The effect of arbuscular mycorrhizae(AM) on the growth of trifoliate orange seeding were investigated in volcanic ash soil. Trifoliate orange is used as a root stock of citrus in Cheju island. Seedings innoculated with AM fungi were grown for 16 weeks in pots of various levels of fertilizer. Growth characteristics and mineral nutrient contents were measured and spores of AM fungi colonized were identified. Seventy % of the replicsted pots of seedings were colonized by AM in the treatment of high level fertilizer and additional phosphate (40g of 21-17-17 complex fertilizer and 50g of fused phosphate added to 50L of soil). In treatments of low levels fertilizer or without fused phosphate addition, the pots colonized were less than 20 %. Colonization of trifoliate seedings with AM fungi greatly increased the growth of seedings. Shoot length and weight of shoot and root positively regressed on AM colonization ratio. AM colonization caused higher concentrations of P, Cu and Mg in plant, and the relations were significant at 5 % level. Contents of N and Zn in plants also tended to increase, while that of Ca to decrease, with increasing colonization ratio. Four species of AM fungi - Glomus deserticola, G. rubiforme, G. vesiculiferum and Acaulospora sp, - were found in the soil where roots of trifoliate orange as an innoculation materials were collected. All of the 4 species were found in the inoculated pot soils after the seedling growth, indicating that these species can be colonized in trifoliate orange roots.
Antagonistic rhizobacteria, more specifically fluorescent pseudomonads and certain species of Bacillus, are known as biocontrol agents of fungal root diseases of agronomic crops. In this study, 144 bacteria were isolated from cucumber rhizosphere and screened as potential biological control agents against Phytophthora drechsleri, the causal agent of cucumber root rot, in vitro condition. Non-volatile compounds of 23 isolates showed noticeable inhibition zone (> 30%) against P. drechsleri, whereas volatile compounds of 7 isolates could prevent more than 30% of the mycelial growth of the fungus. All promising isolates, except of Pseudomonas flourescens V69, promoted significantly plant growth under in vitro condition. P. flourescens CV69 and V11 exhibited the highest colonization on the root. Results of the greenhouse studies showed that a reduction in disease incidence by use of some strains, and particularly use of strains CV6 and V11 as a soil treatment, exhibited a reduction in disease incidence so that suppressed disease by 85.71 and 69.39% respectively. Pseudomonas flourescens CV6 significantly suppressed disease in comparison to Ridomil fungicide. The use of mixture bacterial strains in the soil inoculated by the fungus resulting in falling down the most of the plants which didn't show significant difference with infected control soils without bacteria.

Drought is a major obstacle to high agricultural productivity, worldwide. In drought, it is usually presented by the simultaneous action of high temperature and drying. Also there are negative effects of plant growth under drying conditions. In this study, the effect of Bacillus velezensis YP2 on plant growth-promotion and soil drying stress tolerance of kale plants, Brassica oleracea var. alboglabra Bailey, were investigated under two different conditions; greenhouse and field environments. Root colonization ability of B. velezensis YP2 was also analysed by using plating culture method. As a result of the greenhouse test, the YP2 strain significantly promoted the growth of kale seedlings in increasement of 26.7% of plant height and 142.2% of shoot fresh weight compared to control. B. velezensis YP2 have the mitigation effect of drying injury of kale by decreasing of 39.4% compared to control. In the field test, B. velezensis YP2 strain was also found to be effective for plant growth-promotion and mitigation of drying stress injury on kale plants. Especially, relative water contents (RWC; %) were higher in B. velezensis YP2 treated kales than in control at 7, 10, 14 day after non-watering. The root colonization ability of YP2 strain was continued at least for 21 days after soil drenching treatment of B. velezensis YP2. Our result suggested that enhancement of plant growth and drying injury reduction of kale plants were involved in kale root colonization by B. velezensis YP2, which might be contributed to increasing water availability of plants. Consequentially, the use of B. velezensis YP2 might be a beneficial influence for improving productivity of kale plants under drying stress conditions.

