Screening of Botanicals Against Root Knot Disease Complex in Mulberry (Morus indica L.)

  • For developing an integrated eco-friendly package against root knot disease complex of mulberry caused by the association of Meloidogyne incognita with Fusarium solani and F. oxysporum causing serious loss in terms of leaf yield and quality during cultivation, twenty botanical extracts at 5, 10 & 20% concentrations were screened under in vitro conditions. Among the extracts, Allium sativum followed by Lasownia inermis were found to be effective at 20% concentration against both the virulent fungi and nematode. Both the extracts reduced the mycelial growth of virulent fungi to an extent of 76-100%, inhibited the hatching of nematode eggs by 80-90% and 76-85% larval mortality over the control. The other extracts were found either moderately or poorly effective in reducing the growth of fungi, hatching of nematode eggs and enhancing the mortality of larvae. The two effective botanical extracts, which rated as strong inhibitors against both nematode and virulent fungi, can be utilized in developing an integrated ecofriendly technology for better management of root knot disease complex in mulberry.

약용식물의 뿌리혹선충 발생과 분류동정 (Incidence, and Identification of Three Root-Knot Nematode species Occurring in the Medicinal Herbs)

  • Soil and root samples were collected form the rhizoshpere of 11 different medicinal plants to determine the incidence, density and identification of root-knot nematode species associated with medicinal herbs. About 55% of medicinal herbs examined was found to be infested with root-knot nematodes. As a result of infection casued by three root-knot nematodes, M. hapla recorded 43.3% in medicinal herba whereas M. incognita and M. arenaria showed 7.9% and 3.7%, repectively. Forsythia koreana, Hemerocalis fulva, Hibuscus mutabilis and Petasites japonicus were the most severely infested herbs whereas Acanthopanax sessilflorus was least infested. Population of the second stage younger plants. Meloidogyne hapla, M. incognita and M. arenaria were the species associated with the medicinal herbs. The most abundant nematode observed in medicinal herbs was M. hapla and followed by M. incognita and M. arenaria. M. arenaria was observed firstly on Ficus carica, one of medicinal plant.

Efficacy of Pesticides and Growth Hormones against Root Disease Complex of Mulberry (Morus alba L.)

  • During mulberry cultivation, root disease complex caused by the association of root knot nematode (Meloidogyne incognita) with root rot pathogens like Fusarium solani and Botryodiplodia theobromae poses serious loss in leaf production. Therefore, an attempt was made to assess the efficacy of eight pesticides (Metayalaxyl+Mancozeb, Thiophanate methyl, Mancozeb, Bitertanol, Phenomiphos, Phorate, Thionazin & Carbofuran) and two growth hormones (Salicylic acid and Indole 3 acetic acid) at 0.1 and 0.2% concentrations under in vitro conditions against nematode (hatching of eggs and mortality of larvae) and root rot pathogens (poisoned food technique) for short listing the treatments to develop an IDM strategy. Results revealed that among the pesticides and growth hormones, Carbofuran followed by Salicylic acid were found to be effective at 0.2% concentration against both nematode and pathogenic fungi. Both the chemicals inhibited the hatching of nematode eggs by 83.5-78.9% and 80-76% larval mortality over the control and reduced the mycelial growth of both the pathogenic fungi to an extent of 75.5-77.8%. Though Mancozeb inhibited both the pathogenic fungi strongly (77-80%), it did not show any effectiveness against nematode. The rest of the chemicals were found either moderately or poorly effective in reducing the growth of pathogenic fungi, hatching of nematode eggs and enhancing the mortality of larvae. The two effective chemicals viz., Carbofuran and Salicylic acid, which rated as strong inhibitors against both nematode and pathogenic fungi, can be exploited in developing an IDM package as one of the component for better management of root disease complex in mulberry.

