오미자 추출물을 첨가한 도라지와 연근 정과의 품질특성 (Quality Characteristics of Bellflower and Lotus Root Jeonggwa Added Omija (Schizandra chinensis Baillon) Extract)

  • The purpose of this research is to develop bellflower (Platycodon grandiflorum) root and lotus (Nelumbo nucifera) root Jeonggwa as a health food. The quality characteristics of Jeonggwa to which 0-8% (w/w) of Omija (Schizandra chinensis Baillon) water extract was added were investigated. The moisture contents of bellflower root Jeonggwa were 14.9-18.0%, where as that of lotus root Jeonggwa was 6.9-8.4%. The acidity of bellflower root Jeonggwa was 0.477-0.585% and that of lotus root Jeonggwa was 0.513-0.572%; values increasing levels of Omija extract. The lightness (L) and yellowness (b) values of bellflower root Jeonggwa decreased with increasing amounts of Omija extract, and the lightness (L), redness (a) and yellowness (b) values of lotus root Jeonggwa decreased with increasing Omija extract concentration (p<0.05). In mechanical tests with bellflower root Jeonggwa, the highest values of hardness and strength were seen at 2% (w/w) extract, of cohesiveness were seen with 4-8%, of springiness with 2-6%, of gumminess with 6-8%, and of brittleness with 4-6% (all p<0.01). In mechanical tests with lotus root Jeonggwa, the highest values of hardness and springiness were at 0 and 6% extract, respectively, whereas strength, gumminess and brittleness were at 6-8% Omija extract (all p<0.01). In sensory evaluation, the highest acceptability of bellflower root Jeonggwa was seen with 4% extract and that of lotus root Jeonggwa was seen with 2% extract.

연근(Nelumbo nucifera Root) 추출물의 HT-29 인체 대장암세포 증식 억제 및 사멸 효과 (Effects of Nelumbo nucifera Root Extract on Proliferation and Apoptosis in HT-29 Human Colon Cancer Cells)

  • Our study is investigated the effects of Nelumbo nucifera root extract on HT-29 colon cancer cells. The anti-proliferative effect of 70% ethanol extract from Nelumbo nucifera root on HT-29 colon cancer cells was identified based on cell viability, Hoechst 33342 nuclear staining, apoptosis analysis, Western blotting and RT-PCR analyses. In our study, Nelumbo nucifera root extract inhibited the growth of HT-29 colon cancer cells in a dose-dependent manner. Concomitant activation of the mitochondria-dependent apoptotic pathway of HT-29 colon cancer cells by Nelumbo nucifera root extract occurred via modulation of Bax and Bcl-2 expressions, which activated cleavage of caspases-3 and -9. The findings of this study indicate that Nelumbo nucifera root extract induces apoptosis in HT-29 colon cancer cells, and this phenomenon is occurs via the death receptor-mediated and mitochondria-mediated apoptotic pathways.

Anti-Obesity Effects of Red Beet Extract

  • Obesity is caused by the accumulation of triglycerides in adipocytes by the differentiation and lipid synthesis process of pre-adipocytes, and excessive accumulation of adipocytes by the activated Adipogenesis process within the differentiated cells. Therefore, inhibiting the differentiation of adipocyte cells or controlling the adipogenesis process is known as an effective treatment method for obesity. This study evaluates the inhibition of Red beet root extract on pancreatic lipase and pre-adipocyte cell differentiation. Also it evaluates the Red beet root extract activities on C/EBP-𝛼,𝛽, and PPAR-𝛄. The experiments proved that the Red beet root extract inhibits pancreatic lipase by concentration dependency. Further, in 3T3-L1 inhabitation experiment, it was found Red beet root extract inhibited adipocyte formation. Red beet root extract also inhibits the expression of C/EBP-𝛼, C/EBP-𝛽, and PPAR-𝛾 which effect the process of adipocytic differentiation. We therefore concluded that RBE has a high potential to further studies on anti-obesity effect.

유방암 세포주에서 PMA로 유도된 암세포 침투에 대한 모링가 뿌리 추출물의 억제효과 (Extract of Moringa Root Inhibits PMA-induced Invasion of Breast Cancer Cells)

