탄화수소로부터 균체단백질의 생산에 관한 연구 (Studies on the Production of Microbial Cell Protein from Hydrocarbon)

  • 1) 고온성으로 탄화수소를 자화할 수 있는 균체를 생산하는 목적으로 90여점의 시료로부터 세 균주의 효모를 선별하여 다음의 결과를 얻었다. 2) 선정된 세 균주 효모의 최적생육온도는 4$0^{\circ}C$~45$^{\circ}C$였과. 그리고 이들은 각각 Candida tropicalis (strain A-28), Torulopsis molisichiana (strain B-8) 그리고 Candide krusei (strain C-15)로 각각 동정되었다. 3) 특히 균주 A-28은 tetradecane, hexadecane 과 octadecane을 다같이 쉽게 자화하였고 B-8과 C-15 균주는 다른 n-paraffin 보다 hexadecane을 잘 자화하였다. 4) 선정된 세 균주효모의 세대시간은 3.4.~4.0 시간, 비증식속도는 0.170~0.215이고 그리고 균체수율은 86~98%이였다. 5) 이들 균체의 조단백질은 48.2~61.5%, 지방은 3.7~8.0%, 조섬유는 3.5~4.2%, 회분은 5.6~6.7%, 무질소가용성 물질은 23.5~31.8%였고, thiamine은 0.28~0.93 mg%, riboflavin 6.03~7.3 mg%이었다. 6) 균체단백질의 아미노산조성은 비교적 골고루 함유되었다. 그러나 함황(sulfur containing) amino acid는 극히 적었다.

  • The manufacture of riboflavine fortified Dwen-Jang has been tried with an exceedingly riboflavine productive Aspergillus oryzae #612 mutant which has been developed by the authors. Both the rice and barley koji of this mutant and Aspergillus sojae have been prepared. Their riboflavine production, saccharifying and protease activities have begin compared The riboflavine fortified Dwen-Jang has been manufactured using the barley koji of riboflavine productive mutant. Their riboflavine content and qualities have been studied comparing with an ordinary Dwen-Jang which has been prepared with the barley kojo of A. sojae strain. The following results have been obtained. (1) The baley koji was superior in riboflavine production and protease activity, inferior in saccharifying ability than rice koji both with A. oryzae #612 and A. sojae. (2) In barley koji, the mutant, A. oryzae #612, produces 1.5 times riboflavine than A. sojae and shows stronger saccharifying and protease activities than the latter. (3) The riboflavine fortified Dwen-Jaug manufactured contained $5.2{\gamma}/g$ of riboflavine, about 1.5 times that of A. sojae. The higher contents of free sugar and free amjno nitrogen have been observed than the ordinary Dwen-Jang manufactured with A. sojae.

Mouse cell에서 형개연교탕(荊芥連翹湯)의 iNOS 생성 억제 효과 (The effects of Hyunggaeyungyo-tang of suppression of iNOS production on RAW 264.7cell)

  • Background and Objectives : The aim of this study was to investigate anti-inflammatory and anti-oxidant effects of Hyunggaeyungyo-tang(HYT) on RAW 264.7 cells. Material and Methods : Two types of experiments were implemented for this study: first, the experiment to study the anti-oxidant effect of HYT using riboflavin; second, in vitro experiment to investigate the suppression of NF-${\kappa}$B activation using RAW 264.7 cells (I${\kappa}$B kinase and induce nitric oxide synthase mRNA expression) Results : 1. The anti-oxidant effects of HYT was dose-dependantly increased. 2. The RAW 264.7cells were treated with LPS for 1 hours prior to the addition of indicated concentrations(0.4,-1.0mg/$m\ell$) of HYT, and the cells were further incubated for 24 hours. The LPS-induced IKK & iNOS mRNA expression were dose-dependantly decreased in HYT treated RAW 264.7cells. Conclusion : The results suggest that HYT is significantly effective in the treatment of inflammation through the suppression of NF-${\kappa}$B activation and iNOS production.

