Transformation of Maize Controlling Element Ac and Ds into Armoracia rusticana via, Agrobacterium tumefaciens (Agrobacterium tumefaciens를 매개로 한 옥수수 유동유전자 Ac 및 Ds에 의한 서양고추냉이 (Armoracia rusticana)의 형질전환)

  • For the gene tagging of Armoracia rusticana, maize controlling element Ac and Ds were introduced into A.rusticana via Agrobacterium-mediated transformation method. We established an efficient in via regeneration and transformation system for gene transfer in A. rusticana. The optimum in via regeneration condition has been obtained from leaf, petiole and root organs on modified MS medium supplemented with NAA 0.1 mg/L plus BA 1.0 mg/L for direct shooting and with free growth regulators for root induction for transformation, the leaf, petiole and root explants of A. rusticana were concultivated with Agrobacterium tumefaciens, LBA4404 which carries a binary vector pEND4K containing maize controlling element Ac or Ds, respectively: Selections were performed in the shoot induction medium supplemented with 100 mg/L kanamycin, and 500 mg/L carbenicillin transformation frequency showed about 8 to 10% in case of leaf disks. PCR md Southern blot analyses showed that the Ac and the Ds elements were integrated into the chromosome of donor plants.

Biological Control of Root-Lesion Nematodes(Pratylenchus spp.) by Nematode-Trapping Fungi (선충 포식성 곰팡이를 이용한 뿌리썩이선충(Pratylenchus spp.)의 생물학적 방제)

  • For the biological control of the root-lesion nematodes, Pratylenchus spp., which damage directly and indirectly to the leaf perilla, the nematical effect of three nematode-trapping fungi, Arthrobotrys oligospora, A. conoides and A. dactyloides was evaluated in the field. Three species of Arthrobotrys were isolated from the culture soil of leaf perilla in 1998 and were observed the capture of the root-lesion nematodes, Pratylenchus spp. by adhesive hyphal networks or constricting rings on agar. At 40 days after treatment, the plant-parasitic nematodes and root-lesion nematode populations were approximately increased 3.5 fold in untreated control plot, while the nematode population in fungi treatment plots was similar to initial population. In the A. dactyloides plot, however, the population of plant-parasitic nematodes and Pratylenchus spp. was approximately reduced 65% and 53%, respectively. Thus, the fungus A. dachyloides should provide as biological agent for the control of Pratylenchus spp.

Effects of Some Liming Materials Application on the Alleviation of the Symptoms of Mn Toxicity Inculding Marginal Leaf Chlorosis (석회류제의 시용이 엽연형 황피발생 억제에 미치는 영향)

  • Effect of liming material application on the prevention or cure of Mn toxicity symptoms including marginal leaf chlorosis in 3-year-old ginseng plants grown in acidic soils were investigated. It was found that the ratio of Fe to Mn was above about 0.5 and the ratio of Mn to Fe was below about 2.00 in 4-year old ginseng leaves when liming materials were applied in field experiments. It was apparent that the occurrence of marginal leaf chlorosis was decreasing affected by application of Ca, Mg and Fe. The appearance of marginal leaf chlorosis was decreased to about 78 percent in 4-year-old ginseng and to about 69 percent in 5-year-old ginseng, and then the fresh root weight was increased up to 66 percent in 4-year ginseng plants.

  • Activated carbon(AC) can be utilized as a soil conditioner in agricultural crop areas. This study was conducted to investigate the effect of AC on Leaf and Stem Production of Angelica acutiloba as affected by different amounts of AC. The results obtained are summarized as follows. Growth characteristics including plant height and leaf length were the highest when activated carbon added with 10, suggesting that optimum amount of activated carbon was ranged from 10 to $20\%$. Growth and enlargement of the root were improved by $10\%$ AC.

  • The study has been carried to confirm pathogenesity among the isolates which obtained from leaf blight type symptoms and Kresek type symptoms, and also to observe varietal resistance among 29 varieties and/or lines through the introduction of two different pathotypes, of Xanthomonas oryzae, by the use of root clipping and pin prick methods. There was no significant differences among the isolates when their growth in plant tissue were compared. There was certain tendency, however, that the isolates from Kreseked plants showed longer lesion than those from blight type lesions. Both isolates from blight type and Kresek type induced the same degree of Kresek syptoms when they were introduced into plants by root cutting in the suspension prior to transplant. Varietal resistance to 'Kresek' appeared to be the same with those on leaf blight type though Wase Aikoku group varieties showed Kresek symptoms which is not the case with leaf blight type. IR 20 in Kogyoku group varieties showed highly resistant reaction to Kresek type. Root clipping method induced the more of Kresek than those by pin pricking method.