  • Root-knot nematodes cause billions of dollars in crop losses annually have a broad range of host over 2,000 species of plants. These nematodes are known as obligate, sedentary endo-parasites in a plant host to feed upon to complete their life cycle. To prevent the plant parasitic nematode, methyl bromide was widely applied as a soil fumigant. Other strategies to prevent or control nematodes involve RNAi-mediated suppression, R gene transformation, natural products or chemical treatments, the expression of peptide or proteins in susceptible plants, and others. Over the last decade, the entry in GenBank for Meloidogyne reveals 73,340 ESTs and recently two complete Meloidogyne spp. genomes sequences have simultaneously been presented by two groups. Recent works have demonstrated the effect of RNAi suppression to nematode target genes. These results will provide novel members of genes as a foundation for studies focused on understanding the function of M. incognita nematode genes as well as for the development of novel target genes for parasite control. Thus the successful development of biotechnology-derived plants with nematode resistance will result in large yield benefits for producers as well as environmental benefits and will accelerate the research related to pathogensresistant crops.

작약재배지 뿌리혹선충의 발생생태에 관한 연구 (Annual Phenology of Root-knot Nematode in the Medicinal Herb (Paeonia lactiflora) Field)

  • 경북의성 약용작물재배지에서 뿌리혹선충에 대한 발생생태를 조사하기 위하여 시기별 발생소장과 토성에 따른 선충의 증식정도, 토양심도 다른 뿌리혹선충의 서식밀도를 1991년부터 1992년까지 2개년간 조사한 결과는다음과 같다. 뿌리혹선충(M.hapla)의 발생소장을 조사한 결과 유충발생 최성기는 4월초순, 6월중순, 7월하순, 8월하순 등 4회였다. 토성과 뿌리혹선충과의 관계를 보면 사토와 사양토에서 뿌리혹선충의 난관 유충이 가장 많았고 식토에서는 밀도가 낮았다. 토양심도별 M.hapla 유충밀도는 작약포장에서 토심 15cm까지가 1,064~1,486마리로 가장 높았으며 50cm이상에서도 검출되었다.

Effects of Arugula Vermicompost on the Root-Knot Nematode (Meloidogyne javanica) and the Promotion of Resistance Genes in Tomato Plants

  • Root-knot nematodes are the most important plant-parasitic nematodes worldwide. Many efforts have been made to find non-chemical, risk-free, and environmentally friendly methods for nematode control. In this study, the effects of compost and vermicompost of arugula (Eruca sativa) on Meloidogyne javanica were investigated in three glasshouse experiments. In addition, the expression of the defense-related genes nonexpressor of pathogenesis-related 1 (NPR1) and lipoxygenase 1 (LOX1) was detected in tomato plants treated with vermicompost of arugula at 0, 2, 7, and 14 days after nematode inoculation. The result showed that the vermicompost of arugula significantly reduced the reproduction factor of the nematode by 54.4% to 70.5% in the three experiments and increased the dry weight of shoots of infected tomato plants. Gene expression analysis showed that LOX1 expression increased on the second and seventh day after nematode inoculation, while NPR1 expression decreased. The vermicompost of arugula showed stronger nematode inhibitory potential than the vermicompost of animal manure. The vermicompost of arugula is superior to arugula compost in suppressing the activity of M. javaniva and reducing its impact. It manipulates the expression of resistance genes and could induce systemic resistance against root-knot nematodes.

Root-Knot Nematode (Meloidogyne incognita) Control Using a Combination of Lactiplantibacillus plantarum WiKim0090 and Copper Sulfate

  • Lactic acid bacteria (LAB) exert antagonistic activity against root-knot nematodes, mainly by producing organic acids via carbohydrate fermentation. However, they have not yet been used for root-knot nematode (Meloidogyne incognita) control owing to a lack of economic feasibility and effectiveness. In this study, we aimed to isolate organic acid-producing LAB from kimchi (Korean traditional fermented cabbage) and evaluated their nematicidal activity. Among the 234 strains isolated, those showing the highest nematicidal activity were selected and identified as Lactiplantibacillus plantarum WiKim0090. Nematicidal activity and egg hatch inhibitory activity of WiKim0090 culture filtrate were dose dependent. Nematode mortality 3 days after treatment with 2.5% of the culture filtrate was 100%, with a 50% lethal concentration of 1.41%. In pot tests, the inhibitory activity of an L. plantarum WiKim0090-copper sulfate mixture on gall formation increased. Compared to abamectin application, which is a commercial nematicide, a higher control value was observed using the WiKim0090-copper sulfate mixture, indicating that this combination can be effective in controlling the root-knot nematode.