한국농촌주민(韓國農村住民)의 계절별(季節別) 식품섭취조사연구(食品攝取調査硏究) (A Study on Seasonal Variations of Food Consumption of Korean Farmers)

  • This is the report of a food consumption survey of 193 members of 30 farm families in three rural villages of Korea. From a total of 188 households of the three villages, namely Wolgok in Kyunggi Do Province, Wachon in Kangwon Do Province, and Sobong in Cholla Namdo Province, 30 households were chosen for the survey. Four consecutive seasonal surveys, beginning in the autumn, November 1974 and finishing with the summer, August 1975, were made and each survey covered three consecutive days. The Precise Weighing Method was used in evaluating the kinds of food and nutrient intakes of the subjects. This method entails the accurate weighing of all foods by the investigator just prior to their consumption by the subjects under investigation. This survey was male to determine the actual food eaten by the members of these farm families at each season of the year, and to note any special differences in the foods consumed. The results obtained are summarized as follows: Food Intake The traditional ordinary diet of Korean farmers consists mainly of rice and other cereals as staple foods, and simple subsidary foods. The nutritional shortcomings of rice as a staple food are evident in the subclinical status of nutritional deficiencies. The mean consumption of cereals and their products was very high and it was more than half of the daily total food intake throughout all the seasons. The consumption of animal food, fruits, and fat by the farmers was very low. Fruit intake during winter and spring was almost nil. One of the special foods in the Korean dietary pattern is Kim-chi, a variety of pickled and fermented vegetables. The mean intake of Kim-chi during autumn and winter was very much higher than the other seasons. Korean farmers have difficulties in balanced year-round supply of food due to seasonality of food production and lack of food preservation. As nutrition problems of Korean farmers are highly influenced by seasonal variations, the expansion of an appropriate food preservation and storage programme should be encouraged. Special consideration should be given to new methods of preservation, the types of foods preserved, and production and consumption of nutritionally valuable foods for the improvement of the nutritional status in rural communities. Nutrient Intake Average adult rates for calorie and protein in the three study areas were 0.80 and 0.87 respectively, with little variations by villages. The adult caloric intake in averaged 2,928 kcal for autumn, 2,662 for winter, 2,673 for spring, and 2,760 for summer. Cereals contributed most of the total calories. In terms of the recommended daily dietary allowances for the Korean given by the Korea FAO Association, the diets in this survey were adequate except in the winter and spring. The protein intake averaged from 76.4 grams in autumn to 83.4 grams in summer and was near the recommended allowance, but a relatively large proportion of the total protein came from cereals and their products. The fat intake was unsatisfactory, ranging from 19.2 grams to 29.3 grams for the four seasons, and the consumption during farming season was higher than other seasons. It is apparent that the diet of the Korean farmers should be supplemented by calcium during autumn and winter. The highest calcium intake, with a mean of 583.5 milligrams, was recorded in summer. The mean intake of iron ranged from a low of 9.2 milligrams in the autumn to a high of 15.0 milligrams in the spring and it was satisfactory for all seasons. Among the vitamin group, the daily average intakes of vitamin A, thiamine, riboflavin, and ascorbic acid were lower than the desired intake throughout all the seasons. The mean vitamin A intake as ${\beta}-carotene$ ranged from a low of 2,807.8 IU to a high of 5,221.0 IU. The thiamine intake ranged from 0.98 milligrams to 1.19 milligrams, while mean riboflavin intake ranged from 0.92 milligrams to 1.13 milligrams. Ascorbic acid intake was 37.3 milligrams, the lowest, and the highest 47.8 milligrams. Niacin intake among Korean farmers was higher than the recommended allowance throughout all the seasons, and the mean intake ranged from 18.3 milligrams to 25.8 milligrams. The main character of the Korean farmers' diet has been found low in quality of protein and high in carbohydrate. Attention should be paid to the low intake of vitamin A, thiamine, riboflavin, and ascorbic acid among Korean farmers.

Nutritional Requirements of Actinomyces Isolated from Rumen of Goat

  • The objective of this work was to investigate the nutritional requirements for the growth of Actinomyces sp. 9RCC5 isolated from the rumen of a native goat in Korea. The growth of strain 9RCC5 on the basal medium or the medium minus certain ingredients from the basal medium demonstrated that strain 9RCC5 showed absolute requirement of vitamin B complex mixture, while hemin and volatile fatty acids (VFA) were stimulatory to growth to some extent. The 9RCC5 strain grew well with casein hydrolysate as the sole added nitrogen source. However, neither a complex of 18 amino acids nor ammonium sulfate effectively replaced casein hydrolysate. Vitamins such as riboflavin and pantothenate were essential for growth, while thiamin and biotin were stimulatory. With regard to VFA, the growth was stimulated by acetic acid but inhibited by valeric acid. Relatively large quantities of $Na^+$, $K^+$ and $Ca^{2+}$ were absolutely required for growth. Supplementation of clarified rumen fluid to the basal medium in a range of 0-10% (vol/vol) resulted in an increased rate of growth as well as an increased extent of growth.

