Paenibacillus polymyxa E681, with its plant growth promotion and root colonization ability, has been proven to be a promising biocontrol agent of cucumber and barley. This study investigated the attributes related to the movement of bacteria from the seed to the radicle and to the whole root system. It also illustrated the existing form and population changes of the bacteria on seed and root using the scanning electron microscope and confocal laser scanning microscopy. The bacteria invaded and colonized the inside of the seed coat while the seeds were soaked in bacterial suspension. Almost the same number of bacteria on seed surface invaded the inside of the seed coat right after seed soaking. The population densities of E681 increased greatly inside as well as on the surface of the seed before the radicle emerged. The bacteria attached on the emerging radicle directly affected the initial population of newly emerg-ing root. The colonized cells on the root were arranged linearly toward the elongation of the root axis. In addition to colonizing the root surface, strain E681 was found inside the roots, where cells colonized the inter-cellular space between certain epidermal and cortical cells. When the cucumber seeds were soaked in bacterial suspension and sown in pot, the bacterial populations attached on both the surface and inside of the root were sustained up to harvesting time. This means that E681 successfully colonized the root of cucumber and sustained its population density up to harvesting time through seed treatment.

To determine the degree of variability among the host plant species in their abilities to become colonized by arbuscular mycorrhizal fungi (AMF), the inoculum for AMF was collected from the various sites in Korea and was inoculated to the three horticultural plants; Tagetes patula, Torenia fournieri, and Salvia splendens. After 4-month growth under greenhouse, mycorrhizal root colonization rates and spore density were measured. The roots of T. patula showed higher colonization rate than both plants of T. fournieri and Salvia splendens. The mycorrhizal root colonization was influenced by both of the AM fungal inoculum and the host species or their interactions. The combination of the host and fungal species was suggested to be important for the application of AMF to horticultural crops.
Four arbuscular mycorrhizal fungal(AMF) inocula collected from three arable sites in Korea were used to determine plant growth, mycorrhizal root colonization rate and spore production in three different host plant species; Sorghum bicolor, Allium fistulosum, Tagetes patula. Growth of plant treated with AMF differed from those without AMF. Different AMF inocula showed significantly different root colonization rates and spore production of AMF on the wild plants, A. fistulosum and T. patula, but did not on the cultivated plant, S. bicolor. Results suggested that indigenous mycorrhizal fungal community would be important factors in mycorrhizal symbiosis, and play important roles in the plant succession.
Radionuclides were deposited at forest areas in eastern parts of Japan following the Fukushima Daiichi Nuclear Power Plant incident in March 2011. Ectomycorrhizal (EM) fungi have important effects on radiocaesium dynamics in forest ecosystems. We examined the effect of colonization by the EM fungus Astraeus hygrometricus on the uptake of cesium (Cs) and potassium (K) by Pinus densiflora seedlings. Pine seedlings exhibited enhanced growth after the EM formation due to the colonization by A. hygrometricus. Additionally, the shoot Cs concentration increased after the EM formation when Cs was not added to the medium. This suggests that A. hygrometricus might be able to solubilize Cs fixed to soil particles. Moreover, the shoot K concentration increased significantly after the EM formation when Cs was added. However, there were no significant differences in the root K concentration between EM and non-EM seedlings. These results suggest that different mechanisms control the transfer of Cs and K from the root to the shoot of pine seedlings.
The study was performed to investigate the influence on growth and development of grape-cuttings by arbuscular mycorrhizal (AM) fungi inoculation, AM colonization rate, and the phenomena of mycorrhizal association. Among the grape-cuttings, 'Kyoho' and 'Tamnara' cultivars inoculated with AM fungi showed significantly increase of leaf area, leaf number, total root length and root surface area than non-infected ones. But 'Cambell Early' did not showed any significant difference in total root length and root surface area even after the inoculation. The AM colonization rates in mycorrhizal inoculation treatment were 22.5-32.5% in total average after 8weeks, and were 29.6%, 28.8%, and 48.8% for 'Cambell Early', 'Tamnara', and 'Kyoho' respectively after 12weeks. The AM colonization rate marked very low level in non-colonization control plot.
This study proposed a rapid method to quantify the colonization rate of arbuscular mycorrhizal fungi (AMF) in plant roots. The method involved the use of an image analysis software (WinRHIZO Pro). The colonization rate is defined as the ratio of the fungal body to the plant root area in a micrograph. Three seedlings of Chengiopanax sciadophylloides, a woody species that accumulates radiocesium, were collected from a secondary forest in the Yamakiya district of Kawamata, Fukushima Prefecture during May-September 2014. The colonization of AMF structures was examined under a light microscope, and the percentage of colonization was determined using the WinRHIZO method. The superiority of the new method was verified by comparing with a modified grid-line intersect method. The colonization of AMF was confirmed in all the seedlings, and a significant coefficient of determination ($R^2=0.94$) was found with both the methods. The results suggested that the WinRHIZO method is reliable for estimating the colonization of AMF in C. sciadophylloides.