Occurrence of the Root-Knot Nematode species in Vegetable Crops in Souss Region of Morocco

  • Root-knot nematodes 'Meloidogyne spp' are the most destructive group of plant parasitic nematodes causeing serious losses in vegetables crops and this damages worsened when crops grown under greenhouses conditions. In this sutdy, the distribution and characterization of root-knot nematode species collected from the Souss region of Morocco where vegetables crops intensively cultivated were determined by using both morphological and molecular tools. Out of the 110 samples collected from different greenhouses 91 (81.7%) were found to be infested with root-knot nematodes. Thirty-seven populations of root-knot nematodes were morphologically identified based on perineal patterns as well as molecularlly using species-specific primers. The obtained results indicated that Meloidogyne javanica and M. incognita were identified in 86.4% and 13.5% of the total populations, respectively. The lowest incidence of root-knot nematodes (64%) was found in Toussous, whereas the highest frequencies of 100% and 90% were detected in Taddart and Biogra, respectively. As the majority of the samples have been infested with Meloidogyne species; this indicates that there is an urgent need to provide farmers with a proper control strategy.

지중가온시설을 이용한 고구마뿌리흑선충 방제 (Control of Root-Knot Nematode(Meloidogyne incognita Chitwood) by Root Zone Warming System)

  • 지중가온시설을 이용한 고구마뿌리혹선충(Meloidogyne incognita Chitwood) 방제를 위하여 지하 20cm 깊이의 최저지온을 각각 30. 40, $50^{\circ}C$ 가온구 및 관행의 무지중가온구로 설정하고 담수구와 무담수구로 나누어 8월 1일부터 8월 5일까지 5일간 가온한 결과, 담수구의 경우 지온 4$0^{\circ}C$에서 5일간, 무담수구의경우 지온 $50^{\circ}C$에 5일간 지중가온처리한 구에서 생존한 유충이 발견되지 않아 선충밀도를 경제적 피해수준 이하로 유지할 수 있을 것으로 사료되었다. 지중가온으로 토양내 OM. P$_2$ $O_{5}$, Ca, Mg함량과 EC는 시험전 토양에 비하여 감소하였는데, 특히 담수처리에서 EC가 현저히 저하하였다.

Effects of Heavy Metal Contamination from an Abandoned Mine on Tomato Growth and Root-knot Nematode Development

  • Physicochemical characteristics and heavy metal content of soils located along the drainage way of an abandoned mine at Busan, Korea ($35^{\circ}31'N$, $129^{\circ}22'E$) (contaminated soil; CS) and uncontaminated soils (50-70 m apart from the drainage way) (NS) were examined. Growth of tomato plants (Solanum lycopersicom cv. Rutgers) in CS and NS, development of the root-knot nematode (Meloidogyne incognita) as root-knot gall formation on tomato plants, and non-parasitic nematode populations in soil were also examined. Growth of tomato plants, root-knot gall formation, and non-parasitic nematode populations were significantly reduced in CS with higher As content, lower pH, higher electrical conductivity (EC), and lower available phosphate (av. $P_2O_5$) than in NS. None of the other physicochemical characters examined differed significantly between CS and NS (low and no significance) and were above or below the critical levels detrimental to plant growth and nematode development, suggesting that As may be the primary hazardous heavy metal in CS. The toxicity of As might be enhanced at low pH in CS because exchangeable forms of some heavy metals increase with the decrease of soil pH. The heavy metals, especially As, may have contributed to increasing EC and decreasing av. $P_2O_5$. Therefore, the effects of mine drainage contamination from the abandoned mine were derived primarily from contamination by heavy metals such as As. These may have been enhanced in toxicity (solubility) by the lowered pH, increased soil salinity (EC) and decreased av. $P_2O_5$. Our results suggest synergistic adverse effects on the plant and the nematode by decreasing osmotic potential and nutrient availability.