Mouse cell에서 선방활명음(仙方活命飮)의 항산화작용과 항알러지 및 항염증 효과 (The Effects of Sunbanghwalmyung-eum Extract on Anti-oxidant, Anti-allergic and Anti-inflammatory ability in mouse cell)

  • Background and Objectives : The aim of this study was to investigate the anti-oxidant, anti-allergic and anti-inflammatory ability of the Taglisodog-eum(SHE) extract on the RAW 264.7 and EL4 cells Materials and Methods : Three types of experiments were implemented for this study: first, the experiment to study the anti-oxidant effect of SHE using Riboflavin; second, in vitro experiment to investigate the inhibition of Th 2 cell differentiation by SHE using EL4 cells (IL-4 mRNA expression); third, the suppression of $NF-{\kappa}B$ activation using RAW 264.7 cells (iNOS and COX-2 mRNA expression). Results : The anti-oxidant ability of SHE were dose-dependantly increased. From in vitro, the LPS-induced iNOS and COX-2 mRNA expression were dose-dependantly decreased in the RAW264.7 cells treated with SHE and the PMA-induced IL-4 mRNA expression were also dose-dependantly decreased in EL4 cells. $NF-{\kappa}B$ activation was suppressed, and iNOS & COX-2 production were inhibited by SHE Conclusion : The results suggest that SHE has dose-dependant anti-oxidant ability, and has anti-allergic and anti-inflammatory effects through the suppression of $NF-{\kappa}B$ activation and the inhibition of Th 2 cell differentiation.

The strategies for the supplementation of vitamins and trace minerals in pig production: surveying major producers in China

  • Objective: Adequate vitamin and trace mineral intake for pigs are important to achieve satisfactory growth performance. There are no data available on the vitamin and trace mineral intake across pig producers in China. The purpose of this study was to investigate and describe the amount of vitamin and trace minerals used in Chinese pig diets. Methods: A 1-year survey of supplemented vitamin and trace minerals in pig diets was organized in China. A total of 69 producers were invited for the survey, which represents approximately 90% of the pig herd in China. Data were compiled by bodyweight stages to determine descriptive statistics. Nutrients were evaluated for vitamin A, vitamin D, vitamin E, vitamin K, thiamine, riboflavin, vitamin B6, vitamin B12, pantothenic acid, niacin, folic acid, biotin, choline, copper, iron, manganese, zinc, selenium, and iodine. Data were statistically analyzed by functions in Excel. Results: The results indicated variation for supplemented vitamin (vitamin A, vitamin D, vitamin E, vitamin K, vitamin B12, pantothenic acid, niacin, and choline) and trace minerals (copper, manganese, zinc, and iodine) in pig diets, but most vitamins and trace minerals were included at concentrations far above the total dietary requirement estimates reported by the National Research Council and the China's Feeding Standard of Swine. Conclusion: The levels of vitamin and trace mineral used in China's pig industry vary widely. Adding a high concentration for vitamin and trace mineral appears to be common practice in pig diets. This investigation provides a reference for supplementation rates of the vitamins and trace minerals in the China's pig industry.

담자균(擔子菌)에 관한 연구(硏究)(2) - 느타리와 목이의 진탕 배양(培養)에 의한 균사체(菌絲體) 생산(生産)에 관하여 - (Studies on Basidiomycetes(2) - Production of Mushroom Mycelium(Pleurotus ostreatus and Auricularia auricula-judae) in Shaking Culture -)

