) in the Nursery. Consequently, germination behavior and seedling morphological parameters of H. orixense were assessed. The results revealed that the sand medium of the propagator house (T2) provided the highest germination % (58.57±22.30) and the highest germination energy (11.43±2.43) followed by seedlings growing in 20×15 cm polybags (T1) containing forest topsoil and cow-dung at a ratio of 3:1. Except for germination energy, germination values, and germination capacity, other seed biology parameters, particularly imbibition, germination period, germination rate, and plant survival percent in T1, T2, T3, and T4 were significantly (p<0.05) different from T0. Each phenotypic parameter of seedlings and dry matter of shoot and root significantly differed from control except root length (p<0.992). Based on this study, Polybags of 20×15 cm size are regarded as the best medium for quality seedling development of H. orixense. The nursery bed (T3) had the lowest germination performance and developed more inferior quality seedlings. Thereby, 20×15 cm size of polybags with conventional soil and cow-dung media is recommended for maximum germination and to grow the quality seedlings of H. orixense in the Nursery.

Influence of Bicarbonate Concentrations in Irrigation Solution on Growth of Lettuce and Changes in Chemical Properties of Root Media (원수의 중탄산 농도가 근권 화학성 변화 및 상추의 생장에 미치는 영향)

  • This study was conducted to investigate the influences of bicarbonate ($HCO_3^-$) concentrations in irrigation solution on growth of lettuce and change in chemical properties of root media. The blue leaf and red leaf lettuces with two true leaf stages were transplanted into 10 cm diameter plastic pots filled with perlite. The five treatments were made by dissolving $NaHCO_3$ into irrigation solution to reach 30, 70, 110, 150 and $180mg{\cdot}L^{-1}$ $HCO_3^-$. The crops were fed with fertilizer solution contained $HCO_3^-$ with various concentrations and controlled to $100mg{\cdot}L^{-1}$ in nitrogen concentration. The pH in soil solution of root media 10 weeks after transplant of blue lettuces were 7.04 and 7.10 in the treatments of 30 and $70mg{\cdot}L^{-1}$ of $HCO_3^-$, respectively. But those rose gradually after week 3 and finally reached 7.39, 7.48 and 7.56 at week 10 in the treatments of 110, 150 and $180mg{\cdot}L^{-1}\;HCO_3^-$, respectively. The pH in the treatments of 30 and $70mg{\cdot}L^{-1}\;HCO_3^-$ in cultivation of red leaf lettuce were around 6.65 during week 4 to week 8, but this rose abruptly and reached 6.92 and 7.01 at week 10, respectively. Those in the treatments of 110, 150, and $180mg{\cdot}L^{-1}\;HCO_3^-$ rose gradually and finally reached to 7.49, 7.53, and 7.58, respectively. The EC rose gradually after week 2 in all treatments of blue and red leaf lettuces. The change of macro ion concentrations in both blue and red leaf lettuces showed similar trends. The concentrations of $PO_4-P$, $Ca^{2+}$ and $Mg^{2+}$ increased gradually in all treatments of $HCO_3^-$ during cultivation of blue and red leaf lettuces. As the concentrations of $HCO_3^-$ in irrigation solution were elevated, the concentrations of $PO_4-P$, $Ca^{2+}$ and $Mg^{2+}$ became higher and that of ${SO_4}^{-2}$ became lower in soil solution of root media. The main reason of concentration changes were that $HCO_3^-$ influenced pH and the pH changes also affect the activities of the ions in soil solution of root media.

Effects of White Light and UV Irradiation on Growth and Saponin Production from Ginseng Hairy Root (광 및 UV 조사가 인삼 모상근의 생장 및 사포닌 생합성에 미치는 영향)

  • To investigate the effect of culture conditions on growth and ginsenosides accumulation, we cultured the ginseng hairy root under three different media, white light or ultra-violet irradiation. The MS/B5 medium containning MS basal salt and B5 vitamin was good for the growth and ginsenoside accumulation. The light during the culture period of ginseng hairy root was irradiated. The growth was abundant in the ginseng hairy root cultured in dark. But the ginsenosides accumulation was higher than in the ginseng hairy root cultured in the light irradiation. When the ginseng hairy root was cultured in 20 L bioreactor, the ginsenosides accumulation was observed at 34% higher than the hairy root cultured in dark. UV irradiated the ginseng hairy root during the culture period. The long time irradiation of UV was caused decreasing the growth of ginseng hairy root, but the accumulation of ginsenosidess was increased as to the irradiated time.