Microorganisms are implicated the endodontic treatment failures. Persistent endodontic infection may be the result of retention of microorganisms in the dentin of the root canal walls. Dentinal tubules of the root canal walls have been shown to harbor microorganisms. The purpose of this study was to investigate the invasion of microorganism into the root dentin and dentinal tubules. The effects of irrigation solutions and smear layer on bacterial colonization of root canal were evaluated using a scanning electron microscopy. Canals of extracted human teeth with single and straight canals were stepback prepared using normal saline. Tooth samples were divided into four groups according to the irrigation solutions -5 % sodium hypochlorite and normal saline-and smear layer treatment. The smear layer was removed by 5% NaOCl and 20% EDTA for 10 min respectively. After sterilization, they were incubated with each strains of Streptococcus sanguis, Enterococcus faecalis, Staphylococcus aureus and Escherichia coli. Sodium hypochlorite solution reduced the adhesion of microorganisms effectively compared to normal saline. The smear layer inhibited colonization of E. faecalis, S. aureus and E. coli in the root canals due to their blocking of dentianl tubules. But S. sanguis invaded dentinal tubules in the root canals without smear layer. It was suggested that bacterial attachment might be different according to the strains. Sodium hypochlorite inhibited bacterial attachment in the dentinal tubules dramatically. The absence or presence of smear layer affected bacterial invasion of the dentinal tubules.
This study assessed the possible role of different traits in selected plant growth-promoting rhizobacteria (PGPR) for improving wheat growth and yield under natural conditions. Rhizobacteria exhibiting 1-aminocyclopropane-1-carboxylate (ACC)-deaminase activity were isolated and screened for their growth-promoting activity in wheat under axenic conditions. Five isolates belonging to Pseudomonas and one Burkholderia caryophylli isolate that showed promising performances under axenic conditions were selected and characterized for in vitro ACC-deaminase activity, chitinase activity, auxin production, P solubilization, and root colonization. These isolates were then used as inocula for wheat cultivated under natural conditions in pot and/or field trials. Significant increases in root elongation, root weight, tillers per pot, 1,000-grain weight, and grain and straw yields were observed in response to inoculation with PGPR in the pot trials. Inoculation with these PGPR was also effective under field conditions and increased the wheat growth and yield significantly. However, the efficacy of the strains was inconsistent under the axenic, pot, and field conditions. Pseudomonas fluorescens ($ACC_{50}$), which exhibited a relatively high in vitro ACC-deaminase activity, chitinase activity, auxin production, and P solubilization and more intensive root colonization, was the most efficient isolate under the field conditions. Therefore, these results demonstrated that ACC-deaminase activity is an efficient parameter for the selection of promising PGPR under axenic conditions. However, additional traits of PGPR, including auxin production, chitinase activity, P solubilization, and root colonization, are also important for selecting PGPR as biofertilizers.
Soil or seed applications of plant growth-promoting rhizobacteria (PGPR) have been used to enhance growth of several crops as well as to suppress the growth of plant pathogens. In this study, we selected a PGPR strain, Paenibacillus polymyxa strain E681, out of 3,197 heat-stable bacterial isolates from winter wheat and barley roots. Strain E681 inhibited growth of a broad spectrum plant pathogenic fungi in vitro, and treatment of cucumber seed with E681 reduced incidence of damping-off disease caused by Pythium ultimum, Rhizoctonia solani, or Fusarium oxysporum. When inoculated onto seeds as vegetative cells or as endospores, E681 colonized whole cucumber root systems and root tips. Different temperatures such as $20^{\circ}C\;and\;30^{\circ}C$ did not affect root colonization by strain E681. This colonization was associated with a consistent increase in foliar growth of cucumber in the greenhouse. These results indicate that strain E681 is a promising PGPR strain for application to agricultural systems, particularly during the winter season.
Root colonization by Pseudomonas chlororaphis O6 induces systemic drought tolerance in Arabidopsis thaliana. Microarray analysis was performed using the 22,800-gene Affymetrix GeneChips to identify differentially-expressed genes from plants colonized with or without P. chlororaphis O6 under drought stressed conditions or normal growth conditions. Root colonization in plants grown under regular irrigation condition increased transcript accumulation from genes associated with defense, response to reactive oxygen species, and auxin- and jasmonic acid-responsive genes, but decreased transcription factors associated with ethylene and abscisic acid signaling. The cluster of genes involved in plant disease resistance were up-regulated, but the set of drought signaling response genes were down-regulated in the P. chlororaphis O6-colonized under drought stress plants compared to those of the drought stressed plants without bacterial treatment. Transcripts of the jasmonic acid-marker genes, VSP1 and pdf-1.2, the salicylic acid regulated gene, PR-1, and the ethylene-response gene, HEL, also were up-regulated in plants colonized by P. chlororaphis O6, but differed in their responsiveness to drought stress. These data show how gene expression in plants lacking adequate water can be remarkably influenced by microbial colonization leading to plant protection, and the activation of the plant defense signal pathway induced by root colonization of P. chlororaphis O6 might be a key element for induced systemic tolerance by microbes.