  • 진탕배양에서 느타리와 목이의 균사체(菌絲體) 생산(生産)에 대한 실험(實驗)의 결과(結果)를 요약(要約)하면 다음과 같다. 1. 균사체생산(菌絲體生産)에 양호(良好)한 탄소원(炭素源)은 P. ostreatus경우 glucose와 mannitol이었고 A. auricula-judae는 mannitol과 fructose이었다. 유기산(有機酸)중에서는 양균주(兩菌株) 모두 fumaric acid에서 제일 양호(良好)하였다. 2. 균사체생산(菌絲體生産)에 양호(良好)한 질소원(窒素源)은 P. ostreatus 경우 peptone, urea와 asparagin, DL-threonine이 었고 A. auricula-judae는 peptone, Casamino acid와 L-Serine, DL-threonine이었다. 3. 균사체생산(菌絲體生産)에 양호(良好)한 vitamin은 A. auricula-judae 경우 folic acid, thiamine이었고 A. auricula-judae는 folic acid, inositol, riboflavin이었다. 4.양균주(兩菌株)가 $MgSO_4$는 0.08%, $KH_2PO_4$ 0.2%에서 균사체생산(菌絲體生産)이 양호(良好)하였다. 5. 균사체생산(菌絲體生産)의 최적온도(最適溫度)와 pH는 P. ostreatus에서 $25{\sim}30^{\circ}C$, $pH\;5.5{\sim}6.0$이고 A. auricula-judae에서 $25{\sim}30^{\circ}C$, $pH6.0{\sim}7.0$이었다.

목재(木材)를 이용(利用)한 Alcohol 생산(生産)에 관(關)한 연구(硏究) (Studies on the Production of Alcohol from Woods)