Influence of formulated organic Plant tissue culture medium in the shoot regeneration study of Brassica juncea (l.) - Indian mustard

  • Efficient protocol for plant shoot regeneration of Brassica juncea L. CZERN was established by using organic media components and growth stimulating factors of the vermicompost and coelomic fluids. Formulated organic plant tissue culture media (Vermicompost (30%) extracts supplemented with 20 mL/L coelomic fluid) have shown maximum shoot regeneration when compared with the Murashige and Skoog (MS) medium, which were supplemented with 1 mg/L 6-benzyladenine (BA) and 0.1 mg/L of Naphthaleneacetic acid (NAA). Cotyledon explants produced the highest shoot regeneration frequency from fourday-old germinated seedlings in comparison with non-germinated seedlings. The vermicompost extracts have proved to be the best organic plant growth media to induce shoots from cotyledons compared to the MS media. Statistically significant difference (P = 0.008) for the root length, shoot length (P=0.000350) and the leaves (P=0.375) of the mustard plantlets were analyzed successfully. The survival rate was 98% in the mustard cotyledons on the Vermicompost extract media and 63% on MS media respectively. The coelomic fluid also is much suitable to induce shoots from cotyledons at lower concentrations. It was also shown that the vermicompost extract, which comprised of humic acids along with coelomic fluid, affected shoot regeneration from the cotyledons. An efficient and organic shoot regeneration study was standardized and it can be applicable in the improvement of the economically important crops.

Impact of Pre-planting NH4+:NO3- Ratios in Inert Media on the Growth of Chinese Cabbage Plug Seedlings (혼합상토에 기비로 혼합된 질소의 NH4+:NO3- 비율이 배추의 플러그 묘 생장에 미치는 영향)

  • This research was conducted to evaluate the impact of various pre-planting $NH_4{^+}:NO_3{^-}$ ratios on the growth of plug seedlings of 'Bool-am No.3' Chinese cabbage. With fixation of the pre-planting N concentrations to $300mg{\cdot}kg^{-1}$ in a peatmoss+coir dust+perlite (3.5:3.5:3, v/v/v) medium, the $NH_4{^+}:NO_3{^-}$ ratios were varied to 0:100, 27:73, 50:50, 73:27, 100:0. Then, the each of root media containing various ratios of $NH_4{^+}:NO_3{^-}$ as well as equal concentrations of other essential nutrients was packed into 72-cell plug trays. After seeds of 'Bool-am No.3' Chinese cabbage were sown, the seedling growths were measured 2 and 4 weeks after sowing. The weekly analysis of root media and end-crop tissue analysis for mineral nutrients 4 weeks after seed sowing were also conducted. As the seedlings grew up, the pH of the root media increased, however ECs in all treatments of $NH_4{^+}:NO_3{^-}$ ratios decreased. The concentrations of K, Ca and Mg in root media were higher in the treatments of $NH_4{^+}:NO_3{^-}$ (100:0) and $NH_4{^+}:NO_3{^-}$ (73:27) than those of $NH_4{^+}:NO_3{^-}$ (0:100) and $NH_4{^+}:NO_3{^-}$ (27:73) 2 weeks after seed sowing. But the concentrations of K, Ca, Mg and Zn were get lowered in all treatments and the differences among treatments were not significant 4 weeks after sowing. The highest $NH_4{^+}$ and lowest $NO_3{^-}$ concentrations of the root media were observed in the $NH_4{^+}:NO_3{^-}$ (100:0) among all treatments. Contrary to these, the treatment of $NH_4{^+}:NO_3{^-}$ (0:100) had the lowest $NH_4{^+}$ and highest $NO_3{^-}$ concentrations. The seedling growth in terms of fresh and dry weights of aerial part were the highest in the treatment of $NH_4{^+}:NO_3{^-}$ (23:73) at 2 weeks after sowing and those of $NH_4{^+}:NO_3{^-}$ (50:50) at 4 weeks after sowing. The survival rate of seedlings in $NH_4{^+}:NO_3{^-}$ (100:0) treatment were 19% and the growth of aerial part 4 weeks after sowing was the poorest among all treatments tested. The results mentioned above indicate that the pre-planting $NH_4{^+}$ ratio in inert media should not exceed 25% in plug stage 1 through 3 (until 2 true leaf development) and 50% in plug stage 4 (after 2 true leaves to transplant).