In four citrus grow of Satsuma mandarin (rootstock of trifoliate orange) including two grove of organical management and two groves of conventional management, spores of arbuscular mycorrhizal(AM) fungi were identified and seasonal changes in spore density in soils and AM colonization of citrus roots were investigated. AM colonization in weeds found in the groves were also examined. Three species of Glomus (G.deserticola, G. vesiculiferum, G. rubiforme ) and one unknown species of Acaulospora were observed in all of the groves. Annual mean density of AM fungal spores were in the range of 10,000${\sim}$40,000 per 100g soil with more spores in the organically-managed groves. The least spores were observed in December in all groves, and the most spores in April in the organically-managed groves while in February or April in the conventionally- managed. Annual mean AM colonization more 27% of citrus root were observed in the organically-managed with the high peaks in April and October and the minimum in August, while mean colonization less than 15% in the conventionally-managed with the peak in February and the minimum in different times depending on groves and years. AM colonization corresponded to a sigmoidal curve consisting of a laf phase during winter and a subsequent increase in spring, then succeeded by a maximum, and then a decrease at the end of vegetation. Fungal spore density and AM colonization showed a parallel pattern during the sample period. The seasonality appeared to be related more to the phenology of the plant than to the soil factors. Generally more spore density and AM colonization were found in organically managed groves. AM colonization was not correlated with available P and organic matter content in soil in this field investigation. Among sixteen weed species found in the groves, Astrogalus sinicus of Leguminosae, Portulaca oleracea of Portulacaceae showed high colonization in all groves and they can be considered as a source of inoculumn and host plants for propagation of AM fungi.
Number of bacterial isolates were collected from high mountainous areas at various locations in the whole country to select promising biocontrol agents. Most of selected isolates belonged to fluorescent pseudomonads. Population densities of fluorescent pseudomonads were examined by DLF method. Alll selected isolates rapidly multiplied on spermosphere after seed inoculation upto 24, and then the population abruptly declined. When seeds were germinated fully, bacteria moved to newly emerging radicle. The good root colonizing isolates, B16 and V13 proliferated on the growing root and moved down to the root tip and lateral roots. but the poor root colonizing isolates, MC07 and X01 moved much slower. Scanning electron microscopic observations showed that the cells of the good colonizing isolates were arranged linearly on the growing root and parallel to growing root axis and continuously existed on the root tip, Whereas the cells of poorly colonizing isolates were gathered and scattered randomly on the root surface.
On the instrumented root canal wall, amorphous, irregular smear layer can be observed with Scanning Electron Microscope(SEM). The purpose of this study was to evaluate the effect of the presence or absence of smear layer on the adhesion of Staphylococcus aureus to the dentin of the root canal. Human incisors, extracted within 7 days, with no caries, no fracture, no calcification of canal, were selected. After cutting crown portion at cemento-enamel junction, root canal preparation was done by modified crown-down technique using Profile and Gates - Glidden Drill. During canal preparation, 10ml physiologic saline solution(group1&3) or 10ml 3.5% NaOCl(group2&4) was used as irrigation solution. And 10ml physiologic saline solution(group1&3) or 10ml 0.5M EDTA(group2&4) was applicated for final flush. After vertical sectioning and ethylene oxide gas sterilization, samples(group1&2) were immersed into BHIYHM broth inoculated with Staphylococcus aureus (ATCC 31153) and incubated for 3hrs at $37^{\circ}C$. All samples were prepared for and observed with SEM(JEOL JSM840S). The data were analyzed by Mann-Whitney rank sum test. The conclusions are as follows ; 1. Smear layer covers entire root canal surface after root canal preparation. 2. Smear layer has been removed away and the entrances of dentinal tubules have opened widely, when applying 0.5M EDTA and 3.5% NaOCl. 3. A significantly higher number of bacteria were adhered to the root canal dentin without smear layer(p<0.0001). 4. Smear layer produced during root canal preparation impedes bacterial adhesion and colonization to dentin matrix, therefore inhibits canal reinfection.

The 44 endophytic bacterial strains were isolated from surface-sterilized root of rice cultivated in seven different locations of Chungcheong province, Korea. Each isolate was introduced into rice seedlings grown gnotobiotically by inoculating scissor-cut first true leaf with cell suspensions, and the colonization capacity of each isolate in root tissue was analyzed at 7 days after inoculation. Sixteen out of 44 isolates were re-isolated from root successfully with the frequency of $10^{3-5}$ CFU/g tissue. Interestingly, seven out of 16 isolates were identified as Burkholderia species. The identity between inoculated strains and re-isolates was confirmed by genomic finger-printing and 16S rDNA sequence analysis. By a confocal laser scanning microscopic observation it was revealed that KJ001 strain, one of the sixteen isolates tagged with gfp colonized in root tissue especially around xylem. Six out of seven Burkholderia strains obtained in this study showed antagonizing activities against seven different fungal pathogens, contain nifH gene, and five of them enhanced growth of cucumber over 30%. The isolates showed no hypersensitive response on tobacco leaves and no pathogenecity in rice. From these results it was found that the endophytic Burkholderia strains will be useful in agriculture to develop a biocontrol agent or a bio-fertilizer.
In this study, we examined arbuscular mycorrhizal fungal (AMF) community structure colonizing field-cultivated ginseng roots according of different ages, such as 1- to 5-year-old plant, collected from Geumsan-gun, Korea. A total of seven AMF species namely, Funnelliformis caledonium, F. moseae, Gigaspora margarita, Paraglomus laccatum, P. occultum, Rhizophagus irregularis, and Scutellospora heterogama were identified from the roots using cloning, PCR-restriction fragment length polymorphism and sequence analysis of the large subunit region in rDNA. AMF species diversity in the ginseng roots decreased with the increase in root age because of the decreased species evenness. In addition, the community structures of AMF in the roots became more uniform. These results suggest that the age of ginseng affects mycorrhizal colonization and its community structure.

Root colonization by a rhizobacterium, Pseudomonas chlororaphis O6, elicited induced systemic resistance (ISR) in the leaves of cucumber plants against fungal and bacterial pathogens. To understand the role of unique genes during strain O6-mediated ISR, a suppressive subtractive hybridization method was undertaken and led to isolation of twenty-five distinct genes. The transcriptional levels of all the genes showed an increase much earlier under O6 treatment than in water control plants only after challenge with pathogen, while no difference detected on the plants without pathogen challenge. This suggests that O6-mediated ISR is associated with the priming phenomenon, an enhanced capacity for the rapid and effective activation of cellular defense responses after challenge inoculation.
Four promising biocontrol agents against Phytophthora capsici were selected from 507 bacterial isolates collected from rhizosphere soils and roots of pepper plants. In vitro experiment, these four biocontrol agents inhibited mycelial growth, germination of cystospores, and formation of zoosporangia and zoospores of Phytophthora capsici. In the pot experiment, the four biocontrol agents showed control efficiency higher than 70%. In greenhouse experiment, the isolates G28-6 gave the control value of 79.4%. These four biocontrol agents successfully colonized in the population density beyond 105 cfu/g on roots of pepper in vitro. The isolates G28-6 was identified as Pseudomonas aurantiaca, based on its cultural, morphological, and biochemical characterization and 16S rRNA gene sequence analysis.