  • 침엽수(針葉樹)인 소나무, 리기다소나무, 일본잎갈나무 3수종(樹種)과 활엽수(闊葉樹)인 오리나무, 밤나무, 이태리포푸라 3수종(樹種)의 목재(木材)로부터 alcohol을 생산(生産)하기 위하여 목재(木材)의 화학성분(化學成分) 분석(分析)과 산(酸) 가수분해(加水分解) 조건(條件)을 확립(確立)하고 또한 섬유소(纖維素) 분해력(分解力)이 강(强)한 균주(菌株)를 분리(分離) 동정(同定)하여 균주(菌株)의 효소(酵素) 생산조건(生産條件), 효소(酵素)의 특성(特性) 및 alcohol 발효조건(醱酵條件) 등(等)을 연구조사(硏究調査)한 바 다음과 같은 결과(結果)를 얻었다. 1) 수종별(樹種別) 산(酸) 가수분해(加水分解)에서 HCl은 1.0%와 2.0%, $H_2SO_4$는 2.0%에서 높은 당화율(糖化率)을 보였고 수종(樹種)은 오리나무가 23.4%로 제일 좋았다. 2) HCl과 $H_2SO_4$으로 산(酸) 가수분해(加水分解)할 때 압력(壓力)은 $1.5kg/cm^2$, 산(酸)의 첨가량(添加量)은 30배량(倍量), 분해시간(分解時間)은 HCl 45분과 $H_2SO_4$ 30분에서 당화율(糖化率)이 양호(良好)하였다. 3) 전처리(前處理)에 있어서 농(濃) HCl 보다는 농(濃) $H_2SO_4$이 더 효과적(効果的)이었고 그 농도(濃度)는 50~60%이었으며 열처리(熱處理)는 $190^{\circ}C$에서 30분간(分間) 처리(處理)함으로써 무처리(無處理)보다 당화율(糖化率)이 약(約) 1.5% 증가(增加)하였다. 4) 목분(木粉) 1g에 있어서 산(酸) 가수분해액(加水分解液)의 당조성(糖組成)은 소나무는 glucose 137.78mg, arabinose 68.24mg이었고 오리나무는 glucose 162.22mg, arabinose 65.89mg을 함유(含有)하고 있었으며, xylose와 galactose도 검출(檢出)되었다. 5) 10개(個)의 균주(菌株) 중에 alcohol 효효력(醗酵力)이 왕성한 균주(菌株)는 Sacch. cerevisiae JAFM 101과 Sacch. cerevisiae var. ellipsoideus JAFM 125이었다. 6) 산(酸) 가수분해액(加水分解液)은 $CaCO_3$와 NaOH로 중화(中和)하는 것이 alcohol 생산(生産)과 효모생육(酵母生育)이 양호(良好)하였고 alcohol 생산(生産)은 pH 4.5~5.5, $30^{\circ}C$, 균주생육(菌株生育)에는 pH 5.0~6.0, $24^{\circ}C$에서 발효(醱酵)시키는 것이 좋았다. 7) Alcohol 생산(生産)에는 0.02%의 $(NH_2)_2CO$$(NH_4)_2SO_4$, 0.1%의 $KH_2PO_4$, 0.05%의 $MgSO_4$ 그 밖에 0.025%의 $CaCl_2$, 0.002%의 $MnCl_2$ 첨가(添加)가 효과적(効果的)이었고 효모증식(酵母增殖)에는 0.04~0.06%의 $(NH_2)_2CO$$(NH_4)_2SO_4$, 0.2%의 $K_2HPO_4$$K_3PO_4$, 0.05%의 $MgSO_4$ 그 밖에 0.025%의 $CaCl_2$, 0.002%의 NaCl 첨가(添加)가 효과적(効果的)이다. 8) 여러 가지 vitamin 중에서 alcohol 생산(生産)은 pyridoxine과 riboflavin, 효모증식(酵母增殖)에는 thiamin과 Ca-pantothenate, biotin과 inositol의 첨가(添加)가 좋았고 tannin과 furfural은 0.01% 농도(濃度)에서 alcohol 생산(生産)이 오히려 증가(增加)하였으며 그 이상(以上)의 농도(濃度)에서는 저해(沮害)를 받았다. 9) 발효액(醱酵液) 100ml에서 소나무는 2.201~2.275ml의 alcohol과 168~228mg의 효모(酵母)가 생산(生産)되었고 잔류당(殘溜糖)은 0.55~0.60g이었다. 오리나무는 2.075~2.125ml의 alcohol과 208~256mg의 효모(酵母)를 생산(生産)했으며 잔류당(殘溜糖)은 0.60~0.65g이었고 pH는 3.3~3.6으로 변화(變化)했다. 10) Trichoderma viride JJK107을 섬유소(纖維素) 분해력(分解力)이 강한 균주(菌株)로서 선정(選定)하여 동정(同定)하였다. 11) 효소(酵素) 생산조건(生産條件)으로 CMCase는 pH 6.0, $30^{\circ}C$, xylanase는 pH 5.0, $35^{\circ}C$에서 5일간 배양(培養)할 때 최고(最高)의 효소생산(酵素生産)을 나타냈고 효소작용(酵素作用)의 최적조건(最適條件)은 CMCase는 pH 4.5, $50^{\circ}C$, xylanase는 pH 4.5, $40^{\circ}C$에서 최고(最高)의 활성도(活性度)를 보였다. 12) 목분(木粉)을 peracetic acid로 탈(脫) lignin하고 효소(酵素) 가수분해(加水分解)하여 발효(醱酵)시킬 때가 alcohol 생산(生産)이 좋았으며 peracetic acid 첨가비(添加比)는 10배량(倍量) 이도(移度) 첨가(添加)하는 것이 좋았다. 13) 발효(醱酵)에 있어서 본분(本粉)과 밀기울 Koji의 혼합비율(混合比率)은 10:8일 때에 alcohol 생산(生産)이 좋았고 본분(本粉) 10g에서 소나무 2.01~2.14ml, 오리나무는 2.11~2.20ml의 alcohol을 생산(生産)하였다.

  • 국가적(國家的) 차원에서의 국민영양소요양(國民營養所要量)을 확립하는데 중요한 지표(指標)가 되는 한국인(韓國人) 평균1인1일당(平均1人1日當) 영양소요양(營養所要量)설정의 필요성에 의하여, 1980년도(年度) 경제기획원인구(經濟企劃院人口)센서스 자료(資料)와 FAO한국협회(韓國協會)가 1980년(年) 제(第)3 차 (次) 개정(改定)한 한국인영양권장양으로부터 한국인(韓國人) 평균(平均)1인(人)1일당(日當) 영양소요양(營養所要量)을 계산(計算)하였다. 즉 영양소별(營養素別) 평균(平均)1인(人)1일당(日當) 영양소요양(營養所要量)은 에너지 2,200 kcal, 단백질(蛋白質) 70 g, 칼슘 0.72 g, 철(鐵) 14 mg, 비타민 A 1,900 IU, 비타민 C 50 mg, 비타민$B_{1}$ 0.9 mg, 비타민 $B_{2}$ 1.2 mg 그리고 나이아신당양(當量)은 15 mg이었다.