Studies on the Vegetative Propagation of Korean Ginseng (Panax ginseng C. A. Meyer) 1. Effects of the NAA Concentration and the Rooting Media on the Rooting of the Ginseng Stern Cutting (고려인삼의 영양번식에 관한 연구 제1보 NAA의 농도 및 잡목용배지가 인삼경삽수의 발근에 미치는 영향)

  • To develop a new propagation method of Korean ginseng (Panax ginseng C.A. Meyer) by cutting, stem cuttings with leaflets obtained from ginseng seedlings were planted in rooting media treated with three levels of NAA concentration. NAA solution of 1.0 ppm was the most effective for the rooting of ginseng stem cuttings as well as for root growth after initiation of the roots from cuttings. A mixture of sand and leaf compost as a rooting medium for ginseng stem cuttings showed the best results in rooting percentage of cuttings and root growth after rooting. An acril film cap covering was very effective to prevent water loss by transpiration from the leaflets of stem cuttings and for rooting and root growth after rooting. Cuttings of leaf petiole with 5 leaflets from 2 year old ginseng plants showed good rooting as well as root growth after rooting in a mixture of sand and leaf compost treated with 1.0 ppm NAA.

Anti-Oomycete Activity and Pepper Root Colonization of Pseudomonas plecoglossicida YJR13 and Pseudomonas putida YJR92 against Phytophthora capsici

  • Previously, Pseudomonas plecoglossicida YJR13 and Pseudomonas putida YJR92 from a sequential screening procedure were proven to effectively control Phytophthora blight caused by Phytophthora capsici. In this study, we further investigated the anti-oomycete activities of these strains against mycelial growth, zoospore germination, and germ tube elongation of P. capsici. We also investigated root colonization ability of the bacterial strains in square dishes, including cell motility (swimming and swarming motilities) and biofilm formation. Both strains significantly inhibited mycelial growth in liquid and solid V8 juice media and M9 minimal media, zoospore germination, and germ tube elongation compared with Bacillus vallismortis EXTN-1 (positive biocontrol strain), Sphingomonas aquatilis KU408 (negative biocontrol strain), and MgSO4 solution (untreated control). In diluted (nutrient-deficient) V8 juice broth, the tested strain populations were maintained at >108 cells/ml, simultaneously providing mycelial inhibitory activity. Additionally, these strains colonized pepper roots at a 106 cells/ml concentration for 7 days. The root colonization of the strains was supported by strong swimming and swarming activities, biofilm formation, and chemotactic activity towards exudate components (amino acids, organic acids, and sugars) of pepper roots. Collectively, these results suggest that strains YJR13 and YJR92 can effectively suppress Phytophthora blight of pepper through direct anti-oomycete activities against mycelial growth, zoospore germination and germ tube elongation. Bacterial colonization of pepper roots may be mediated by cell motility and biofilm formation together with chemotaxis to root exudates.

Effects of Auxin-induced Ethylene on Growth and Development of Adventitious Roots of Panax ginseng C.A. Meyer (IBA와 NAA 처리에 의해 생성된 Ethylene이 인삼(Panax ginseng C.A. Meyer) 부정근의 생장과 발달에 미치는 영향)

  • The effect of IBA and NAA on adventitious root cultures of Panax ginseng C.A. Mater were investigated. Results indicated differences in growth and development of the roots according to 5mg/L IBA and 2mg/L NAA. IBA resulted in a normal root development and a higher growth compared to NAA. The roots formed on NAA-containing media were shorter and thicker than those in IBA, showing a hypertrophy of the root tip. NAA induced more than 1.6 times higher ethylene production compared to IBA, which caused inhibition of the root growth. Under the ventilation, in the other hand, on difference was observed in ethylene concentration and the root growth between IBA and NAA treatments. Under ventilation ethylene production was not detected until 10 days of culture, while detected from the initial stage under on ventilation. The results suggested the importance of ventilation during the culture for the growth and development of ginseng adventitious roots.

In Vitro Germination and Propagation by Embryo Culture of Taxus cuspidata for the Taxol Production

  • To develop an efficient propagation method for yew tree, zygotic embryos were cultured under various conditions. When dissected embryos were cultured on GA$_3$ containing media, the highest germination frequency was observed on WPM medium contaning 1.0 mg/L GA$_3$. For germination of the embryos, two different conditions were compared; culturing embryos with endosperm (Method I), and 2) culturing embryos only (Method II). Maximum germination was achieved in 0.5 mg/L GA$_3$ when embryos with endosperm were cultured on the media. Of the media tested, White and WPM medium were the most suitable on germination of embryos. The abnormality of yew embryos found was observed when it cultured on GA$_3$ or culture media. About 40% of the precociously germinated embryos could be developed into full seedlings. Seedlings contained taxol in high quantity (535 $\mu\textrm{g}$/g dry weight). In vitro techniques will be sewed as a useful tool for the development of transformed root cultures and biosynthesis studies.