  • 여성에게 특히 높은 발병률을 보이는 유방암은 다양한 인자에 의해 암세포 침투와 전이가 발생한다. 그중에서 MMP-9은 유방암에서 높은 발현율을 나타내며, 유방암 세포의 침투와 전이에 주요하게 작용한다. MMP-9은 다양한 성장인자와 사이토카인, PMA 등에 의해 발현이 유도되며 세포 내 자연적으로 발생된 내생적 억제인자인 TIMP-1과 -2에 의해 그 활성 및 발현이 조절된다. MMP-9의 활성 및 발현을 저해하기 위해 사용한 물질은 모링가 추출물로 모링가의 모든 부위가 다양한 질환을 치료하는 것으로 알려져 있다. 이 연구에서는 대표적으로 모링가씨, 뿌리, 잎, 열매 추출물질을 이용하여 MMP-9의 활성억제 유무를 연구하였으며, MCF-7 세포에서 모링가 뿌리추출물이 MMP-9을 가장 효과적으로 억제하였다. 또한 MDA-MB-231을 이용한 비교실험에서도 세포특이성 없이 MMP-9의 활성 및 발현이 모링가 뿌리 추출물에 의해 억제되었고 단백질과 mRNA 발현에서도 동일한 결과가 도출되었다. 뿐만 아니라 암세포 침투실험에서도 모링가 뿌리 추출물에 의해 암세포 침투가 50% 이상 감소 되었으나, TIMP-1과 -2 mRNA 발현은 모링가 뿌리 추출물에 의해 저해되지 않았으므로 모링가 뿌리 추출물이 MMP-9 활성 및 발현 저해를 통해 암세포 침투를 저해시키는 것으로 예상된다. 그리고 MMP-9 활성 및 발현 조절기전을 확인하기 위해 MAP kinase의 인산화를 확인한 실험에서 모링가 뿌리 추출물이 ERK와 JNK의 인산화를 억제시킴을 확인하였다. 결론적으로 이 실험을 통해 모링가 추출물 중 뿌리 추출물이 MMP-9 활성 및 발현저해에 가장 효과적이며, ERK와 JNK의 인산화 조절을 통해 MMP-9의 활성 및 발현을 직접적으로 조절함을 확인할 수 있었다.

청미래덩굴뿌리 추출물 첨가 장국용 조미간장 개발을 위한 관능평가 (Sensory Evaluation of Cheongmirae (Smilax china) Root Extract for Soy Sauce Development)

  • The objective of this study is to investigate the sensory quality attributes for the development of soy sauce containing Cheongmirae (Smilax china) root. Aqueous extracts of Cheongmirae root and Cheongmirae-soy sauce were used for determining the antioxidant effect by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and sensory evaluation. Chengmirae-soy sauce was prepared by mixing Chengmiae extract (50%) and soy sauce (50%). Cheongmirae extract and Cheongmirae-soy sauce had strong DPPH scavenging effects, compared to that of general soy sauce. In sensory analysis, low intensities of smell, taste and overall preference were observed in Cheongmirae root extract. Various seasoning items, such as anchovy, dried-pollack, katsuobushi, shiitake, radish, and kelp, were used to determine the suitable type of soy sauce containing Cheongmirae root extract. The anchovy-kelp-radish and anchovy-shiitake-kelp-radish among the seasoning items were well-matched with Cheongmirae root extract. From these results, soy sauce containing Cheongmirae root extract may be used as a soup seasoning for Korean style noodles.

LPS로 자극한 Raw 264.7 cell에서 황금(黃芩)의 type 1 interferon 억제 효과 (The inhibitory effect of Scutellaria baicalensis on type 1 interferon production in Raw 264.7 cells)

  • Objective : The present study was designed to investigate whether the water extract of the root of Scutellaria baicalensis could regulate lipopolysaccharide (LPS)-induced type 1 interferon. Methods : To evaluate of type 1 interferon inhibitory effect of the root of Scutellaria baicalensis, we examined type 1 interferon in LPS-stimulated RAW264.7 cells. Furthermore, Interleukin (IL)-10 and interferon regulatory factor (IRF) - 1, 7 expression level were examined to study the inhibition mechanisms. Results 1. Extract from the root of Scutellaria baicalensis didn't have any cytotoxity itelf. 2. Extract from the root of Scutellaria baicalensis inhibited interferon-a,b in dose dependant- and type 1 interferon production in time dependant manner. 3. Extract from the root of Scutellaria baicalensis reduced IL-10 and IRF-1, 7 production in LPS-stimulated RAW 264.7 cells. Conclusion : The extract from the root of Scutellaria baicalensis down-regulated LPS-induced type 1 interferon through suppression of IL-10 and IRF-1, 7 expression. This results suggested that the extract from the root of Scutellaria baicalensis may be a beneficial drug against inflammatory diseases.