Asexual Propagation of Kalopanax pictus by Root Cutting (근삽에 의한 음나무의 무성번식)

  • This research was carried out to establish the large quantity propagation system for Kalopanax pictus using root cutting. The results are as follows. As there was no significant difference in the promotion of adventitious shoot development from the root cuttings of Kalopanax pictus among kinds (IBA, NAA and Kinetin) and concentrations (0 to 2000 mg/L) of growth regulators, no application is recommended, which showed more than 90% of shoot development. Horizontal and vertical placement of rooting showed more than 93% of shoot occurrence, but the latter showed more desirable growth traits in the number of shoot development, number of leaf, petiole length, and leaf length and leaf width Polarity was observed and thus normal vertical placement of root cutting is preferred. However, root cuttings placed upside down showed the shoot development at the distal end, which indicates further sophisticated test is necessary to investigate this phenomenon.

Induction and Proliferation of Adventitious Roots in Codonopsis spp. (더덕 속 식물의 부정근 유도와 증식)

  • We have established adventitious root culture systems of Codonopsis lanceolata, Codonopsis pilosula and Codonopsis ussuriensis. Root segments of C. lanceolata were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with $0.5mg/{\ell}$ NAA and produced $18.8{\pm}1.9$ roots per explant. Root segments of C. pilosula were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with $1.0mg/{\ell}$ NAA and produced $8.5{\pm}1.8$ roots per explant. Leaf segments of C. ussuriensis were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with $0.5mg/{\ell}$ NAA and produced $7.8{\pm}0.4$ roots per explant. In liquid culture, the best production of adventitious root (fresh weight) was obtained in 1/2 MS medium with $1.0mg/{\ell}$ NAA. This study demonstrated for the first time to produce adventitious roots in C. pilosula and C. ussuriensis.

Germination and Seedling Growth Response of Sprouts and Leafy Vegetables after Applying Oxygen Nanobubble Water

  • Background and objective: The nanobubbles remain stable in water, and it increased dissolved oxygen (DO) in the water that promotes the seed germination and the plant growth. We evaluated the seed germination and growth of sprouts (radish, wheat, and barley) and leafy vegetables (red mustard and pak choi) when irrigated with various DO of nanobubble water (NB). Methods: The oxygen NB was generated by surface friction and treated in 4 levels: NB 0% (control, DO 9.21 mg·L-1), NB 20% (DO 15.40 mg·L-1), NB 33% (DO 20.93 mg·L-1), and NB 100% (DO 39.29 mg·L-1). Results: The root length of radish and wheat increased more in NB 33% than the control plot. The fresh weight increased in NB 33% compared to the control plot in radish and wheat, and both fresh and dry weight increased more in NB 20%, NB 33%, and NB 100% than the control plot of barley. The leaf length and width of red mustard decreased more in NB 33% and NB 100% than the control plot and NB 20%, which indicated the leaf compactness. The fresh and dry weight of shoot and root increased more in NB 100% than the control plot in red mustard. In pak choi, the shoot fresh weight increased more in NB 100% than the control plot. In leafy vegetables, the germination rate of red mustard in NB 100% was higher than the control plot, however, it was not significantly different between oxygen NB plots in sprout vegetables. Conclusion: The results showed that the root growth and biomass increased after applying NB 33% in sprout vegetables. The leaf growth properties as the number of leaves and leaf size were not significantly different or decreased in NB treatments compared to control plots, but NB 100% (DO 39.29 mg·L-1) effectively increased the root growth and plant biomass in leafy vegetables.

Effects of Plant Growth Regulators on Bulblets Regeneration of Liliem cernum K. (솔나리의 인편 재분화에 미치는 식물생장조절제 효과)

  • The purpose of this study was carried out to investigate the effects of plant growth regulators on in vitro propagation of Liliem cernum Komarov. Small bulblets were poliferated from callus explants after 2 weeks and leaf, root and bulb were formed after 4 weeks culture. Leaf differentiation was promoted vigorously by the combination of TDZ 0.1 mg/L and NAA 0.01 mg/L(87.5%). The rate of root differentiation was the greatest at BA 0.2 mg/L alone(81.8%). The rate of callus formation was the high in medium containing TDZ. The number of bulblets and leaves formed in bulb scales was the greatest at TDZ 0.1 mg/L(5.7). Also, the longest length of total length, leaf and root length were in Zeatin 1.0 mg/L + NAA 0.1 mg/L(10.5 cm). However the longest bulblet was in TDZ 0.1 mg/L(1.4 cm).