부추뿌리 추출물의 생리활성 (Physiological activities of ethanol extract of Allium tuberosum root)

  • 부추뿌리 추출물(Allium tuberosum root extract)에 대한 다양한 생리활성을 조사하여 천연물소재 가능성을 시험하였다. 부추뿌리 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타내었으며 세포독성이 거의 없는 농도에서 부추뿌리 추출물 처리 시, 산화방지 활성(ID50, 6.2 ㎍/mL), 타이로시네이즈 활성저해(ID50, 115.4 ㎍/mL) 및 멜라닌 합성 저해(ID50, 31.5 ㎍/mL)를 나타내었다. 부추뿌리 추출물 농도 의존적으로 타이로시네이즈 발현이 억제가 관찰되었으며, 부추뿌리 추출물이 타이로시네이즈 단백질 발현을 억제시킴으로써 타이로시네이즈 활성저해 및 멜라닌 합성을 저하시키는 효과가 나타난 것으로 판단된다. 또한, 부추뿌리 추출물이 피부탄력 유지에 관련된 엘라스테이즈 활성저해(ID50, 145.1 ㎍/mL)를 보여주었으며 이소케르세틴(isoquercitrin)이 유효성분으로 확인되었다. 이와 같은 결과로 미루어 볼 때 부추뿌리 추출물은 기능성 소재에 활용하기 위한 효과적인 천연물 재료가 될 수 있다고 판단된다.

Comparison of Physiological Properties of Gamma-Irradiated Root and Stolen Extracts of Gamcho (Licorice, Glycyrrhiza uralensis Fischer)

  • Physiological properties of root and stolen of Gamcho (Licorice, Glycyrrhiza uralensis Fischer) were compared following irradiation at 20 kGy. The root and stolen of Gamcho were extracted with 70 % ethanol, irradiated and stoved in a 4"C refrigerator. Irradiation induced color changes, electron donating ability (EDA), and tyro-sinase inhibition effect (TIE) were investigated. The color of the non-irradiated stolen extract was darker than the non-irradiated root extract (p<0.05), but irradiation eliminated color differences between stolen and root extracts. Generally, irradiation did not affect EDA and TIE of either of the extracts. However, EDA and TIE were higher in stolen extract than in root extract, when the higher dilution factor was considered. These results indicate that the stolen of Gamcho, which is mostly wasted, is a valuable source of phytochemicals with greater EDA and TIE activities than Gamcho root.root.

토복령 추출물을 첨가한 소스의 항산화 활성 및 저장 중 품질 특성 (Antioxidant Activity and Quality Characteristics of Stew Sauce Mixed with Smilax china L. Extract During Storage)

  • This study investigates the effects of a China root (Smilax china L.) extract on the chemical and microbiological characteristics and antioxidant activity of the sausage stew sauce and the soft-tofu stew sauce over a 5-week storage period. Commercial sauces were obtained from the market, and samples were prepared using four different concentrations of the China root extract (0% (control), 0.5%, 1.0%, and 1.5%) and stored at $5^{\circ}C$ and $20^{\circ}C$. Over the 5-week storage period, pH and salinity showed slight changes in both the sausage and soft-tofu stew sauces, but there was no significant difference (p<0.05) regardless of the extract amount and temperature. At 5 weeks, there were significant decreases in the total microbial count in groups with the China root extract (p<0.05) for both the sausage and soft-tofu stew sauces at $5^{\circ}C$. Over the whole storage period, no coliform, yeast, and mold were detected in any sample. S. aureus counts were not detected in 1.0% and 1.5% China root groups at $5^{\circ}C$, but 1.00-1.60 log CFU/g was found in the control and 0.5% groups. DPPH radical scavenging activity at 5 weeks showed an increase with an increase in the amount of the China root extract in both sauces. These results indicate that the China root extract inhibited microbial growth during storage as well as scavenging activity and thus that it can be considered to prolong the shelf life of commercial sauces.

황련추출액의 항균활성과 항균성물질의 동정 (Antifungal Activity or Coptis japonica Root-stem extract and Identification of Antifungal Substances)

  • 황련(Coptis japonica)의 추출액을 공시하여 Phytophthora capsici, Fusarium oxysporum, Colletotrichum dematium, C. truncatum, Botrytis cinerea, Botryosphaeria dothidea과 Alternaria porri에 대해 항균 활성 검정, 몇가지 작물에 대한 병해 방제 효과를 조사하고 항균 활성 물질을 동정하였다. 황련 추출액은 P. capsici, F. oxysporum, C. dematium, B. cinerea, B. dothidea 및 A. porri에 대해서 항균력이 우수하였다. Methanol을 용매로 추출하여 분리된 물질과 berberine-Cl 표준품의 항균활성을 비교할 때, 황련추출액으로부터 분리된 물질은 표준품 berberine-Cl과 비슷하게 항균력을 나타내었다. 황련 추출액은 고추 역병(83.3%), 참깨 모잘록병 (92.0%) 및 파 검은무늬병(87.5%)에 대해서 방제효과가 인정되었다. 고추 잎과 과실, 파 잎 및 딸기 과실에는 약해가 나타나지 않았으나 고농도에서는 고추 및 참깨의 발아를 억제하거나 유근의 신장을 억제하는 것으로 나타났다. Methanol로 추출한 황련 추출액으로부터 황색 분말 4.24g/100g을 얻을 수 있었다. 이들 물질을 HPLC로 분석한 결과 항균성을 나타내는 물질은 berberine-Cl로 동정되었다.
